Multiscale Mechanics Of Bone: Examining The Influence Of Genetic Alterations, Brendyn James Miller

This MSc thesis investigates how genetic alterations involved in the synthesis of proteins or in the regulation of tissue remodeling affects the composition, structure and mechanics of bone at the multiscale levels. This was accomplished by performing two mouse model studies. In the first study, bone properties of TIMP-3 knock-out (Timp-3⁻ˊ⁻) mice were examined by performing mechanical testing, µCT analysis, and FTIR analysis. Results demonstrated that the mechanical integrity of the Timp-3⁻ˊ⁻ bone was compromised. Decreased values for the yield and ultimate load, stress capacity, and fracture toughness were observed in both the TIMP-3 deficient bones. These reductions in the …

Launch vehicles are in an ever-decreasing weight trend, with bodies becoming more slender to reduce aerodynamic loading. Mass and stiffness tend to be opposing concepts when designing launch vehicles: a stiffer structure means it is usually heavier. Lower stiffness on launch vehicles may produce excessive vibrations during flight, reducing the performance, operating characteristics and safety margins of the vehicle. It also impacts flight dynamics: as the vehicle flies, the orientation of its thrusters and actuators are impacted by the vibrations, creating potential instabilities in the control schemes. Exploring The Role Of Biofilms In The Transport And Establishment Of Invasive Marine Macrofoulers, Lynn Holly Sweat

Biofilms dominated by bacteria and diatoms universally colonize submerged surfaces and mediate larval recruitment in benthic marine systems. The ability for macrofoulers to be transported by vessels and become globally established is likely affected by cues from the biofilm organisms they encounter. This study investigates how macrofoulers respond to biofilms shaped by local geography, transport processes and relocation. Biofilm assemblages in neighboring ports were largely unique to the site at which they were developed. Bacteria were primary drivers of variations among sites, and overall bacterial richness was an order of magnitude higher than has been reported for other marine environments. …

Poorly coordinated care transitions result in nearly half of discharged patients’ experiencing at least 1 medication error and 1 in 5 Medicare beneficiaries readmitted within 30 days. Repercussions of suboptimal transitions of care greatly impact the nation’s economy costing between $12 billion and $44 billion annually. Ineffective handoffs between providers at hospital discharge contribute to suboptimal patient outcomes; therefore, strategies to improve transitions of care are necessary to provide quality care while decreasing health care expenditure. The current study examined the relationship between attachment style, self-esteem, and narcissism as they pertain to behavioral tendencies termed Love-Bombing behaviors among a sample of young adult Millennials. Love-Bombing was identified as the presence of excessive communication at the beginning of a relationship in order to passively obtain power and control over another’s life as a means of narcissistic self-enhancement. The sample consisted of 484 college students from a large southern university who ranged in age 18-30. The results indicated that Love-Bombing was positively correlated with narcissistic tendencies, avoidant attachment, anxious attachment, and negatively correlated with self-esteem. The gene expression profile of a eukaryotic cell is responsive to a variety of extracellular stimuli, including nutrient availability, which allows cells to toggle between anabolism and catabolism based on the favorability of their environment. Much of this information is relayed through signaling complexes, such as the target of rapamycin complex 1 (TORC1), to downstream chromatin modifying enzymes. These enzymes impact the gene regulatory process through altered histone post-translation modifications, changes in chromatin structure, and docking of chromatin regulatory complexes. Several important antifungal drugs inhibit the synthesis of ergosterol, a lipid that modulates the thickness, fluidity and permeability of fungal cell membranes. These include the azole antifungals, which block ergosterol biosynthesis by inhibiting lanosterol demethylase (Erg11p). The resulting depletion of cellular ergosterol and the accumulation of ‘toxic’ sterol intermediates are both thought to cause plasma membrane dysfunction and ultimately growth arrest. However, the effects of ergosterol depletion upon the function of intracellular membranes and organelles are not well described. Mammalian UNC-51–like kinases 1 and 2 (ULK1 and ULK2), Caenorhabditis elegans UNC-51 and Drosophila melanogaster Atg1 are redundant serine/threonine kinases that regulate flux through the autophagy pathway in response to various types of cellular stress. C. elegans UNC-51 and D. melanogaster Atg1 also promote axonal growth and defasciculation, and disruption of these genes results in defects in axon guidance in invertebrates. Germline Ulk1/2-deficient mice die perinatally. Therefore, we used a conditional-knockout approach to investigate the roles of ULK1/2 in the brain. Protein Kinase D1 (PKD1) is a serine threonine kinase which is downregulated in Prostate, Breast and Colon Cancer. It functions as a tumor suppressor in different cancer cells. Downregulation of PKD1 is known to be associated with aggressiveness of the cancer. PKD1 is known to regulate many key oncogenic signaling pathways such as E-cadherin, β-catenin and Androgen Receptor signaling pathways. Aberrant expression of these oncogenic pathways leads to transformation of cells from normal to malignant phenotype, thereby leading to increased proliferation, growth and metastasis to distant organs of these cancer cells. In recent decades the prevalence of childhood and adult depression and obesity has increased worldwide. Multiple factors influence the onset of both depression and obesity. Individual manifestations of depressive symptomatology and health-related problems associated with obesity vary; however, genetics and environmental and psychological factors have been implicated as correlates. Nucleoli are nuclear membrane-less organelles that are the sites for ribosome biogenesis and serve as sensors of cellular stress. Weak, multivalent protein-protein interactions and interactions between disordered, low complexity domains (LCDs) and rRNA have been shown to promote liquid-liquid phase separation (LLPS) in vitro, suggesting a basis for the liquid-like behavior of nucleoli. Nucleophosmin (NPM1), a multifunctional and highly abundant nucleolar protein, exhibits structural features associated with LLPS suggesting a role in nucleolar organization. Specifically, NPM1 forms a pentamer through its N-terminal oligomerization domain and can bind to rRNA through its C-terminal nucleic acid binding domain. Multiple acidic tracts throughout …

The article explores Chief Justice John Roberts' judicial approach, characterizing it as "maximinimalism"—a blend of narrow decision-making with deep, impactful reasoning. While Roberts is often seen as a minimalist for his emphasis on deciding cases narrowly, his rulings in significant cases like Citizens United v. FEC, Shelby County v. Holder, and NFIB v. Sebelius reveal a deeper, more substantive approach that challenges the law, thus embodying maximalist tendencies. This duality allows the Court to influence the law profoundly while maintaining judicial restraint.

This thesis examines the effects of shade on ‘Prime-Ark 45’ blackberries (Rubus spp.) in greenhouse (GH) and field (FD) experiments aimed at improving fruit production in the southern United States Ozark Plateau region. Primocane blackberry production in the southern United States is limited in acreage of production as well as low yields by high temperatures during the bloom and early fruiting period, resulting in poor fruit set and poor fruit quality. Synthetic cannabinoids are man-made mind-alternating chemicals. Law enforcement and legislation have attempted to classify many of these synthetic cannabinoids as schedule I controlled substances, however, they are continuously being modified by dealers at the retail end of the distribution chain. Addiction and moderate use of illicit drugs have been identified as major reasons for non-adherence to antiretroviral therapy among HIV patients. However, there is no data regarding the impact of synthetic cannabinoid exposure in this population and how this affects their compliancy with taking antiretroviral therapy. Objectives To evaluate characteristics and prognostic factors of open globe injuries (OGI) presented to King Fahad Specialist Hospital in the Jazan region, Saudi Arabia. Methods This study is a retrospective review of medical records of OGI patients who underwent operative repair of their injuries in King Fahad Central Hospital, Jazan, Saudi Arabia between January 2011 and December 2013. Demographic information, eye injury, preoperative, and postoperative visual acuity were collected. The Leadership in Energy and Environmental Design (LEED) certification program serves as a benchmark for ‘green’ construction in the U.S. Although considerable research has investigated the marketability of LEED-certified buildings, no direct attention has been given to assessing if the benefits of LEED certification filter down to the profitability of products manufactured in those buildings. To investigate this potential influence, I conducted a large-scale survey of regional owners of the Volkswagen Passat, which is manufactured in the world's first automobile plant to achieve LEED Platinum status.
