ETSU Faculty Works

Endothelial damage is a critical mediator of myocardial ischemia/reperfusion (I/R) injury. HSPA12B is an endothelial-cell-specifically expressed heat shock protein. However, the roles of HSPA12B in acute myocardial I/R injury is unknown. Here we reported that myocardial I/R upregulated HSPA12B expression in ventricular tissues, and endothelial overexpression of HSPA12B in transgenic mice (Tg) limited infarct size, attenuated cardiac dysfunction and improved cardiomyocyte survival compared with their wild type littermates. These improvements were accompanied with the diminished myocardial no-reflow phenomenon, decreased microvascular leakage, and better maintained endothelial tight junctions. The I/R-evoked neutrophil infiltration was also suppressed in Tg hearts compared with its …

Myelinating Glia Differentiation Is Regulated By Extracellular Matrix Elasticity, Mateusz M. Urbanski, Lyle Kingsbury, Daniel Moussouros, Imran Kassim, Saraf Mehjabeen, Navid Paknejad, Alicia Meléndez

Publications and Research

The mechanical properties of living tissues have a significant impact on cell differentiation, but remain unexplored in the context of myelin formation and repair. In the PNS, the extracellular matrix (ECM) incorporates a basal lamina significantly denser than the loosely organized CNS matrix. Inhibition of non-muscle myosin II (NMII) enhances central but impairs peripheral myelination and NMII has been implicated in cellular responses to changes in the elasticity of the ECM. To directly evaluate whether mechanotransduction plays a role in glial cell differentiation, we cultured Schwann cells (SC) and oligodendrocytes (OL) on matrices of variable elastic modulus, mimicking either their …

MLKL, a key component downstream of RIPK3, is suggested to be a terminal executor of necroptosis. Genetic studies have revealed that Ripk3 ablation rescues embryonic lethality in Fadd- or Caspase-8-deficient mice. Given that RIPK3 has also been implicated in non-necroptotic pathways including apoptosis and inflammatory signaling, it remains unclear whether the lethality in Fadd(-/-) mice is indeed caused by necropotosis. Here, we show that genetic deletion of Mlkl rescues the developmental defect in Fadd-deficient mice and that Fadd(-/-)Mlkl(-/-) mice are viable and fertile. Mlkl(-/-)Fadd(-/-) mice display significantly accelerated lymphoproliferative disease characterized by lymphadenopathy and splenomegaly when compared to Ripk3(-/-)Fadd(-/-) mice. …

Using Geographic Information Systems To Identify Habitat For The Golden-Winged Warbler (Vermivora Chrysoptera) In Western New York, Jesse Michael Rubenstein Sep 2016

Using Geographic Information Systems To Identify Habitat For The Golden-Winged Warbler (Vermivora Chrysoptera) In Western New York, Jesse Michael Rubenstein

Theses

The Golden-winged Warbler (Vermivora chrysoptera) is a passerine species whose populations have decreased internationally by approximately 66 percent since the 1960s, and by approximately 50 percent in New York. Major causes for declines are from loss of shrubland habitat and through hybridization with the Blue-winged Warbler (Vermivora cyanoptera). This study utilized published data and expert opinion on Golden-winged Warbler (GWWA) habitat requirements to create habitat suitability models within the Western Finger Lakes Region of NY (NYSDEC Region 8) using a geographic information system. This region is an important area for migratory birds and is previously unstudied for GWWA. The concentration …

Cell Culture Investigation Of The Effects Of Aerogel Surface Topography And Dc Electric Field Stimulation On Pc12 Cell Neurite Growth, Kyle Jeremiah Lynch

Electronic Theses and Dissertations Archive

Polyurea crosslinked silica aerogel (PCSA) is an open pore light-weight and topographically rich material with tunable physical and chemical properties. Because of the unique properties of this material, and the ability to tailor the properties to specific needs and applications, PCSA has attracted significant interest from the biophysical and biomedical fields in recent years. Recent advances in cell biology and biophysics have demonstrated that cells respond to their micro- and nanoscale environmental cues in the presence and/or absence of electrical cues and stimuli. Aerogels offer a unique platform for studying the effects of topography, substrate stiffness, and porosity on cellular …

Heterogeneity Among Isolates Reveals That Fitness In Low Oxygen Correlates With Aspergillus Fumigatus Virulence, Caitlin H. Kowalski, Sarah R. Beattie, Kevin K. Fuller, Elizabeth A. Mcgurk, Yi-Wei Tang, Tobias Hohl, Joshua Obar, Robert Cramer Jr. Sep 2016

Heterogeneity Among Isolates Reveals That Fitness In Low Oxygen Correlates With Aspergillus Fumigatus Virulence, Caitlin H. Kowalski, Sarah R. Beattie, Kevin K. Fuller, Elizabeth A. Mcgurk, Yi-Wei Tang, Tobias Hohl, Joshua Obar, Robert Cramer Jr.

Dartmouth Scholarship

Previous work has shown that environmental and clinical isolates of Aspergillus fumigatus represent a diverse population that occupies a variety of niches, has extensive genetic diversity, and exhibits virulence heterogeneity in a number of animal models of invasive pulmonary aspergillosis (IPA). However, mechanisms explaining differences in virulence among A. fumigatus isolates remain enigmatic. Here, we report a significant difference in virulence of two common lab strains, CEA10 and AF293, in the murine triamcinolone immunosuppression model of IPA, in which we previously identified severe low oxygen microenvironments surrounding fungal lesions. Therefore, we hypothesize that the ability to thrive within these lesions …

The Fly

Children's Book and Media Review

This poor housefly doesn’t seem to understand why nobody likes him. First, the boy in the story tries to swat him just for wanting some of his breakfast. Then, even animals can’t seem to get along with him—cows are annoyed with him, frogs try to eat him, and birds try to catch him. When the fly returns home for another meal, the boy yet again flaps his swatter with the hopes of swatting the poor fly.

Patterns And Predictors Of Recent Forest Conversion In New England, Alexandra M. Thorn, Jonathan R. Thompson, Joshua S. Plisinski

New Hampshire EPSCoR

New England forests provide numerous benefits to the region’s residents, but are undergoing rapid development. We used boosted regression tree analysis (BRT) to assess geographic predictors of forest loss to development between 2001 and 2011. BRT combines classification and regression trees with machine learning to generate non-parametric statistical models that can capture non-linear relationships. Based on National Land Cover Database (NLCD) maps of land cover change, we assessed the importance of the biophysical and social variables selected for full region coverage and minimal collinearity in predicting forest loss to development, specifically: elevation, slope, distance to roads, density of highways, distance …

My Love/Hate Relationship With The County Fair, Laura Hanby Hudgen

Erma Bombeck Writers' Workshop Blog

Anyone even remotely familiar with small towns knows that that the biggest event of the year (next to Friday Night Football) is the County Fair. For one week, normally sensible parents forego bedtimes, healthy eating, proper hygiene and a whole lot of money in favor of the carnival life. Every year I look forward to the fair. And every year I dread it. I have a love/hate relationship with the County Fair.

Four-Dimensional (4d) Subharmonic Aided Pressure Estimation For Monitoring Neoadjuvant Chemotherapy Response Of Breast Cancer, Kibo Nam, Maria Stanczak, Anush Sridharan, Adam C. Berger, Tiffany Avery, John R. Eisenbrey, Flemming Forsberg

Department of Radiology Posters

Introduction

Neoadjuvant chemotherapy is a well-established treatment option for patients with non-metastatic breast cancer. The patient response is correlated with survival. However, the optimal method for monitoring neoadjuvant therapy response has not been established. One factor that may affect the response of neoadjuvant therapy is the interstitial fluid pressure (IFP). Increased IFP prevents an effective delivery of therapeutic agents and reduces the efficacy of the therapy. Recently, subharmonic-aided pressure estimation (SHAPE) using contrast-enhanced ultrasound (CEUS) has been developed and its potential was demonstrated in animals as a non-invasive technique for IFP measurements. The SHAPE method estimates IFP based on the …

Intenciones Enmascaradas En La Pantalla Plateada. El Santo Y El Mimetismo Imperial, David S. Dalton

Alambique.
