Center for Molecular Medicine and Genetics

Gene-by-environment (GxE) interactions determine common disease risk factors and biomedically relevant complex traits. However, quantifying how the environment modulates genetic effects on human quantitative phenotypes presents unique challenges. Environmental covariates are complex and difficult to measure and control at the organismal level, as found in GWAS and epidemiological studies. An alternative approach focuses on the cellular environment using in vitro treatments as a proxy for the organismal environment. These cellular environments simplify the organism-level environmental exposures to provide a tractable influence on subcellular phenotypes, such as gene expression. The proper establishment of epithelial polarity allows cells to sense and respond to signals that arise from the microenvironment in a spatiotemporally controlled manner. Atypical PKCs (aPKCs) are implicated as key regulators of epithelial polarity. However, the molecular mechanism underlying the negative regulation of aPKCs remains largely unknown. In this study, we demonstrated that PH domain leucine-rich repeat protein phosphatase (PHLPP), a novel family of Ser/Thr protein phosphatases, plays an important role in regulating epithelial polarity by controlling the phosphorylation of both aPKC isoforms. Interleukin-17a Promotes Cd8 T Cell Cytotoxicity To Facilitate West Nile Virus Clearance, Amber M. Paul, Dhiraj Acharya, Penghua Wang, Jianfeng Dai, David Gate

CD8 T cells are crucial components of immunity and play a vital role in recovery from West Nile virus (WNV) infection. Here, we identify a previously unrecognized function of interleukin-17A (IL-17A) in inducing cytotoxic-mediator gene expression and promoting CD8 T cell cytotoxicity against WNV infection in mice. We find that IL-17A-deficient (Il17a/) mice are more susceptible to WNV infection and develop a higher viral burden than wild-type (WT) mice. Interestingly, the CD8 T cells isolated from Il17a/ mice are less cytotoxic and express lower levels of cytotoxic-mediator genes, which can be restored by supplying recombinant IL-17A in vitro and in …

Better Late Than Never: Effective Air-Borne Hearing Of Toads Delayed By Late Maturation Of The Tympanic Middle Ear Structures, Molly C. Womack, Jakob Christensen-Dalsgaard, Kim L. Hoke

Most vertebrates have evolved a tympanic middle ear that enables effective hearing of airborne sound on land. Although inner ears develop during the tadpole stages of toads, tympanic middle ear structures are not complete until months after metamorphosis, potentially limiting the sensitivity of post-metamorphic juveniles to sounds in their environment. We tested the hearing of five species of toads to determine how delayed ear development impairs airborne auditory sensitivity. We performed auditory brainstem recordings to test the hearing of the toads and used micro-computed tomography and histology to relate the development of ear structures to hearing ability. This paper is about memory, the elusive process of remembering and of an encounter between a researcher and a participant who after five years reunited to remember. The object under study is a high school social justice curriculum with a central focus on the development of social action projects. In What Sense Are You A Person?, Pamela Barone, Antoni Gomila

Animal Sentience

According to Rowlands, personhood in nonhuman animals calls for a unified mental life and pre-reflective self-awareness provides this. The concept of “person” is fuzzy. Any attempt to define it with necessary and sufficient conditions faces the problem of borderline cases satisfying only some of the conditions to varying degrees. We ask about the implications of a metaphysical sense of personhood for its moral and legal sense. Finally, we address Rowlands’s reliance on pre-reflective self-awareness and present our own criteria for personhood.

A New Species Of Centruroides (Scorpiones: Buthidae) From Quiché, Northwestern Guatemala, Rony E. Trujillo, Luis F. De Armas

A new species of the genus Centruroides Marx, 1890 is described from Sotzil Village (15.61775°N, -91.09745°W, 1173 m a.s.l.), Chajul Municipality, Quiché Department, Guatemala, on basis of one female and one male. It closely resembles Centruroides caral Armas et Trujillo, 2013 from northeastern Guatemala (240 Km southeast of Chajul), but male differs by having a most globose pedipalp manus, as well as metasoma clearly more attenuate (len-gth/width ratio: segments II–IV = 2.4, 2.7 and 1.4, respectively; 1.7, 2.2 and 2.5 in the holotype of C. caral, the only known specimen of this taxon). “Our Common Future” harmonized development policies around a new sustainable development (SD) paradigm, and experts also emphasize the importance of a democratic and equitable approach to define and achieve sustainable development. However, SD targets and indicators are often defined by a suite of experts or a few stakeholder groups, far removed from on-the-ground conditions. The most common expert-led development framework, the United Nations’ Millennium Development Goals (MDGs), promoted one set of targets and indicators for all developing countries. The historiography of the Philippines has been largely bounded by the nation-state, which has defined how its past has been conceived and to whom its peoples are mainly compared. A more transnational environmental history, however, seeks to situate the archipelago within the context of the daily threats that its peoples have to face. The scientific endeavors of the Observatorio Meteorológico de Manila in the nineteenth century brought about major changes in colonial science in the Philippines. The observatory pioneered the modern study of weather, climate, and natural hazards through the use of instruments and apparatus it invented and produced. Philippine Studies: Historical and Ethnographic Viewpoints

In contrast to the literature’s dominant focus on Western constructions of tropicality, thisarticle explores representations of the tropics by the colonized, specifically the climatological conditions of the Philippines as portrayed in the late nineteenth century by the Europe-based native intellectuals known as ilustrados. Their anticolonial sentiment was intertwined with visceral estrangement from Spain and idealized views of the tropics, which reversed the colonizers’ racial-geographic prejudice and asserted an identity as a civilizable tropical people capable of genius. Rizal’s return visit to the homeland in 1887, however, made him agree with the Spanish premise about the climate in order to argue …

Ischemic stroke is a leading cause of adult disability, including cognitive impairment. Our laboratory has previously shown that treatment with function-blocking antibodies against the neurite growth inhibitory protein Nogo-A promotes functional recovery after stroke in adult and aged rats, including enhancing spatial memory performance, for which the hippocampus is critically important. Since spatial memory has been linked to hippocampal neurogenesis, we investigated whether anti-Nogo-A treatment increases hippocampal neurogenesis after stroke. Adult rats were subject to permanent middle cerebral artery occlusion followed 1 week later by 2 weeks of antibody treatment. Cellular proliferation in the dentate gyrus was quantified at the …
