Biomedical Engineering ETDs

Poisoning by Botulinum Neurotoxins (BoNTs) leads to the severe neuroparalytic illness known as botulism, which is characterized by flaccid paralysis, respiratory failure, and death. BoNTs are the most acutely toxic substances on the planet, so detection at physiologically relevant concentrations is difficult. Because current treatments for botulism cannot reverse existing paralysis, rapid detection is key, and current diagnostic techniques are not meeting this need. We have developed a flow-based assay platform that allows for detection of the Light Chain of BoNT/A as well as a means to study the action of the toxin on its protein substrate in a system …

Emerging evidence demonstrates that the DNA repair kinase DNA-PKcs exerts divergent roles in transcriptional regulation of unsolved consequence. Here, in vitro and in vivo interrogation demonstrate that DNA-PKcs functions as a selective modulator of transcriptional networks that induce cell migration, invasion, and metastasis. Accordingly, suppression of DNA-PKcs inhibits tumor metastases. Clinical assessment revealed that DNA-PKcs is significantly elevated in advanced disease and independently predicts for metastases, recurrence, and reduced overall survival. Further investigation demonstrated that DNA-PKcs in advanced tumors is highly activated, independent of DNA damage indicators. Combined, these findings reveal unexpected DNA-PKcs functions, identify DNA-PKcs as a potent driver …

Faculty Publications

Introduction

The growth and expression of cancer stem cells (CSCs) depend on many factors in the tumor microenvironment. The objective of this work was to investigate the effect of cancer cells’ tissue origin on the optimum matrix stiffness for CSC growth and marker expression in a model polyethylene glycol diacrylate (PEGDA) hydrogel without the interference of other factors in the microenvironment.

Methods

Human MCF7 and MDA-MB-231 breast carcinoma, HCT116 colorectal and AGS gastric carcinoma, and U2OS osteosarcoma cells were used. The cells were encapsulated in PEGDA gels with compressive moduli in the 2-70 kPa range and optimized cell seeding density …

The Mouse That Trolled: The Long And Tortuous History Of A Gene Mutation Patent That Became An Expensive Impediment To Alzheimer's Research, Tania Bubela, Saurabh Vishnubhakat, Robert Cook-Deegan

Faculty Scholarship

This case study presents the tale of the academic discovery of a rare mutation for early-onset Alzheimer's disease that was patented by a sole inventor and licensed to a non-practicing entity (NPE), the Alzheimer's Institute of America (AIA). Our aims are (1) to relate this story about patents, research tools, and impediments to medical progress, and (2) to inform ongoing debates about how patents affect research, disposition of university inventions, and the distribution of benefits from publicly funded research. We present an account of the hunt for Alzheimer's genes, their patenting, assignment, and enforcement based on literature, litigation records and …

The Role Of Philornis Downsi In Avian Disease Transmission In The Galápagos Islands, Courtney Lauren Pike Jul 2015

The Role Of Philornis Downsi In Avian Disease Transmission In The Galápagos Islands, Courtney Lauren Pike

Theses

Philornis downsi is a dipteran fly species known to parasitize passerine birds in the Neotropics. P. downsi is speculated to be a recently introduced species in Galápagos Islands, so it is important to understand its effects on the archipelago’s native and endemic avian species. Avian parasites and pathogens have been previously reported in the Galápagos, leading the Galápagos National Park and the Charles Darwin Foundation to focus efforts on the study and management of potential avian disease vectors. The main goal of this study was to determine whether P. downsi is a host for avian parasites and pathogens found in …

Dumbbell-Pcr: A Method To Quantify Specific Small Rna Variants With A Single Nucleotide Resolution At Terminal Sequences., Shozo Honda, Yohei Kirino

Computational Medicine Center Faculty Papers

Recent advances in next-generation sequencing technologies have revealed that cellular functional RNAs are not always expressed as single entities with fixed terminal sequences but as multiple isoforms bearing complex heterogeneity in both length and terminal sequences, such as isomiRs, the isoforms of microRNAs. Unraveling the biogenesis and biological significance of heterogenetic RNA expression requires distinctive analysis of each RNA variant. Here, we report the development of dumbbell PCR (Db-PCR), an efficient and convenient method to distinctively quantify a specific individual small RNA variant. In Db-PCR, 5'- and 3'-stem-loop adapters are specifically hybridized and ligated to the 5'- and 3'-ends of …

Aspirin Triggered Resolution Phase Interaction Product D1: A Novel Treatment For Hyperoxic Acute Lung Injury, Ruan Rollin Cox, Jr.

USF Tampa Graduate Theses and Dissertations

Acute Lung injury (ALI) and the more severe acute respiratory distress syndrome (ARDS) are respiratory maladies that present immense clinical challenges. ALI affects 200,000 individuals annually and features a 40% mortality rate. ALI can be initiated by both pathogenic and sterile insults originating locally in the lungs or systemically. While immense research has been poured into this disease in an effort to find a therapeutic strategy, the heterogeneously diffuse nature of the disease has not yielded a cure for the disease. Death from this disease is strongly attributed to reduced gas exchange from a severely compromised alveolar-capillary barrier. The only …

Microwave-Assisted Synthesis Of Ss-Amino Alcohols, Hinalbahen Sanket Desai Jul 2015

Microwave-Assisted Synthesis Of Ss-Amino Alcohols, Hinalbahen Sanket Desai

Master's Theses and Doctoral Dissertations

ß-amino alcohols are common substructures in many biologically active compounds and can also be used as catalysts for asymmetric reactions. One common method for forming these moieties is by ring opening of unsymmetrical epoxides using amines via a substitution reaction. However, the majority of these methods requires excess of reagents, inorganic initiators, and/or extended reaction times. We have developed an efficient, regioselective route to synthesize amino alcohols via microwave-assisted aminolysis of several hindered and unhindered epoxides using amine nucleophiles of varying strengths. Microwave reactions can be done without Lewis acids or promoters, even for the most hindered trisubstituted epoxides. In …

Nervous System Development In Lecithotrophic Larval And Juvenile Stages Of The Annelid Capitella Teleta, Néva P. Meyer, Allan Carrillo-Baltodano, Richard E. Moore, Elaine C. Seaver

Biology

Background: Reconstructing the evolutionary history of nervous systems requires an understanding of their architecture and development across diverse taxa. The spiralians encompass diverse body plans and organ systems, and within the spiralians, annelids exhibit a variety of morphologies, life histories, feeding modes and associated nervous systems, making them an ideal group for studying evolution of nervous systems. Results: We describe nervous system development in the annelid Capitella teleta (Blake JA, Grassle JP, Eckelbarger KJ. Capitella teleta, a new species designation for the opportunistic and experimental Capitella sp. I, with a review of the literature for confirmed records. Zoosymposia. 2009;2:25-53) using …

Faculty Articles, Papers, and Other Scholarship

Background Plastome sequences for 18 species of the PACMAD grasses (subfamilies Panicoideae, Aristidoideae, Chloridoideae, Micrairoideae, Arundinoideae, Danthonioideae) were analyzed phylogenomically. Next generation sequencing methods were used to provide complete plastome sequences for 12 species. Sanger sequencing was performed to determine the plastome of one species, Hakonechloa macra, to provide a reference for annotation. These analyses were conducted to resolve deep subfamilial relationships within the clade. Divergence estimates were assessed to determine potential factors that led to the rapid radiation of this lineage and its dominance of warmer open habitats. Results New plastomes were completely sequenced and characterized for 13 PACMAD …

The Intersection Of Art And Medicine: Tanya Sheehan Breaks Down The Boundaries Between Disciplines, Gerry Boyle

Colby Magazine

Tanya Sheehan arrived at Georgetown University intending to become a medical doctor. She aced her organic chemistry lab section, eventually was turned off by the rote memorization the program emphasized, and then discovered art history and film studies. "That changed everything," she said, "but I never forgot about science and medicine."
