Photoacoustic Microscopy And Photoacoustic Computed Tomography Using High-Frequency Linear Array Ultrasonic Transducers, Guo Li Aug 2015

Photoacoustic Microscopy And Photoacoustic Computed Tomography Using High-Frequency Linear Array Ultrasonic Transducers, Guo Li

McKelvey School of Engineering Graduate Student Theses & Dissertations

Photoacoustic tomography (PAT) is a highly promising imaging technology which forms images by detecting the induced pressure waves resulting from pulsed light absorption in biological tissues. Because the excitation source is light, PAT is a very safe, non-ionizing, and non-carcinogenic imaging technology. In biomedicine, PAT has the unique advantage of probing endogenous optical absorbers at different length scales with 100% relative sensitivity. With such scalability, PAT can image anatomical, functional, metabolic, molecular, and genetic contrasts of vasculature, hemodynamics, oxygen metabolism, biomarkers, and gene expression. Among several implementations of PAT, optical-resolution photoacoustic microscopy (OR-PAM) and photoacoustic computed tomography (PACT) are two …

Using Fluorescence – Polarization Endoscopy In Detection Of Precancerous And Cancerous Lesions In Colon And Pancreatic Cancer, Tauseef Charanya

McKelvey School of Engineering Graduate Student Theses & Dissertations

Colitis-associated cancer (CAC) arises from premalignant flat lesions of the colon, which are difficult to detect with current endoscopic screening approaches. We have developed a complementary fluorescence and polarization reporting strategy that combines the unique biochemical and physical properties of dysplasia and cancer for real time detection of these lesions. Utilizing a new thermoresponsive sol-gel formulation with targeted molecular probe allowed topical application and detection of precancerous and cancerous lesions during endoscopy. Incorporation of nanowire-filtered polarization imaging into NIR fluorescence endoscopy served as a validation strategy prior to obtaining biopsies.

In order to reduce repeat surgeries arising from incomplete tumor …

Online Modeling And Tuning Of Parallel Stream Processing Systems, Jonathan Curtis Beard Aug 2015

Online Modeling And Tuning Of Parallel Stream Processing Systems, Jonathan Curtis Beard

McKelvey School of Engineering Graduate Student Theses & Dissertations

Writing performant computer programs is hard. Code for high performance applications is profiled, tweaked, and re-factored for months specifically for the hardware for which it is to run. Consumer application code doesn't get the benefit of endless massaging that benefits high performance code, even though heterogeneous processor environments are beginning to resemble those in more performance oriented arenas. This thesis offers a path to performant, parallel code (through stream processing) which is tuned online and automatically adapts to the environment it is given. This approach has the potential to reduce the tuning costs associated with high performance code and brings …

Synthesis Of Clickable Poly(Ethylene Glycol) Derivatives For Fabrication Of Modular Microsphere-Based Scaffolds To Promote Vascularization, Peter Nguyen Aug 2015

Synthesis Of Clickable Poly(Ethylene Glycol) Derivatives For Fabrication Of Modular Microsphere-Based Scaffolds To Promote Vascularization, Peter Nguyen

McKelvey School of Engineering Graduate Student Theses & Dissertations

Vascularization plays an important role in supporting transplanted tissues and cells in tissue engineering applications. Most tissues require access to blood vessels for the delivery of oxygen and nutrients, as well as the removal of carbon dioxide and cellular waste products. Without an adequate blood supply, cells within tissue-engineered constructs and scaffolds lose viability and fail to perform their intended functions. The goal of this dissertation was to design scaffolds that can promote vascularization of biomaterial implants for biomedical applications. In order to accomplish this goal, clickable poly(ethylene glycol) (PEG) derivatives were synthesized in order to fabricate modular microsphere-based scaffolds …

Directed Differentiation And Characterization Of Spinal V3 Interneurons From Mouse Embryonic Stem Cells, Hao Xu Aug 2015

Directed Differentiation And Characterization Of Spinal V3 Interneurons From Mouse Embryonic Stem Cells, Hao Xu

McKelvey School of Engineering Graduate Student Theses & Dissertations

Neuronal populations involved in locomotion controlling central pattern generators within the spinal cord hold great potential for spinal cord injury therapy. Spontaneous recovery in rodent models suggests that a population that can reorganize around an injury site could be useful for functional recovery therapeutics after spinal cord injury. The glutamatergic, commissural, long-extending V3 interneurons shown to balance locomotor rhythm regularity and robustness within central pattern generators in vivo are both an ideal population for spinal cord injury therapeutics and a vital population to study as a part of locomotor circuitry. Unfortunately, due to the scarcity of these cells in the …

Estimated Mouse Population At The Pennypack Ecological Restoration Trust Vs. Reported Incidence Of Lyme Disease In Pennsylvania, Erika Clark

The Compass

The purpose of this research paper is to determine if the mouse population at the PennyPack Trust is correlated with occurrence of Lyme disease state wide.

Mark-recapture studies of white footed mice, Peromyscus Leucopus, were held at the PennyPack Ecological Restoration Trust in Huntingdon, Pennsylvania in order to estimate the population sizes over six years. The common method of ear tagging has been shown not to interfere with the daily activities of mice, or increase their chances of mortality.

White-footed mice are primary hosts of the black-legged tick, Ixodes scapularis, which is the vector of Borrelia burgdorferi, the bacteria that …

From Orphans To Families In Crisis: Parental Rights Matters In Maine Probate Courts, Deirdre M. Smith Aug 2015

From Orphans To Families In Crisis: Parental Rights Matters In Maine Probate Courts, Deirdre M. Smith

Faculty Publications

This Article examines the sources of the contemporary problems associated with the adjudication of parental rights matters in Maine’s probate courts and identifies specific reforms to address both the structural and substantive law problems. The Article first reviews the development of Maine’s probate courts and their jurisdiction over parental rights matters. It traces the expansion of jurisdiction over children and families from a limited role incidental to the administration of a decedent’s estate to the current scope: a range of matters that may result in the limitation, suspension, or termination of the rights of living parents. Maine probate courts now …

An Examination Of Net Primary Production In Southern Appalachian Wetlands, Mike Maguigan Aug 2015

An Examination Of Net Primary Production In Southern Appalachian Wetlands, Mike Maguigan

Theses and Dissertations

Southern Appalachian wetlands have yet to be studied in terms of net primary production (NPP), thus few studies have been conducted to examine what environmental factors have relationships with NPP. To that end, this research investigates several facets of southern Appalachian wetland production. The research was divided into three studies. The first study was conducted to answer the question of what environmental factors have relationships with NPP. It appears that stream discharge and annual precipitation had the strongest relationships with NPP (r = 0.91, p <0.05 and r = 0.81, p <0.05, respectively), yet both factors showed multicolinearity (r = 0.97, p <0.05). The strong relationships between hydrologic factors and NPP is similar to montane wetlands in the western United States. The second study was conducted to examine the relationship between water chemistry and NPP. Calcium (Ca), Magnesium (Mg), and pH were examined in order to determine if any of the aforementioned factors had a relationship with NPP. Neither Ca (r = -0.34, p = 0.0835) nor Mg (r =-0.38, p = 0.0535) had strong relationships with NPP, though pH (r = -0.66, p <0.05) had a strong negative relationship with NPP. The acidity of the stream water, driven by the acid rain in the southern Appalachians, creates enhanced conditions for wetland plants to grow. The third study was conducted to establish which vegetation index was best for estimating NPP from proximally and remotely sensed data. The findings suggest that VARIRed Edge was best for examining NPP at the in situ level, NDVI was best for examining NPP at the airborne level, and the DVI was the best for examining NPP at the satellite level. NPP in southern Appalachian wetlands is driven by the chemistry, specifically the pH, of stream discharge and annual precipitation and can be monitored by NDVI using NAIP data or DVI using Landsat data. The examination of NPP in southern Appalachians in response to environmental factors and water chemistry along with the examination of vegetation indices at three levels of platforms will help to monitor and manage these rare and unique ecosystems in the future.

Transcriptional And Post-Transcriptional Regulation Of Hepcidin And Iron Metabolism By Lipid Signaling In The Liver, Sizhao Lu Aug 2015

Transcriptional And Post-Transcriptional Regulation Of Hepcidin And Iron Metabolism By Lipid Signaling In The Liver, Sizhao Lu

Theses & Dissertations

Although iron is required for essential biological processes, excess iron is detrimental due to oxidative damage induced by iron-mediated Fenton reactions, which promote tissue injury. Cellular iron uptake, transport and storage must therefore be tightly regulated. This task is accomplished mainly through hepcidin, the key iron-regulatory hormone. Hepcidin is synthesized primarily in hepatocytes as a circulatory antimicrobial peptide. It controls iron metabolism by inhibiting iron absorption from the duodenum and iron release from reticuloendothelial macrophages. Besides synthesizing hepcidin, the liver plays an important role in maintaining iron homeostasis by serving as the main storage organ for excess iron. Patients with …

Variables In The Introduction Of The Species, Streptocephalus Sealii, Spinytail Fairy Shrimp Into A Local Seasonal Wetland, Jane Anne Porterfield

School of Education and Leadership Student Capstone Theses and Dissertations

The research question addressed in this capstone is what factors play a role in the introduction of a wingless aquatic invertebrate to a recently restored mitigation wetland. The motivation for this capstone was the discovery of a fairy shrimp, Streptocephalus sealii in a Rosemount, Minnesota wetland in 2012. This organism was previously recorded from two other sites in Minnesota at least 35 miles from this location. Its appearance in a mitigation wetland generated questions as to whether restoration materials brought in the species or the mitigation created the environment in which the organism could thrive. This study investigates both of …

Improving Antibiotic Resistant Infection Transmission Situational Awareness In Enclosed Facilities With A Novel Graphical User Interface For Tactical Biosurveillance, Valeriya V. Kettelhut

Theses & Dissertations

Serious challenges associated with antibiotic resistant infections (ABRIs) force healthcare practitioners (HCP) to seek innovative approaches that will slow the emergence of new ABRIs and prevent their spread. It was realized that traditional approaches to infection prevention based on education, retrospective reports, and biosurveillance often fail to ensure reliable compliance with infection prevention guidelines and real-time problem solving. The objective of this original research was to develop and test the conceptual design of a situational awareness (SA)-oriented information system for coping with healthcare-associated infection transmission.

Constantly changing patterns in spatial distribution of patients, prevalence of infectious cases, clustering of contacts, …

Development Of Polymer Peptide Conjugates For Enhanced Pancreatic Cancer Imaging, Wen Shi Aug 2015

Development Of Polymer Peptide Conjugates For Enhanced Pancreatic Cancer Imaging, Wen Shi

Theses & Dissertations

Pancreatic ductal adenocarcinoma (PDAC) is the fourth leading cause of cancer-related deaths in the US with very poor prognosis. All clinically available biomarkers and diagnostic tools either fail to detect early stage PDAC or suffer from low specificity and sensitivity. There is an urgent need for diagnostic agents with greater efficacy for PDAC detection and staging. Nanomaterials such as N-(2-Hydroxypropyl) methacrylamide (HPMA) copolymers can effectively target tumors, and offer novel opportunities for the development of effective diagnostic agents for cancer. However, a major problem of many nanomaterials-based diagnostics is their opsonization and sequestration by the mononuclear phagocyte system (MPS), leading …

The Role Of Glutaminase 1 In Hiv-1 Associated Neurocognitive Disorders And In Brain Development, Yi Wang Aug 2015

The Role Of Glutaminase 1 In Hiv-1 Associated Neurocognitive Disorders And In Brain Development, Yi Wang

Theses & Dissertations

Glutaminase is the enzyme that converts glutamine into glutamate, which serves as a key excitatory neurotransmitter and one of the energy providers for cellular metabolism. Glutamate is essential for proper brain functioning but at excess levels, it is neurotoxic and has a key role in the pathogenesis of various neurodegenerative diseases, including HIV-1 associated neurocognitive disorders (HAND). However, the detailed mechanism of glutamate-mediated neurotoxicity remains unclear. In part I, we identified the regulation of glutaminase 1 (GLS1) in the central nervous system (CNS) of HAND animal models including HIV-Tat transgenic (Tg) mouse and HIVE-SCID mouse, since GLS1 is the dominant …

Whether for cultural, spiritual, economic, or other purposes, the use of traditional and local knowledge of habitat and resource management is slowly making its way into the modern environmental movement, and is being incorporated into the dominant conservation paradigms. These managed areas, known as Indigenous and Community-Conserved Territories and Areas, or ICCAs, are defined by the International Union for Conservation of Nature as “natural and/or modified ecosystems containing significant biodiversity …

Factors Influencing Honey Bee Abundance Across Agricultural Landscapes In The Midsouth, Daniel Adam Whalen Aug 2015

Factors Influencing Honey Bee Abundance Across Agricultural Landscapes In The Midsouth, Daniel Adam Whalen

Theses and Dissertations

Populations of honey bees have declined worldwide in recent years. One suspected cause is the widespread use of pesticides in agriculture. Experiments were conducted to examine potential exposure routes of pesticides to honey bees in the Midsouth. Neonicotinoid seed treatment compounds were studied to determine the rate at which they drift during planting and the rate at which they diminish in crop tissue during crop development. Honey bee foraging activity in Midsouth crops was observed to determine when and at what densities foraging honey bees could be active during pesticide applications. This project was designed to aid in understanding the …

Allelopathy In The Non-Native Macrophyte, Myriophyllum Spicatum And Its Influence On Trophic Dynamics In Aquatic Systems, Daniel J. Sullivan Aug 2015

Allelopathy In The Non-Native Macrophyte, Myriophyllum Spicatum And Its Influence On Trophic Dynamics In Aquatic Systems, Daniel J. Sullivan

Theses and Dissertations

Non-native macrophytes structurally impact aquatic assemblages, yet little is known regarding how they influence energy pathways in freshwater ecosystems. Allelopathy in Eurasian watermilfoil- Myriophyllum spicatum has been shown to target basal epiphytic organisms resulting in differences in assemblage structure of colonizing epiphyton between M. spicatum and native M. sibiricum. I conducted a growth chamber experiment to investigate the hypothesis that differences in assemblage structure of colonizing epiphyton between these two macrophytes influence trophic dynamics within aquatic systems. My data suggest M. spicatum produces higher concentrations of allelochemicals, resulting in a more diverse epiphytic assemblage compared to M. sibiricum. This could …

The Involvement Of Let-7 Mirna In The Regulation Of Retinal Progenitor Cells, Xiaohuan Xia Aug 2015

The Involvement Of Let-7 Mirna In The Regulation Of Retinal Progenitor Cells, Xiaohuan Xia

Theses & Dissertations

Emerging evidence has shown that miRNA-mediated gene regulation plays an important role in the development of the central nervous system (CNS). One of the developmental mechanisms may involve let-7 miRNAs and their up-stream regulator, Lin28a and Lin28b in regulating neural stem cells (NSCs). Recently, let-7 has been observed to influence the differentiation of NSCs along neuronal versus glial lineage. However, there is also a report suggesting that the neurogliogenic decision is independent of let-7. These paradoxical results might represent contextual roles of let-7 and lin28 during the CNS development. We have tested this premise in the mammalian retina, a reliable …

Investigation Of Behavioral And Cellular Changes In The Maternal Immune Activation Model Of Autism Spectrum Disorders, Shreya Roy Aug 2015

Investigation Of Behavioral And Cellular Changes In The Maternal Immune Activation Model Of Autism Spectrum Disorders, Shreya Roy

Theses & Dissertations

Maternal infection during pregnancy, which leads to maternal immune activation (MIA), is an environmental risk factor for autism spectrum disorders (ASD). MIA can be induced in mice and their offspring exhibit behaviors that model the core symptoms of ASD. One of the core behavioral symptoms in ASD patients is presence of increased repetitive behavior, which is modeled by an increase in marble burying in MIA mice. It has been shown that the deficits seen in MIA mice are associated with the dysregulation of cytokine levels in the developing brain, specifically an increase in pro-inflammatory cytokines. In this thesis, I tested …

Impact Of Estrogen Receptor Alpha On Sle1-Induced Loss Of Tolerance, Shayla D. Yoachim Aug 2015

Impact Of Estrogen Receptor Alpha On Sle1-Induced Loss Of Tolerance, Shayla D. Yoachim

Theses & Dissertations

The autoimmune disease lupus shows a significant female sex bias. This sex bias may be due to the ability of estrogens to promote loss of tolerance to chromatin, the initial loss of tolerance event in lupus. Previously, we demonstrated that the ability of estrogens to promote lupus in (NZBxNZW) F1 mice is dependent on signaling via estrogen receptor alpha (ERα). The Sle1 lupus susceptibility allele controls loss of tolerance to chromatin, and C57BL/6 (B6) mice carrying the Sle1 lose tolerance and develop anti-chromatin autoantibodies and spontaneously activated immune cells. Loss of tolerance occurs earlier and with a higher penetrance in …
