Training "In A Good Way": Evaluating The Effect Of A Culturally Responsive Pre-Training Intervention On Learning And Motivation, Adam Thomas Murry
Dissertations and Theses
Employee Training and Development (T&D) is a crucial component to an organization’s success and its ability to remain competitive. Although researchers in the field have discovered ways to enhance the effectiveness of training programs through the design, delivery, and evaluation process, research has not provided empirically-based recommendations for how to best train individuals whose cultural backgrounds may influence receptiveness of training curriculum. This is particularly relevant for employees whose cultural groups have been historically discriminated against, where cultural norms implicit in the training design may be met with resistance on behalf of the trainees. In the field of multicultural education, …

Sialic acids (SAs) are widely expressed on immune cells and their levels and linkages named as sialylation status vary upon cellular environment changes related to both physiological and pathological processes. In this study, we performed a global profiling of the sialylation status of macrophages and their release of SAs in the cell culture medium by using flow cytometry, confocal microscopy and liquid chromatography tandem mass spectrometry (LC-MS/MS). Both flow cytometry and confocal microscopy results showed that cell surface α-2,3-linked SAs were predominant in the normal culture condition and changed slightly upon treatment with atorvastatin for 24 h, whereas α-2,6-linked SAs …

Rna-Seq Analysis Of Differential Splice Junction Usage And Intron Retentions By Dexseq, Yafang Li, Xiayu Rao, William W. Mattox, Christopher I. Amos, Bin Liu
Dartmouth Scholarship
Alternative splicing is an important biological process in the generation of multiple functional transcripts from the same genomic sequences. Differential analysis of splice junctions (SJs) and intron retentions (IRs) is helpful in the detection of alternative splicing events. In this study, we conducted differential analysis of SJs and IRs by use of DEXSeq, a Bioconductor package originally designed for differential exon usage analysis in RNA-seq data analysis. We set up an analysis pipeline including mapping of RNA-seq reads, the preparation of count tables of SJs and IRs as the input files, and the differential analysis in DEXSeq. We analyzed the …

Intracellular Na+ Concentration ([Na+]I) Is Elevated In Diabetic Hearts Due To Enhanced Na+–Glucose Cotransport, Rebekah Lambert, Sarah Srodulski, Xiaoli Peng, Kenneth B. Margulies, Florin Despa, Sanda Despa
Pharmacology and Nutritional Sciences Faculty Publications
BACKGROUND: Intracellular Na+ concentration ([Na+]i) regulates Ca2+ cycling, contractility, metabolism, and electrical stability of the heart. [Na+]i is elevated in heart failure, leading to arrhythmias and oxidative stress. We hypothesized that myocyte [Na+]i is also increased in type 2 diabetes (T2D) due to enhanced activity of the Na+-glucose cotransporter.
METHODS AND RESULTS: To test this hypothesis, we used myocardial tissue from humans with T2D and a rat model of late-onset T2D (HIP rat). Western blot analysis showed increased Na+-glucose cotransporter expression in failing hearts from T2D patients compared …

Coordination Between Autophagy And The Heat Shock Response: Evidence From Exercise In Animals And Humans, Nathan H. Cole
Coordination Between Autophagy And The Heat Shock Response: Evidence From Exercise In Animals And Humans, Nathan H. Cole
Health, Exercise, and Sports Sciences ETDs
Proteins play a critical role in nearly every biological activity. In consequence, organismal health and homeostasis often hinges on the ability of intracellular regulatory systems to sustain the quality and function of these diverse, structurally complex macromolecules. Correct protein function depends on correct form, and during periods of destabilizing cellular stress, protein quality is managed in part by the heat shock response, which acts to support, isolate, and reform new or damaged proteins, and in part by the autophagic recycling of abnormal proteins, cytotoxic protein aggregates, and terminally damaged organelles. We conducted a pooled analysis of available research in humans …

A Multiple Case Examination Of Sexuality In Counselor Education: Pedagogy, Ideology, And Discourse, Megan Speciale
Individual, Family, and Community Education ETDs
In this study, I explored the role and function of counselor educators' ideology in their sexuality counseling pedagogy, as demonstrated by teaching philosophy, curriculum, and teaching strategies. In line with my theoretical framework, queer feminist pedagogy, I employed qualitative multicase methodology with six counselor educators, bounded by the shared phenomena of teaching sexuality-focused counseling curricula. I performed critical discourse analysis on the dialogic and textual discourse generated from interviews and curricular document mining to identify ideologies embedded in educators' discourse. The analysis revealed the considerable influence of personal ideology on educators' sexuality counseling pedagogy, which constituted the following themes: (a) …

Quinol Oxidase Encoded By Cyoabcd In Rhizobium Etli Cfn42 Is Regulated By Actsr And Is Crucial For Growth At Low Ph Or Low Iron Conditions, Zachary Ryan Lunak, K. Dale Noel
Biological Sciences Faculty Research and Publications
Rhizobium etli aerobically respires with several terminal oxidases. The quinol oxidase (Cyo) encoded by cyoABCD is needed for efficient adaptation to low oxygen conditions and cyo transcription is upregulated at low oxygen. This study sought to determine how transcription of the cyo operon is regulated. The 5′ sequence upstream of cyo was analysed in silico and revealed putative binding sites for ActR of the ActSR two-component regulatory system. The expression of cyo was decreased in an actSR mutant regardless of the oxygen condition. As ActSR is known to be important for growth under low pH in another rhizobial species, the …

UNLABELLED: The bone is a preferred site for metastatic homing of prostate cancer cells. Once prostate cancer patients develop skeletal metastases, they eventually succumb to the disease; therefore, it is imperative to identify key molecular drivers of this process. This study examines the involvement of protein kinase C epsilon (PKCε), an oncogenic protein that is abnormally overexpressed in human tumor specimens and cell lines, on prostate cancer cell bone metastasis. PC3-ML cells, a highly invasive prostate cancer PC3 derivative with bone metastatic colonization properties, failed to induce skeletal metastatic foci upon inoculation into nude mice when PKCε expression was silenced …

Less Is More: The Physiological Basis For Tapering In Endurance, Strength, And Power Athletes, Kevin A. Murach, James R. Bagley
Physical Therapy Faculty Publications
Taper, or reduced-volume training, improves competition performance across a broad spectrum of exercise modes and populations. This article aims to highlight the physiological mechanisms, namely in skeletal muscle, by which taper improves performance and provide a practical literature-based rationale for implementing taper in varied athletic disciplines. Special attention will be paid to strength- and power-oriented athletes as taper is under-studied and often overlooked in these populations. Tapering can best be summarized by the adage “less is more” because maintained intensity and reduced volume prior to competition yields significant performance benefits.
The New Peonage, Tamar R. Birckhead
The New Peonage, Tamar R. Birckhead
Washington and Lee Law Review
Although the Thirteenth Amendment to the Constitution formally abolished slavery and involuntary servitude in 1865, the text created an exception for the punishment of crimes “whereof the party shall have been duly convicted.” Two years later, Congress passed The Anti-Peonage Act in an attempt to prohibit the practice of coerced labor for debt. Yet, in the wake of the Civil War, Southern states innovated ways to impose peonage but avoid violations of the law, including criminal surety statutes that allowed employers to pay the court fines for indigent misdemeanants charged with minor offenses in exchange for a commitment to work. …

Testosterone Might Not Be Necessary To Support Female Aggression In Incubating Northern Cardinals, M. Susan Devries, Caitlin P. Winters, Jodie M. Jawor
Faculty Publications
Testosterone's (T) influence on male aggression has been well established in many vertebrate species, but the impact of T on female aggressive behaviour is poorly understood. Among birds, a link between T and female aggression is plausible, as females of many species exhibit a seasonal peak in T concentrations at the onset of breeding when social instability is greatest and they may have circulating T through much of the breeding season. However, investigations examining the relationship between T and female aggression are few and have yielded conflicting results, with experimentally or endogenously elevated T supporting aggressive behaviour in females of …
