Lipin1, an intracellular protein, plays critical roles in controlling lipid synthesis and energy metabolism through its enzymatic activity and nuclear transcriptional functions. Several mouse models of skeletal muscle wasting are associated with lipin1 mutation or altered expression. Recent human studies have suggested that children with homozygous null mutations in the LPIN1 gene suffer from rhabdomyolysis. However, the underlying pathophysiologic mechanism is still poorly understood. In the present study we examined whether lipin1 contributes to regulating muscle regeneration. We characterized the time course of skeletal muscle regeneration in lipin1-deficient fld mice after injury. We found that fld mice exhibited smaller regenerated …

This study investigates the use of saliva, as an emerging diagnostic fluid in conjunction with classification techniques to discern biological heterogeneity in clinically labelled gingivitis and periodontitis subjects (80 subjects; 40/group) A battery of classification techniques were investigated as traditional single classifier systems as well as within a novel selective voting ensemble classification approach (SVA) framework. Unlike traditional single classifiers, SVA is shown to reveal patient-specific variations within disease groups, which may be important for identifying proclivity to disease progression or disease stability. Salivary expression profiles of IL-1ß, IL-6, MMP-8, and MIP-1α from 80 patients were analyzed using four classification …

Influence Of Ph And Bulk Density On Carbon Dioxide Efflux In Three Urban Wetland Types, Christopher Chappell, Andra Johnson
Professional Agricultural Workers Journal
Abstract
The aim of this study was to understand soil carbon dioxide (CO2) efflux of three different urban wetlands and how pH and bulk density relate to soil CO2 efflux of each wetland. The three wetlands were bottomland, upland, and shrub/scrub. The study was conducted over a twenty week period using the Li-Cor 8100 “closed chamber” method to measure soil CO2 efflux. The findings show that the bottomland wetland efflux was significantly higher than the shrub/scrub and upland wetland. The pH of shrub/scrub was significantly higher than the upland. The bulk density of the bottomland was …

Fatty Acid Transport Protein-2 Inhibitor Grassofermata/Cb5 Protects Cells Against Lipid Accumulation And Toxicity, Nipun Saini, Paul N. Black, David Montefusco, Concetta Dirusso
Department of Biochemistry: Faculty Publications
The inhibition of the fatty acid uptake into non-adipose tissues provides an attractive target for prevention of lipotoxicity leading to obesity-associated non-alcoholic fatty liver disease and type 2 diabetes. Fatty acid transport proteins (FATPs) are bifunctional proteins involved in the uptake and activation of fatty acids by esterification with coenzyme A. Here we characterize Grassofermata/CB5, previously identified as a fatty acid uptake inhibitor directed against HsFATP2. The compound was effective in inhibiting the uptake of fatty acids in the low micro-molar range (IC50 8–11μM) and prevented palmitate-mediated lipid accumulation and cell death in cell lines that are models for …

Heterogeneous Oxidation Of Catechol, Elizabeth A. Pillar, Ruixin Zhou, Marcelo I. Guzman
Chemistry Faculty Publications
Natural and anthropogenic emissions of aromatic hydrocarbons from biomass burning, agro-industrial settings, and fossil fuel combustion contribute precursors to secondary aerosol formation (SOA). How these compounds are processed under humid tropospheric conditions is the focus of current attention to understand their environmental fate. This work shows how catechol thin films, a model for oxygenated aromatic hydrocarbons present in biomass burning and combustion aerosols, undergo heterogeneous oxidation at the air–solid interface under variable relative humidity (RH = 0–90%). The maximum reactive uptake coefficient of O3(g) by catechol γO3 = (7.49 ± 0.35) × 10–6 occurs for …

Genomic Deoxyuracil: Targeting, Regulation And Repair, Nate Warren Holton
Theses and Dissertations
Maintaining genomic integrity is critical to avoid sequence mutations and genetic disease. However the immune system requires genomic rearrangements in order to produce novel antibodies and illicit a full immunological response. The expression of DNA modifying enzymes is essential for antibody production but the off target effects are not well understood. The work presented here was performed to elucidate molecular mechanisms responsible for the regulation and targeting of DNA editing enzymes, as well as characterize repair of these lesions at off target loci.

A Pilot Study Of Demographic And Dopaminergic Genetic Contributions To Weight Change In Kidney Transplant Recipients, Ansley Stanfill, Donna Hathaway, Ann Cashion, Ramin Homayouni, Patricia Cowan, Carol Thompson, Behrouz Madahian, Yvette Conley
Nursing Faculty Publications
Kidney transplant recipients often experience a significant amount of weight gain in the first year post-transplantation. While demographic factors such as age, race, and sex have been associated with weight gain in this population, these factors do not explain all of the variability seen. A number of studies have suggested that genetics also plays a critical role in weight changes. Recently, alterations in the activity of the neurotransmitter dopamine have been associated with weight change, and gene expression studies in kidney transplant recipients have supported this association. The purpose of this pilot study is to first examine the feasibility and …

Introduction: Central American countries, like many others, face a shortage of rural health physicians. Most medical schools in this region are located in urban areas and focus on tertiary care training rather than on community health or primary care, which are better suited for rural practice. However, many countries require young physicians to do community service in rural communities to address healthcare provider shortages. This study aimed to: (a) synthesize what is known about the current state of medical education preparing physicians for rural practice in this region, and (b) identify common needs, challenges and opportunities for improving medical education …

School Science Capacity: A Study Of Four Urban Catholic Grade Schools, Lara K. Smetana, Elizabeth R. Coleman
School Science Capacity: A Study Of Four Urban Catholic Grade Schools, Lara K. Smetana, Elizabeth R. Coleman
Journal of Catholic Education
Working from the view of schools as a system, and of school improvement as an ongoing journey (Hallinger & Heck, 2011; Jackson, 2000), this study investigated the perspectives of teacher leaders and principals from four metropolitan Catholic grade schools engaged in efforts to improve their school science programs. Built upon existing conceptualizations of school capacity, a model of school science capacity is presented and used as a framework for the development of four case studies. Findings gleaned from collecting and analyzing reports of teacher leaders’ and principals’ interpretations of their school systems illustrate how elements of the school science capacity …
When Did It Begin? Catholic And Public School Classroom Commonalities, Richard T. Ognibene
When Did It Begin? Catholic And Public School Classroom Commonalities, Richard T. Ognibene
Journal of Catholic Education
Catholic educational historians note that although preserving Catholic identity has been a constant in the mission of Catholic schools, their curriculum and instructional practices evolved in ways that were similar to public schools, thus enabling Catholic parents to select schools that were both faith based and modern. Since there is an absence of information about when and how this change in Catholic education began, this article documents its origin in the 1940s when Catholic educators joined a public school reform movement called Life Adjustment Education. Once begun, there was no turning back, and Catholic educators participated in the major reforms …

Modeling Wood Fibre Length In Black Spruce (Picea Mariana (Mill.) Bsp) Based On Ecological Land Classification, Elisha Townshend, Bharat Pokharel, Art Groot, Doug Pitt, Jeffery P. Dech
Agricultural and Environmental Sciences Faculty Research
Effective planning to optimize the forest value chain requires accurate and detailed information about the resource; however, estimates of the distribution of fibre properties on the landscape are largely unavailable prior to harvest. Our objective was to fit a model of the tree-level average fibre length related to ecosite classification and other forest inventory variables depicted at the landscape scale. A series of black spruce increment cores were collected at breast height from trees in nine different ecosite groups within the boreal forest of northeastern Ontario, and processed using standard techniques for maceration and fibre length measurement. Regression tree analysis …

Theory And Simulations Of Macromolecular Soft Materials: Linking Molecular Design To Macroscale Morphology And Function, Arthi Jayaraman
Amgen Seminar Series in Chemical Engineering
We use theory and simulation techniques to connect molecular features of macromolecular materials, specifically polymers, to their morphology and macroscopic properties, thereby guiding synthesis of materials for various applications in the energy and biomedical fields.
In the first part of my talk I will present our recent theory and simulation studies of polymer functionalized nanoparticles in polymer nanocomposites. The goal of this work is to control spatial arrangement of nanoparticles in a polymer nanocomposite so as to engineer materials with target mechanical or optical properties. One can tailor the inter-particle interactions and precisely control the assembly of the particles in …

Dry eye is a common disorder caused by inadequate hydration of the ocular surface that results in disruption of barrier function. The homeostatic protein clusterin (CLU) is prominent at fluid-tissue interfaces throughout the body. CLU levels are reduced at the ocular surface in human inflammatory disorders that manifest as severe dry eye, as well as in a preclinical mouse model for desiccating stress that mimics dry eye. Using this mouse model, we show here that CLU prevents and ameliorates ocular surface barrier disruption by a remarkable sealing mechanism dependent on attainment of a critical all-or-none concentration. When the CLU level …

A spindle detection method was developed that: (1) extracts the signal of interest (i.e., spindle-related phasic changes in sigma) relative to ongoing "background" sigma activity using complex demodulation, (2) accounts for variations of spindle characteristics across the night, scalp derivations and between individuals, and (3) employs a minimum number of sometimes arbitrary, user-defined parameters. Complex demodulation was used to extract instantaneous power in the spindle band. To account for intra- and inter individual differences, the signal was z-score transformed using a 60 s sliding window, per channel, over the course of the recording. Spindle events were detected with a z-score …
