Synthesis Of Unnatural Amino Acids For Genetic Encoding By The Pyrrolysyl-Trna/Rna Synthetase System, William A. Knight Jan 2015

Synthesis Of Unnatural Amino Acids For Genetic Encoding By The Pyrrolysyl-Trna/Rna Synthetase System, William A. Knight

Theses and Dissertations

The complexity of all biomolecules in existence today can be attributed to the variation of the amino acid repertoire. In nature, 20 canonical amino acids are translated to form these biomolecules, however, many of these amino acids have revealed posttranslational modifications (i.e. acetylation, methylation) after incorporation. Amino acids that exhibit PTM are known for their involvement in cellular processes such as DNA repair and DNA replication; these PTMs are commonly found on histones within the chromatin complex. Utilization of in vivo site-specific incorporation has recently reported functionality of post-translationally modified amino acids.1 xii Here we report the synthesis and in …

Regulation Of Eps8 Dependent Pathways By Src In Head And Neck Squamous Cell Carcinoma, Dhwani Patel Jan 2015

Regulation Of Eps8 Dependent Pathways By Src In Head And Neck Squamous Cell Carcinoma, Dhwani Patel

Theses and Dissertations

Head and neck squamous cell carcinoma (HNSCC) is a type of cancer that begins in the epithelial cells that line the mucosal surfaces of the head and neck, including the oral cavity, pharynx, larynx, paranasal sinuses, nasal cavity, and salivary glands. Head and neck cancer is the sixth most common type of cancer with a 5-year survival rate of 60% for all cases. Over the past few years, a subset of cells with stem-like properties, called cancer stem cells, are believed to have tumor-initiation capabilities and are responsible for maintaining on-going tumor growth. Previous data from our lab suggested that …

Polycultural Interactions: Fuzzy Identity, Xi Lu Jan 2015

Polycultural Interactions: Fuzzy Identity, Xi Lu

Theses and Dissertations

“Robin D.G. Kelley coined the term polyculturalism as an alternative to multiculturalism, ‘since the latter often implies that cultures are fixed, discrete entities that exist side by side—a kind of zoological approach to culture.’ ”

Polyculturalism assumes the whole world’s cultures are interactive and fluid instead of independent and static, and individuals’ relationship to cultures are complex and cannot be categorized. Yet an individual constitutes multiple cultures, and individual identity embraces the various forms of culture in all aspects of one’s life.

My research examines how polyculturalism affects aspects of communications among people who hold a multiplicity of voices. It …

Applications Of Stability Analysis To Nonlinear Discrete Dynamical Systems Modeling Interactions, Jonathan L. Hughes Jan 2015

Applications Of Stability Analysis To Nonlinear Discrete Dynamical Systems Modeling Interactions, Jonathan L. Hughes

Theses and Dissertations

Many of the phenomena studied in the natural and social sciences are governed by processes which are discrete and nonlinear in nature, while the most highly developed and commonly used mathematical models are linear and continuous. There are significant differences between the discrete and the continuous, the nonlinear and the linear cases, and the development of mathematical models which exhibit the discrete, nonlinear properties occurring in nature and society is critical to future scientific progress. This thesis presents the basic theory of discrete dynamical systems and stability analysis and explores several applications of this theory to nonlinear systems which model …

Determinants Of Stress And Effects On Performance In Internal Medicine Residents, Sarah Braun Jan 2015

Determinants Of Stress And Effects On Performance In Internal Medicine Residents, Sarah Braun

Theses and Dissertations

The purpose of the present study was to assess: a) perceived stress, burnout, depression, and empathy at three time points in internal medicine residents, b) the role of gender and trait mindfulness in stress response during residency and c) to evaluate the impact these variables have on performance evaluations. Additionally, specific tasks of the residency that may contribute to the experience of stress and burnout were evaluated to test a model of job strain. Stress predicted subsequent burnout and depression. Burnout predicted subsequent depression, and stress mediated this relationship. Women reported higher mean levels of empathy and burnout than men. …

Prediction Of Protein-Protein Interactions In Escherichia Coli From Experimental Data In Treponema Pallidum, Marco A. Abreu Jan 2015

Prediction Of Protein-Protein Interactions In Escherichia Coli From Experimental Data In Treponema Pallidum, Marco A. Abreu

Theses and Dissertations

Protein – Protein interactions (PPIs) are thought to be conserved between species, although this has not been systematically investigated. This problem was explored in Escherichia coli from experimental data in Treponema pallidum by predicting PPIs, focusing on protein domains of little or unknown function. The comparison of T. pallidum to a model organism such as E. coli can not only reveal additional data about T. pallidum but also reveals how E. coli is similar to this distantly related, obligate parasite. A set of novel T. pallidum interactions, enriched for proteins of unknown function, were the basis of over 23,000 predicted …

Cute As A Button, Marta R. Finkelstein Jan 2015

Cute As A Button, Marta R. Finkelstein

Theses and Dissertations

Cute As A Button explores powerlessness, vulnerability, illness and addiction all wrapped up in tender buttons and a cute, cuddly creature. Using animation, sculpture, sound and an intimate space, I surround the viewer in a saccharine nightmare, one that references the dark underbelly of the cute and the sweet. The visual and aural elements are representative of the psychological and emotional states of powerlessness, which are overcome by the act of making and exploring a medium over which I can have complete control.

The Orphanage Of Things: A Narrative Of Abandonment, Malaz Elgemiabby Jan 2015

The Orphanage Of Things: A Narrative Of Abandonment, Malaz Elgemiabby

Theses and Dissertations

In Sudan, 110 babies are abandoned in the streets of Khartoum every month. The majority of abandoned children are born out of wedlock. Young women with illegitimate pregnancies are often ostracized by their families and society, and the lack of emotional, financial and legal support has led many to take desperate measures, including the abandonment of their children. Relinquishing mothers exist like ghosts in Sudanese society. The only evidence of the mother’s experience is her anonymous, abandoned child. In order to understand and examine this phenomenon, I used ethnographic performance art informed by design research practice (Performative Research Design). I …

Inhibition Of Mtor Signaling Protects Against Myocardial Reperfusion Injury, Acute Myocardial Infarction, Scott M. Filippone Jan 2015

Inhibition Of Mtor Signaling Protects Against Myocardial Reperfusion Injury, Acute Myocardial Infarction, Scott M. Filippone

Theses and Dissertations

Acute myocardial infarction (AMI) is the leading cause of death worldwide. Currently, the best method of treating cardiac ischemia is early reperfusion which, itself, induces myocardial damage. The mTOR complex is a key regulator of cardioprotection against cell stressors. We hypothesized that reperfusion therapy with Rapamycin, a potent mTOR inhibitor, would reduce infarct size in adult mouse hearts. Rapamycin was administered at the onset of reperfusion following 30 min in situ LAD ligation. After 24 hours of reperfusion, myocardial infarct size and apoptosis were significantly reduced in rapamycin-treated mice compared to control. Rapamycin inhibited pro-apoptotic protein Bax and phosphorylation of …

Cyclic Gmp: A Satiety Signal In C. Elegans, Ji S. Park Jan 2015

Cyclic Gmp: A Satiety Signal In C. Elegans, Ji S. Park

Theses and Dissertations

Appetite control and satiety mechanisms help animals maintain energy homeostasis; however, these mechanisms can be misregulated, leading to overweight and obesity. Caenorhabditis elegans is an excellent model system to study appetite and satiety because of its conserved behavioral aspects of satiety and conserved molecular mechanisms. ASI senses nutrition and its activity is required for the behavioral state of satiety quiescence. The purpose of this thesis project was to elucidate the function of cGMP signaling in ASI by looking at behavioral effects from the pharmacological use of sildenafil (Viagra), a PDE inhibitor, and the effects on ASI activation from mutating guanylyl …

A Continuous Process Towards The Synthesis Of Quinolones, Stevara N. Clinton Jan 2015

A Continuous Process Towards The Synthesis Of Quinolones, Stevara N. Clinton

Theses and Dissertations

Abstract

The development of quinolones is described from the first quinolone to the latest fluoroquinolones. Quinolones have generated considerable interest since their discovery because of their antibacterial capabilities. Analogs incorporating the 4-quinolone ring system comprise a largely expanding group of synthetic compounds. The development of antibacterial resistance has created the need for an efficient synthesis of quinolones that can be easily adapted toward the assembly of quinolone based antibacterial drugs.

There are several reported approaches to the 4-quinolone ring system. Many of these methods use expensive starting materials, require the removal of high boiling solvents, or use high temperature conditions …

Postcession, Evan D. Pomerantz Jan 2015

Postcession, Evan D. Pomerantz

Theses and Dissertations

This is a series of daily writings. Each day consists of a new topic and is closed at the end of the day. The ideas presented are philosophical, humorous, rambling, lamentations, incantations, doubt-ridden, aesthetic pep talks which combine into an affective representation of my studio practice’s becoming. There will be little congruency, some stories, and a lot of parallels because that is who I am.

Investigation Into The Specification Of Nurf Recruitment To The Genome, Marissa Mack Jan 2015

Investigation Into The Specification Of Nurf Recruitment To The Genome, Marissa Mack

Theses and Dissertations

The nucleosome remodeling factor (NURF) is a mutli-protein complex that plays a role in the regulation of gene expression through its ability to remodel nucleosomes. The largest subunit of this complex, Bptf (Bromodomain PHD Finger Transcription Factor) is important for many cellular processes as a transcriptional regulator and improper function results in disease or malignancy. To further understand the genome-wide recruitment of the NURF complex, the interaction partner for the N-terminal PHD finger domain of Bptf was investigated through pull down assays followed by mass spectrometry. It was determined that this domain does not recognize histones; instead it recognizes a …

The Object Inbetween, Lucy L. Derickson Jan 2015

The Object Inbetween, Lucy L. Derickson

Theses and Dissertations

My mission is to bring attention to the unusual, undefined, and fluid connections we have to objects through various avenues of experience. Things, objects, and devices have shaped our culture, environment, and personal philosophies. We live, grow, learn, and mourn with objects. We worship, sympathize, and relate to objects. But what I am most interested in is a human tendency to retreat to objects, allowing them to mediate experiences of friendship and intimacy between people.

During my graduate studies, I have been evaluating my personal and professional connections with others in order to understand how technology such as smart phones …

Development Of Novel Chimeric Epitope Based Diagnostic Antigens And Vaccines For Lyme Disease, Lee D. Oliver Jr. Jan 2015

Development Of Novel Chimeric Epitope Based Diagnostic Antigens And Vaccines For Lyme Disease, Lee D. Oliver Jr.

Theses and Dissertations

Lyme disease (LD) is the leading arthropod-borne disease in North America with 300-600,000 cases each year. There are currently no approved human LD vaccines. Outer surface protein C (OspC) has emerged as a leading vaccine candidate and an attractive diagnostic marker due to its antigenicity and expression early in infection. Several chimeric, epitope based OspC derived proteins were generated. The constructs were found to be highly immunogenic in mice and vaccination induced complement-dependent bactericidal antibodies. These results suggest that a broadly protective polyvalent OspC epitope based vaccine can be produced. Currently, LD diagnostic approaches are unreliable and unable to differentiate …

Polysaccharide-Based Shear Thinning Hydrogels For Three-Dimensional Cell Culture, Vasudha Surampudi Jan 2015

Polysaccharide-Based Shear Thinning Hydrogels For Three-Dimensional Cell Culture, Vasudha Surampudi

Theses and Dissertations

The recreation of the complicated tissue microenvironment is essential to reduce the gap between in vitro and in vivo research. Polysaccharide-based hydrogels form excellent scaffolds to allow for three-dimensional cell culture owing to the favorable properties such as capability to absorb large amount of water when immersed in biological fluids, ability to form “smart hydrogels” by being shear-thinning and thixotropic, and eliciting minimum immunological response from the host. In this study, the biodegradable shear-thinning polysaccharide, gellan-gum based hydrogel was investigated for the conditions and concentrations in which it can be applied for the adhesion, propagation and assembly of different mammalian …

Reconnect: Designing To Touch, Dana Seros Rohani Jan 2015

Reconnect: Designing To Touch, Dana Seros Rohani

Theses and Dissertations

Our everyday lives are surrounded by gadgets and digital devices that help us perform our daily chores with ease and efficiency. However, these digital devices can also separate us from what we should do ourselves. Although children who are exposed to high levels of technology might become attuned to the latest and the best gadgets, they might not learn to use their physical abilities. Another implication of a child’s over dependence on technology is that parental interactions such as encouragement, tutoring and reinforcement are provided by gadgets rather than living, breathing parents. Research done by Padma Ravichandran and Brandel France …

A Measure Of Social Behavior In Team-Based, Multiplayer Online Games: The Sociality In Multiplayer Online Games Scale (Smog), Chelsea M. Hughes

Theses and Dissertations

Video games have become a new platform for social interaction. I review the sociality of video games and the relationship between virtual- and real-world behaviors. I review and address the pros and cons of methods of measuring social behavior. Finally, I present two studies drawn from internet populations. In Study 1 (N = 250), I develop a scale, The Sociality in Multiplayer Online Games Scale (SMOG), which measures the frequency of social gaming behaviors in team-based, multiplayer online games. I hypothesized these to align on dominance and affiliation dimensions of social interaction (Kiesler, 1982). In Study 2 (N …

The field of entrepreneurship is rapidly advancing and matures as a discipline that receives substantial amount of attention. One popular area of research in the discipline of entrepreneurship is to investigate one’s intent to start a business, which is entrepreneurial intention. This is an important construct that warrants ongoing research because entrepreneurial intention is not only a great predictor of entrepreneurial behavior but also an important step in the process of becoming an entrepreneur. The present study, based on a sample of 321 subjects along with 264 observers, makes five contributions to the entrepreneurship literature. First, I examined the psychometric …

Fallen/Lifted, Hillary Waters Fayle Jan 2015

Fallen/Lifted, Hillary Waters Fayle

Theses and Dissertations

I use found botanical material such as leaves, seedpods, and branches to explore human connection to the physical world. By combining these organic objects with the rich traditions of needlecraft, I bind nature and the human touch. Both tender and ruthless, this intricate stitch work communicates the idea that our relationship with the natural world is both tenuously fragile and infinitely complex.

The way I think about and make art mirrors the way I think about my life and how I walk through the world. What I do is about elevating details. It is about noticing cycles and connections. It …

Structural Determinants Of Abuse-Related Neurochemical And Behavioral Effects Of Para-Substituted Methcathinone Analogs In Rats, Julie S. Bonano Jan 2015

Structural Determinants Of Abuse-Related Neurochemical And Behavioral Effects Of Para-Substituted Methcathinone Analogs In Rats, Julie S. Bonano

Theses and Dissertations

Methcathinone (MCAT) is the β-ketone analog of methamphetamine, and like its amphetamine analog, MCAT functions as a monoamine releaser that selectively promotes the release of dopamine (DA) and norepinephrine (NE) over serotonin (5-HT). MCAT produces amphetamine-like psychostimulant effects and is classified as a Schedule I drug of abuse by the United States Drug Enforcement Administration (DEA). Recently, synthetic MCAT analogs have emerged as designer drugs of abuse in Europe and the United States and have been marketed under deceptively benign names like “bath salts” in an attempt to evade legal restriction. These dangerous, recently emergent and novel drugs of abuse …

Colloidal Synthesis And Optical Characterizations Of Semiconductor Nanocrystals From Nontoxic Elements, Minh Q. Ho

Theses and Dissertations

To date, the search efforts have shifted from the toxic II-VI, III-V and IV-VI semiconductors to more environmentally friendly materials. Among Group II-V semiconductors, Zn3P2 has shown to be a more benign option, similar to Group IV (Ge, Si) materials, for future applications in photovoltaics and optoelectronics. This work is dedicated to the development of wet-chemical synthetic routes of (1) Zn3P2 and (2) Group IV (Ge, Si, Si1-xGex) nanocrystals with precise control over composition, crystal structure, size and dispersity by adjusting different reaction parameters such as temperature, time and solvent …

Cell Cycle Regulation Of Retinal Progenitors; A Role For The Nance-Horan Syndrome Protein In Retinogenesis, Paul J. Vorster Jan 2015

Cell Cycle Regulation Of Retinal Progenitors; A Role For The Nance-Horan Syndrome Protein In Retinogenesis, Paul J. Vorster

Theses and Dissertations

The Nance-Horan syndrome gene (NHS) plays a role in lens, eye and brain development. To date, the function of NHS remains unclear. Recent evidence showed that p53 isoform, Δ113p53, inhibits abnormal cell growth during organogenesis. We show that NHS is expressed in the retinas of Danio rerio and Xenopus tropicalis during key stages of retinogenesis, and that knockdown of the gene resulted in a small eye phenotype in both species. Initially, knockdown of nhsb in zebrafish had no visible defects at 24hpf. But examination of the retina at 48hpf, we see a marked difference in size compared to control embryos. …

A Comparison Of Obesity Interventions Using Energy Balance Models, Marcella Torres Jan 2015

A Comparison Of Obesity Interventions Using Energy Balance Models, Marcella Torres

Theses and Dissertations

An energy balance model of human metabolism developed by Hall et al. is extended to compare body composition outcomes among standard and proposed obesity interventions. Standard interventions include a drastic diet or a drastic diet with endurance training. Outcomes for these interventions are typically poor in clinical studies. Proposed interventions include a gradual diet and the addition of resistance training to preserve lean mass and metabolic rate. We see that resistance training, regardless of dietary strategy, achieves these goals. Finally, we observe that the optimal obesity intervention for continued maintenance of a healthy body composition following a diet includes a …

The Effects Of Nicotine Sequestration On The Dynamics Of Hyperparasitism In A Stage-Structured Model Of Manduca Sexta And Its Related Parasitoid Wasps, Mark P. Zimmerman Jan 2015

The Effects Of Nicotine Sequestration On The Dynamics Of Hyperparasitism In A Stage-Structured Model Of Manduca Sexta And Its Related Parasitoid Wasps, Mark P. Zimmerman

Theses and Dissertations

Two proposed models will be used to help answer a long observed question in the dynamics of \textit{Manduca sexta} and its related parasitoid wasps-Why is there a large difference in diversity in hyperparasitoid species between tobacco and other related plants such as tomato? Two stage structured differential equation models are presented. The first is a single patch model to study the changes in dynamics that occur between hosts, parasitoids, and hyperparasitoids as the amount of nicotine in the plant increases. The second is a two patch model that allows hyperparasitoids to choose between patches that are nicotine negative (i.e. tomato …

A Quinol Oxidase, Encoded By Cyoabcd, Is Utilized To Adapt To Lower O2 Concentrations In Rhizobium Etli Cfn42, Zachary Ryan Lunak, K. Dale Noel

Biological Sciences Faculty Research and Publications

Bacteria have branched aerobic respiratory chains that terminate at different terminal oxidases. These terminal oxidases have varying properties such as their affinity for oxygen, transcriptional regulation and proton pumping ability. The focus of this study was a quinol oxidase encoded by cyoABCD. Although this oxidase (Cyo) is widespread among bacteria, not much is known about its role in the cell, particularly in bacteria that contain both cytochrome c oxidases and quinol oxidases. Using Rhizobium etli CFN42 as a model organism, a cyo mutant was analysed for its ability to grow in batch cultures at high (21 % O2) and low …

Genetic Algorithm Optimization Of Point Charges In Force Field Development: Challenges And Insights, Maxim Vadimovich Ivanov, Marat R. Talipov, Qadir K. Timerghazin Jan 2015

Genetic Algorithm Optimization Of Point Charges In Force Field Development: Challenges And Insights, Maxim Vadimovich Ivanov, Marat R. Talipov, Qadir K. Timerghazin

Chemistry Faculty Research and Publications

Evolutionary methods, such as genetic algorithms (GAs), provide powerful tools for optimization of the force field parameters, especially in the case of simultaneous fitting of the force field terms against extensive reference data. However, GA fitting of the nonbonded interaction parameters that includes point charges has not been explored in the literature, likely due to numerous difficulties with even a simpler problem of the least-squares fitting of the atomic point charges against a reference molecular electrostatic potential (MEP), which often demonstrates an unusually high variation of the fitted charges on buried atoms. Here, we examine the performance of the GA …

Liana Competition With Tropical Trees Varies Seasonally But Not With Tree Species Identity, Leonor Álvarez-Cansino, Stefan A. Schnitzer, Joseph P. Reid, Jennifer S. Powers Jan 2015

Liana Competition With Tropical Trees Varies Seasonally But Not With Tree Species Identity, Leonor Álvarez-Cansino, Stefan A. Schnitzer, Joseph P. Reid, Jennifer S. Powers

Biological Sciences Faculty Research and Publications

Lianas in tropical forests compete intensely with trees for above‐ and belowground resources and limit tree growth and regeneration. Liana competition with adult canopy trees may be particularly strong, and, if lianas compete more intensely with some tree species than others, they may influence tree species composition. We performed the first systematic, large‐scale liana removal experiment to assess the competitive effects of lianas on multiple tropical tree species by measuring sap velocity and growth in a lowland tropical forest in Panama. Tree sap velocity increased 60% soon after liana removal compared to control trees, and tree diameter growth increased 25% …
