Nanocomposite materials are well characterized using a variety of advanced microscopy and spectroscopy techniques. In particular, the interfaces in the nanocomposites are often tailored (in engineered composites) and evolved (in biological nanocomposites) to unique molecular characteristics that provide optimized and superior properties to the nanocomposites. In this presentation, we will describe a multiscale mechanics perspective on role of molecular interactions as well as the nano and microstructures on the mechanics of nanocomposites. Specific examples including biological and synthetic nanocomposites will be presented. The biological material systems promise the possibility of developing innovative materials that simultaneously self-assembled, self-organized and self-regulated; characteristics that are difficult to achieve in purely synthetic systems. Proteins play an essential role in fabrication of biological materials due to their diverse functions ranging from structural to biochemical. The ability to mimic any of these functions can be a game changer in designing new biomaterials. There are several challenges in these strategies including replicating the hierarchical organization of biological materials, organization that provides multi-scale structure/property interdependence. Cancer cells are adept at invading, migrating to and colonizing on an organ away from its origin resulting in metastatic cancer. The process through which cancer cells colonize on a distant organ is known as metastasis. It is estimated that about 90% of the deaths associated with cancer are attributed to metastasis, yet the fundamental mechanisms of cancer metastasis are unknown. Several cancers are known to metastasize to bone. Of these, the most prolific are prostate, breast and colon cancers. Combining soft and hard building blocks is a widely used approach to fabricate composite materials exhibiting mechanical properties that are not achievable by their single components alone. Remarkably, the design strategies applied in engineering and biological processes lead to striking differences in the nano/microstructure and mechanical performance of such composite materials. The silkmoth chorion was studied extensively by F.C. Kafatos’ group for almost 40 years. However, the complete structure of the chorion locus was not obtained in the genome sequence of Bombyx mori published in 2008 due to repetitive sequences, resulting in gaps and an incomplete view of the locus. To obtain the complete sequence of the chorion locus, expressed sequence tags (ESTs) derived from follicular epithelium cells were used as probes to screen a bacterial artificial chromosome (BAC) library. Seven BACs were selected to construct a contig which covered the whole chorion locus. By Sanger sequencing, we successfully obtained complete …

Despite more than 40 years of intense study, essential features of the silkmoth chorion (eggshell) are still not fully understood. To determine the precise structure of the chorion locus, we performed extensive EST analysis, constructed a bacterial artificial chromosome (BAC) contig, and obtained a continuous genomic sequence of 871,711 base pairs. We annotated 127 chorion genes in two segments interrupted by a 164 kb region with 5 non-chorion genes, orthologs of which were on chorion bearing scaffolds in 4 ditrysian families. Detailed transcriptome analysis revealed expression throughout choriogenesis of most chorion genes originally categorized as “middle”, and evidence for diverse …

An instrumented NASA P-3B aircraft was used for airborne sampling of trace gases in a plume that had emanated from a small forest understory fire in Georgia, USA. The plume was sampled at its origin for deriving emission factors and followed ~ 13.6 km downwind for observing chemical changes during the first hour of atmospheric aging. The P-3B payload included a proton-transfer-reaction time-of-flight mass spectrometer (PTR-ToF-MS), which measured non-methane organic gases (NMOGs) at unprecidented spatio-temporal resolution (10m/0.1 s). The molecular mechanisms by which aging affects stem cell number and function are poorly understood. Murine data have implicated cellular senescence in the loss of muscle stem cells with aging. Here, using human cells and by carrying out experiments within a strictly pre-senescent division count, we demonstrate an impaired capacity for stem cell self-renewal in elderly muscle. We link aging to an increased methylation of the SPRY1 gene, a known regulator of muscle stem cell quiescence. Replenishment of the reserve cell pool was modulated experimentally by demethylation or siRNA knockdown of SPRY1. We propose that suppression of SPRY1 by age-associated …

Vertebrates exhibit substantial diversity in genome size, and some of the largest genomes exist in species that uniquely inform diverse areas of basic and biomedical research. For example, the salamander Ambystoma mexicanum (the Mexican axolotl) is a model organism for studies of regeneration, development and genome evolution, yet its genome is ~10× larger than the human genome. As part of a hierarchical approach toward improving genome resources for the species, we generated 600 Gb of shotgun sequence data and developed methods for sequencing individual laser-captured chromosomes. Based on these data, we estimate that the A. mexicanum genome is ~32 Gb. …

Cytomegalovirus (CMV) is a herpesvirus that persists for life and maintains extremely large numbers of T cells with select specificities in circulation. However, it is unknown how viral persistence impacts T cell populations in mucosal sites. We found that many murine (M)CMV-specific CD8s in mucosal tissues became resident memory T cells (TRM). These cells adopted an intraepithelial localization in the salivary gland that correlated with, but did not depend on, expression of the integrin CD103. MCMV-specific TRM cells formed early after infection, and spleen-localized cells had reduced capacities to become TRM at late times. Surprisingly, however, small numbers of new …

Wendy Harmon grew up with the mountains, rivers, lakes, and streams of western North Carolina at her back door. Some of her fondest memories are of outdoor exploratory excursions with her parents, Tommy and Darlene Harmon, where she literally first got her feet wet in ecology — the study of how organisms interact with their environments. Her mother was a Gardner-Webb graduate, but Harmon wanted to make her own decision about where to go to college.

Background: The evolution and development of sexual dimorphism illuminates a central question in biology: How do similar genomes produce different phenotypes? In an XX/XO system especially the state of a sexually dimorphic trait is determined by differences in gene expression, as there are no additional genetic loci in either sex. Here, we examine the XX/XO ostracod crustacean species Euphilomedes carcharodonta. This species exhibits radical sexual dimorphism of their lateral eyes, females have only a tiny simple lateral eye while males have elaborate ommatidial eyes. Quantifying the mechanical forces produced by fluid flows within the ocean is critical to understanding the ocean’s environmental phenomena. Such forces may have been instrumental in the origin of life by driving a primitive form of self-replication through fragmentation. Among the intense sources of hydrodynamic shear encountered in the ocean are breaking waves and the bursting bubbles produced by such waves. On a microscopic scale, one expects the surface-tension–driven flows produced during bubble rupture to exhibit particularly high velocity gradients due to the small size scales and masses involved. Silicosis is a lung disease characterized by pulmonary fibrosis that arises from chronic inhalation of silica dust in various occupational settings. Inhaled silica particles get deposited in the alveoli where macrophages engulf them leading to inflammation and macrophage cell death. The presence of apoptotic macrophages in the lungs is sufficient to cause inflammation and fibrosis whereas; inhibition of macrophage cell death prevents fibrosis. The goal of this study is therefore to identify the processes within an alveolar macrophage leading to cell death after uptake of silica particles. The present study examines middle school teachers’ beliefs about seven learning outcomes related to a project that involves developing and examining the effects of a set of engineering design modules constructed for use by middle school math and science teachers. Overall, the teachers involved in the intervention appear to believe they have the instructional skills, professional development, and resources to carry out the modules. Teachers from all of the schools (both intervention and comparison schools) for the most part valued the outcomes as important. This paper is based on the presentations and discussions that have been guided by the Chair and numerous stakeholders such as university researchers, Non Governmental Organizations, and federal, state, and local governments. SURA’s workshop series promotes collaboration and fully-integrated processes, and it should be noted that the actual workshop is only a first step; the follow-up work is likely to continue for years. In this article, we explain how mindfulness can enhance a democratic way of being, connecting practices of awareness, reflection, dialog, and action to democratic citizenship and social arrangements. We begin by sharing our understanding of democracy as a philosophy and a political system. We then provide a background for the concept of mindfulness as it is used by those in the field of education and health care and as we connect it to democracy and democratic education. Recent advancements in nonlinear dynamical systems theory have led to the development of a methodology, called geodesic eddy detection, for the objective (i.e., frame-independent) framing and tracking of coherent Lagrangian eddies in two-dimensional unsteady flows. Such eddies have material boundaries that do not stretch or fold over long periods of time, thereby providing an explicit mechanism of transport of fluid and distinguished properties, such as temperature and salinity. Eddies identified from Eulerian criteria (e.g., as regions enclosed by streamlines where rotation dominates over strain instantaneously) do not possess this property as such criteria either depend on the choice of reference …

Since 2002, the Vision Care Technician program has continually worked to recruit bright young women from rural villages to engage in comprehensive training in ophthalmic nursing. Two Global Social Benefit Fellows collected both qualitative and quantitative data by interviewing a large cohort of student and staff Vision Care Technicians in different phases of the program. In order to document the social impact of two cohorts, this comparative analysis intends to further understand how the Vision Care Technician program has impacted women’s lives.
