This Ph.D. dissertation presents a process-oriented study with two primary objectives: 1) to obtain a physical understanding of the impacts of local and remote forcing, including basin-scale natural climate variability, on the interannual variability of along-shelf transport and water properties of the Middle Atlantic Bight (Chapter 1); 2) to explore the impacts of physical processes on the seasonal variation of phytoplankton biomass in Rhode Island coastal waters (Chapter 2). To achieve the two objectives, we apply a numerical modeling approach employed by the Regional Ocean Modeling System (ROMS) together with substantive analyses of both in-situ and remote observations.

The synthesis of carbon-nitrogen (C-N) bonds is an area of research that has garnered significant interest due to the ubiquity of C-N bonds in pharmaceuticals and natural products. Various methods exist for forming C-N bonds, most recognizably Buchwald-Hartwig amination, but these reactions typically require functionalization of either the C-H or N-H bond resulting in the formation of harmful byproducts. As a result, the need to explore alternative means for synthesizing C-N bonds merits consideration. Educators are asked by employers and policy makers to provide instruction which would develop student skills in both the “hard” (academic and technical) and “soft” (personality traits and habits) skills required to be workplace-ready. Solid tumors have a microenvironment that is inherently acidic and hypoxic. Hypoxia is caused by leaky blood vessels and large diffusion distances from cells to them. It is heterogeneous throughout the tumor and while all solid tumors are hypoxic to a degree, it is difficult to predict invasiveness based on it. However, acidity is a near ubiquitous characteristic of tumors with more aggressive tumors producing greater acidity. Endocytic trafficking of platelet surface receptors plays a role in the accumulation of granule cargo (i.e. fibrinogen and VEGF) and thus could contribute to hemostasis, angiogenesis, or inflammation. However, the mechanisms of platelet endocytosis are poorly understood. The small GTP-binding protein, ADP-ribosylation factor 6 (Arf6), regulates integrin trafficking in nucleated cells; therefore, we posited that Arf6 functions similarly in platelets. To address this, we generated platelet-specific, Arf6 knockout mice. Arf6-/- platelets had a storage defect for fibrinogen but not other cargo, implying Arf6’s role in integrin αIIbβ3 trafficking. Additionally, platelets from Arf6-/- mice injected with biotinylated-fibrinogen, showed …

Taxane based chemotherapy is an effective treatment for castration-resistant prostate cancer (CRPC) via stabilization of microtubules. Progression to castration-resistant prostate cancer is characterized by increased androgen receptor (AR), elevated intra-prostatic androgens and activated AR signaling despite castrate levels of androgens.
Previous studies identified that the inhibitory effect of microtubule targeting chemotherapy on AR activity was conferred by interfering with AR intracellular trafficking. The N-terminal domain (NTD) of AR was identified as a tubulin interacting domain that can be effectively targeted by the novel small molecular inhibitor, EPI. Although a widely used practice, it was previously unknown whether disciplinary segregation is actually effective at modifying prison behavior. This quantitative, retrospective observational study tested deterrence theory and explored the effectiveness of disciplinary segregation in deterring subsequent prison inmate misconduct among those subjected to it (N = 228). It compared a cohort of male inmates incarcerated by the Oregon Department of Corrections who had spent time in disciplinary segregation in 2011 and/or 2012 with a comparison cohort who had not spent any time in disciplinary segregation. The goal of this study was to identify critical regions of a novel gene, Nance-Horan syndrome-like 1b (nhsl1b). It was previously discovered that C-terminal truncation of the Nhsl1b protein in nhsl1b mutants resulted in a loss of migration in the facial motor neurons of the hindbrain (Walsh et al. 2011). As nhsl1b expresses many isoforms, multiple targets were investigated in order to determine which transcript bears the largest impact on the motor neurons. This study demonstrates original findings of statin effects on IL-33 stimulated mast cells. Statins are a class of drugs used to lower cholesterol production by targeting HMG CoA reductase. These commonly prescribed drugs have been shown to be immunomodulatory. In this study, we have found that pretreatment with statins has a variety of effects on IL-33 stimulated mast cells. Atorvastatin suppresses TNF and IL-6 production, while fluvastatin significantly enhances release of these proinflammatory cytokines in BMMCs. Although they have differing effects on cytokine production, both statins lowered ST2 expression on the cell surface, decreased cell viability, and enhanced expression of …

Algal blooms are directly related to human-caused nutrient enrichment of water bodies. The cyanobacteria Microcystis aeruginosa produces microcystin (MC), a toxin that has been linked with mortalities and illness of many organisms. We show that MC is not constrained by the aquatic-terrestrial ecotone. MC was detected in a primary consumer and emerging aquatic invertebrate (Hexagenia Mayfly), a terrestrial insect and predator of emerging aquatic invertebrates (Tetragnathidae Spider), and a vertebrate consumer (Prothonotary Warbler). Mayfly and spider MC levels varied across the blooming period. MC levels in prothonotary warbler livers varied by age class; nestlings having the highest levels. MC levels …

Genetic engineering of microbes has developed rapidly along with our ability to synthesize DNA de novo. Yet, even with decreasing DNA synthesis costs there remains a need for inexpensive, rapid and reliable methods for assembling synthetic DNA into larger constructs or combinatorial libraries. While technological advances have resulted in powerful techniques for in vitro and in vivo assembly of DNA, each suffers inherent disadvantages. Analysis of variance (ANOVA) is a robust test against the normality assumption, but it may be inappropriate when the assumption of homogeneity of variance has been violated. Welch ANOVA and the Kruskal-Wallis test (a non-parametric method) can be applicable for this case. In this study we compare the three methods in empirical type I error rate and power, when heterogeneity of variance occurs and find out which method is the most suitable with which cases including balanced/unbalanced, small/large sample size, and/or with normal/non-normal distributions.

Smith-Magenis Syndrome (SMS; OMIM #182290) is a multiple congenital abnormality and intellectual disability (ID) disorder caused by either an interstitial deletion of the 17p11.2 region containing the retinoic acid induced-1 (RAI1) gene or a mutation of the RAI1 gene. Individuals diagnosed with SMS typically present characteristics such as ID, self-injurious behavior, sleep disturbance, ocular and otolaryngological abnormalities, craniofacial and skeletal abnormalities, neurological and behavioral abnormalities, as well as other systemic defects and manifestations. Organizational leaders are unaware of the gender-specific leadership skillsets women possess to increase organizational effectiveness and how to address potential barriers for assuring these skillsets are recognized as effective. Of the estimated 69,000 police officers serving in Canada, approximately 14,000 are women. Of those 14,000, only 10% hold a senior rank, and less than 3% hold the position of Chief of Police. Though a host of analgesics have been developed to alleviate pain, especially acute pain, significant side effects and a lack of long-term efficacy have encouraged research attempts to pursue novel targets that may be associated with fewer side effects or a more sustained efficacy. The tumor suppressor TP53 is the most frequently altered gene in human cancers. The growth-promoting complex, mTORC1 plays a part of the oncogenic profile caused by dysfunctional p53. mTORC1 sits downstream of AMPK and other crucial tumor suppressors/oncogenes, PTEN, LKB1, and Akt. The antifolate pemetrexed was found by this laboratory to activate AMPK via the inhibition of the enzyme AICART in de novo purine synthesis. This work presents a mechanism of mTORC1 activation with p53 loss, as well as of mTORC1 inhibition by pemetrexed-induced AMPK. The initiation or progression of periodontitis might involve a local renin-angiotensin system (RAS) in periodontal tissue. The aim of this study was to further characterize the local RAS in human and rat periodontal tissues between healthy and periodontally-affected tissue. Components of the RAS were investigated using in vitro, ex vivo and in vivo experiments involving both human and Wistar rat periodontium. Although not upregulated when challenged with P. gingivalis-lipopolysaccharide, human gingival and periodontal ligament fibroblasts expressed RAS components. Obesity is a global epidemic, exacting an enormous human and economic toll. In the absence of a comprehensive global governance strategy, states have increasingly employed a wide array of legal strategies targeting the drivers of obesity. This Article identifies recent global trends in obesity-related legislation and makes the normative case for an updated global governance strategy. National governments have responded to the epidemic both by strengthening traditional interventions and by developing novel legislative strategies. Vulvodynia is a chronic, heterogeneous, and multifactorial disease. This condition may affect up to 18 percent of the female population including Caucasians, African Americans, Africans and Hispanics particularly those sexually active at child bearing age. The etiology of this condition is complex and multifactorial and it is frequently accompanied by physical disabilities, psychological distress and sexual dysfunction. Clinically, vulvodynia can be generalized or localized and pain can be provoked or unprovoked. Patients may also describe vulvar paresthesias or dysesthesias that may last hours. Gold nanorods (GNRs) due to their tunable optical properties within the near infrared (NIR) region have been identified as strong candidates for new nano-based biomedical applications. Unfortunately, many have reported GNR cytotoxicity and it is still unclear how GNR aspect ratio (AR), surface charge and surface chemistry contribute to cellular association and cytotoxicity. The tuberous sclerosis complex 1 and 2 (TSC1 and TSC2) genes and their products hamartin and tuberin, respectively, are involved in several regulatory cellular mechanisms, including the mTOR pathway that is important for proliferation, cell migration, and protein synthesis. In addition, it has been suggested that this pathway plays a role in regulating Na+-K+-2Cl- and K+-Cl- cotransporters (NKCC and KCC, respectively) [41, 42, 43]. Therefore, the TSC1 and TSC2 genes are expected to play a significant role in regulating cell volume, and K+ and Cl- homeostasis.
