Heterozygosity For A Hypomorphic Polβ Mutation Reduces The Expansion Frequency In A Mouse Model Of The Fragile X-Related Disorders, Rachel Adihe Lokanga, Ali Senejani, Joann B. Sweasy, Karen Usdin

Biology and Environmental Science Faculty Publications

The Fragile X-related disorders (FXDs) are members of the Repeat Expansion Diseases, a group of human genetic conditions resulting from expansion of a specific tandem repeat. The FXDs result from expansion of a CGG/CCG repeat tract in the 5’ UTR of the FMR1 gene. While expansion in a FXD mouse model is known to require some mismatch repair (MMR) proteins, our previous work and work in mouse models of another Repeat Expansion Disease show that early events in the base excision repair (BER) pathway play a role in the expansion process. One model for repeat expansion proposes that a non-canonical …

Effect Of Bay (Laurus Nobilis) Essential Oil On Physicochemical Properties Of Alaska Pollock (Theragra Chalgoramma) Surimi Nutrified With Salmon And Flaxseed Oils Under Refrigerated Storage, Ugochukwu C. Anyanwu

Theses

Surimi seafood products are widely accepted and consumed in the U.S. and other parts of the world. In recent time, there has been increased demand for surimi seafood products nutrified with ω-3 polyunsaturated fatty acids (PUFAs) as a result of their reported health benefits, but this may have a negative effect on the storage time of the product due to lipid oxidation of fatty acids (FA). Bay (Laurus nobilis) essential oil (BEO) which has demonstrated antioxidant properties was incorporated to the surimi seafood to reduce lipid oxidation thereby extending storage time. The objectives of this study were to (1) evaluate …

Mechanisms Of Pluripotency And Epigenetic Reprogramming In Primordial Germ Cells: Lessons For The Conversion Of Other Cell Types Into The Stem Cell Lineage, Suresh Palamadai Krishnan

Turkish Journal of Biology

Primordial germ cells (PGCs) provide an excellent tool to better understand ancestor?descendent relationships as well as the efficiency and molecular mechanisms governing pluripotency in the reprogramming of somatic cells, since the latter type of cells have a relatively lower efficiency of conversion to pluripotent cells. This kind of comparison has gained credence from the commonalities regarding the expression of key transcription factors such as octamer-binding transcriptionhttp://en.wikipedia.org/wiki/Transcription_factor factor-4 (Oct3/4), SRY-related HMG box (Sox2), myelocytomatosis (c-Myc), and Nanog, as well as redundancy in terms of Kruppel-like factor 2 (Klf2), Kruppel-like factor 5 (Klf5), estrogen-related receptor beta (Esrrb), and estrogen-related receptor gamma (Esrrg) …

The mu-opioid receptor 1 (OPRM1) binds endogenous opioids. Increasing evidence suggests that endogenous OPRM1 agonists released at the time of trauma may contribute to the development of posttraumatic musculoskeletal pain (MSP). In this prospective observational study, we evaluated the hypothesis that individuals with an AG or GG genotype at the OPRM1 A118 G allele, which results in a reduced response to opioids, would have less severe MSP 6 weeks after motor vehicle collision (MVC). Based on previous evidence, we hypothesized that this effect would be sex-dependent and most pronounced among women with substantial peritraumatic distress. European American men and women …

Journal Articles

Background/Aims: Current staging guidelines for small intestinal neuroendocrine tumors (SI-NETs) differentiate between the presence (N1) and absence (N0) of lymph node (LN) metastases. However, the prognostic significance of the extent of LN involvement remains unknown. In this study, we used data from a population-based cancer registry to examine whether involvement of a higher number of LNs is associated with worse survival. Methods: We used the Surveillance, Epidemiology, and End Results (SEER) database to identify patients with histologically confirmed, surgically resected SI-NETS diagnosed between 1988 and 2010. Patients were classified into three groups by the LN ratio (number of positive LNs/number …

Hypothesis: Neuroendocrine Mechanisms (Hypothalamus-Growth Hormone-Stat5 Axis) Contribute To Sex Bias In Pulmonary Hypertension, P. B. Sehgal, Y. M. Yang, E. J. Miller Jan 2015

Hypothesis: Neuroendocrine Mechanisms (Hypothalamus-Growth Hormone-Stat5 Axis) Contribute To Sex Bias In Pulmonary Hypertension, P. B. Sehgal, Y. M. Yang, E. J. Miller

Journal Articles

Pulmonary hypertension (PH) is a disease with high morbidity and mortality. The prevalence of idiopathic pulmonary arterial hypertension (IPAH) and hereditary pulmonary arterial hypertension (HPAH) is approximately 2-4-fold higher in women than in men. Paradoxically, there is an opposite male bias in typical rodent models of PH (chronic hypoxia or monocrotaline); in these models administration of estrogenic compounds [e.g. estradiol-17beta (E2)] is protective. Further complexities are observed in humans ingesting anorexigens (female bias), and in rodent models such as after hypoxia plus SU5416/Sugen (little sex bias) or involving serotonin transporter overexpression or dexfenfluramine administration (female bias). These complexities in sex …

Acute brain diseases such as acute strokes and transit ischemic attacks are the leading causes of mortality and morbidity worldwide, responsible for 9% of total death every year. "Time is brain" is a widely accepted concept in acute cerebrovascular disease treatment. Efficient and accurate computational framework for hemodynamic parameters estimation can save critical time for thrombolytic therapy. Meanwhile the high level of accumulated radiation dosage due to continuous image acquisition in CT perfusion (CTP) raised concerns on patient safety and public health. However, low-radiation leads to increased noise and artifacts which require more sophisticated and time-consuming algorithms for robust estimation. …

BACKGROUND: Relatively poor penetration and retention in tumor tissue has been documented for large molecule drugs including therapeutic antibodies and recombinant immunoglobulin constant region (Fc)-fusion proteins due to their large size, positive charge, and strong target binding affinity. Therefore, when designing a large molecular drug candidate, smaller size, neutral charge, and optimal affinity should be considered. METHODS: We engineered a recombinant protein by molecular engineering the second domain of VEGFR1 and a few flanking residues fused with the Fc fragment of human IgG1, which we named HB-002.1. This recombinant protein was extensively characterized both in vitro and in vivo for …

ATP-binding cassette subfamily G member 2 (ABCG2) is a member of the ABC transporter superfamily proteins, which has been implicated in the development of multidrug resistance (MDR) in cancer, apart from its physiological role to remove toxic substances out of the cells. The diverse range of substrates of ABCG2 includes many antineoplastic agents such as topotecan, doxorubicin and mitoxantrone. ABCG2 expression has been reported to be significantly increased in some solid tumors and hematologic malignancies, correlated to poor clinical outcomes. In addition, ABCG2 expression is a distinguishing feature of cancer stem cells, whereby this membrane transporter facilitates resistance to the …
