Gastric cancer (GC) is a major cause of global cancer mortality. Genetic variations in DNA repair genes can modulate DNA repair capability and, consequently, have been associated with risk of developing cancer. We have previously identified a T to C point mutation at nucleotide 889 (T889C) in DNA polymerase beta (POLB) gene, a key enzyme involved in base excision repair in primary GCs. The purpose of this study was to evaluate the mutation and expression of POLB in a larger cohort and to identify possible prognostic roles of the POLB alterations in GC. Primary GC specimens and their matched normal …

Pleuropulmonary blastoma (PPB) is the most frequent pediatric lung tumor and often the first indication of a pleiotropic cancer predisposition, DICER1 syndrome, comprising a range of other individually rare, benign and malignant tumors of childhood and early adulthood. The genetics of DICER1-associated tumorigenesis are unusual in that tumors typically bear neomorphic missense mutations at one of five specific “hotspot” codons within the RNase IIIb domain of DICER 1, combined with complete loss of function (LOF) in the other allele. We analyzed a cohort of 124 PPB children for predisposing DICER1 mutations and sought correlations with clinical phenotypes. Over …

Beyond Cd19: Opportunities For Future Development Of Targeted Immunotherapy In Pediatric Relapsed-Refractory Acute Leukemia., Haneen Shalabi, Anne Angiolillo, Terry J Fry

Pediatrics Faculty Publications

Chimeric antigen receptor (CAR) T cell therapy has been used as a targeted approach in cancer therapy. Relapsed and refractory acute leukemia in pediatrics has been difficult to treat with conventional therapy due to dose-limiting toxicities. With the recent success of CD 19 CAR in pediatric patients with B cell acute lymphoblastic leukemia (ALL), this mode of therapy has become a very attractive option for these patients with high-risk disease. In this review, we will discuss current treatment paradigms of pediatric acute leukemia and potential therapeutic targets for additional high-risk populations, including T cell ALL, AML, and infant ALL.

CD24 is overexpressed in nearly 70% human cancers, whereas TP53 is the most frequently mutated tumour-suppressor gene that functions in a context-dependent manner. Here we show that both targeted mutation and short hairpin RNA (shRNA) silencing of CD24 retard the growth, progression and metastasis of prostate cancer. CD24 competitively inhibits ARF binding to NPM, resulting in decreased ARF, increase MDM2 and decrease levels of p53 and the p53 target p21/CDKN1A. CD24 silencing prevents functional inactivation of p53 by both somatic mutation and viral oncogenes, including the SV40 large T antigen and human papilloma virus 16 E6-antigen. In support of the …

We have previously mapped the interval on Chromosome 4 for a major polycystic kidney disease modifier (Mpkd) of the B6(Cg)-Cys1cpk/J mouse model of recessive polycystic kidney disease (PKD). Informatic analyses predicted that this interval contains at least three individual renal cystic disease severity-modulating loci (Mpkd1-3). In the current study, we provide further validation of these predicted effects using a congenic mouse line carrying the entire CAST/EiJ (CAST)-derived Mpkd1-3 interval on the C57BL/6J background. We have also generated a derivative congenic line with a refined CAST-derived Mpkd1-2 interval and demonstrated its dominantly-acting disease-modulating effects (e.g., 4.2-fold increase in total cyst area; …

Effective protection against pathogens requires the host to produce a wide range of immune effector proteins. The Sp185/333 gene family, which is expressed by the California purple sea urchin Strongylocentrotus purpuratus in response to bacterial infection, encodes a highly diverse repertoire of anti-pathogen proteins. A subset of these proteins can be isolated by affinity to metal ions based on multiple histidines, resulting in one to four bands of unique molecular weight on standard Western blots, which vary depending on the individual sea urchin. Two dimensional gel electrophoresis (2DE) of nickel-isolated protein samples followed by Western blot was employed to detect …

Wilms tumour is an embryonal tumour of childhood that closely resembles the developing kidney. Genomic changes responsible for the development of the majority of Wilms tumours remain largely unknown. Here we identify recurrent mutations within Wilms tumours that involve the highly conserved YEATS domain of MLLT1 (ENL), a gene known to be involved in transcriptional elongation during early development. The mutant MLLT1 protein shows altered binding to acetylated histone tails. Moreover, MLLT1-mutant tumours show an increase in MYC gene expression and HOX dysregulation. Patients with MLLT1-mutant tumours present at a younger age and have a high prevalence of precursor intralobar …

Long-Term Effects Of An Invasive Shore Crab On Cape Cod, Massachusetts, Christopher P. Bloch, Kevin D. Curry, John C. Jahoda

Biological Sciences Faculty Publications

Invasive species can cause dramatic changes in the structure of intertidal communities. In some systems, however, abundance or impacts of invaders may peak 10–20 years after invasion and decline thereafter. Hemigrapsus sanguineus (Asian Shore Crab) has been established at Sandwich, MA, on the north side of Cape Cod, since the mid-1990s. This study documented population dynamics of the Asian Shore Crab and 3 species of prey or competitors (Carcinus maenas [Green Crab], Mytilus edulis [Blue Mussel], and Littorina littorea [Common Periwinkle]) over 10 years. An additional goal of the study was to determine whether population growth of the Asian …

Sri Lanka has a rich assemblage of gekkonid fauna. Among Sri Lankan geckos, rare species such as Hemiphyllodactylus typus and Lepidodactylus lugubris are poorly studied; both are considered vulnerable in national conservation assessments. Detailed ecological studies are needed for robust conservation assessments of these species, especially with the focus on island-wide distribution and microhabitat requirements. This study was conducted via patch sampling to record relative abundance and distribution of H. typus and L. lugubris based on random walks to 82 locations representing the three major bioclimatic zones of Sri Lanka. Morphological characteristics, behavior, and habitat use of the focal species …

College of Natural and Applied Sciences

Management of silver carp Hypophthalmichthys molitrix has become a growing concern for multiple state and federal entities. Commercial fishing may have the greatest potential to control silver carp. However, for a management action to be successful, the level of exploitation required to reduce silver carp populations must be quantified. Therefore, silver carp were collected from Midwestern U.S. rivers (i.e., Upper, Middle, and Lower Mississippi, Missouri, Illinois, Ohio, and Wabash rivers) to obtain population dynamics (i.e., recruitment, growth and mortality). Parameters obtained from population demographics were used to simulate exploitation levels using a spawning potential ratio (SPR) approach to determine target …

Fluorescent Image Analysis Of Hiv-1 And Hiv-2 Uncoating Kinetics In The Presence Of Old World Monkey Trim5Α, Eric Takeda, Ken Kono, Amy E. Hulme, Thomas J. Hope, Emi E. Nakayama, Talsuo Shioda Jan 2015

Fluorescent Image Analysis Of Hiv-1 And Hiv-2 Uncoating Kinetics In The Presence Of Old World Monkey Trim5Α, Eric Takeda, Ken Kono, Amy E. Hulme, Thomas J. Hope, Emi E. Nakayama, Talsuo Shioda

College of Health and Human Services

Uncoating of Human Immunodeficiency Virus type 1 (HIV-1) and type 2 (HIV-2) conical cores is an important early step for establishment of infection. In Old World Monkey (OWM) cells, the TRIM5α cellular factor potently suppresses an early step of infection by HIV-1. Previously, biochemical studies using whole cell lysates of infected cells revealed that OWM TRIM5α accelerates the uncoating of HIV-1, leading to premature reverse transcription. In the present study, we re-evaluated uncoating kinetics of HIV-1 in the presence of OWM TRIM5α by using an in situ uncoating assay, which allowed us to differentiate productive HIV-1 entry from simple (non-productive) …

Cardiovascular Patterning As Determined By Hemodynamic Forces And Blood Vessel Genetics, Gregory A. Anderson, Ryan S. Udan, Mary E. Dickinson, R. Mark Henkelman

College of Natural and Applied Sciences

Background: Vascular patterning depends on coordinated timing of arteriovenous specification of endothelial cells and the concomitant hemodynamic forces supplied by the onset of cardiac function. Using a combination of 3D imaging by OPT and embryo registration techniques, we sought to identify structural differences between three different mouse models of cardiovascular perturbation. Results: Endoglin mutant mice shared a high degree of similarity to Mlc2a mutant mice, which have been shown to have a primary developmental heart defect causing secondary vessel remodeling failures. Dll4 mutant mice, which have well-characterized arterial blood vessel specification defects, showed distinct differences in vascular patterning when compared …

Cellulose synthases (CESAs) synthesize the β-1,4-glucan chains that coalesce to form cellulose microfibrils in plant cell walls. In addition to a large cytosolic (catalytic) domain, CESAs have eight predicted transmembrane helices (TMHs). However, analogous to the structure of BcsA, a bacterial cellulose synthase, predicted TMH5 in CESA may instead be an interfacial helix. This would place the conserved FxVTxK motif in the plant cell cytosol where it could function as a substrate-gating loop as occurs in BcsA. To define the functional importance of the CESA region containing FxVTxK, we tested five parallel mutations in Arabidopsis thaliana CESA1 and Physcomitrella patens …

Oceanic and coastal waters are acidifying due to processes dominated in the open ocean by increasing atmospheric CO2 and dominated in estuaries and some coastal waters by nutrient-fueled respiration. The patterns and severity of acidification, as well as its effects, are modified by the host of stressors related to human activities that also influence these habitats. Temperature, deoxygenation, and changes in food webs are particularly important co-stressors because they are pervasive, and both their causes and effects are often mechanistically linked to acidification. Development of a theoretical underpinning to multiple stressor research that considers physiological, ecological, and evolutionary perspectives …

Endocardial and myocardial progenitors originate in distinct regions of the anterior lateral plate mesoderm and migrate to the midline where they coalesce to form the cardiac tube. Endocardial progenitors acquire a molecular identity distinct from other vascular endothelial cells and initiate expression of specific genes such as nfatc1. Yet the molecular pathways and tissue interactions involved in establishing endocardial identity are poorly understood. The endocardium develops in tight association with cardiomyocytes. To test for a potential role of the myocardium in endocardial morphogenesis, we used two different zebrafish models deficient in cardiomyocytes: the hand2 mutant and a myocardial-specific genetic …

The Use Of Biomarkers In Outdoor Education Research: Promises, Challenges, And The Development Of Evidence, Alan Ewert

Research in Outdoor Education

While widely used in research connected with outdoor education, self-report data can be subject to a number of issues related to validity and generalizability. This paper argues that biomarkers present another type of evidence that is equally or even more rigorous than self-report data. The paper describes several types of biomarkers that are commonly measured in other disciplines along with how data from those biomarkers are collected. Data from a recent study using two biomarkers commonly used for measuring stress is presented as an example of how the use of biomarkers can broaden the body of evidence being developed in …

The enhanced greenhouse effect is currently considered to be our most important global environmental problem. While the magnitude of radiation absorbed by greenhouse gases is known to a high certainty, the absorption of radiation by atmospheric aerosol particles is not. In the initial Visiting Faculty Program application, we proposed the use of an ultraviolet-visible (UV/Vis) spectrometer equipped with a liquid waveguide capillary flow cell to determine the extent to which secondary organic aerosol particles (SOA) absorb visible light. Early in the research period, the UV/Vis technique was optimized for three solvent systems (methanol, water and 0.1 M hydrochloric acid). Using …

Etiology Beliefs Moderate The Influence Of Emotional Self-Control On Willingness To See A Counselor Through Help-Seeking Attitudes Among Asian American Students, Paul Youngbin Kim, Dana L. Kendall

SPU Works

To identify correlates of Asian American professional help-seeking, we tested a mediation model describing Asian American help-seeking (Asian value of emotional self-control → help-seeking attitudes → willingness to see a counselor; Hypothesis 1) in a sample of Asian American college students from the Pacific Northwest region of the United States (N = 232). We also examined biological and spiritual etiology beliefs as moderators of the mediation model (Hypotheses 2a & 2b). Our findings indicated that help-seeking attitudes significantly mediated the relation between emotional self-control and willingness to see a counselor, consistent with our mediation hypothesis. Furthermore, biological and spiritual …
