Classification Of Breast Cancer Patients Using Somatic Mutation Profiles And Machine Learning Approaches, Suleyman Vural

The high degree of heterogeneity observed in breast cancers makes it very difficult to classify cancer patients into distinct clinical subgroups and consequently limits the ability to devise effective therapeutic strategies. In this study, we explore the use of gene mutation profiles to classify, characterize and predict the subgroups of breast cancers. We analyzed the whole exome sequencing data from 358 ethnically similar breast cancer patients in The Cancer Genome Atlas (TCGA) project. Identified somatic and non-synonymous single nucleotide variants were assigned a quantitative score (C-score) that represents the extent of negative impact on the function of the gene. Using …

The Patient Protection and Affordable Care Act of 2010 ushered in a new era of fiscal accountability for healthcare organizations. Healthcare organizations and providers are now jointly held responsible for the improved quality of patient care and sustained reductions in patient care events termed healthcare-acquired conditions. To ensure compliance with this newly enacted legislation, the Centers for Medicare and Medicaid Services (CMS) began penalizing hospitals for targeted conditions leading to 30-day readmissions beginning in October 2012. The demands of WWII mobilization led to the creation of the first standing women's army in the US known as the Women's Army Corps (WAC). An unintended consequence of this was that the WAC provided queer women with an environment with which to explore their gender and sexuality while also giving them the cover of respectability and service that protected them from harsh societal repercussions. They could eschew family for their military careers. They could wear masculine clothing, exhibit a masculine demeanor, and engage in a homosocial environment without being seen as subversive to the American way of life. Quite …

Research on electrochemical disinfection of municipal wastewater has been conducted at the University of New Orleans using a continuous flow electrochemical reactor connected to a direct current (DC) power supply changing its polarity and varying the electrode distance. Bacterial inactivation and chlorine production were the main parameters that were recorded.

After months of research, it was determined that the electrochemical disinfection reactor is efficient and has a great potential for the future. There is no need to use chlorine and it has low operation costs.

Since the 1990s, academic physicians have been subjected to increased requirements for disclosure in their roles as educators and researchers and for conflict of interest (COI) resolution in their financial relationships with pharmaceutical, medical device, and biotech companies, collectively referred to as industry. The requirements are the result of the convergence of federal regulations, accreditation guidelines, professional and industry codes of ethics and conduct, and institutional policies. The disclosure and COI resolution requirements are managed and resolved by a review of forms and compliance with relevant guidance documents and policies. Where and how to house the urban poor remains a controversial issue. Public housing residents are particularly vulnerable. Issues of race, class and gender intersect in their lives. Public-private partnerships in urban redevelopment projects and a focus on issues that arise from concentrated poverty gave rise to HOPE VI policy aimed at deconcentrating poverty via public housing demolition and redevelopment. Second Lieutenant Charles Scheffel, B Company Platoon Leader, 39th Infantry Regiment, 9th Infantry Division modified existing methods of venereal disease control in Algeria, North Africa during Operation Torch after being ordered to reduce the venereal disease rate by his regimental commander, Colonel William Ritter. Tasked with defeating the Germans first, Scheffel learned other enemies lurked as well, and he instituted an illegal policy to solve the problem as fast and as effectively as possible. Official United States policy on the eve of World War Two prohibited the establishment and operation of a brothel. Scheffel operated this brothel as …

This thesis is concerned with the question of how America’s citizen soldiers are remembered and how their services can be interpreted through monuments and memorials. The paper discusses the concept of memory and the functions of memorialization. It explores whether and how monuments and memorials portray the difficulties, hardships, horror, costs, and consequences of armed combat. The political motivations behind the design, formation and establishment of the edifices are also probed. The paper considers the Vietnam War monuments and memorials erected by Americans and Vietnam expatriates in New Orleans, Louisiana, and examines their illustrative and educational usefulness. Heavy metal contamination in food is a worldwide concern. Man-made ponds are domestic sites in the production of Procambarus clarkii and Procambarus zonangulus, two edible species of crayfish. Ponds may be constructed in former sugar cane or rice fields. Crayfish farming is an ancillary seasonal business within the rice-growing season. The use of products to control insects, pests, and weeds in rice and sugar cane production, may cause an accumulation of heavy metals in the crayfish tail within pond structures. Despite her immense popularity in the nineteenth century, Constance Fenimore Woolson's reputation dwindled substantially in the decades which followed. While her works have been rediscovered over the past thirty years, they are often categorized as regionalist writing or, in the case of her penultimate novel, Jupiter Lights, melodrama. What many fail to consider, however, is that Woolson very much considered herself a realist author, and may have been remembered as such were it not for the influence of William Dean Howells and his peers, whose very narrow parameters for literary realism excluded Woolson, among others. Huntington’s disease is an incurable, progressive neurological disorder characterized by loss of motor control, psychiatric dysfunction, and eventual dystonia leading to death. Despite the fact that this disorder is caused by a mutation in one single gene, there is no cure. The mutant Huntingtin (mHtt) protein is expressed ubiquitously throughout the brain but frank cell death is limited to the striatum. Recent work has suggested that Rhes, Ras homolog enriched in striatum, which is selectively expressed in the striatum, may play a role in Huntington’s disease neuropathology. Much of the research examining emotion induction, regulation, and suppression considers solely the visual modality (e.g., pictures of faces) for emotion elicitation. In reality, emotions are cued, expressed, and interpreted through multiple modalities by employing the extensive use of auditory stimuli in addition to visual stimuli. There have been some recent efforts to offset this imbalance in modality preference by using emotional auditory stimuli alone or in addition to visual stimuli. Follicular lymphoma (FL) is the second most common lymphoma in the United States. Although it is generally an indolent lymphoma, FL is not curable, and, in about 30% of patients, the FL undergoes transformation into an aggressive lymphoma (tFL) with marked worsening of prognosis. To identify mutations preferentially present in tFL, we performed whole exome sequencing (WES) on paired FL and tFL arising in the same patients and developed a mutational analysis pipeline. Over activation of the sympathetic nervous system is prevalent in many forms of cardiovascular disease such as chronic heart failure (CHF) and hypertension. Although increased neuronal renin-angiotensin system activity in presympathetic neurons has been well implicated in mediating this sympatho-excitation, many of the neuronal effects of angiotensin II (Ang II) signaling remain poorly understood. One particular mechanism of Ang II-mediated increases in presympathetic neuronal activity is through reductions in voltage-gated K+ currents. The epidermal growth factor receptor (EGFR) family member ErbB2 (Her2) is overexpressed in 20 -30% of invasive breast cancers and this overexpression correlates with poor prognosis and shorter overall as well as disease-free survival. Aberrant expression of ErbB2 through gene amplification, transcriptional deregulation and/or altered endocytic trafficking results in overexpression of ErbB2 at the plasma membrane and biases ErbB2 from primarily ligand-driven hetero-dimerization under normal expression conditions to increased ligand-independent homo-dimer and hetero-dimer formation and consequent activation. The language barrier may severely restrict how severe weather warnings are received and responded to by Hispanics/Latinos in rural Nebraska, a state well known for frequent, volatile weather patterns. Nearly 50% of Spanish speaking Nebraskans rated their English abilities as “less than very well” (US Census Bureau, 2013). The estimated number of Hispanics/Latinos with limited English proficiency (LEP) in Nebraska equates to approximately 57,000 people. Endocytic trafficking is an essential process in eukaryotic cells, specifically for the transport of nutrients, membrane components, and receptors. Cargo destined for endocytic traffic is internalized at the cell surface via clathrin-dependent and clathrin-independent pathways, and brought to the early or sorting endosomes. From there, cargo is further trafficked to lysosomes for degradation, trafficked to other compartments in the cell, or recycled back to the cell surface (either directly or via the endocytic recycling compartment).

Membrane trafficking is a basic cell biological process that controls the distribution of proteins and lipids. Our lab is particularly interested in delineating the cellular functions as well as the molecular mechanisms that regulate the C-terminal Eps15 Homology Domain (EHD) protein family of adenosine-5’-triphosphatases in mammalian cells. EHD1-4 are ubiquitously expressed in mammalian tissues and serve partially overlapping but also distinct functions in regulating membrane shaping and fission along the endocytic pathway. This dissertation examines the role of CXCR2, a seven transmembrane G- protein coupled receptor, in mediating autocrine as well as paracrine mechanisms during pancreatic cancer progression. Data presented in the initial section demonstrates the aberrant expression of the CXCR2 biological axis in human pancreatic cancer tissue specimens. A study performed within the first section of this dissertation investigates the contribution of CXCR2 signaling in pancreatic cancer initiation. These studies have identified a novel role of CXCR2 in mediating KRAS(G12D) -induced autocrine growth transformation of pancreatic cancer cells. Children who are more fearful and inhibited during early childhood are at greater risk for social problems (e.g., loneliness, social isolation) and clinically significant internalizing disorders during adolescence and adulthood (e.g., Rubin, Chen, McDougall, Bowker, & McKinnon, 1995; Williams et al., 2009). While the impact of fearful temperament on adjustment indices are regularly the focus of study, less well understood are biological and social processes that may affect the development of fearful temperament. The present study considered the role of the 5-HTTLPR polymorphism and parenting on change in fearful and inhibited temperamental characteristics during early childhood.
