Identification Of Candidate Genes For Craniosynostosis, Karen Rymer Jan 2015

Identification Of Candidate Genes For Craniosynostosis, Karen Rymer

Theses and Dissertations

Craniosynostosis is a disorder characterized by the premature fusing of cranial sutures in an infant. Premature closure of these sutures can lead to detrimental consequences on the development of a child. The two broad categories of craniosynostosis are classified as syndromic and nonsyndromic. Nonsyndromic craniosynostosis involves only the fusion of one or more sutures, whereas syndromic craniosynostosis involves other abnormalities throughout the body of the affected individual. Two of the families analyzed in this study were of the syndromic nature, and known FGFR mutations were discovered. However, phenotypical features documented in association with these mutations differed from our individuals. Two …

The application of FSLs in medical therapeutics is a recent development, and although …

Incorporating Public Perceptions Into The Selection Of A Nuclear Fuel Cycle, John M. Swanson Jan 2015

Incorporating Public Perceptions Into The Selection Of A Nuclear Fuel Cycle, John M. Swanson

Theses and Dissertations

The final disposal location for used nuclear fuel in the U.S. remains unresolved. A major complication in resolving this issue has historically been the lack of public acceptance. This motivates the creation of a decision making model for selecting a nuclear fuel cycle in the U.S. that incorporates the preferences of the public. A model based on the Analytic Hierarchy Process (AHP) was created, tested, and shown to be problematic in incorporating public opinion into decision objectives. A new model based on Multi-Attribute Utility Theory (MAUT) has been created. This model contains the fundamental objectives for both technical and non-technical …

A Cross-Sectional Analysis Of Health Impacts Of Inorganic Arsenic In Chemical Mixtures, Paul Hargarten Jan 2015

A Cross-Sectional Analysis Of Health Impacts Of Inorganic Arsenic In Chemical Mixtures, Paul Hargarten

Theses and Dissertations

Drinking groundwater is the primary way humans accumulate arsenic. Chronic exposure to inorganic arsenic (iAs) (over decades) has been shown to be associated with multiple health effects at low levels (5-10 ppb) including: cancer, elevated blood pressure and cardiovascular disease, skin lesions, renal failure, and peripheral neuropathy. Using hypertension (or high blood pressure) as a surrogate marker for cardiovascular disease, we examined the effect of iAs alone and in a mixture with other metals using a cross-sectional study of adults in United States (National Health and Examination Survey, NHANES, 2005-2010) adjusting for covariates: urinary creatinine level (mg/dL), poverty index ratio …

Social Performance And Reticence: Mental Negotiations In Austen, Brontë, And Eliot, Meredith L. Spencer

Theses and Dissertations

This thesis examines how three nineteenth-century British novels purvey and critique contemporary standards regarding social performance and reticence and the strains such standards place on those whose dispositions disincline them to conform to the regulations for decorum articulated in conduct books of the time. Utilizing the psychological lens of introversion and extroversion alongside the cognitive narrative theories of Alan Palmer and Lisa Zunshine, this thesis investigates the construction of individual character identities through the reading of interactions among multiple fictional minds in Charlotte Brontë’s Villette (1853), Jane Austen’s Pride and Prejudice (1813), and George Eliot’s The Mill on the Floss …

The Effects Of 7,8-Dihydroxyflavone (7,8-Dhf) On Neuroprotection And Neuroplasticity Follwing A Traumatic Brain Injury, Jennifer M. Romeika

Theses and Dissertations

Aside from preventing traumatic brain injuries (TBIs) altogether, treatment options for TBI typically focus on the secondary biochemical processes that occur in response to the primary mechanical insult. These secondary injuries can lead to apoptosis and necrosis in the days and weeks that follow a TBI. Therefore, finding a treatment that can prevent, reduce, and repair secondary damage is instrumental in the recovery of TBI patients. The flavonoid 7,8-dihydroxyflavone (7,8-DHF) has been identified as a TrkB agonist that mimics the effects of brain derived neurotrophin factor (BDNF). Upon binding to the TrkB receptor, signaling cascades are initiated that can promote …

Investigating The Role Of Bptf In Immunoediting In Breast Cancer And Melanoma, Kristen N. Peterson

Theses and Dissertations

In this study, we explore the effects of NURF depletion on the growth of tumors in immune-competent mice. NURF depletion in tumors results in reduced tumor growth in immune-competent mice, suggesting enhanced anti-tumor immunity. Analysis of the tumor microenvironment by flow cytometry revealed a significantly elevated CD8 and progressively elevated activated CD8 phenotype in Bptf KD tumors, possibly contributing to the increase in cell death and decrease in tumor weight observed. Examination of antigen presentation was evaluated using the OT-1 and Pmel-17 models, though no significant difference in cytotoxicity was observed as measured by LDH and/or IFNγ assays. This indicates …

Prediction Of Protein-Protein Interactions In Escherichia Coli From Experimental Data In Treponema Pallidum, Marco A. Abreu Jan 2015

Prediction Of Protein-Protein Interactions In Escherichia Coli From Experimental Data In Treponema Pallidum, Marco A. Abreu

Theses and Dissertations

Protein – Protein interactions (PPIs) are thought to be conserved between species, although this has not been systematically investigated. This problem was explored in Escherichia coli from experimental data in Treponema pallidum by predicting PPIs, focusing on protein domains of little or unknown function. The comparison of T. pallidum to a model organism such as E. coli can not only reveal additional data about T. pallidum but also reveals how E. coli is similar to this distantly related, obligate parasite. A set of novel T. pallidum interactions, enriched for proteins of unknown function, were the basis of over 23,000 predicted …

Cute As A Button, Marta R. Finkelstein Jan 2015

Cute As A Button, Marta R. Finkelstein

Theses and Dissertations

Cute As A Button explores powerlessness, vulnerability, illness and addiction all wrapped up in tender buttons and a cute, cuddly creature. Using animation, sculpture, sound and an intimate space, I surround the viewer in a saccharine nightmare, one that references the dark underbelly of the cute and the sweet. The visual and aural elements are representative of the psychological and emotional states of powerlessness, which are overcome by the act of making and exploring a medium over which I can have complete control.

The Orphanage Of Things: A Narrative Of Abandonment, Malaz Elgemiabby Jan 2015

The Orphanage Of Things: A Narrative Of Abandonment, Malaz Elgemiabby

Theses and Dissertations

In Sudan, 110 babies are abandoned in the streets of Khartoum every month. The majority of abandoned children are born out of wedlock. Young women with illegitimate pregnancies are often ostracized by their families and society, and the lack of emotional, financial and legal support has led many to take desperate measures, including the abandonment of their children. Relinquishing mothers exist like ghosts in Sudanese society. The only evidence of the mother’s experience is her anonymous, abandoned child. In order to understand and examine this phenomenon, I used ethnographic performance art informed by design research practice (Performative Research Design). I …

Inhibition Of Mtor Signaling Protects Against Myocardial Reperfusion Injury, Acute Myocardial Infarction, Scott M. Filippone Jan 2015

Inhibition Of Mtor Signaling Protects Against Myocardial Reperfusion Injury, Acute Myocardial Infarction, Scott M. Filippone

Theses and Dissertations

Acute myocardial infarction (AMI) is the leading cause of death worldwide. Currently, the best method of treating cardiac ischemia is early reperfusion which, itself, induces myocardial damage. The mTOR complex is a key regulator of cardioprotection against cell stressors. We hypothesized that reperfusion therapy with Rapamycin, a potent mTOR inhibitor, would reduce infarct size in adult mouse hearts. Rapamycin was administered at the onset of reperfusion following 30 min in situ LAD ligation. After 24 hours of reperfusion, myocardial infarct size and apoptosis were significantly reduced in rapamycin-treated mice compared to control. Rapamycin inhibited pro-apoptotic protein Bax and phosphorylation of …

Cyclic Gmp: A Satiety Signal In C. Elegans, Ji S. Park Jan 2015

Cyclic Gmp: A Satiety Signal In C. Elegans, Ji S. Park

Theses and Dissertations

Appetite control and satiety mechanisms help animals maintain energy homeostasis; however, these mechanisms can be misregulated, leading to overweight and obesity. Caenorhabditis elegans is an excellent model system to study appetite and satiety because of its conserved behavioral aspects of satiety and conserved molecular mechanisms. ASI senses nutrition and its activity is required for the behavioral state of satiety quiescence. The purpose of this thesis project was to elucidate the function of cGMP signaling in ASI by looking at behavioral effects from the pharmacological use of sildenafil (Viagra), a PDE inhibitor, and the effects on ASI activation from mutating guanylyl …

Place-Based Scholarship Program Design, Context, And Intergenerational Mobility: A Case Study Of The Kalamazoo Promise Scholarship Program, Arielle F. Niemeyer

Theses and Dissertations

Earning a college degree correlates with achieving financial security. Thus, improving an individual’s access to college is a key tactic used to mitigate poverty and foster intergenerational mobility. Despite the recognized value of higher education, earning a degree remains unattainable for many because of financial constraints. However, research definitively demonstrates that financial aid overcomes that obstacle. It also reveals that some program designs are more effective than others. The Kalamazoo Promise is a place-based scholarship program that offers four-year, full-tuition scholarships to residents who graduate from a Kalamazoo public high school. It is characterized by first-dollar and universal eligibility features, …

One Script, Two Perspectives: Generation Me And The Staging Of Really Really, Kirstin R. Hensley Jan 2015

One Script, Two Perspectives: Generation Me And The Staging Of Really Really, Kirstin R. Hensley

Theses and Dissertations

Really, Really, by Paul Downs Colaizzo, is loosely based on the 2006 Duke University sexual assault scandal resulting from what proved to be a false accusation of rape made against three members of the men’s lacrosse team. After individually reading the play, Jorge Burmudez and I both arrived at differing opinions of who was the true victim, in turn inspiring this thesis, which is an exploration of two separate productions of the play demonstrating contrasting perspectives/outlooks regarding sex crimes within sports organizations in the university setting. It will describe our process from conceptualization to a post-mortem talking about particular …

The Object Inbetween, Lucy L. Derickson Jan 2015

The Object Inbetween, Lucy L. Derickson

Theses and Dissertations

My mission is to bring attention to the unusual, undefined, and fluid connections we have to objects through various avenues of experience. Things, objects, and devices have shaped our culture, environment, and personal philosophies. We live, grow, learn, and mourn with objects. We worship, sympathize, and relate to objects. But what I am most interested in is a human tendency to retreat to objects, allowing them to mediate experiences of friendship and intimacy between people.

During my graduate studies, I have been evaluating my personal and professional connections with others in order to understand how technology such as smart phones …

Development Of Novel Chimeric Epitope Based Diagnostic Antigens And Vaccines For Lyme Disease, Lee D. Oliver Jr. Jan 2015

Development Of Novel Chimeric Epitope Based Diagnostic Antigens And Vaccines For Lyme Disease, Lee D. Oliver Jr.

Theses and Dissertations

Lyme disease (LD) is the leading arthropod-borne disease in North America with 300-600,000 cases each year. There are currently no approved human LD vaccines. Outer surface protein C (OspC) has emerged as a leading vaccine candidate and an attractive diagnostic marker due to its antigenicity and expression early in infection. Several chimeric, epitope based OspC derived proteins were generated. The constructs were found to be highly immunogenic in mice and vaccination induced complement-dependent bactericidal antibodies. These results suggest that a broadly protective polyvalent OspC epitope based vaccine can be produced. Currently, LD diagnostic approaches are unreliable and unable to differentiate …

Reconnect: Designing To Touch, Dana Seros Rohani Jan 2015

Reconnect: Designing To Touch, Dana Seros Rohani

Theses and Dissertations

Our everyday lives are surrounded by gadgets and digital devices that help us perform our daily chores with ease and efficiency. However, these digital devices can also separate us from what we should do ourselves. Although children who are exposed to high levels of technology might become attuned to the latest and the best gadgets, they might not learn to use their physical abilities. Another implication of a child’s over dependence on technology is that parental interactions such as encouragement, tutoring and reinforcement are provided by gadgets rather than living, breathing parents. Research done by Padma Ravichandran and Brandel France …

Community Collaboration In Virginia Legal Aid Programs: A Constructivist Grounded Theory Investigation, Andrew C. Schoeneman

Theses and Dissertations

Legal aid programs comprise a robust national infrastructure attempting to alleviate and reduce poverty. Since their proliferation as part of the War on Poverty, these organizations have provided individual civil legal assistance and engaged in collective legal and political strategies to advance systemic change. Starting in the 1980s, however, public policies have been enacted to cut funding and restrict the ability of federally funded legal aid programs to engage in collective and systemic advocacy. As a result, the ability of programs to work alongside low-income communities has been compromised. The histories and core commitments of legal aid and social work …
