A Non-Invasive Fluorescence-Based Oxygen Sensor And Platform For Studying Cell Responses To Metabolic Agents In Real-Time, Koutilya Reddy Buchapudi

Doctoral Dissertations

A fluorescence-based sensor in a transverse flow/stop measurement platform has been developed to determine real-time changes in oxygen consumption rates for cell metabolic studies. The oxygen sensitive fluorophore platinum octaethylporphyrin was embedded in a cellulose acetate matrix and affixed to a fiber optic bundle, which provided for transmission of the excitation and emission wavelengths of the film. The fiber optic bundle was sealed in a sensor head that can be used in standard 24-well plates common to research labs. The utility of the sensor and sensing platform were determined by measuring the changes in oxygen consumption rates of Candida albicans …

Fret Study Of Ligand Binding And Exchange Kinetics On The Surface Of Cdse/Zns Quantum Dots, Randee Jean Mcbride Jul 2015

Fret Study Of Ligand Binding And Exchange Kinetics On The Surface Of Cdse/Zns Quantum Dots, Randee Jean Mcbride

Graduate Theses and Dissertations

Quantum dots are a valuable tool in many research applications and commercial development. Their applications are far reaching and as such have become a topic of great interest. One of their greatest assets is the ability to utilize them as water soluble fluorescent labels. Rendering the nanocrystals water soluble can be accomplished in several ways, but one very popular method is by capping them with water-soluble multi-functional organic ligands. However, these ligands, when attached to the quantum dot surface, are labile and can be exchanged which can be problematic when using the quantum dots in research applications. This projects investigates …

Bears In Pakistan: Distribution, Population Biology And Human Conflicts, Fakhar -I- Abbas, Zahid Iqbal Bhatti, Jibran Haider, Afsar Mian

Journal of Bioresource Management

We conducted questionnaire based interviews (n = 1873) of respondents coming from 258 localities about bear tracts in northern parts of Pakistan in 2012-2014 to study Himalyan brown (U. arctos isalbellinus) and Himalayan black (U. t. laniger) bears. Brown bears were more frequent in northern latitudes (northern Chitral, Ghizer, Gilgit and Skardu), while black bears were widely distributed in southern latitudes (Battagram). Both brown and black bears are present in central latitudes (Astor, Diamir, Kohistan and Mansehra). We identified 34 populations of brown bears; a large population in the Deosai Plateau and small to very small …

Synthesis, Characterization, Thermal Analysis And Structural Studies Of New Complexes With Tetradentate Ligand, Abbas A. Salih Al-Hamdani, Sahar S. Hassan Jun 2015

Synthesis, Characterization, Thermal Analysis And Structural Studies Of New Complexes With Tetradentate Ligand, Abbas A. Salih Al-Hamdani, Sahar S. Hassan

Baghdad Science Journal

Some new complexes of 4-(5-(1,5-dimethyl-3-oxo-2-phenyl pyrazolidin-4- ylimino)-3,3-dimethyl cyclohexylideneamino) -1,5- dimethyl-2- phenyl -1H- pyrazol -3(2H) –one (L) with Mn(II), Fe(III), Co(II), Ni(II), Cu(II), Pd(II), Re(V) and Pt(IV) were prepared. The ligand and its metal complexes were characterized by phisco- chemical spectroscopic techniques. The spectral data were suggested that the (L) as a neutral tetradentate ligand is coordinated with the metal ions through two nitrogen and two oxygen atoms. These studies revealed Octahedral geometries for all metal complexes, except square planar for Pd(II) complex. Moreover, the thermodynamic activation parameters, such as ΔE*, ΔH,ΔS,ΔG and K are calculated from the TGA curves using …

God's Eye News: The Use Of Drones In Journalism, A Documentary Film, Robert Lance Carroll Jun 2015

God's Eye News: The Use Of Drones In Journalism, A Documentary Film, Robert Lance Carroll

Theses and Dissertations

This thesis uses the format of documentary film to investigate the aesthetic, legal, and ethical issues surrounding the use of Unmanned Aircraft Systems, commonly known as drones, in journalism. Particularly important are the topics of public perception, safety, freedom of speech, and privacy. Do journalists' First Amendment protections extend to the right to gather images using drones? How will the privacy of citizens be protected against aerial cameras that can go virtually unnoticed? Can drones be safely integrated into the National Airspace System? The goal of the documentary is not necessarily to answer these questions, but to gather opinions from …

Molecular Dynamics Investigation Of The Arabinan-Cellulose Interface For Cellulose Nanocomposite Applications, Luke Thornley Jun 2015

Molecular Dynamics Investigation Of The Arabinan-Cellulose Interface For Cellulose Nanocomposite Applications, Luke Thornley

Materials Engineering

Atom level computer simulations of the arabinan and cellulose interface were performed to better understand the mechanisms that give arabinan-cellulose composites (ArCCs) their strength with the goal to improve man-made ArCCs. The molecular dynamics (MD) software LAMMPS was used in conjunction with the ReaxFF/c force field to model the bond between cellulose and arabinan. A cellulose nanocrystal with dimensions 51 x 32 x 8 Å was minimized with various weight percent of water, 0%, 3%, 5%, 8%, 10%, and 12%. After the system was equilibrated for at least 100,000 femtoseconds, an arabinan molecule composed of 8 arabinose rings was added …

Geographically Isolated Wetlands: Rethinking A Misnomer, David M. Mushet, Aram J. K. Calhoun, Laurie C. Alexander, Matthew J. Cohen, Edward S. Dekeyser, Laurie Fowler, Charles R. Lane, Mark C. Rains, Susan C. Walls Jun 2015

Geographically Isolated Wetlands: Rethinking A Misnomer, David M. Mushet, Aram J. K. Calhoun, Laurie C. Alexander, Matthew J. Cohen, Edward S. Dekeyser, Laurie Fowler, Charles R. Lane, Mark C. Rains, Susan C. Walls

School of Geosciences Faculty and Staff Publications

We explore the category “geographically isolated wetlands” (GIWs; i.e., wetlands completely surrounded by uplands at the local scale) as used in the wetland sciences. As currently used, the GIW category (1) hampers scientific efforts by obscuring important hydrological and ecological differences among multiple wetland functional types, (2) aggregates wetlands in a manner not reflective of regulatory and management information needs, (3) implies wetlands so described are in some way “isolated,” an often incorrect implication, (4) is inconsistent with more broadly used and accepted concepts of “geographic isolation,” and (5) has injected unnecessary confusion into scientific investigations and discussions. Instead, we …

The Human Ecology Dialectic: Culture As A Behavioral Adaptation, Marcia S. Taylor May 2015

The Human Ecology Dialectic: Culture As A Behavioral Adaptation, Marcia S. Taylor

The Hilltop Review

Contrary to popular thought, which implicated the disengagement of Marx from anthropological theory, his dialectical concept will be applied to explain human ecology in a Boasian historical particularism milieu. Environmental conditions will be the common denominator for the Darwinian biogeography theory, intertwined with the cultural adaptation theory of Boas within the Human Ecology Dialectic (Taylor, 2014). Anthropologically, in a biological context the theories will explain the behavioral adaptation process demonstrated culturally by the prehistoric ancestors of the Inuit of the Canadian Central Arctic in their development and essential engagement in the creation of art; a pursuit that became a cultural …

Comparison Of Quantitative Analysis To Qualitative Analysis For Interpretation Of Lower Limb Lymphoscintigraphy, Edward Chege Nganga May 2015

Comparison Of Quantitative Analysis To Qualitative Analysis For Interpretation Of Lower Limb Lymphoscintigraphy, Edward Chege Nganga

Master of Medicine in Imaging and Diagnostic Radiology

Background: Lymphoscintigraphy is now the primary imaging modality used in determining a diagnosis in patients with suspected extremity lymphedema. However, analysis is mainly by visual qualitative analysis which is subjective and has a wide inter-observer variability. Subtle differences in ilioinguinal uptake between normal and abnormal limbs may be missed with visual qualitative analysis. This study seeks to compare quantitative analysis to qualitative analysis by computing percentage ilioinguinal nodes uptake at 1.5 hours. Objectives: To compare quantitative analysis to qualitative analysis of lower limb lymphoscintigraphy in the diagnosis of lymphedema. To determine the diagnostic accuracy of quantitative lymphoscintigraphy analysis. Methodology: 52 …

Using Otolith Strontium Isotopes To Elucidate Population Structure And Movements Of Bering Cisco (Coregonus Laurettae), Andrew John Padilla May 2015

Using Otolith Strontium Isotopes To Elucidate Population Structure And Movements Of Bering Cisco (Coregonus Laurettae), Andrew John Padilla

Marine Biology

Methods for stock discrimination and tracking the movements and distribution of fishes have often involved expensive field logistics, a problem compounded in remote regions such as Alaska. An alternative approach is to use the chemical signatures preserved in otoliths, or ear bones, of teleost fishes to discriminate stocks or to track the movement history of fish. Currently, a commercial fishery targeting the anadromous Bering cisco Coregonus laurettae is occurring in the Yukon River, Alaska. There are only three known Bering cisco spawning rivers worldwide, the Yukon, South Fork Kuskokwim (Kuskokwim), and Susitna rivers. Managers and researchers believed that two of …

Multiblock Discriminant Analysis For Integrative Genomic Study, Mingon Kang, Dong-Chul Kim, Chunyu Liu, Jean Gao May 2015

Multiblock Discriminant Analysis For Integrative Genomic Study, Mingon Kang, Dong-Chul Kim, Chunyu Liu, Jean Gao

Computer Science Faculty Publications

Human diseases are abnormal medical conditions in which multiple biological components are complicatedly involved. Nevertheless, most contributions of research have been made with a single type of genetic data such as Single Nucleotide Polymorphism (SNP) or Copy Number Variation (CNV). Furthermore, epigenetic modifications and transcriptional regulations have to be considered to fully exploit the knowledge of the complex human diseases as well as the genomic variants. We call the collection of the multiple heterogeneous data “multiblock data.” In this paper, we propose a novel Multiblock Discriminant Analysis (MultiDA) method that provides a new integrative genomic model for the multiblock analysis …

Development And Application Of Mass Spectrometry-Based Biophysical Approaches, Ying Zhang May 2015

Development And Application Of Mass Spectrometry-Based Biophysical Approaches, Ying Zhang

Arts & Sciences Graduate Student Theses and Dissertations

Mass spectrometry (MS)-based biophysical approaches are new "tools" for protein characterization owing to its capability to analyze proteins and protein complexes that range in molecular weight from kDa to MDa. Protein characterization requires more than identifying the primary structure. More importantly, protein high order structures (i.e., secondary, tertiary and quaternary structures) are needed for biological studies. MS has become the major tool in studies of protein primary structure and post translational modifications (PTMs) over the past two decades. Because MS has high sensitivity and fast turnaround, more and more biophysical approaches rely on MS to generate information for protein higher …

The Roles Of Mist1 And Rab26 In Zymogenic (Chief) Cell Differentiation And Subcellular Organization, Ramon Jin May 2015

The Roles Of Mist1 And Rab26 In Zymogenic (Chief) Cell Differentiation And Subcellular Organization, Ramon Jin

Arts & Sciences Graduate Student Theses and Dissertations

Little is known about how differentiating cells reorganize their cellular structure to perform specialized physiological functions. Specifically, exocrine cells like pancreatic acinar and zymogenic chief cells have a highly developed secretory architecture that is rapidly established upon differentiation. What is known in these cells is that the evolutionarily conserved transcription factor MIST1 is required for final maturation. We hypothesized that MIST1 directly regulates specific subcellular components that facilitate functional maturation of secretory cells. We show that MIST1 binds to conserved CATATG E-boxes to activate transcription of 6 genes, including the small GTPases RAB26 and RAB3D. We next demonstrate that RAB26 and …

Climate Change And Phenology: Empoasca Fabae (Hemiptera: Cicadellidae) Migration And Severity Of Impact, Mitchell B. Baker, P. Dilip Venugopal, William O. Lamp

Publications and Research

Climate change can benefit individual species, but when pest species are enhanced by warmer temperatures agricultural productivity may be placed at greater risk. We analyzed the effects of temperature anomaly on arrival date and infestation severity of potato leafhopper, Empoasca fabae Harris, a classic new world long distance migrant, and a significant pest in several agricultural crops. We compiled E. fabae arrival dates and infestation severity data at different states in USA from existing literature reviews and agricultural extension records from 1951–2012, and examined the influence of temperature anomalies at each target state or overwintering range on the date of …

Gender And The History Of Philosophy: An Analysis Of Essentialism And Gender Disempowerment, Forrest T. Deacon May 2015

Gender And The History Of Philosophy: An Analysis Of Essentialism And Gender Disempowerment, Forrest T. Deacon

Mahurin Honors College Capstone Experience/Thesis Projects

In this project, I examine the philosophical theories of truth, gender, and power, and the parallels between each theory. I argue that both Friedrich Nietzsche and William James advanced theories that deconstructed the idea that human beings, or “man” and “woman,” were bound by an essential nature or innate characteristics that determined their social role. Though this critique was robust, I argue that it enforces gender disempowerment on a number of platforms since the theories did not analyze gender, but rather truth and value. Simone de Beauvoir, I argue, expanded Nietzsche’s and James’ thought, but included a critical analysis of …

The Synthesis And Study Of The Biological And Colloidal Properties Of Bolaamphiphiles, Louis M. Damiano May 2015

The Synthesis And Study Of The Biological And Colloidal Properties Of Bolaamphiphiles, Louis M. Damiano

Senior Honors Projects, 2010-2019

Over the past decade, antibiotic resistant bacteria have caused infections in patients throughout the world.[1] The rise in antibiotic resistance is primarily due to the misuse and overuse of antibiotics. [1] To counter the increase in antibiotic resistance, infection control mechanisms have been aggressively researched in recent years. In particular, drug delivery has become a focal point to fight antibiotic resistant infections.[2] Amphiphiles have a wide range of applications in the clinical setting, including the ability to inhibit bacterial transference because of their bactericidal activity. [3] Bolaamphiphiles are a subclass of amphiphiles that possess two or more hydrophilic …

Synthesis And Characterization Of C8 And N2 2'-Deoxyguanosine Adducts Of 6-Nitrochrysene, A Cancer-Causing Agent, Kimberly R. Rebello May 2015

Synthesis And Characterization Of C8 And N2 2'-Deoxyguanosine Adducts Of 6-Nitrochrysene, A Cancer-Causing Agent, Kimberly R. Rebello

Master's Theses

Nitropolycyclic aromatic hydrocarbons are ubiquitous in the environment, and many compounds of this family are carcinogenic. 6-Nitrochrysene, a representative member of this group, is a suspected human carcinogen that has been shown to cause lung, breast, and other forms of cancer in rodents. It is believed that this derivative of chrysene causes cancer by forming DNA adducts, which, in turn, induce mutations in critical gene sequences. The goal of my research is to synthesize one or more of the major DNA adducts formed by 6-nitrochrysene, which will be introduced in DNA in order to study their biological effects.

The initiation …
