Foodborne illness continues to be a substantial public health issue in the United States, with fresh produce being one of the leading causes of outbreaks. Understanding routes of contamination of fresh produce and how pathogens survive on plant surfaces is paramount in improving food safety and reducing risk to public health. The objectives of this study were to select environmental E.coli isolates as pathogen surrogates of Salmonella typhimurium and E.coli O157:H7, assess lettuce plant contamination by spray irrigation water, and evaluate a common industry quality control (QC) E.coli strain (ATCC 25922). Background

Clustered regularly interspaced short palindromic repeats (CRISPR) and CRISPR-associated genes (cas) are widely distributed among bacteria. These systems provide adaptive immunity against mobile genetic elements specified by the spacer sequences stored within the CRISPR.

Methods

The CRISPR-Cas system has been identified using Basic Local Alignment Search Tool (BLAST) against other sequenced and annotated genomes and confirmed via CRISPRfinder program. Using Polymerase Chain Reactions (PCR) and Sanger DNA sequencing, we discovered CRISPRs in additional bacterial isolates of the same species of Bordetella. Transcriptional activity and processing of the CRISPR have been assessed via RT-PCR.

Running buffalo clover (Trifolium stoloniferum Muhl. ex A. Eaton) is a species of clover that is federally listed as endangered. Trifolium stoloniferum requires mesic habitats with partially filtered light and will be outcompeted without periodic disturbance, such as grazing, mowing, or trampling. The purpose of this study was to understand rates of flowering and clonal reproduction associated with different growth stages of T. stoloniferum. During this study I visited 6 restoration sites of T. stoloniferum once each week from April to October, marking new individuals with a unique numbered metal tag and assessing the growth stage of individuals. …

Cardiovascular homeostasis is regulated in large part by the rostral ventrolateral medulla (RVLM) in mammals. Projections from the RVLM to the intermediolateral column of the thoracolumbar spinal cord innervate preganglionic neurons of the sympathetic nervous system causing elevation of blood pressure and heart rate. A large proportion, but not all, of the neurons in the RVLM contain the enzymes necessary for the production of epinephrine and are identified as the C1 cell group. Aging is the major risk factor for neurodegenerative diseases such as Alzheimer's disease, but little is known about the processes that lead to age-related decline of brain structures and function. Here we use RNA-seq in combination with high resolution histological analyses to show that aging leads to a significant deterioration of neurovascular structures including basement membrane reduction, pericyte loss, and astrocyte dysfunction. Neurovascular decline was sufficient to cause vascular leakage and correlated strongly with an increase in neuroinflammation including up-regulation of complement component C1QA in microglia/monocytes. Importantly, long-term aerobic exercise from midlife to old age prevented this age-related neurovascular decline, …

Pharmacology, Physiology and Toxicology

Cisplatin is a commonly used drug for cancer treatment by crosslinking DNA, leading to apoptosis of cancer cells, resistance to cisplatin treatment often occurs, leading to relapse. Therefore, there is a need for the development of more effective treatment strategies that can overcome chemoresistance. Myricetin is a flavonoid from fruits and vegetables, showing anticancer activity in various cancer cells. In this study, we found myricetin exhibited greater cytotoxicity than cisplatin in two cisplatin-resistant ovarian cancer cell lines, OVCAR-3 and A2780/CP70, and it was less cytotoxic to the normal ovarian cell line IOSE-364. Insect performance is limited by the temperature of the environment, and in temperate, polar, and alpine regions, the majority of insects must face the challenge of exposure to low temperatures. The physiological response to cold exposure shapes the ability of insects to survive and thrive in these environments, and can be measured, without great technical difficulty, for both basic and applied research. For example, understanding insect cold tolerance allows us to predict the establishment and spread of insect pests and biological control agents. This essay emerges from a joint symposium of the Hastings Business Law Journal and the Hastings Science and Technology Law Journal entitled “Regulating the Disruption Economy: Tech Startups as Regulatory Reformers.” The symposium featured panels on virtual currency, crowdfunding, and the sharing economy.

The pathophysiology of Type 2 Diabetes Mellitus (T2DM) is modelled using a coupled system of non-linear deterministic differential equations. An attempt is made to construct to a clinically plausible mathematical model that incorporates the homeostasis associated with endocrinological regulation of glucose and glycogen levels in the human body, by the hormones, insulin and glucagon. The model variables include the concentrations of glucose in the venous blood plasma, the concentration of glycogen in the liver/tissues, the concentration of the hormone glucagon, and the concentration of insulin in the venous blood plasma. Infection with the human liver fluke Opisthorchis viverrini induces cancer of the bile ducts, cholangiocarcinoma (CCA). Injury from feeding activities of this parasite within the human biliary tree causes extensive lesions, wounds that undergo protracted cycles of healing, and re-injury over years of chronic infection. We show that O. viverrini secreted proteins accelerated wound resolution in human cholangiocytes, an outcome that was compromised following silencing of expression of the fluke-derived gene encoding the granulin-like growth factor, Ov-GRN-1. Recombinant Ov-GRN-1 induced angiogenesis and accelerated mouse wound healing. Ov-GRN-1 was internalized by human cholangiocytes and induced gene and protein expression changes associated …

Medulloblastoma (MB), a primitive neuroectodermal tumor, is the most common malignant childhood brain tumor and remains incurable in about a third of patients. Currently, survivors carry a significant burden of late treatment effects. The p53 tumor suppressor protein plays a crucial role in influencing cell survival in response to cellular stress and while the p53 pathway is considered a key determinant of anti-tumor responses in many tumors, its role in cell survival in MB is much less well defined. Assessing The Effects Of An Std Educational Intervention On Greek Society College Students’ Knowledge And Attitudes Toward Safe Sex Behaviors, Jennifer W. Goldsberry

Doctor of Nursing Practice (DNP) Translational and Clinical Research Projects

This study measured the effectiveness of an evidence-based educational intervention targeting STD prevention in Greek society members attending college and examined relationships between sexual health knowledge, attitudes toward safe sex behaviors, and sexual demographics. One hundred thirty-two Greek society members attending college participated. Instruments measured demographic characteristics, knowledge regarding sexually transmitted diseases (STDs), and attitudes toward practicing safe sex behaviors at baseline and one week post-intervention. There was a significant increase in STD knowledge from baseline (M=13.03, SD=6.5) to one week (M=20.27, SD=4.9) t (131) = -13.53, p = .000. Fraternity members were more …
