The Role Of The B-Type Phospholipases In S. Cerevisiae: Function, Regulation, And Physiological Relevance In Lipid Homeostasis, Beth A Surlow Jan 2014

The Role Of The B-Type Phospholipases In S. Cerevisiae: Function, Regulation, And Physiological Relevance In Lipid Homeostasis, Beth A Surlow

Electronic Theses and Dissertations

Membrane phospholipid synthesis and turnover is a continual process during normal cell growth. The turnover of the glycerophospholipids by B-type phospholipases (PLBs) in Saccharomyces cerevisiae results in the formation glycerophosphodiesters through a deacylation reaction. Here, I address several aspects of the glycerophospholipid deacylation, transport, and reutilization pathway in S. cerevisiae. First, I show the RAS GTPase-activating proteins, Ira1 and Ira2, are required for utilization of the glycerophosphodiester - glycerophosphoinositol (GroPIns) - as a phosphate source. Second, I demonstrate loss of the cell surface associated PLBs, Plb1-3, and/or utilization of GroPIns causes actin cytoskeleton defects and an increased cell size. For …

Crisis Clinician's Lived Experience Of Clinical Supervision, Elizabeth A. Sysak Jan 2014

Crisis Clinician's Lived Experience Of Clinical Supervision, Elizabeth A. Sysak

Electronic Theses and Dissertations

Crisis intervention first responders experience both small and large-scale disasters that can leave lasting negative impacts on crisis clinicians. However, little is known about how clinicians receive support for their personal and professional well being in clinical supervision. This hermeneutic phenomenological study explored the lived experience of crisis clinicians to understand how and if they receive support through clinical supervision in their work in crisis intervention. This study found that crisis clinicians struggle to manage the complex emotions that accompany the unique work of crisis intervention. This study also found that crisis clinicians are not receiving the support they need …

Are Community Food Assistance Programs Meeting The Needs Of Food-Insecure Households In Pittsburgh's North Side Neighborhoods?, Ryan M. Van Dinter Jan 2014

Are Community Food Assistance Programs Meeting The Needs Of Food-Insecure Households In Pittsburgh's North Side Neighborhoods?, Ryan M. Van Dinter

Electronic Theses and Dissertations

Semi-structured interviews with three householders and three community food program managers were conducted to explore if food assistance programs meet the needs of residents in Pittsburgh's North Side neighborhoods. Findings show that insufficient income is the main factor contributing to household food insecurity. North Side residents also have difficulty obtaining nutritious and affordable foods because fresh produce is often too expensive and supermarket food quality is considered low. Farm stands and other food access programs are well received, but only provide access to nutritious foods during the summer. Limited income and limited food access increase reliance on food pantries and …

Communication And Response-Ability: Levinas And Kierkegaard In Conversation, Beth A. Walter Jan 2014

Communication And Response-Ability: Levinas And Kierkegaard In Conversation, Beth A. Walter

Electronic Theses and Dissertations

This project contends that hope for ethical communication in a postmodern age lies in the ability to rethink ethics in terms of "existential pathos." To that end, this study locates communicative responsibility in the responsive element of the self-other relation by relying primarily on the work of the twentieth-century Lithuanian-born French philosopher Emmanuel Levinas. I maintain that Levinas's disruption of the philosophical tradition informs a communication ethic comprised of dialectical, dialogical, and rhetorical modes of interpersonal interaction that are fundamentally rooted in an existential understanding of human striving. Further, I assert that these dialectical, dialogical and rhetorical components are best …

Modulation Of Immunity Via Antibodies Displayed On In Situ Forming Peptide Co-Assemblies, Yi Wen Jan 2014

Modulation Of Immunity Via Antibodies Displayed On In Situ Forming Peptide Co-Assemblies, Yi Wen

Electronic Theses and Dissertations

Many diseases are associated with disruption of immune homeostasis. Monoclonal antibodies (mAb) targeting soluble antigens or immune cell surface molecules have been developed to modulate immunity. However, systemic administration can lead to severe immune related side effects. This dissertation investigated a versatile platform to display antibodies locally in diseased tissues to minimize unwanted systemic exposure. This system is a stimuli-responsive and injectable formulation utilizing in situ forming peptide co-assemblies and two linker proteins. The self-assembling peptides EAK16-II and EAKIIH6 co-assemble into fibrous structures with accessible His-tags when exposed to salts. Anti-His-tag antibodies (αH6-IgG) bind to these His-tags and provide fragment …

The Genetics Of Conservation Translocations: A Comparison Of North American Golden Eagles (Aquila Chrysaetos Canadensis) And Bald Eagles (Haliaeetus Leucocephalus), Maria Wheeler Jan 2014

The Genetics Of Conservation Translocations: A Comparison Of North American Golden Eagles (Aquila Chrysaetos Canadensis) And Bald Eagles (Haliaeetus Leucocephalus), Maria Wheeler

Electronic Theses and Dissertations

In the past half-century, two raptors with similar life histories, bald eagles (Haliaeetus leucocephalus) and golden eagles (Aquila chrysaetos canadensis) were the subjects of either broad-scale reintroduction or translocation projects in North America. These two different conservation approaches provided the framework for a retrospective natural experiment. The goal of my research was to determine the effects of these two conservation management approaches on the genetic population structure of each species. Methods included sequencing cyt b mtDNA and genotyping 10 microsatellite loci for contemporary and historic golden eagles and bald eagles. Contemporary samples were collected from captive and wild birds, and …

Parent-Teacher Engagement During Child-Centered Pedgogial Change In Elementary School: The Lived Experiences Of Teachers And Involved Parents, Cristiana P. White Jan 2014

Parent-Teacher Engagement During Child-Centered Pedgogial Change In Elementary School: The Lived Experiences Of Teachers And Involved Parents, Cristiana P. White

Electronic Theses and Dissertations

With the advance of technology as a manifestation of our rapidly changing world, we often hear how difficult it is to be a parent with school-age children. The more knowledge and access parents have to technology, the better position they are in to collaborate with teachers and to help their children academically and behaviorally. However, the lack of updated technology may be a serious concern for children living in poverty. For these and many other reasons, it is imperative that educators provide parents with the knowledge base and the resources needed to facilitate parent engagement in helping their children with …

A Decentralized Approach To Confront The Debasement Of Black Male Student Athletes Who Attend Pwi's, Ronald William Whitaker Ii Jan 2014

A Decentralized Approach To Confront The Debasement Of Black Male Student Athletes Who Attend Pwi's, Ronald William Whitaker Ii

Electronic Theses and Dissertations

The debasement and "niggering " of Black male student athletes who attend PWI's is an issue that has been well documented since the mid to late sixties (Edwards, 1969, Olson, 1968). This population has been negatively stereotyped as being superior athletically, but inferior intellectually (Edwards, 1984). Both scholars and practitioners have focused on other pervasive issues including: (1) the academic achievement gap that exist between Black male student athletes and their White counterparts (Harper, 2013), (2) the commercialization of college athletics (Beamon, 2008; Donnor, 2005), and (3) negative stereotypes that continue to plague Black male student athletes (Hodge, Burden, Robinson, …

The Race For Privilege In Blackness: Transformative Leadership In Traversing Space In Search Of Black Queer Cool, Tiffany Wilkins Jan 2014

The Race For Privilege In Blackness: Transformative Leadership In Traversing Space In Search Of Black Queer Cool, Tiffany Wilkins

Electronic Theses and Dissertations

This portraiture project, incorporating auto-ethnographic elements, investigating how African American educational systems became systems of oppression faced by lesbian, gay, bisexual, transgender and questioning (LGBTQ) youth of color, intentionally blends artist in creative glance with researcher in systemic inquiry and knowledge gathering (Lawrence-Lightfoot & Hoffman-Davis, 1997; Ellis et al., 2011).

An aspiring African American, gender non-conforming, lesbian leader is an auto-ethnographic portraiture exemplar: a counter-narrative of transcending obstacles overshadowing accessing college, graduate school, career success in education; doctoral ambitions with audacity within classes of pervasive racism, heterosexism and homophobia. Parallel instances of critical confrontations in predominantly Black secondary schools and …

Lipoprotein Subclass Measurements By An Updated Nuclear Magnetic Resonance Spectroscopy Method In The Prediction Of Coronary Artery Disease In Type 1 Diabetes, Qing Xu Jan 2014

Lipoprotein Subclass Measurements By An Updated Nuclear Magnetic Resonance Spectroscopy Method In The Prediction Of Coronary Artery Disease In Type 1 Diabetes, Qing Xu

Electronic Theses and Dissertations

The role of low density lipoprotein (LDL) cholesterol as a major risk factor for the development of cardiovascular disease is widely accepted. Indeed, major health organizations, such as the American Diabetes Association and the American Heart Association, recommend increased efforts to keep LDL concentrations within target as a means of subsequently reducing the incidence of atherosclerosis, especially among individuals with diabetes1.

However, the role of high density lipoprotein (HDL) cholesterol in the development of cardiovascular disease is not as clear as that of LDL. Although earlier studies have suggested an inverse association between HDL cholesterol concentrations and CAD incidence2, more …

The Application Of Mass Spectrometric Measurement Techniques For The Evaluation And Assessment Of Autism Spectrum Disorders, Gregory M. Zinn

Electronic Theses and Dissertations

Autism spectrum disorders (ASDs) are neurological developmental disorders that affect communication and social interaction. They are becoming more important to society as their rate has risen dramatically over the past 30 years. Presently, there is no known cure for ASDs and the current research focuses on improvements in diagnosis and different types of treatments. The most significant contribution of the research described in this dissertation is the application of advanced mass spectrometric measurement techniques that provide higher accuracy and precision to the diagnosis and treatment of ASDs. The treatment of ASDs was assessed through the evaluation of elemental contamination in …

The Role Of G-Quadruplex Rna Motif In Fragile X Syndrome, Yang Zhang Jan 2014

The Role Of G-Quadruplex Rna Motif In Fragile X Syndrome, Yang Zhang

Electronic Theses and Dissertations

Fragile X syndrome (FXS), the most common cause of inherited mental impairment, is caused by the loss of expression of the fragile X mental retardation protein (FMRP). As an RNA binding protein, FMRP has been proposed to regulate the transport and translation of specific message RNA (mRNA). It has been reported that FMRP uses its RGG box domain to bind mRNA targets that form a G-quadruplex structure, structure believed to be important for FMRP recognition of at least a subclass of its mRNA targets. We have hypothesized that the interaction of FMRP with selected relevant mRNA targets occurs in a …

Max Scheler's Critical Theory: The Idea Of Critical Phenomenology, Eric J. Mohr Jan 2014

Max Scheler's Critical Theory: The Idea Of Critical Phenomenology, Eric J. Mohr

Electronic Theses and Dissertations

This work explores the way core elements of phenomenology map on to the critical theory program in order to demonstrate phenomenology's relevance for ideology critique. "Critical phenomenology" means putting the findings of phenomenology to work for the sake of social critique. I argue that phenomenology gains a critical edge precisely where many critical theorists suggest phenomenology withdraws from a critical function: on the basis of their theory of intuition. While Adorno takes phenomenological intuition to be another version of identity philosophy, he overlooks the significance of the way in which phenomenological givenness is incommensurable with, and at best only symbolized …

Transparency, Robin Feldman Jan 2014

Transparency, Robin Feldman

The patent regime is a quintessential notice system. Implicit in its design is the concept that one attempting to license a patent can identify those who hold the requisite rights and the territory that the patent holders claim. The modern system, however, bears little resemblance to the idealized form. In the last decade, an entire Hobbit’s world has been created under the foliage with little information available to inform the market. To address the problem, this Article suggests borrowing from doctrines related to disclosure in the realm of corporate securities, molding those doctrines to particular patent concerns. As a government …
