Effects Of Selected Commercial Diets And Yeast Substitution On The Growth And Associated Microbiota Of Rotifer (Brachionus Plicatilis), Ryan D. Lind
Open Access Theses
The rotifer (Brachionus plicatilis) is an essential feed organism for successful marine larviculture. Cost effective feeding regimes and the control of harmful bacteria are critical components of rotifer production. Supplementation of commercial diets with yeast can reduce costs, but its effects on bacteria such as Vibrio spp. are not well documented. Two experiments were conducted to identify a practical culture diet. The first experiment evaluates different commercial diets on rotifer growth and total Vibrio. The second experiment determines a practical level of yeast substitution, with regard to rotifer growth, culture Vibrio loads, and cost. The diets evaluated in the first …

The success in recovering genetic profiles from aged and degraded biological samples is diminished by fundamental aspects of DNA extraction, as well as its long-term preservation that are not well understood. While numerous studies have been conducted to determine whether one extraction method performed superior to others, nearly all of them were initiated with no knowledge of the actual starting DNA quantity in the samples prior to extraction, so they ultimately compared the outcome of all methods relative to the best. Using quantitative PCR (qPCR) to estimate the copy count of synthetic standards before (i.e., “copies in”) and after (i.e., …
A Conversation With D. Nurkse, Josh Brewer
A Conversation With D. Nurkse, Josh Brewer
Booth
Comfortably ensconced in a breakfast nook, poet D. Nurkse discussed the architecture of Butler’s Efromyson Center for Creative Writing (one way of talking about the relationship between labor and form), literary tradition, falling snow (visible through the window), and his parents leaving Europe as it disintegrated into World War II. We also discussed his use of baseball in poetry. Wizened and soft-spoken, he sipped coffee and reflected upon several decades of work. Nurkse’s career could have been different.
A Harvard graduate and son of an esteemed economist, he turned down an opportunity to pursue an international academic career. Instead, he …

K20 is a novel amphiphilic antifungal aminoglycoside that is synthetically derived from the antibiotic kanamycin A. Reported here are investigations of K20′s antimicrobial activities, cytotoxicity, and fungicidal mechanism of action. In vitro growth inhibitory activities against a variety of human and plant pathogenic yeasts, filamentous fungi, and bacteria were determined using microbroth dilution assays and time-kill curve analyses, and hemolytic and animal cell cytotoxic activities were determined. Effects on Cryptococcus neoformans H-99 infectivity were determined with a preventive murine lung infection model. The antifungal mechanism of action was studied using intact fungal cells, yeast lipid mutants, and small unilamellar lipid …

A Third Species Of The Endemic Floridian Milliped Genus Floridobolus Causey, 1957 (Spirobolida: Spirobolidae: Floridobolinae), Rowland Shelley, Gary J. Phillips, Ernest C. Bernard
Insecta Mundi
Characterized by small body size, apically rounded/lobed anterior gonopod telopodites, long slender posterior gonopod telopodites, and torsion in the cyphopod receptacles, Floridobolus floydi, n. sp., is described from the southern sector of the Brooksville Ridge in northwestern peninsular Florida. It inhabits sandy “Big Scrub” environments like F. penneri Causey, 1957, and F. orini Shelley, 2014, and is documented from the sector’s center and northern periphery, in Hernando and Citrus Counties, respectively, with a sight record from the eastern periphery. Its discovery supports the thesis that each sand ridge in peninsular Florida may harbor a unique species of this …
Avian Diversity Across Three Distinct Agricultural Landscapes In Guadalupe, Chiriquí Highlands, Panama, Jarred Jones
Avian Diversity Across Three Distinct Agricultural Landscapes In Guadalupe, Chiriquí Highlands, Panama, Jarred Jones
Independent Study Project (ISP) Collection
Deforestation of tropical forest for agricultural purposes threatens habitat loss of native species. The value of various agricultural landscapes in conserving avian populations is useful in determining diversity-conscious development plans. However, generalized results from regionalscale studies cannot be implemented to insular avian habitats. This study serves as the only current avian diversity study of the Chiriquí Highlands. To determine the effect of agricultural land use within an insular avian habitat, I compared avian diversity and site population similarity in Guadalupe, Chiriquí Highlands of the Talamanca Range, Panama. I hypothesized that avian diversity is greatest at forest edge followed by forest …

Mating Dynamics Of The Calanoid Copepods Acartia Tonsa And Acartia Hudsonica, Zair P Lojkovic Burris
Doctoral Dissertations
Copepods are the most abundant metazoans in the ocean, link the microbial plankton to upper trophic levels, and affect ecosystem function through their role in biogeochemical cycles. This thesis focuses on an understudied area in copepods-- factors that determine mating success. First, I tested the hypothesis that female-biased adult sex ratios of Acartia tonsa may be explained by ratios already skewed at birth. Offspring of field-caught females were raised to adulthood and the corresponding adult sex ratios were used as proxies for birth ratios. Almost one-fifth of mothers produced significantly female-biased sex ratios, suggesting that secondary processes commonly used to …

Caveolin-1 (CAV1) is a vital scaffold protein heterogeneously expressed in both healthy and malignant tissue. We focus on the role of CAV1 when overexpressed in T-cell leukemia. Previously, we have shown that CAV1 is involved in cell-to-cell communication, cellular proliferation, and immune synapse formation; however, the molecular mechanisms have not been elucidated. We hypothesize that the role of CAV1 in immune synapse formation contributes to immune regulation during leukemic progression, thereby warranting studies of the role of CAV1 in CD4+ T-cells in relation to antigen-presenting cells. To address this need, we developed a computational model of a CD4+ …

We investigate the vibrational modes of quasi-two-dimensional disordered colloidal packings of hard colloidal spheres with short-range attractions as a function of packing fraction. Certain properties of the vibrational density of states (vDOS) are shown to correlate with the density and structure of the samples (i.e., in sparsely versus densely packed samples). Specifically, a crossover from dense glassy to sparse gel-like states is suggested by an excess of phonon modes at low frequency and by a variation in the slope of the vDOS with frequency at low frequency. This change in phonon mode distribution is demonstrated to arise largely from localized …

Famine In Context: Possible Long-Term Effects Of Exposure To Starvation And Malnutrition In Early Life And Subsequent Implications For The Current Crisis In South Sudan, Jenna Mae Stoken
Honors Projects
Central focus
The central focus of this thesis is to examine the historical and sociopolitical roots of famine. In examining the root causes of famine, we can more carefully analyze its effects on the human condition. More specifically, in analyzing the pathologies associated with food deprivation and malnutrition at an early age, we can examine the possible long-term consequences of malnutrition on human health. In doing so, we can apply this understanding to the current crisis in South Sudan, to consider the implications that the current political situation presents for future generations of South Sudanese people.
Thesis rationale
This thesis …

Combating Eutrophication: An Ecosystem Scale Analysis Of Aluminum Sulfate (Alum) Effectiveness Among Lakes, With Comparison To Alum And Biomanipulation Dual Treatment, Christa M. Webber
School of Natural Resources: Dissertations, Theses, and Student Research
Eutrophic conditions in lakes and reservoirs in agricultural regions often drive summer blooms of toxic cyanobacteria. Aluminum sulfate (alum) applications are commonly used to control cyanobacteria blooms and restore water quality in eutrophic lakes. However, studies of alum treatments often lack true replication, comparison to reference lakes, or comparison to other restoration techniques, such as an alum and biomanipulation combined or “dual” treatments. Without these comparisons, the variation of treatment response between replicate lakes and restoration techniques remains uncertain. Therefore, I sought to assess how water quality is affected by multiple restoration techniques among geographically proximate (1.4 km2) …

BACKGROUND: Early detection of ovarian cancer remains a challenge due to widespread metastases and a lack of biomarkers for early-stage disease. This study was conducted to identify relevant biomarkers for both laparoscopic and serum diagnostics in ovarian cancer.
METHODS: Bioinformatics analysis and expression screening in ovarian cancer cell lines were employed. Selected biomarkers were further validated in bio-specimens of diverse cancer types and ovarian cancer subtypes. For non-invasive detection, biomarker proteins were evaluated in serum samples from ovarian cancer patients.
RESULTS: Two kallikrein (KLK) serine protease family members (KLK6 and KLK7) were found to be significantly overexpressed relative to normal …

Generalized arterial calcification of infancy (GACI), an autosomal recessive disorder caused by mutations in the ENPP1 gene, manifests with extensive mineralization of the cardiovascular system. The affected individuals in most cases die within the first year of life, and there is currently no effective treatment for this disorder. In this study, we characterized a spontaneous mutant mouse, asj-2J, as a model for GACI. These mice were identified as part of a phenotypic deviant search in a large-scale production colony of BALB/cJ mice at The Jackson Laboratory. They demonstrated a characteristic gait due to stiffening of the joints, with phenotypic similarity …

A Descriptive Mixed-Methods Study Examining Resilience And Transitioning To Adulthood Among Emerging Adults With Disabilities, Jennifer E. Mannino
Theses & Dissertations
Transitioning to adulthood is not without challenges. The move away from family influence towards independence and self-determination is filled with uncertainty. As emerging adults (18-29) transition to adulthood they will encounter various challenges related to their new adult roles (Arnett & Tanner, 2005; Jensen & Arnett, 2012). Increased challenges and vulnerabilities in transitioning are evident among emerging adults with disabilities because they face additional challenges related to their disability over and above what all others of this developmental stage experience (Betz & Redcay, 2002; Blomquist, 2007; Faux & Nehring, 2010; King, Baldwin, Currie & Evans, 2005; Murray, 2003; National Research …

Status, Distribution, And Conservation Of Scripps's Murrelet At Santa Catalina Island, California, Darrell L. Whitworth, Harry R. Carter, Tyler M. Dvorak, Linda S. Farley, Julie L. King
Monographs of the Western North American Naturalist
The small population of Scripps’s Murrelets (Synthliboramphus scrippsi) at Santa Catalina Island, California, has been restricted for at least several millennia to isolated pairs nesting in cliff and shoreline habitats that are mostly inaccessible to island fox (Urocyon littoralis catalinae). Prior to 1994, the only evidence of murrelets breeding at Catalina was a single nest reportedly found on Bird Rock in 1967. In 1994–1995, a larger and more widespread population estimated at 25–75 pairs was indicated through vocal detection surveys of murrelets attending nocturnal at-sea congregations in nearshore waters near breeding areas. Murrelets were heard at …
Evaluating Management Strategies For Urban Stormwater Runoff, Corrina M. Fleischmann
Evaluating Management Strategies For Urban Stormwater Runoff, Corrina M. Fleischmann
Doctoral Dissertations
Urban stormwater runoff, a leading cause of waterway impairment, has become a focal point of urban stormwater management strategies. As urbanization increases, regulations demanding preservation of pre-development peak flow rates or runoff volumes have been implemented and low impact development (LID) is encouraged as a strategy to achieve this stormwater runoff reduction requirement. While the success of LID has been proven at the site-scale, limited watershed-scale assessment of LID has been conducted. This research explores the potential benefit of watershed-scale LID implementation on two common urban stormwater issues: degraded stream health and combined sewer overflows (CSOs). Assessment results indicate that …

Since the early 2000s, educators, administrators, politicians, and researchers have given increased attention to the potential affordances of video games for educating K-12 and university learners. This has led to the instantiation of numerous game-based learning (GBL) journals, investigations of efficacy, achievement, and motivation using multimillion dollar tools, and a federally-funded competition for science, technology, engineering, and mathematics game development. Yet, little is known about the way particular game mechanics, narrative structures, and community-driven tools (e.g., forums, cheat guides, mods) influence the skills needed to be a successful 21st century learner. Few studies have catalogued the particular actions and …
