Dartmouth Scholarship

The TLO genes are a family of telomere-associated ORFs in the fungal pathogens Candida albicans and C. dubliniensis that encode a subunit of the Mediator complex with homology to Med2. The more virulent pathogen C. albicans has 15 copies of the gene whereas the less pathogenic species C. dubliniensis has only two (CdTLO1 and CdTLO2). In this study we used C. dubliniensis as a model to investigate the role of TLO genes in regulating virulence and also to determine whether TLO paralogs have evolved to regulate distinct functions. A C. dubliniensis tlo1Δ/tlo2Δ mutant is unable …

A Decision Support Model For Personalized Cancer Treatment, Florentino Antonio Rico-Fontalvo Oct 2014

A Decision Support Model For Personalized Cancer Treatment, Florentino Antonio Rico-Fontalvo

USF Tampa Graduate Theses and Dissertations

This work is motivated by the need of providing patients with a decision support system that facilitates the selection of the most appropriate treatment strategy in cancer treatment. Treatment options are currently subject to predetermined clinical pathways and medical expertise, but generally, do not consider the individual patient characteristics or preferences. Although genomic patient data are available, this information is rarely used in the clinical setting for real-life patient care. In the area of personalized medicine, the advancement in the fundamental understanding of cancer biology and clinical oncology can promote the prevention, detection, and treatment of cancer diseases.

The objectives …

Quantitative Real-time PCR (qRT-PCR) is a powerful technique to investigate comparative gene expression. In general, normalization of results using a highly stable housekeeping gene (HKG) as an internal control is recommended and necessary. However, there are several reports suggesting that regulation of some HKGs is affected by different conditions. The western corn rootworm (WCR), Diabrotica virgifera virgifera LeConte (Coleoptera: Chrysomelidae), is a serious pest of corn in the United States and Europe. The expression profile of target genes related to insecticide exposure, resistance, and RNA interference has become an important experimental technique for study of western corn rootworms; however, lack …

Reproducible Protein Nmr Data Analysis, Matthew Fenwick Oct 2014

Reproducible Protein Nmr Data Analysis, Matthew Fenwick

Doctoral Dissertations

Nuclear Magnetic Resonance (NMR) spectroscopy is a technique for studying biological molecules such as proteins at the atomic level. The information obtained from NMR is used to identify binding partners, locate active sites and binding pockets, and obtain structural and dynamics information which can be used in drug design. During the analysis process, large amounts of data and meta data are generated. However, much of this is not recorded and thus does not show up in archives such as the Biological Magnetic Resonance Data Bank (BMRB). This raises serious reproducibility concerns, since the data and meta data describing how the …

Wastewater Nutrient Recovery Using Anaerobic Membrane Bioreactor Permeate For Hydroponic Fertigation, Jorge Luis Calabria Oct 2014

Wastewater Nutrient Recovery Using Anaerobic Membrane Bioreactor Permeate For Hydroponic Fertigation, Jorge Luis Calabria

USF Tampa Graduate Theses and Dissertations

The imbalance between global population growth and resource consumption is indicative of unsustainable practices and foreshadows a grim future of continued resource depletion, food and water scarcity, social inequality, and deteriorating public and environmental health. Meanwhile, the urban centers of the world continue to experience exponential growth resulting in overwhelmed food, water, and sanitation infrastructure. Decentralized and satellite wastewater treatment technologies capable of resource recovery, such as anaerobic membrane bioreactors (AnMBR), foster synergistic opportunities to help manage the food, energy, and water sectors of urban environments. Specifically, the nutrient concentration and high effluent quality of permeate produced by AnMBR systems …

The Influence Of Social Environment And Hypercholesterolemia On Sympathetic Nervous System Innervation Of Vascular Tissue In Rabbits, Crystal M. Noller

Open Access Dissertations

Behavioral and psychosocial factors have been shown to influence cardiovascular disease. While interventions targeting these risk factors demonstrate clinical improvement, mechanisms underlying these effects remain to be determined. Research has also defined a relationship between psychosocial stress and immune function, and revealed stress related increases in lymphatic sympathetic nerve density. Considering that inflammation characterizes the various stages of heart disease, the current study assessed whether social stress could influence vascular sympathetic innervation in the presence or absence of hypercholesterolemia. We found dense sympathetic innervation extending into the vascular media and intima throughout the aortic arch and thorax in diseased as …

The soil organic carbon (SOC) model that we used was parameterized with data from arable land under normal farming conditions in North America, Europe, Africa and Asia, but the equation is insensitive to changes in tillage, soil texture and moisture. The model has reasonable accuracy, however, in predicting changes in SOC, residue remaining and CO2 emissions from initial SOC, carbon inputs from residue, and daily temperature; the shoot-to-root ratio used in the geospatial simulation was 0.29 (that is, root carbon is 29% of total aboveground carbon), which did not underestimate carbon input to soil (Supplementary Figure 2 in Ref. …

Lipophilic cationic technetium-99m-complexes are widely used for myocardial perfusion imaging (MPI). However, inherent uncertainties in the supply chain of molybdenum-99, the parent isotope required for manufacturing 99Mo/99mTc generators, intensifies the need for discovery of novel MPI agents incorporating alternative radionuclides. Recently, germanium/gallium (Ge/Ga) generators capable of producing high quality 68Ga, an isotope with excellent emission characteristics for clinical PET imaging, have emerged. Herein, we report a novel 68Ga-complex identified through mechanism-based cell screening that holds promise as a generator-produced radiopharmaceutical for PET MPI.

  1. The relationship between abiotic stress and fitness in an individual is usually described by the intensity and duration of stress. Yet in natural systems, variability in abiotic stress is common. Since individuals have physiological and fitness responses to single bouts of stress, frequency of stress may also determine the lifetime success of an organism. However, the majority of laboratory studies have focused only on the effects of single stress events.
  2. We investigated the relative importance of stress parameters including duration, intensity and number of cold events on the short-term physiology and long-term fitness in the freeze-avoiding eastern spruce budworm Choristoneura …

At its best, a university is a collection of individuals ‐‐ students and faculty ‐‐ focused on learning and discovering new knowledge. For this goal to be realized, a critical element is having faculty members deeply engaged in such discovery within their disciplines. Scholarship, in the form of journal articles, book chapters, monographs and similar endeavors, creative activity which can take an even wider range of forms, and funded research which explores the boundaries of disciplines all contribute to such engagement. Through such participation, faculty members stay at the growing edges of their fields, and in so doing, they enrich …

Scientists are increasingly engaging the web to provide formal and informal science education opportunities. Despite the prolific growth of web-based resources, systematic evaluation and assessment of their efficacy remains limited. We used clickstream analytics, a widely available method for tracking website visitors and their behavior, to evaluate 60,000 visits over three years to an educational website focused on ecology. Visits originating from search engine queries were a small proportion of the traffic, suggesting the need to actively promote websites to drive visitation. However, the number of visits referred to the website per social media post varied depending on the social …
