Design And Synthesis Of Pyrimidine Fused Heterocycles As Single Agents With Combination Chemotherapy Potential, Roheeth Pavana

This dissertation describes the design, synthesis and biological evaluation of pyrimidine fused heterocycles as single agents with combination chemotherapy potential. Major limitations of cancer chemotherapy include dose limiting toxicities of clinically used agents and the development of multidrug resistance by the tumor. Agents that interfere with microtubules are important antitumor agents. Tumor angiogenic mechanisms that are vital for tumor growth and metastasis are targeted by antiangiogenic agents. Antiangiogenic agents are usually not tumoricidal but are mainly cytostatic. Combination chemotherapy with antiangiogenic and cytotoxic agents have shown significant promise and several studies with such combinations are in progress in the clinic. …

The coordination chemistry of dithiolene complexes has attracted enormous interest by inorganic chemists due to their presence in biological systems and possible applications in material science. Dithiolene units have been identified at the active site of metalloenzymes, containing molybdenum and tungsten. Model complexes have played a significant role in understanding the structural and reactivity of the active site of these enzymes.

Numerous in vitro and in vivo studies implicate that certain ligands that interact with DAT, nAChRs, and mGlu2 have tremendous potential as anti-addiction therapeutics. However, understanding how these promising anti-addiction compounds interact with their major drug targets at the molecular level is limited because of the absence of human DAT, nAChRs, and mGlu2 x-ray crystal structures. The United States is undergoing a period in which the demographics of the population are shifting drastically. The profession of counseling is tasked with providing services to diverse groups of clients, many of whom may have experienced racism, prejudice, and oppression as a result of their cultural or ethnic identity. Numerous behavioral studies indicate that dopamine transporter (DAT) inhibitors and metabotropic glutamate receptor 5 (mGlu5) negative allosteric modulators (NAMs) possess promising anti-addiction therapeutic properties. Obesity is a sensitive, global health problem that impacts individuals of all ages and places individuals at risk for various chronic health problems. Nurses caring for hospitalized obese clients are in a prime position to provide them with timely information on weight management. This study utilized an exploratory sequential mixed method design to examine medical-surgical nurses' intentions to provide weight management education to hospitalized obese adults and the factors that influence nurses' intentions.

This portraiture project, incorporating auto-ethnographic elements, investigating how African American educational systems became systems of oppression faced by lesbian, gay, bisexual, transgender and questioning (LGBTQ) youth of color, intentionally blends artist in creative glance with researcher in systemic inquiry and knowledge gathering (Lawrence-Lightfoot & Hoffman-Davis, 1997; Ellis et al., 2011).

Marx W. Wartofsky was born in Brooklyn and received his bachelor’s, master’s, and doctoral degrees at Columbia University. He was a professor at Boston University (where he taught for twenty-six years) and then at Baruch College and the Graduate Center of the City University of New York. He was long-time editor of the Philosophical Forum, which he founded in 1970. He also co-founded with Robert Cohen the Boston University Center for Philosophy and History of Science in 1960. Objective. To investigate the joint effects of the single nucleotide polymorphisms (SNPs) of genes in the folic acid pathway on homocysteine (Hcy) metabolism. Methods. Four hundred women with normal pregnancies were enrolled in this study. SNPs were identified by MassARRAY. Serum folic acid and Hcy concentration were measured. Analysis of variance (ANOVA) and support vector machine (SVM) regressions were used to analyze the joint effects of SNPs on the Hcy level. Results. Sequential Activation Of Elk-1/Egr-1/Gadd45Α By Arsenic, Qiwen Shi, Vijaykumar Sutariya, Anupam Bishayee, Deepak Bhatia

Long-term exposure to arsenic, an environmental contaminant, leads to increased risks of cancers. In the present study, we investigated the sequential regulation of Elk-1 and Egr-1 on As3+-induced GADD45α, an effector of G2/M checkpoint. We found that As3+ transcriptionally induced both Elk-1 and Egr-1, and NF-κB binding site was necessary for As3+-induced Egr-1 promoter activity. However, specific inhibition of JNK, ERK, and Elk-1 inhibited Egr-1 induction. Furthermore, silencing of Egr-1 downregulated As3+-induced expression of GADD45α and ChIP assay confirmed the direct binding of Egr-1 to GADD45α promoter. Dysregulation of miRNAs is involved in osteosarcoma (OS). Here, we demonstrate that miR-382 is decreased in specimens of OS patients with a poor chemoresponse compared to those with a good chemoresponse. In addition, our clinical data show that decreased miR-382 was associated with poor survival in OS patients. Overexpression of miR-382 inhibited cell growth and chemoresistance by targeting KLF12 and HIPK3, respectively. In contrast, inhibition of miR-382 or overexpression of target genes stimulated OS cell growth and chemoresistance both in vitro and in vivo. Optimizing anticancer therapeutics needs to account for variable drug responses in heterogeneous cell populations within the tumor as well as in organs of toxicity. To address cell heterogeneity, we propose a multiscale modeling approach—from in vitro to preclinical and clinical studies—to develop cell-type–specific pharmacokinetic–pharmacodynamic (PK-PD) models. A physiologically based mechanistic modeling approach integrating data from aqueous solutions, U87 glioma cells, mice, and cancer patients was utilized to characterize the brain disposition of temozolomide (TMZ), the cornerstone of chemotherapy against glioblastoma multiforme. Assessing The Impact Of Parental Depressive Symptoms On Offspring Temperament And Development In Infancy, Nancy Huynh, Jackie Finik, Jenny Ly, Yoko Nomura

The study prospectively followed 135 women during their pregnancy and their offspring till 6 months of age, to examine the roles of maternal and paternal depression during pregnancy on offspring neurobehavioral development as measured by their early temperament. Maternal and paternal depression statuses were ascertained during the third trimester, and infant temperament was evaluated at 6 months, via mothers self-report. Multivariable general linear model was used to assess 1) the main effects of maternal and paternal depression on infant temperament and 2) the interaction effect between maternal and paternal depression on infant temperament. Background. In stressful situations, decision making processes related to informed consent may be compromised. Given the profound levels of distress that surrogates of children in pediatric intensive care units (PICU) experience, it is important to understand what factors may be influencing the decision making process beyond the informed consent. The purpose of this study was to evaluate the role of clinician influence and other factors on decision making regarding participation in a randomized clinical trial (RCT). Method.
