United States Department of Agriculture-Agricultural Research Service / University of Nebraska-Lincoln: Faculty Publications

Macrophage migration inhibitory factor (MIF) of Ostertagia ostertagi, an abomasal parasite of cattle, was characterised in the present study. Phylogenetic analysis identified at least three O. ostertagi MIFs (Oos- MIFs), each encoded by a distinct transcript: Oos-MIF-1.1, Oos-MIF-1.2 and Oos-MIF-2. Oos-MIF-2 is only distantly related to Oos-MIF-1s, but has higher sequence similarity with the Caenorhabditis elegans MIF2. Oos-MIF-1.1 and Oos-MIF-1.2 are similar (93%) and thus collectively referred to as Oos-MIF-1 when characterised with immunoassays. Recombinant Oos-MIF-1.1 (rOos-MIF-1.1) is catalytically active as a tautomerase. A mutation (rOos-MIF-1.1P1G) or duplication of Pro1 residue (rOos-MIF-1.1P1+P) resulted in reduced oligomerisation and loss of tautomerase …

The Molecular Biology And Treatment Of Malignant Melanoma With Brafv600 Mutations, Michael P. Mullane Feb 2014

The Molecular Biology And Treatment Of Malignant Melanoma With Brafv600 Mutations, Michael P. Mullane

Journal of Patient-Centered Research and Reviews

Since 2011, the treatment options for metastatic malignant melanoma have significantly changed. In that year, ipilimumab, an anti-CTLA4 monoclonal antibody, and vemurafenib, a potent inhibitor of mutated-BRAF (V600E and V600K), were approved by the U.S. Food and Drug Administration (FDA). In 2013, dabrafenib, another inhibitor of mutated-BRAF, and trametinib, a MEK inhibitor, were approved by the FDA. Most recently, combination therapy with dabrafenib and trametinib was approved. This article will describe a patient with metastatic malignant melanoma with BRAFV600E who has responded very well to vemurafenib monotherapy. We will then explore the molecular basis, pharmacologic development and clinical outcomes …

Trans-Substantivity And The Processes Of American Law, David Marcus Feb 2014

Trans-Substantivity And The Processes Of American Law, David Marcus

BYU Law Review

The term “trans-substantive” refers to doctrine that, in form and manner of application, does not vary from one substantive context to the next. Trans-substantivity has long influenced the design of the law of civil procedure, and whether the principle should continue to do so has prompted a lot of debate among scholars. But this focus on civil procedure is too narrow. Doctrines that regulate all the processes of American law, from civil litigation to public administration, often hew to a trans-substantive norm. This Article draws upon administrative law, the doctrine of statutory interpretation, and the law of civil procedure to …

Ocean Acidification Impairs Vermetid Reef Recruitment., Marco Milazzo, Riccardo Rodolfo-Metalpa, Vera Bin San Chan, Maoz Fine, Cinzia Alessi, Vengatesen Thiyagarajan, Jason M. Hall-Spencer, Renato Chemello Feb 2014

Ocean Acidification Impairs Vermetid Reef Recruitment., Marco Milazzo, Riccardo Rodolfo-Metalpa, Vera Bin San Chan, Maoz Fine, Cinzia Alessi, Vengatesen Thiyagarajan, Jason M. Hall-Spencer, Renato Chemello

School of Biological and Marine Sciences

Vermetids form reefs in sub-tropical and warm-temperate waters that protect coasts from erosion, regulate sediment transport and accumulation, serve as carbon sinks and provide habitat for other species. The gastropods that form these reefs brood encapsulated larvae; they are threatened by rapid environmental changes since their ability to disperse is very limited. We used transplant experiments along a natural CO2 gradient to assess ocean acidification effects on the reef-building gastropod Dendropoma petraeum. We found that although D. petraeum were able to reproduce and brood at elevated levels of CO2, recruitment success was adversely affected. Long-term exposure to acidified conditions predicted …

An Updated And Annotated Checklist Of The Larger Butterflies (Papilionoidea) Of Trinidad, West Indies: Papilionidae, Pieridae And Nymphalidae, Matthew J. W. Cock

Insecta Mundi

A revised checklist for the butterfly families Papilionidae, Pieridae and Nymphalidae of Trinidad (Trinidad and Tobago) is presented, bringing nomenclature in line with modern usage, indicating synonyms from earlier lists and adding new records since the last checklist was published in 1970. Migrant and vagrant species are provisionally recognised, and records considered incorrect are discussed. The checklist includes 204 species: 15 Papilionidae, 29 Pieridae and 160 Nymphalidae. The only taxonomic change is to treat Hamadryas feronia insularis (Fruhstorfer) as a synonym of H. f. feronia (Linnaeus), syn. nov., and not as a synonym of H. feronia farinulenta (Fruhstorfer).

Department of Medicine Faculty Papers

RATIONALE: Anti-inflammatory and vascular protective actions of adiponectin are well recognized. However, many fundamental questions remain unanswered.

OBJECTIVE: The current study attempted to identify the adiponectin receptor subtype responsible for adiponectin's vascular protective action and investigate the role of ceramidase activation in adiponectin anti-inflammatory signaling.

METHODS AND RESULTS: Adiponectin significantly reduced tumor necrosis factor (TNF)α-induced intercellular adhesion molecule-1 expression and attenuated TNFα-induced oxidative/nitrative stress in human umbilical vein endothelial cells. These anti-inflammatory actions were virtually abolished by adiponectin receptor 1 (AdipoR1-), but not AdipoR2-, knockdown (KD). Treatment with adiponectin significantly increased neutral ceramidase (nCDase) activity (3.7-fold; P87% of adiponectin-induced nCDase …
