Preventive Effect Of Non-Mitogenic Acidic Fibroblast Growth Factor On Diabetes-Induced Testicular Cell Death., Melissa Skibba 1990- May 2014

Preventive Effect Of Non-Mitogenic Acidic Fibroblast Growth Factor On Diabetes-Induced Testicular Cell Death., Melissa Skibba 1990-

Electronic Theses and Dissertations

Fibroblast growth factor (FGF)-1 was found to protect the heart from oxidative damage, but clinically its long-term use was restricted for its undesirable proliferating activity on cells. Thus a cluster of amino acids responsible for the proliferation were deleted in the native FGF-1 to create a non-mitogenic FGF-1 (nmFGF-1). To assess whether nmFGF-1 protects male germ cells from diabetes-induced apoptotic death, the study employed a murine model of diabetes using multiple low-doses of streptozotocin, followed by nmFGF-1 treatment for 6 months. Diabetic mice showed a decrease in testicular weight and an increase in apoptotic cell death. Treatment with nmFGF-1 alleviated …

Inflammation-Resolving Lipid Mediators Promote Revascularization To Enhance Wound Healing., Michael Jingyuan Zhang May 2014

Inflammation-Resolving Lipid Mediators Promote Revascularization To Enhance Wound Healing., Michael Jingyuan Zhang

Electronic Theses and Dissertations

Wound healing is a highly concerted cellular process that begins with inflammation and proceeds to resolution to revascularize the site of injury. Although inflammation is essential to revascularization during wound healing, it is now recognized that resolution is an active process that is equally important. Other investigations have implicated a beneficial effect of resolving inflammation and promoting resolution in the remission of inflammatory pathologies. Recently investigations have yielded a novel class of ?-3 fatty acid derived lipid mediators, biosynthesized by leukocytes, which are capable of resolving inflammation and promoting resolution. We therefore hypothesized that these leukocyte-derived pro-resolving lipid mediators can …

Epigenetic Modifications Associated With Aortic Remodeling In Hyperhomocysteinemia., Nithya Narayanan May 2014

Epigenetic Modifications Associated With Aortic Remodeling In Hyperhomocysteinemia., Nithya Narayanan

Electronic Theses and Dissertations

Background: Hyperhomocysteinemia (HHcy) is prevalent in hypertensive patients and is an independent risk factor for aortic pathologies. HHcy is known to cause an imbalance between matrix metalloproteinases (MMPs) and tissue inhibitors of metalloproteinases (TIMPs) leading to accumulation of collagen in the aorta resulting in stiffness and development of hypertension. Although the exact mechanism of extracellular matrix remodeling (ECM) is unclear, emerging evidence implicates epigenetic regulation involving DNA methylation. The purpose of the study was to investigate whether inhibition of DNA methylation reduces high blood pressure by regulating aortic ECM remodeling in HHcy. Methods: In the first set of experiments, we …

Markers And Mechanisms Of Metabolic Aging., Peter Dziewornu Ahiawodzi May 2014

Markers And Mechanisms Of Metabolic Aging., Peter Dziewornu Ahiawodzi

Electronic Theses and Dissertations

Oxidative stress has been implicated in aging and age-related diseases. Using three distinct studies, this dissertation examines associations between lifestyle factors and oxidative stress, and their effects on advanced glycation end products, telomere length, and type II diabetes. Carboxymethyl-lysine (CML), an advanced glycation end-product, has been linked to oxidative stress, cardiovascular and other age-related diseases. The first study investigates the association between sleep disordered breathing (SDB), a source of oxidative stress, and CML. 1002 participants with sleep data from the Cardiovascular Health Study (CHS) were analyzed. SDB was not significantly associated with CML, but after stratification by gender and hemoglobin …

The Role Of Pesticides In Non-Alcoholic Fatty Liver Disease (Nafld)., Laila Al-Eryani 1986- May 2014

The Role Of Pesticides In Non-Alcoholic Fatty Liver Disease (Nafld)., Laila Al-Eryani 1986-

Electronic Theses and Dissertations

NAFLD, the most common form of liver disease worldwide, is caused by various factors including industrial chemicals and pesticides exposure. Annually, 5.2 billion pounds of pesticides are used worldwide and can contribute to liver disease, but their role is modestly studied. We hypothesize that pesticides contaminating food supply can worsen diet-induced steatosis via xenobiotic receptor activation. Two human and two rodent databases were utilized and 85% of the 330 chemicals identified associated with NAFLD were pesticides. Eight were selected for evaluating hepatic receptor activation in vitro. The majority including DDT activated hPXR/CAR and mPXR. DDT (100 mg/kg) was studied in …

Neutrophils Alter Epithelial Response To Porphyromonas Gingivalis In A Gingival Crevice Model., Jessica Lynn Bondy-Carey 1987- May 2014

Neutrophils Alter Epithelial Response To Porphyromonas Gingivalis In A Gingival Crevice Model., Jessica Lynn Bondy-Carey 1987-

Electronic Theses and Dissertations

A gingival crevice model (epithelial cell-Porphyromonas gingivalis-neutrophil) was established and used to profile gingipain, matrix metalloproteinase (MMP), MMP mediators [neutrophil gelatinase-associated lipocalin (NGAL) and tissue inhibitor of metalloproteinases 1 (TIMP-1)] and cytokine networks. Smoking is the primary environmental risk factor for periodontitis. Therefore, the influence of cigarette smoke extract (CSE) was also monitored in the same model. Porphyromonas gingivalis alone induced low levels of interleukin-1b and interleukin-8 from epithelial cells, but high levels of both cytokines were produced on the addition of neutrophils. Exposure to CSE (100 and 1000 ng mL1 nicotine equivalency) significantly compromised P. gingivalis-induced cytokine secretion (both …

Effects Of Distillers Grains Plus Solubles And Monensin Supplementation On Yearlings Grazing Smooth Bromegrass, Tyler L. Hasenauer

Department of Animal Science: Dissertations, Theses, and Student Research

Supplementing cattle on grass is an effective way of increasing animal efficiency and grass utilization. Distiller grains plus solubles (DGS) supplementation has been repeatedly proven as an effective supplement by providing ruminally undegradable protein (RUP), fat, and highly digestible fiber. The effects of monensin supplementation on grazing cattle are variable. Although some research has shown a decrease in forage organic matter intake (FOMI), the popular belief is monensin increases ADG while maintaining DMI in a grazing situation. Two experiments were designed to observe the effects of DGS and monensin supplementation on cattle grazing smooth bromegrass. In the first study, ADG …

Genomic Predictions For Age At Puberty And Reproductive Longevity In Sows Using Bayesian Methods, Katherine Lindsay Lucot May 2014

Genomic Predictions For Age At Puberty And Reproductive Longevity In Sows Using Bayesian Methods, Katherine Lindsay Lucot

Department of Animal Science: Dissertations, Theses, and Student Research

Including marker-assisted selection in breeding programs is potentially more efficient than traditional selection for improving traits that are expensive or difficult to measure. One of the challenges of genomics is the lack of robustness of marker effects across populations and over time (generations) and the cost to commercial producers of high-density arrays. The objective of this study was to analyze differences in the proportion of phenotypic variation explained by different fractions of major 1 Mb windows and SNPs. Using a population of Nebraska Index Line and commercial Large White x Landrace females (n = 1,234) generated in 11 batches, …

An Examination Of The Feasiblity Of A Food Hub For The Pee Dee Region, Emily Purcell

All Theses

Horry County, the home of Myrtle Beach, South Carolina, attracts a large level of spending by visitors who are helping to fuel growing demand for locally produced food. Regional growers are interested in meeting this demand but are limited by retail requirements with respect to lot size, timing, and quality (including food safety aspects such as Good Agricultural Practices (GAP) certification). A number of communities have turned to food hubs as a means of aggregating production by local growers and providing appropriate marketing functions, such as storage and meeting standards. Hence, food hubs can fill a gap between producers and …

Comprehensive Cancer Care: An Outpatient Treatment Facility, Braden Abrams Reid May 2014

Comprehensive Cancer Care: An Outpatient Treatment Facility, Braden Abrams Reid

All Theses

A cancer diagnosis can result in a loss of decision-making and control for patients. The majority of clinical treatments fail to adequately address corollary symptoms and needs that arise both from treatment and from the disease itself. Thus, the architectural problem identified in this thesis inquiry is how can an outpatient cancer centre treat the whole patient comprehensively, while at the same time instilling control and re-empowering the patient? This thesis project proposes a patient-centered environment designed to support the myriad and nuanced needs of the cancer patient, as well as a site-specific proposal for a comprehensive outpatient cancer treatment …

An Evaluation Of The East Indianapolis Food Desert, Angelina Qin May 2014

An Evaluation Of The East Indianapolis Food Desert, Angelina Qin

Undergraduate Honors Thesis Collection

The Indy East Food Desert (IEFD) appears to suffer from many common conditions noted by various scholars as food desert identifiers. Yet, its situation still remains unique. Though there is a prevalence of minority populations, low-income households, and low-education attainment levels, there are also factors of low-access, poor food options (within the limited food outlets in the neighborhoods), and poor eating habits which shape the situation of the community. In an attempt to alleviate the food desert problems, the Indy East Food Desert Coalition (IEFDC) was formed. IEFDC partnered with Butler University's Center for Urban Ecology in order to assess …

Publications

Unlike other important vectors such as mosquitoes and sandflies, genetic and genomic tools for Culicoides biting midges are lacking, despite the fact that they vector a large number of arboviruses and other pathogens impacting humans and domestic animals world-wide. In North America, female Culicoides sonorensis midges are important vectors of bluetongue virus (BTV) and epizootic hemorrhagic disease virus (EHDV), orbiviruses that cause significant disease in livestock and wildlife. Libraries of tissue-specific transcripts expressed in response to feeding and oral orbivirus challenge in C. sonorensis have previously been reported, but extensive genome-wide expression profiling in the midge has not. Here, we …

Lowering Somatic Cell Count With Best Management Practices, Stephen F. Higgins, Kylie Schmidt, Sarah Wightman May 2014

Lowering Somatic Cell Count With Best Management Practices, Stephen F. Higgins, Kylie Schmidt, Sarah Wightman

Agriculture and Natural Resources Publications

As health and food safety concerns grow, dairy producers are facing more stringent regulations. In 2010, the European Union (EU) set the somatic cell count (SCC) upper limit, an indicator of milk quality, for exported milk at 400,000 cells per milliliter. However, the current U.S. SCC limit is 750,000 cells per milliliter. As of January 2012, any U.S. milk used in export markets must meet the EU standards. It is projected that US milk processors will gradually adopt the EU upper limit, making it difficult for dairy producers to sell milk containing more than 400,000 somatic cells per milliliter. Dairy …
