UF Law Faculty Publications

This Article proposes a novel approach to addressing global climate change's impacts on agricultural production and food security. The climate change crisis is the most significant environmental issue facing our planet. The changes predicted to occur as the earth's climate warms include significant impacts to agriculture. At the same time that the planet is undergoing dramatic climatic changes, the global population is increasing, and economic development in many parts of the world is exerting increased demand for a greater and more diverse supply of food.

The relationship between climate change and agriculture is a close and complex one, as the …

Physical And Chemical Attributes Of A Genetically Modified Fruit Pectin, Carl Patrick Littrell Jul 2014

Physical And Chemical Attributes Of A Genetically Modified Fruit Pectin, Carl Patrick Littrell

Open Access Theses

Pectin is an important polymer used in the food industry as a thickening and gelling agent. Though pectin is ubiquitous in plants, chemical and structural differences among pectin molecules prevent most from being viable for industrial use. Enzymes found naturally in fruit cell walls during the ripening process impair many desirable attributes of fruit pectins, rendering them unsuitable for industrial applications. Pectin methylesterase (PME) is one such enzyme whose expression can be altered during ripening through the use of recombinant genetic engineering. Reduction in levels of PME results in increased degree of methylation and molecular size of pectin, greatly increasing …

Management Of Root Knot Nematode (Meloidogyne Incognita ) In Indiana Soybeans, David Edgardo Perla Martinez

Open Access Theses

The aim of this project was to evaluate different strategies for management of Root Knot Nematode (RKN) on soybean and tomato in Indiana. Seed treatments were evaluated under field and greenhouse conditions, but no effect on RKN populations was observed. Soybean lines evaluated for resistance to RKN under greenhouse conditions showed that six lines may be resistant to RKN. Four different commercial mustard cover crops were evaluated for their bio-fumigant impact on RKN populations in the production of tomato. Euruca sativa, Cv. Nemat was a poor host of RKN. A positive impact on the vigor of the tomato plants followed …

Ems Induced Mutations In Dhurrin Metabolism And Their Impacts On Sorghum Growth And Development, Jenae Lavon Skelton Jul 2014

Ems Induced Mutations In Dhurrin Metabolism And Their Impacts On Sorghum Growth And Development, Jenae Lavon Skelton

Open Access Theses

Sorghum is the fifth most important grain crop in the world. It is a staple food, feed, and silage crop in many developing countries in the semi-arid tropics. One factor that can impact sorghum forage quality is dhurrin content. Dhurrin is a cyanogenic glucoside naturally produced in the plant. When tissues containing dhurrin are crushed, hydrogen cyanide (HCN) is released during dhurrin decomposition. HCN is toxic to humans and livestock. While there is natural genetic diversity for the concentration of dhurrin within sorghum lines, there have been no naturally occurring dhurrin-free genotypes identified to date. ^ We have identified ethyl …

Fact Or Fiction: Random Mating In Field Populations Of Western Corn Rootworm (Diabrotica Virgifera Virgifera Leconte) Emerging On Bt And Refuge Corn Plants, Steven Joel Smith

Open Access Theses

The western corn rootworm, or WCR, (Diabrotica virgifera virgifera LeConte) is the most significant pest of field corn (Zea mays) in the United States, and has recently expanded its range into Europe. Since 2004, hybrid corn containing Bt toxins targeting the corn rootworm complex have been heavily adopted and are now the primary control measure for this pest in North American corn production. ^ The evolution of resistance is an ongoing concern, and to ensure Bt products will retain their usefulness, insect resistance management (IRM) tactics using various refuge structures have been adopted. One of the key …

The Effectiveness Of Virtual And On-Site Dairy Farm Field Trips To Increase Student Knowledge In Science, Social Studies, And Health And Wellness Standards, Abby M. Sweet

Open Access Theses

The integration of agriculture into the curriculum can be difficult for educators when materials and resources are not aligned with state learning standards. However, online educational tools, such as virtual field trips that are aligned with state learning standards, can allow for educators to bring the experience of a field trip to the classroom, as well as complement curriculum in the classroom. Virtual field trips also assist with removing many barriers that prevent educators from taking their students on an on-site field trip. ^ This study sought to describe the effectiveness of a virtual and on-site dairy farm field trip …

Iron Nutriture Following Roux-En-Y Gastric Bypass Surgery, Breanne N. Wright Jul 2014

Iron Nutriture Following Roux-En-Y Gastric Bypass Surgery, Breanne N. Wright

Open Access Theses

Roux-en-Y gastric bypass surgery (RYGB) is effective for weight loss, but is commonly associated with iron deficiency and its clinical manifestation, anemia. Diagnosing iron deficiency is complex because iron status depends on other nutrients; additionally, anemia following surgery is not specifically due to deficiency in iron, as it can be due to deficiencies in other nutrients including zinc, copper, vitamin B6, folate, and vitamin B 12. In patients who have undergone RYGB, our aims were to 1) conduct a comprehensive assessment of nutrients involved in iron homeostasis, 2) determine the contribution of dietary intake to iron deficiency, …

1,25-Dihydroxyvitamin D Alters Lipid Metabolism And Epithelialto- Mesenchymal Transition In Metastatic Epithelial Breast Cancer Cells, Alle Nicole Barnard

Open Access Theses

Evidence suggests that high vitamin D status (marked by serum 25-hydroxyvitamin D, 25(OH)2 D) is associated with a decreased risk of breast cancer. It has been established that 1,25-dihydroxyvitamin D (1,25(OH) 2 D) can alter glycolysis and the Krebs cycle of breast cancer cells (Jiang et al., 2010; Zheng et al., 2013) but little information is available on 1,25(OH) 2 D's alterations of lipid metabolism in breast cancer cells. Thus, the current research investigates if there was an effect of 1,25(OH) 2 D on proteins that regulate lipid metabolism in MCF10A, MCF10A-ras, MCF10CA1h, and MCF10CA1a epithelial breast cancer cells. While …

Agronomic And Genetic Characterization Of Aluminum Tolerance In A Recombinant Inbred Population Of Sorghum [Sorghum Bicolor (L.) Moench], Amanda Christine Easterly

Open Access Theses

Acid soils are estimated to cover up to 30% of arable soils globally and lead to significant limitations on agricultural productivity, primarily through aluminum toxicity. In sorghum [Sorghum bicolor (L.) Moench], significant genetic variation exists for tolerance to phytotoxic species of aluminum; tolerance is conferred through the exudation of citrate at the root tip, binding aluminum in the soil rhizosphere. A gene in the multidrug and toxic compound extrusion (MATE) family is the primary tolerance locus in sorghum. The purpose of this research was to evaluate a recombinant inbred line (RIL) population of sorghum segregating for SbMATE to establish …

Dental Fluctuating Asymmetry As A Measure Of Environmental Stress In Nasca, Shawna L. Follis Jul 2014

Dental Fluctuating Asymmetry As A Measure Of Environmental Stress In Nasca, Shawna L. Follis

Open Access Theses

This thesis evaluates how environmental stressors affected three groups (Nasca, Loro, and Chakipampa) that lived in Nasca during the Early Intermediate Period (ca. A.D. 1-750) and the Middle Horizon (ca. A.D. 750-1000). Using fluctuating asymmetry analysis as a proxy for developmental instability, biological evidence is assessed for differential stress levels incurred by groups occupying the Peruvian south coast. This study found high levels of stress in the Middle Horizon, supporting the hypothesis that populations living in Nasca were unfavorably affected by Wari colonizers. However, stress was found to be highest among the Chakipampa. This is attributed to Wari imperialistic occupation …

Separation Of On-Column Labeled Model Proteins With Packed Capillary Electrophoresis, Yingxu Hao Jul 2014

Separation Of On-Column Labeled Model Proteins With Packed Capillary Electrophoresis, Yingxu Hao

Open Access Theses

Protein drugs are increasingly developed in the pharmaceutical company. Under the regulation of FDA, high purity of therapeutic proteins needs to be maintained. Before putting those drugs in the market, fast and efficient method is in need to achieve homogeneity. Traditionally, polyacrylamide gel electrophoresis (PAGE), capillary electrophoresis (CE), and size-exclusion chromatography (SEC) are used for the purification process. These methods have the disadvantages of low time and cost efficiency, and this quality assurance process has become the bottleneck of production. In our group, sub-micron silica colloidal particles with polyacrylamide layer on the surface are packed inside capillaries to increase the …

Biological Sciences Faculty Research and Publications

Writing assignments, including note taking and written recall, should enhance retention of knowledge, whereas analytical writing tasks with metacognitive aspects should enhance higher-order thinking. In this study, we assessed how certain writing-intensive “interventions,” such as written exam corrections and peer-reviewed writing assignments using Calibrated Peer Review and including a metacognitive component, improve student learning. We designed and tested the possible benefits of these approaches using control and experimental variables across and between our three-section introductory biology course. Based on assessment, students who corrected exam questions showed significant improvement on postexam assessment compared with their nonparticipating peers. Differences were also observed …

Environmental Management Of Threatening Government Public Policy, Jess Mikeska Jul 2014

Environmental Management Of Threatening Government Public Policy, Jess Mikeska

College of Business: Dissertations, Theses, and Student Research

This dissertation is the first to propose a test of environmental management theory through a reflective, managerial scale, and does so through a public policy lens. It matches 76 managerial respondents’ (17 firms) perceptions of environmental management with objective, secondary data ranging between 2002 and 2014 so as to reflect longitudinal changes in marketplace activities which influence end consumers (e.g., pricing, promotional activity). Such changes are artifacts of firms’ environmental management of threatening government public policy.

The findings of two studies, qualitative interviews based on a transcendental phenomenological design and an online survey matched to secondary marketplace data utilizing Hierarchical …

Using a non-experimental design, the researcher tested two hypotheses using a mixed factorial analysis of variance (ANOVA). The independent variables for the two hypotheses were change over time with two levels measured in school years (2008 versus 2009 versus 2010 versus 2011) and the two NSLA funding levels. The …

Functional Genomics Of Maize Endosperm Maturation And Protein Quality, Lingling Yuan Jul 2014

Functional Genomics Of Maize Endosperm Maturation And Protein Quality, Lingling Yuan

Department of Agronomy and Horticulture: Dissertations, Theses, and Student Research

Maize is one of the most important cereal crops and widely cultivated throughout the world. The study on maize kernel development including protein quality improvement is essential for removing dietary protein deficiency because of the lack of essential amino acids, especially lysine and tryptophan, in maize kernel. Quality Protein Maize (QPM) is a hard kernel variant of the high-lysine mutant, opaque-2. We created opaque QPM variants to identify opaque-2 modifier genes and to investigate deletion mutagenesis combined with Illumina sequencing as a maize functional genomics tool. A K0326Y-QPM deletion mutant, line 107, was null for the 27- and 50-kD …

Non-Digestible Oligosaccharides: Anti-Adherence And Other Biological Properties, Maria I. Quintero-Villegas Jul 2014

Non-Digestible Oligosaccharides: Anti-Adherence And Other Biological Properties, Maria I. Quintero-Villegas

Department of Food Science and Technology: Dissertations, Theses, and Student Research

Bacterial adherence is the first and one of the most important steps in bacterial pathogenesis. Adherence to host cell surfaces requires that the bacteria recognize specific receptors in the surface of epithelial cells. Therefore, agents that act as molecular decoys to interfere with adherence could be useful prophylactic treatments to prevent or mitigate the onset of infections. The goal of this research was to assess the ability of several food grade non-digestible oligosaccharides (NDOs) to act as molecular decoys and prevent bacterial adherence in vitro as well as in vivo. First, the antiadherence effect of specific species chitooligosaccharides (CHOS) …

Characterization Of Genetically Modified High Biomass Producing Tobacco Plant, Pankaj Singh Kuhar Jul 2014

Characterization Of Genetically Modified High Biomass Producing Tobacco Plant, Pankaj Singh Kuhar

Department of Agricultural and Biological Systems Engineering: Dissertations, Theses, and Student Research

Global warming and peak oil has clouded our energy security. In light of this situation, bioethanol as emerged as one of the most amenable solutions to the problem. However bioethanol has its own shortcomings and transgenics seem imperative to exploit its full potential. A high biomass producing line in transgenic tobacco (Nicotiana tabacum cv. Xanthi) was identified during a routine genetic transformation, termed giant recombinant (GR). To characterize the phenotype of the giant line, growth rate and lignocellulosic composition was analyzed relative to the non-transgenic control line. The GR line accounted for 240% more biomass than the untransformed line …
