Novel Serum Biomarkers For Lung Cancer Early Diagnosis And Clinical Outcome, Fanmao Zhang

Dissertations and Theses (Open Access)

The five-year survival rate for all stages of lung cancer combined is only 17%, which has changed little over the past 40 years. Despite the tremendous efforts made, serum biomarkers with clinical utility for lung cancer early detection and clinical outcome prediction are still lacking. Metabolic alterations have been recognized as an emerging hallmark of cancer. We aimed to investigate the metabolic changes associated with lung cancer and to identify novel clinically applicable serum biomarkers for lung cancer early diagnosis and clinical outcome.

Serum metabolites are potential biomarkers for lung cancer early detection. We first performed global metabolomic profiling followed …

Genetic Predictors Of Metabolic Side Effects Of Diuretic Therapy, Jorge L. Del Aguila Aug 2014

Genetic Predictors Of Metabolic Side Effects Of Diuretic Therapy, Jorge L. Del Aguila

Dissertations and Theses (Open Access)

Thiazide diuretics are a recommended first-line monotherapy for hypertension (i.e.SBP>140 mmHg or DBP>90 mmHg). Even so, diuretics are associated with adverse metabolic side effects, such as hyperlipidemia, hyperglycemia and hypokalemia which increase the risk of developing type II diabetes. This thesis used three analytical strategies to identify and quantify genetic factors that contribute to the development of adverse metabolic effects due to thiazide diuretic treatment. I performed a genome-wide association study (GWAS) and meta-analysis of the change in fasting plasma glucose and triglycerides in response to HCTZ from two different clinical trials: the Pharmacogenomic Evaluation of Antihypertensive Responses …

DISSECTING THE ROLES OF TRIM24 IN REGULATION OF HEPATIC LIPID

METABOLISM AND INFLAMMATION

Lindsey Cauthen Minter, B.S., B.A.

Advisory Professor: Michelle C. Barton, Ph.D.

In this dissertation, I report the characterization of a new mouse model that recapitulates development of hepatocellular carcinoma (HCC) following spontaneous hepatic lipid accumulation, inflammation, and damage of liver tissue, due to complete loss of Trim24 expression. In human HCC and other cancers, TRIM24 expression is aberrantly high, while deletion of TRIM24 in the mouse has been shown to act as a liver specific tumor suppressor. The hypothesis tested here was that TRIM24, the E3 ubiquitin …

Angiomotin Is A Novel Cadherin-11 Interacting Protein That Mediates Migration In Prostate Cancer Cells, Angelica Ortiz

Dissertations and Theses (Open Access)

Prostate cancer (PCa), the second leading cause of cancer-related deaths among men in the United States, has the proclivity to metastasize to bone resulting in sclerotic lesions. These cancer induced bone growths cause bone pain and fractures. Therefore, understanding the molecular mechanisms contributing to PCa bone metastasis is required in order to find better prognostic tools and suitable targets for metastasis treatment and/ or prevention. Previous work in our laboratory showed increased expression of cadherin-11 (Cad11), a mesenchymal cadherin, during PCa progression. Furthermore, Cad11 expression endows PCa cells with increased migratory potential and metastasis to bone. Deletion of the Cad11 …

Energy Stress Causes Chaperones To Assemble Into Cytoplasmic Complexes, Kimberly J. Cope Aug 2014

Energy Stress Causes Chaperones To Assemble Into Cytoplasmic Complexes, Kimberly J. Cope

Dissertations and Theses (Open Access)

The majority of proteins require molecular chaperones to assist their folding into tertiary and quaternary structures. Certain stresses can compromise the weak hydrophobic forces responsible for these structures and lead to protein unfolding, misfolding, and aggregation. Aggregates of proteins are hallmarks of devastating diseases such as Alzheimer’s, Parkinson’s, and Huntington’s diseases. Fortunately, bacteria, plants, and fungi have a potent disaggregase, named Hsp104 in Saccharomyces cerevisiae. Recently, heat-induced aggregates, termed Q-bodies, were found to contain three molecular chaperones: Hsp70, Hsp104, and Hsp42. Their coalescence from small puncta into larger inclusions required Hsp104. During glucose deprivation, a stress that isn’t known to …

An Agent-Based Model Of Ant Colony Energy And Population Dynamics: Effects Of Temperature And Food Fluctuation, Guo Xiaohui Aug 2014

An Agent-Based Model Of Ant Colony Energy And Population Dynamics: Effects Of Temperature And Food Fluctuation, Guo Xiaohui

Electronic Theses and Dissertations

The ant colony, known as a self-organized system, can adapt to the environment by a series of negative and positive feedbacks. There is still a lack of mechanistic understanding of how the factors, such as temperature and food, coordinate the labor of ants. According to the Metabolic Theory of Ecology (MTE), the metabolic rate could control ecological process at all levels. To analyze self-organized process of ant colony, we constructed an agent-based model to simulate the energy and population dynamics of ant colony. After parameterizing the model, we ran 20 parallel simulations for each experiment and parameter sweeps to find …

Analysis Of High Fat Diet Induced Genes During Mammary Gland Development: Identifying Role Players In Poor Prognosis Of Breast Cancer, Raquel C. Martinez-Chacin, Megan Keniry, Robert Dearth

School of Integrative Biological & Chemical Sciences (Formerly Dept. of Biology)

Background

Epidemiological studies have shown that consumption of a high-fat diet (HFD) increases the risk of developing breast cancer (BC). Studies in rodents have shown HFD causes changes in the genetic programming of the maturing mammary gland (MG) increasing the susceptibility of developing the disease. Less is known about how HFD induced genes impact BC development. HFD exposure two weeks before conception to six weeks of age was previously shown to dramatically change MG gene expression in 10 week old mice. Therefore, we investigated these differentially expressed HFD-induced genes for their expression in BC using the NKI 295 breast tumor …

Effects Of Water Parameters On Container Mosquito (Diptera: Culicidae) Oviposition And Performance, Stephanie Sue Schelble Aug 2014

Effects Of Water Parameters On Container Mosquito (Diptera: Culicidae) Oviposition And Performance, Stephanie Sue Schelble

Master's Theses

Water body parameters have a considerable effect on the communities that develop within them. In small container habitats like tires, the depth, surface area, and volume effect the development and success of the mosquito communities. This study investigated the choices of the adult female mosquitoes, Aedes albopictus and Culex quinquefasciatus, between different depths and surface areas. In addition, larval performance was determined under differing depth and larvae densities. Oviposition studies showed that Ae. albopictus had a preference for deeper habitats (χ2= 14.2902, p= 0.0139) but did not prefer any surface areas (χ2= 7.2321, p= 0.0649) though …

How Microlending Affects Innovation And Entrepreneurship: Evidence From Ethiopia, Robert Hirth

Graduate Theses and Dissertations

Advocates of microlending suggest it is a sustainable intervention that reaches the poor directly and offers them the means to invest and improve their incomes (Khavul, 2010; Morduch, 1999; Yunus, 2007); yet, impact studies of these interventions have suggested they often have little or even a detrimental impact on borrowers (Van Rooyen, Stewart & De Wet, 2012). This dissertation examines the efforts to promote entrepreneurship and alleviate poverty in developing countries through microlending. I begin by reviewing the microlending literature, and in particular, impact studies of the effect microlending is having in developing countries. Next, I review theory and empirical …
