Comparing Population Density Estimation Techniques For Columbia Black-Tailed Deer (Odocoileus Hemionus Columbianus): Fecal Standing Crop And Distance Sampling Methods, Jubilee D. Brenneman
Honors Projects
I estimated deer density and population size on Blakely Island, WA, using the fecal standing crop (FSC) method described by Martin et al. (2011). I compared the FSC method with a well established and broadly applicable estimation method, distance sampling. Additionally, architecture of a commonly browsed shrub, Ocean Spray (Holodiscus discolor), was measured as an indicator of browsing pressure. Fecal standing crop was estimated by counting pellet groups along 26, 100 x 2m line transects. Deer density was estimated using these data and the equation derived by Martin et al. (2011). Distance sampling consisted of recording perpendicular distance …

Engineering An In Vitro Metastasis Platform, Justus Carlisle, Mark-Phillip Pebworth
Bioengineering Senior Theses
Although metastasis is the primary cause of cancer deaths and results in 90% of all cancer fatalities currently, all attempts to discover anti-metastatic drugs have failed. Many of the traditional methods of studying cell migration, using two-dimensional (2D) platforms, only study two-dimensional migration, which is fundamentally different and more multifaceted than the three-dimensional migration that occurs in vivo. To more accurately capture this in vivo variation, we developed a two-and-a-half-dimensional (2.5D) cell culture platform to better study three-dimensional (3D) migration. This platform consists of a layer of alginate on top of a monolayer of cells grown on tissue culture polystyrene …

Genetic engineering has always held great opportunity for the field of gene therapy. If the cause of a genetic disease can be determined, correction of this gene would allow for an efficient and permanent cure. However, current technologies utilizing engineered retroviruses have serious drawbacks that significantly limit their practical applications for gene therapy. In this project we investigate a novel genetic editing technology called TALENs. By utilizing a modular protein isolated from a plant pathogen that can be quickly and efficiently redesigned to recognize and bind to any desired sequence of DNA combined with a nuclease, we can target a …

Ecto-Protein Kinases And Phosphatases: An Emerging Field For Translational Medicine, Garif Yalak, Yigal H. Ehrlich, Bjorn R. Olsen
Publications and Research
Progress in translational research has led to effective new treatments of a large number of diseases. Despite this progress, diseases including cancer and cardiovascular disorders still are at the top in death statistics and disorders such as osteoporosis and osteoarthritis represent an increasing disease burden in the aging population. Novel strategies in research are needed more than ever to overcome such diseases. The growing field of extracellular protein phosphorylation provides excellent opportunities to make major discoveries of disease mechanisms that can lead to novel therapies. Reversible phosphorylation/dephosphorylation of sites in the extracellular domains of matrix, cell-surface and trans-membrane proteins is …

Early Back-To-Africa Migration Into The Horn Of Africa, Jason A. Hodgson, Connie J. Mulligan, Ali Al-Meeri, Ryan L. Raaum
Early Back-To-Africa Migration Into The Horn Of Africa, Jason A. Hodgson, Connie J. Mulligan, Ali Al-Meeri, Ryan L. Raaum
Publications and Research
Genetic studies have identified substantial non-African admixture in the Horn of Africa (HOA). In the most recent genomic studies, this non-African ancestry has been attributed to admixture with Middle Eastern populations during the last few thousand years. However, mitochondrial and Y chromosome data are suggestive of earlier episodes of admixture. To investigate this further, we generated new genome-wide SNP data for a Yemeni population sample and merged these new data with published genome-wide genetic data from the HOA and a broad selection of surrounding populations. We used multidimensional scaling and ADMIXTURE methods in an exploratory data analysis to develop hypotheses …

Recent research advancements in induced pluripotent stem cells (iPSCs) have broadened their capacity for research in specific disease models3 need for effective and robust techniques to detect cell differentiation. Two techniques currently exist to determine cell type; morphology and immunostaining. Although they have been used by researchers for many years, these methods can be unreliable, time-consuming, and very uneconomical. We seek to create a technology that will be more accurate and efficient in detecting differentiation of iPSCs. This will allow researchers greater speed in screening for specific differentiation products, further increasing the utility of iPSCs in medical research. We will …

Reverse Protein Engineering Of Firefly Luciferase, Kahler Bugtong, Skyler Herczeg, Abraham Munoz, Alexandra Obata
Bioengineering Senior Theses
Firefly luciferase is a bioluminescent protein commonly used as a bioluminescent tag in biological studies and applications. However, because the protein is fairly large in size, it is sometimes larger than the molecules it is intended to measure and is therefore not a sufficient tag in smaller applications. The active site of firely luciferase is also not well understood, making it difficult to engineer the protein without affecting its bioluminescent activity. In this paper, we discuss the experimental methods of Reverse Protein Engineering: a bioengineering technology that reduces the size of a protein while retaining its original function. This involves …

The sperm or eggs of sexual organisms go through a series of cell divisions from the fertilized egg; mutations can occur at each division. Mutations in the lineage of cells leading to the sperm or eggs are of particular importance because many such mutations may be shared by somatic tissues and also may be inherited, thus having a lasting consequence. For decades, little has been known about the pattern of the mutation rates along the germline development. Recently it was shown from a small portion of data that resulted from a large-scale mutation screening experiment that the rates of recessive …

Evaluating The Efficacy Of Shaping With A Percentile Schedule To Increase The Duration Of Sustained Interaction Following A Bid For Joint Attention In Children With Autism, Therese Gutbrod
USF Tampa Graduate Theses and Dissertations
This study examined the use of shaping with a percentile schedule to increase the duration of the interaction following a bid for joint attention in children with autism. Specifically, the therapist initiated a bid for joint attention and reinforced longer successive approximations in seconds of sustained interaction with the therapist and activity. A percentile schedule ranked the most recent 10 observations and reinforcement was provided if the current observation equaled the sixth ranking. Most-to-least prompting was used if the child failed to meet the calculated criterion. Shaping with a percentile schedule of reinforcement was effective at increasing the duration of …

Pre -Service School Psychologists' Racial And Weight-Related Biases And The Relationship That Taking Diversity Courses Has On These Biases, Shaerica Jackson
Theses and Dissertations
Past research has shown that racial and weight-related biases are present in school settings and can have a negative impact on students. The purpose of this study was to assess the impact of racial and weight-related biases upon judgments that school psychology graduate students make about Black and overweight students who were having problems in school. The problematic behaviors exhibited by the student in the case study were based on symptoms often seen in children with Attention-deficit/Hyperactivity Disorder (ADHD). We also examined the extent to which implicit and/or explicit attitudes moderated the degree of school psychology graduate students' attributions of …
Body Shape Divergence In Invasive Round Goby, Cat Collins
Body Shape Divergence In Invasive Round Goby, Cat Collins
DePaul Discoveries
The round goby, Neogobius melanostomus, originally native to the Black and Caspian seas, was introduced into the Great Lakes via ballast water in the 1990’s. Since then, the species has spread to all of the Great Lakes, thriving in the Lake Michigan region and spreading to surrounding bays and rivers. Invasive species are considered to have a high evolutionary potential. Differences in environmental conditions between native and introduced ranges stimulate adaptive evolution. Multiple introductions of an exotic species can result in separate instances of founder effects, further increasing the chance of evolutionary change. A total of 267 round goby specimens …
Arthropod Communities And Red-Headed Woodpecker (Melanerpes Erythrocephalus) Habitat Selection, Michael C. Lordon
Arthropod Communities And Red-Headed Woodpecker (Melanerpes Erythrocephalus) Habitat Selection, Michael C. Lordon
DePaul Discoveries
Red-headed woodpecker populations are near-threatened. Their habitat selection and reproductive success may be related to the availability of arthropods. We sampled the arthropod community within 0.04 ha plots surrounding known nests and compared these findings to plots without nests in the same fragments of forest or park in Cook County, IL. After 14 days, the traps were recovered, yielding close to 45,000 arthropods across 10 orders. Nest and control site differences were not statistically significant. Differences between park and forest diversity were not statistically significant either, but the greater diversity values in forests were consistent with expectations. We conclude that …
Vitamin A Deficiency Causes Ovulation Abnormalities In Mice, Rebecca Ursin
Vitamin A Deficiency Causes Ovulation Abnormalities In Mice, Rebecca Ursin
DePaul Discoveries
Retinoic acid (RA) is an active metabolite of vitamin A (VA) and is involved in tissue organization, patterning, and growth. RA has been shown to regulate male reproduction, however information on its role in ovary development is limited. To investigate the functions of RA in the ovary, we examined its role in ovary development and ovulation using an in vivo dietary VA-deprivation animal model. Our preliminary results have shown that VA deficiency causes a variety of ovarian pathologies, including reduced numbers of total follicles and corpus lutea, formation of hemorrhagic and atretic follicles, and formation of bursa and follicular cysts. …

Holding Back The Genes: Limitations Of Research Into Canine Behavioural Genetics, Diane Van Rooy, Elizabeth R. Arnott, Jonathan B. Early, Paul Mcgreevy, Claire M. Wade
Genetics Collection
Canine behaviours that are both desirable and undesirable to owners have a demonstrable genetic component. Some behaviours are breed-specific, such as the livestock guarding by maremmas and flank sucking seen in Dobermanns. While the identification of genes responsible for common canine diseases is rapidly advancing, those genes underlying behaviours remain elusive. The challenges of accurately defining and measuring behavioural phenotypes remain an obstacle, and the use of variable phenotyping methods has prevented meta-analysis of behavioural studies. International standardised testing protocols and terminology in canine behavioural evaluations should facilitate selection against behavioural disorders in the modern dog and optimise breeding success …
