Understanding how cytotoxic T cells (CTL) kill is of fundamental importance to the field of immunology. In the current model for how CTL kill target cells there is a critical dependence on lymphocyte-function associated antigen 1 (LFA-1, αLβ2) to tightly bind its ligand, intercellular adhesion molecule 1 (ICAM-1), following T cell receptor (TCR) recognition of cognate antigen (Dustin 2009). Recently, another member of the of integrin family, CD103 (integrin αE) has been characterized and data suggest that it may play an important role in killing ligand bearing targets (Feng, Wang et al. 2002; Gorfu, Rivera-Nieves et al. 2009). CD103 is …

The Role Of The Testa During Development And In Establishment Of Dormancy Of The Legume Seed, Petr Smýkal, Vanessa Vernoud, Matthew W. Blair, Aleš Soukup, Richard D. Thompson

Agricultural and Environmental Sciences Faculty Research

Timing of seed germination is one of the key steps in plant life cycles. It determines the beginning of plant growth in natural or agricultural ecosystems. In the wild, many seeds exhibit dormancy and will only germinate after exposure to certain environmental conditions. In contrast, crop seeds germinate as soon as they are imbibed usually at planting time. These domestication-triggered changes represent adaptations to cultivation and human harvesting. Germination is one of the common sets of traits recorded in different crops and termed the “domestication syndrome.” Moreover, legume seed imbibition has a crucial role in cooking properties. Different seed dormancy …

Bartonella bacilliformis is the bacterial agent of Carrión's disease and is presumed to be transmitted between humans by phlebotomine sand flies. Carrión's disease is endemic to high-altitude valleys of the South American Andes, and the first reported outbreak (1871) resulted in over 4,000 casualties. Since then, numerous outbreaks have been documented in endemic regions, and over the last two decades, outbreaks have occurred at atypical elevations, strongly suggesting that the area of endemicity is expanding. Approximately 1.7 million South Americans are estimated to be at risk in an area covering roughly 145,000 km2 of Ecuador, Colombia, and Peru. Although disease …

The Trade-Off Between Dietary Salt And Cardiovascular Disease; A Role For Na/K-Atpase Signaling?, Joe X. Xie, Anna Pearl Shapiro, Joseph I. Shapiro Md

Biochemistry and Microbiology

It has been postulated for some time that endogenous digitalis-like substances, also called cardiotonic steroids (CTS), exist, and that these substances are involved in sodium handling. Within the past 20 years, these substances have been unequivocally identified and measurements of circulating and tissue concentrations have been made. More recently, it has been identified that CTS also mediate signal transduction through the Na/K-ATPase, and consequently been implicated in profibrotic pathways. This review will discuss the mechanism of CTS in renal sodium handling and a potential “trade-off” effect from their role in inducing tissue fibrosis.

Contributions to Books

Published as Chapter 6 in Environmental Law and Contrasting Ideas of Nature: a Constructivist Approach, Keith H. Hirokawa, ed.

Whereas a large and growing scholarly literature is dedicated to studying human populations in the city, not much has been written about nonhuman animals in this space. This essay explores the presence of nonhuman animals in the American city through a legal lens. I begin with a few general contemplations about the legal classification of animals in American cities, and then move to explore specific legal classifications of animals in cities: domestic and companion animals, agriculture or livestock animals, wild animals, …

Evidence that brain edema and aquaporin-4 (AQP4) water channels have roles in experimental binge ethanol-induced neurodegeneration has stimulated interest in swelling/edema-linked neuroinflammatory pathways leading to oxidative stress. We report here that neurotoxic binge ethanol exposure produces comparable significant effects in vivo and in vitro on adult rat brain levels of AQP4 as well as neuroinflammation-linked enzymes: key phospholipase A2 (PLA2) family members and poly (ADP-ribose) polymerase-1 (PARP-1). In adult male rats, repetitive ethanol intoxication (3 gavages/d for 4 d, ∼ 9 g/kg/d, achieving blood ethanol levels ∼ 375 mg/dl; "Majchrowicz" model) significantly increased AQP4, Ca+2-dependent PLA2 GIVA (cPLA2), phospho-cPLA2 GIVA …

The Wonderfully Made Design Of The Skin And Its Microbiome, Alan L. Gillen

Faculty Publications and Presentations

Many microbes live in a mutualistic relationship with the human body, make up the human microbiome, and play a role in our health by modulating the immune system. Man is “covered” inside and outside his body with millions of microbes to maintain normal bodily functions and sustain life in our changing world. The skin is the largest organ in the human body and is colonized by millions of microbes. This external colonization of the integumentary system is termed our skin microbiome. Man cannot see it (except with a microscope), but we need it for normal functioning, certainly in a pathogenic …

Set Based Association Testing In High Dimensional Genomic Studies, Xueyuan Cao

Electronic Theses and Dissertations Archive

The last decade has ushered in an era of high dimensional, high volume data. In particular with the biotechnological revolution of the era, high-dimensional genomic studies of various designs have provided investigators with the tools to study thousands or even millions of genomic features simultaneously. These studies have shed new light on the underlying mechanisms of complex diseases. The accumulated knowledge of these complex relationships between genes has led scientists to formalize pathways and graphical networks that visually and succinctly give descriptions of the geometry of these relationships. With such knowledge, it has become possible to develop procedures for statistical …

Glutamate Dysfunction Associated With Developmental Cerebellar Damage: Relevance To Autism Spectrum Disorders, Eric Mckimm Jul 2014

Glutamate Dysfunction Associated With Developmental Cerebellar Damage: Relevance To Autism Spectrum Disorders, Eric Mckimm

Electronic Theses and Dissertations Archive

Neural abnormalities commonly associated with autism spectrum disorders include prefrontal cortex (PFC) dysfunction and cerebellar pathologies in the form of Purkinje cell loss and cerebellar hypoplasia. It has been shown that cerebellar Purkinje cell loss and dysfunction results in abnormal dopamine neurotransmission in the PFC. With use of a mouse model, we explored the effects of Purkinje cell loss on the disruption of glutamatergic projections to the PFC that ultimately influences dopamine (DA) release. Glutamate release was evoked by localized electrical stimulation coupled with fixed potential amperometry in combination with a glutamate selective enzyme-based recording probe in urethane anesthetized Lurcher …

An Integrated Approach To Understanding The Structure And Function Of The Large-Conductance, Voltage- And Calcium-Activated Potassium Channel, Jacob E. Mcmillan Jul 2014

An Integrated Approach To Understanding The Structure And Function Of The Large-Conductance, Voltage- And Calcium-Activated Potassium Channel, Jacob E. Mcmillan

Electronic Theses and Dissertations Archive

The large-conductance, voltage- and calcium-activated potassium channel (BKCa channel) is an important transmembrane ion channel involved in many physiological and pathophysiological processes. However, little is known about its atomic-level three-dimensional structure. The functional channel requires four alpha subunits assembled as a potassium pore and is typically associated with four beta subunits that regulate function in various ways depending on the isoform of the beta subunit is interacting with the channel. Currently, the only major high-resolution atomic structure available is of the cytosolic tail domain of the alpha subunit. In the absence of structural information, electrophysiology has allowed for extensive characterization …

Judgment Day For Fraud-On-The-Market: Reflections On Amgen And The Second Coming Of Halliburton, Donald C. Langevoort

Georgetown Law Faculty Publications and Other Works

The Supreme Court has reaffirmed the "fraud on the market" presumption of reliance, facilitating large scale class actions for this kind of securities fraud. This essay traces the road from its decision last year in Amgen to this year's reaffirmation in Halliburton II, and considers some of the issues that will emerge as lower courts struggle with Halliburton II's secondary holding--that the issue of "price impact" is crucial to class certification, even if the burden of proof is on the defendants.

Learning To Generate Understandable Animations Of American Sign Language, Matt Huenerfauth Jul 2014

Learning To Generate Understandable Animations Of American Sign Language, Matt Huenerfauth

Annual Effective Access Technology Conference

Motivations & Methods - Standardized testing has revealed that many deaf adults in the U.S. have lower levels of English literacy; providing American Sign Language (ASL) on websites can make information more accessible. Unfortunately, video recordings of human signers are difficult to update when information changes, and there is no way to support just-in-time generation of web content from a query.

Self-Adjusting Biofeedback With A Dynamic Feedback Signal Set (Dyfss), Laurence I. Sugarman, Brian L. Garrison, Anna E. Hope

Annual Effective Access Technology Conference

Abstract - A lack of control over their autonomic nervous system presents a major challenge for many children with Autism Spectrum Disorder (ASD). Autonomic biofeedback training is a promising treatment for managing anxiety and ASD symptoms more generally. We describe software that tunes four autonomic measurements to the best abilities and needs of each individual patient. Using this dynamic feedback signal set (DyFSS), a strength-based, self-customizing algorithm, we aim to address the autonomic heterogeneity of youth with ASD. The DyFSS may improve autonomic biofeedback training for the user by making it more understandable and easier to accomplish. Because it is …

Learning To Generate Understandable Animations Of American Sign Language, Matt Huenerfauth

Annual Effective Access Technology Conference

Abstract - Standardized testing has revealed that many deaf adults in the U.S. have lower levels of written English literacy; providing American Sign Language (ASL) on websites can make information and services more accessible. Unfortunately, video recordings of human signers are difficult to update when information changes, and there is no way to support just-in-time generation of web content from a query. Software is needed that can automatically synthesize understandable animations of a virtual human performing ASL, based on an easy-to-update script as input. The challenge is for this software to select the details of such animations so that they …

Parts Of The Whole: Strategies For The Spread Of Quantitative Literacy: What Models Can Tell Us, Dorothy Wallace Jul 2014

Parts Of The Whole: Strategies For The Spread Of Quantitative Literacy: What Models Can Tell Us, Dorothy Wallace

Numeracy

Two conceptual frameworks, one from graph theory and one from dynamical systems, have been offered as explanations for complex phenomena in biology and also as possible models for the spread of ideas. The two models are based on different assumptions and thus predict quite different outcomes for the fate of either biological species or ideas. We argue that, depending on the culture in which they exist, one can identify which model is more likely to reflect the survival of two competing ideas. Based on this argument we suggest how two strategies for embedding and normalizing quantitative literacy in a given …
