Proximal spinal muscular atrophy (SMA) is an early onset, autosomal recessive motor neuron disease caused by loss of or mutation in SMN1 (survival motor neuron 1). Despite understanding the genetic basis underlying this disease, it is still not known why motor neurons (MNs) are selectively affected by the loss of the ubiquitously expressed SMN protein. Using a mouse embryonic stem cell (mESC) model for severe SMA, the RNA transcript profiles (transcriptomes) between control and severe SMA (SMN2+/+;mSmn-/-) mESC-derived MNs were compared in this study using massively parallel RNA sequencing (RNA-Seq). Previous studies have reported that chromosome synteny in Lepidoptera has been well conserved, yet the number of haploid chromosomes varies widely from 5 to 223. Here we report the genome (393Mb) of the Glanville fritillary butterfly (Melitaea cinxia; Nymphalidae), a widely recognized model species in metapopulation biology and eco-evolutionary research, which has the putative ancestral karyotype of n¼31. Using a phylogenetic analyses of Nymphalidae and of other Lepidoptera, combined with orthologue-level comparisons of chromosomes, we conclude that the ancestral lepidopteran karyotype has been n¼31 for at least 140My. Prior to, or concurrent with, the encoding of concepts into speech, the individual faces decisions about whether, what, when, how, and with whom to communicate. Compared to the existing wealth of linguistic knowledge however, we know little of the mechanisms that govern the delivery and accrual of information. Here we focus on a fundamental issue of communication: The decision whether to deliver information. Specifically, we study spontaneous confession to a victim. Given the costs of social devaluation, offenders are hypothesized to refrain from confessing unless the expected benefits of confession (e.g. enabling the victim to remedially modify their course of …

The present study analysis has been taken into account in the filed of mobile technology research output using Engineering Index database for selected period of ten years from 2003 to 2012. This study also explores the trend in authorship pattern and collaborative in mobile technology publications with sample of 1, 44,567 research articles during the period from 2003 to 2012. The findings of the study shows that in the year 2011 was the most productive year with 20318 articles (14.05%) published. Among the top ten country wise authors, the highest numbers of articles (22.83%) were contributed by the authors from …

The Companion Animal Parasite Council hosted a meeting to identify quantifiable factors that can influence the prevalence of tick-borne disease agents among dogs in North America. This report summarizes the approach used and the factors identified for further analysis with mathematical models of canine exposure to tick-borne pathogens.

Approximately 4,000 children in the United States are diagnosed annually with a brain tumor. Brain cancers are the deadliest of all pediatric cancers as they have survival rates of less than 20%. Although surgery and radiation therapy are widely used to treat adult patients, chemotherapy is the primary therapeutic option for children. One important chemotherapeutic agent is temozolomide, an alkylating agent that causes cell death by damaging DNA. In this project, we tested the ability of a specific non-natural nucleoside developed in our lab, designated 5-NIdR, to increase the efficacy of temozolomide against brain cancer. It is well documented that open reading frames containing high GC content show poor expression in A+T rich hosts. Specifically, G+C-rich codon usage is a limiting factor in heterologous expression of Mycobacterium avium subsp. paratuberculosis (MAP) proteins using Lactobacillus salivarius. However, re-engineering opening reading frames through synonymous substitutions can offset codon bias and greatly enhance MAP protein production in this host. In this report, we demonstrate that codon-usage manipulation of MAP2121c can enhance the heterologous expression of the major membrane protein (MMP), analogous to the form in which it is produced natively by MAP bacilli. Induction of fetal hemoglobin (HbF) has therapeutic importance for patients with sickle cell disease (SCD) and the beta-thalassemias. It was recently reported that increased expression of LIN28 proteins or decreased expression of its target let-7 miRNAs enhances HbF levels in cultured primary human erythroblasts from adult healthy donors. Here LIN28A effects were studied further using erythrocytes cultured from peripheral blood progenitor cells of pediatric subjects with SCD. Transgenic expression of LIN28A was accomplished by lentiviral transduction in CD34(+) sickle cells cultivated ex vivo in serum-free medium. Introduction:
Ethanol has been used for years in neonatal and infant liquid medications, yet the pharmacokinetics, pharmacodynamics, and safety of ethanol in this vulnerable population have not been well characterized. The purpose of this review is to raise awareness of ethanol use as an excipient in neonatal and infant medications and to provide insight, based on the available evidence, into clearance rates of ethanol in babies. We also discuss ethanol pharmacokinetics in adults, theoretical pharmaco-kinetic changes in neonates and infants as it may apply to ethanol disposition, and case reports involving ethanol exposure in neonates and infants.

© 2014 Jiang et al.; licensee BioMed Central Ltd. Background: During mammalian pre-implantation embryonic development dramatic and orchestrated changes occur in gene transcription. The identification of the complete changes has not been possible until the development of the Next Generation Sequencing Technology.Results: Here we report comprehensive transcriptome dynamics of single matured bovine oocytes and pre-implantation embryos developed in vivo. Surprisingly, more than half of the estimated 22,000 bovine genes, 11,488 to 12,729 involved in more than 100 pathways, is expressed in oocytes and early embryos. Overexpression of the histone methyltransferase MMSET in t(4;14)+ multiple myeloma patients is believed to be the driving factor in the pathogenesis of this subtype of myeloma. MMSET catalyzes dimethylation of lysine 36 on histone H3 (H3K36me2), and its overexpression causes a global increase in H3K36me2, redistributing this mark in a broad, elevated level across the genome. Here, we demonstrate that an increased level of MMSET also induces a global reduction of lysine 27 trimethylation on histone H3 (H3K27me3). Despite the net decrease in H3K27 methylation, specific genomic loci exhibit enhanced recruitment of the EZH2 histone methyltransferase and become hypermethylated on …

This project was designed to evaluate the effect of ruminant grazing practices at Hickory Hill Farm on the surrounding environment by measuring soil nutrients and runoff chemistry. Three pastures on the farm (Goat, Cattle and Control) were selected for soil sampling and nutrient analyses were recorded. Physical water quality parameters were conducted on the runoff collected from the farm after Hurricane Sandy. The sites with animal activity had higher levels of sulfate, phosphate, nitrate, Mehlich 3 phosphorus and conductivity when compared to control site. The age-old connection between the people and the land is something that seven women, who have organized themselves into a loosely fitted cooperative, are reviving. In the process of growing together as members of a cooperative, these women in Macon County, Alabama, are not only promoting healthy living and nurturing community, but are also addressing the issue of food security by making nutritious affordable produce and other agricultural products available to their community. Additionally, they are illustrating alternative strategies of community and economic development.
