We describe and characterize a method for estimating the pressure field corresponding to velocity field measurements such as those obtained by using particle image velocimetry. The pressure gradient is estimated from a time series of velocity fields for unsteady calculations or from a single velocity field for quasi-steady calculations. The corresponding pressure field is determined based on median polling of several integration paths through the pressure gradient field in order to reduce the effect of measurement errors that accumulate along individual integration paths. Integration paths are restricted to the nodes of the measured velocity field, thereby eliminating the need for …

Animal propulsors such as wings and fins bend during motion and these bending patterns are believed to contribute to the high efficiency of animal movements compared with those of man-made designs. However, efforts to implement flexible designs have been met with contradictory performance results. Consequently, there is no clear understanding of the role played by propulsor flexibility or, more fundamentally, how flexible propulsors should be designed for optimal performance. The Ozobranchidae family is the smallest and least studied hirudinean taxon. Our research includes the largest molecular dataset yet reported for marine ozobranchids (Ozobranchus margoi and Ozobranchus branchiatus) with the most number of documented turtle hosts (57) from the Atlantic and Pacific Oceans to date of any marine turtle epibiont study. Turtle species sampled in this study include loggerheads (Caretta caretta), hawksbill (Eretmochelys imbricata), olive ridley (Lepidochelys olivacea), and green turtles (Chelonia mydas). Phylogenetic analyses of mitochondrial (COI) and nuclear ribosomal (18S and 28S) genes all support the monophyly of marine Ozobranchidae leeches with speciation occurring over an extensive period …

Angiotensin Type 1 (AT1) receptors are involved in cardiovascular pathology. Losartan, the AT1 receptor blocker, in combination with exercise has been shown to be effective in improving cardiac performance. The goal of the study was to investigate the role of AT1a receptors on cardiac function, and exercise tolerance in response to aerobic exercise using AT1a receptor knockout (AT1aKO) mice. Cytokine activation of macrophages leads to "macrophage polarizatio" The M1 subset is polarized by exposure to lipopolysaccharide (LPS) and interferon-gamma (IFN-γ) and M2 polarized cells develop after exposure to interleukin-4 (IL-4) or IL-13. In this study, the RAW 264.7 murine macrophage cell line was used to study the effect herpes simplex virus-Type I (HSV-1) and Dengue virus serotype-2 (DENV2) infections have on cultures of unpolarized (M0) or polarized subsets. The macrophage subsets were characterized using cluster of differentiation markers CD14 and CD86. It has previously been reported that AMP-activated protein kinase (AMPK) may be critical for hypoxic chemotransduction in carotid body type I cells (Wyatt et al, 2007). This study sought to determine the importance of the regulatory upstream kinase of AMPK, CamKKβ, in the acute response to hypoxia in isolated mouse type I cells.
Initial studies indicated several previously unreported artefacts associated with using the CamKKβ inhibitor STO-609 and calcium imaging techniques. Strong STO-609 autofluorescence meant that Fura-2 could not be used and X-Rhod-1 imaging revealed that STO-609 quenched fluorescence even in the absence of intracellular Ca2+. The goal of this study is to validate the accuracy of the SC/Tetra CFD code by modeling four cases: a 2D zero pressure gradient flat plate, an axisymmetric separated boundary layer, flow over an NACA 0012 airfoil, and flow over an NACA 4412 airfoil trailing edge separation. The results will be compared to experimental data and CFL3D results. Historically, the value of a woman has been based on her ability to produce healthy and successful (male) children, leaving little room for worth based on intelligence or other achievements. Second wave feminists made significant strides for women's reproductive choices, acknowledging that motherhood is not the only way for a woman to have fulfillment or value. While women throughout the United States have more access, opportunity, and choice, women in United States prisons have not benefited from the same advancements in reproductive freedom. Circulating microvesicles (cMVs) are the extracellular MVs released from the cells in the blood and on the vascular wall. Our previous study demonstrates that cMVs of diabetic mouse are detrimental to endothelial progenitor cells (EPCs), which are known to be very important for maintaining normal endothelial function and structure. In this study, we compared the levels of circulating EPCs and EPC-derived MVs (EPC-MVs) in diabetic and healthy subjects. With the inspiration of owl's silent flight, a traditional airfoil S833 equipped with flexible fringes on the trailing edge is investigated through numerical simulation and experiments in a wind tunnel. The newly constructed airfoil is modeled and numerically investigated. An incompressible, 2D and viscous flow solver in the Computational Fluid Dynamics (CFD) software FLUENT is utilized to conduct the numerical simulation on the vortex flow feature in the wake of the airfoil. Animal shelter housing is highly stressful for a dog, compromising welfare and leading to undesirable behaviors and unknown health consequences. We documented the changes in circulating numbers of white blood cells, plasma cortisol, and fecal parasite shedding of dogs housed for 10 days at a county animal shelter. White blood cell changes were most prominent on Day 10 after arrival to the shelter. Changes included increased total leukocytes, mature neutrophils, and lymphocytes, with less consistent increases in monocytes and neutrophil to lymphocyte ratio (N:L). Fecal parasite shedding was elevated and not affected by day. Cortisol levels of shelter dogs declined …

Previous studies show a fluoxetine/simvastatin drug combination successfully reduced infarct size and increased functional recovery when administered 20-26 hours post stroke. This project tested the hypothesis that earlier drug delivery at 6-12 hours post stroke will improve functional recovery and decrease infarct size, and that limb rehabilitation will improve functional recovery.
Pre-stroke function was determined in rats, a stroke was induced, and daily treatment began 6-12 hours post stroke. Baseline functional deficit was established, and additional testing over 90 days monitored functional recovery. Limb rehabilitation was provided for designated animals, and brain analysis measured infarct size.

The nervous system is faced with perturbations in activity levels throughout development and in disease or injury states. Neurons need to adapt to these changes in activity, but also need to maintain circuit firing within a normal range to stabilize the network from becoming too excited or too depressed. Homeostatic synaptic plasticity, the compensatory increase or decrease in synaptic strength as a result of excessive circuit inhibition or excitation, is a mechanism that the nervous system utilizes to keep network activity at normal levels. Assortative fertilization refers to the species-specific interactions between sperm and oocytes that affect the success of fertilization. One type of interaction is chemotaxis of sperm to oocytes. In Caenorhabditis elegans, amoeboid sperm must crawl along the uterine lining towards the spermathecae in response to oocyte-derived prostaglandin signals for fertilization. This chemotactic signaling system likely operates in other species of the Elegans-Group of Caenorhabditis as sperm of C. briggsae and C. remanei do localize to the C. elegans spermathecae. Muscle spindles (MS) and Golgi tendon organs (GTOs) are encapsulated, stretch-activated sensory receptors housed within skeletal muscles, yet their functional contributions to proprioceptive monitoring of limb movements are quite different. For example, MS are responsible for detecting stretch of muscle fibers, and GTOs provide feedback regarding muscle tension during contraction. Peripheral axons extending from proprioceptive sensory neurons (PSNs) in the dorsal root ganglia supply MS and GTOs. MS are innervated by Group Ia and II sensory fiber endings, while GTOs are innervated by a single Group Ib sensory fiber. The developmental processes guiding axons from PSNs to their targets and …

Crystal Lake showed distinct biogeochemical patterns, which were governed by thermal stratification, photosynthesis, and sulfur redox processes. Field parameter measurements and water and Planktothrix rubescens samples were collected in May, August, and October, 2013 at the deepest point in the lake, to better understand these processes. At the metalimnion-hypolimnion boundary, P. rubescens produced chlorophyll and turbidity maxima. Photosynthesis produced oxygen-rich and nutrient-poor surface waters. The decay of organic matter produced anoxic, nutrient-rich water in the hypolimnion. Sulfate concentrations were high in the epilimnion and metalimnion, with a maximum at the layer of P. rubescens, and decreased with depth in the …

The distribution of mercury (Hg) in the ocean is complex as a result of in situ chemical transformations and inputs from natural and anthropogenic sources. Within the ocean, inorganic Hg is methylated to monomethylmercury (MMHg), which bioaccumulates and biomagnifies in marine food webs and poses a health risk to humans who eat fish. The Slc12a5 gene encodes the neuron-specific electroneutral K+Cl- cotransporter-2 (KCC2), a constitutively active Cl- extruder. In mature neurons, KCC2 functionally predominates over other members of the Slc12a family, particularly the Na+K+2Cl- cotransporter-1 (NKCC1), a ubiquitous Cl- loader. Therefore, KCC2 keeps and maintains the intracellular chloride concentration ([Cl-]i) below values predicted by thermodynamic equilibrium and in doing so, KCC2 plays a key role in neuron physiology: it makes possible the inhibitory action of neurotransmitters such as gamma-aminobutyric acid (GABA) and glycine. Neonates are at a rapid stage of development from birth throughout childhood. Impairments to certain cortical areas can result in long lasting neuro-cognitive dysfunctions. Disorders to the swallowing neural pathways can have detrimental effects throughout life course since methods to provide nutrition can be comprised. Dysphagia, or the inability to swallow, can be caused by a multitude of reasons, chiefly neurological, but the underlying disruptions to the neural pathways are not defined. This study presents novel predictive equations for von Mises stresses and deflection of bones in the frontal and lateral regions of the skull. The equations were developed based on results of a finite element model developed here. The model was validated for frontal and lateral loading conditions with input values mimetic to fall scenarios. Using neural network processing of the information derived from the model achieved R2 values of 0.9990 for both the stress and deflection. Based on the outcome of the fall victims, a threshold von Mises stress of 40.9 to 46.6 MPa was found to indicate skull fracture …

Patent trolling is a serious legal problem. In addressing patent trolling, disclosure requirements and periodic reporting standards will be critical to minimizing the costs of this controversial practice.
Patent trolling, at its most problematic, generally refers to patent infringement allegations made by non-practicing entities (NPEs) which produce essentially no products or services except in connection with the buying and selling of patent rights. Nativists have postulated fundamental geometric knowledge that predates linguistic and symbolic thought. Central to these claims is the proposal for an isolated cognitive system dedicated to processing geometric information. Testing such hypotheses presents challenges due to difficulties in eliminating the combination of geometric and non-geometric information through language. We present evidence using a modified matching interference paradigm that an incongruent shape word interferes with identifying a two-dimensional geometric shape, but an incongruent two-dimensional geometric shape does not interfere with identifying a shape word. With previous indication of the Group VIA phospholipase A2 (iPLA2β) enzyme regulating ER-stress induced apoptosis in β-cells by regulating the anti-apoptotic protein Bcl-xL via alternative splicing, our lab postulated iPLA2β to be utilizing a similar mechanism to regulate apoptosis in mice lung. Our previous lab work has shown implications of lung function compromise in iPLA2β-/- mice, and we speculated the cause to be due altered lung architecture stemming from the attenuation of apoptosis. During the summer of 1934, over 70% of western North America experienced extreme drought, placing this summer far outside the normal range of drought variability and making 1934 the single worst drought year of the last millennium. Alcoholism is a complex neurological disorder characterized by loss of control in limiting intake, compulsion to seek and imbibe ethanol, and chronic craving and relapse. It is suggested that the characteristic behaviors associated with the escalation of drug use are caused by long-term molecular adaptations precipitated by the drug’s continual administration. These lasting activity-dependent changes that underlie addiction-associated behavior are thought, in part, to depend on new protein synthesis and remodeling at the synapses. Transcranial direct current stimulation (tDCS) has been utilized in human studies to modulate a multitude of psychological, cognitive, and psychiatric disorders. There have been positive behavioral results in human subjects but our knowledge of biological processes occurring during stimulation to elicit behavioral outcomes is limited. Our study utilizes a rodent tDCS (R-tDCS) model in which Sprague Dawley rats receive tDCS in order to examine whether tDCS affects neuronal activation. We examined two immediate early genes (IEG's), cFos and zif268, in order to discern if tDCS affects neuronal activation. Our findings indicate that tDCS does affect neuronal activation by means of …

White-tailed deer directly impact vegetation structure and species composition through selective foraging, and indirectly impact other species by altering habitat, food-web interactions, and microclimate. I examined the direct effects of deer exclusion on vegetation communities, and indirect effects on beetle, spider, and web-building spider (WBS) assemblages. Forb and woody plant percent cover were higher in exclosures, while graminoid cover was higher in controls. There were no differences in beetle and spider assemblages between browsed and protected areas. The absence of differences could be attributed to legacy effects, or alternatively high vagility of individuals. WBS assemblages were more abundant and diverse …
