Cellular Processes Regulating Cytoskeletal Signaling, Insulin Resistance And Calcium Signaling Are Commonly Misregulated In Type 1 And Type 2 Myotonic Dystrophy Patients, Nripesh Prasad
Dissertations
Myotonic dystrophy (DM) is a mutisystemic neurological disorder with an autosomal dominant inheritance involving skeletal and cardiac muscles. There are two different types of DM identified; type1 myotonic dystrophy (DM1) and type2 myotonic dystrophy (DM2). In this study I undertook an intense unbiased investigative approach by profiling the global mRNA, miRNA and protein expression landscape in skeletal muscle biopsies from DM1 and DM2 patients. I also compared the mRNA expression of three transgenic mouse models (DM2-KI, HSA-CTG and HSA-CCTG) with human DM patient muscle biopsy samples. This paper offers a genealogy of dialogic education, tracing its origins in Romantic epistemology and corresponding philosophy of childhood, and identifying it as a counterpoint to the purposes and assumptions of universal, compulsory, state-imposed and regulated schooling. Dialogic education has historically worked against the grain of standardized mass education, not only in its view of the nature, capacities and potentialities of children (and therefore of adults as well), but in its economic, political and social views, for which childhood is understood as a promissory condition. Across the animal kingdom it has been observed that outcomes of conflicts are influenced by past experiences, whereby previous winners are likely to keep winning and losers are likely to lose again. These so-called “winner and loser effects” are hypothesized to result from factors such as information acquisition and endocrine responses following the initial bouts. This paper applies the understanding of this phenomenon to a novel domain: patterns of winning and losing in Major League Baseball (MLB) double headers. Litigation often pivots on the testimony of fact witnesses. Unfortunately, serving as a witness may take people away from their jobs or interrupt their lives. It is therefore understandable that fact witnesses may want to be paid for devoting time to litigation.
It was once the rule that fact witness compensation was limited to statutory witness fees. That limitation rested on several factors, including the concern that greater payments could entice fact witnesses to perjure themselves, might simply influence witnesses to shape their testimony in ways favorable to the parties paying them, couldprice justice out of the reach of some …

A wide array of questions arises from global change to confront environmental law. The IPCC has examined social decisions affecting the climate in the design of human settlements, transport systems, industrialisation, agriculture and silviculture, waste management, provisions for energy, and virtually all other socio-economic dimensions of human life. The AR-5, too, cannot avoid raising issues of human ethics and values at local and regional scales. Such issues reach environmental policy and law directly. The IPCC’s AR-5 report furthers widespread public debate about the human dimensions of climate change, and how social theory relates to environmental change. Already, climate change has …

Carnivores substantially impact humans, but are elusive and difficult to monitor. Although less precise than intensive methods (e.g., capture-recapture), indices of relative abundance are widely used to monitor carnivore numbers. This study assessed public sightings and hunter surveys as approaches to monitoring the distribution and relative abundance of bobcats (Lynx rufus) in New Hampshire. To validate indices, I used a telemetry-based model of habitat suitability and information from camera surveys conducted by volunteers in three study areas. Bobcats were found widely distributed in New Hampshire with lower abundance in northern and mountainous regions. Public sightings and hunter surveys (both effort-corrected) …

In cattle sub-acute inflammatory disorders are difficult to assess because visual symptoms of diseases are not easily detectable with large numbers of dairy cattle in loose housing. In addition, inflammation is not always followed by leukocytosis in cattle, which makes the use of common blood tests like total white blood cell counts less reliable as a diagnostic tool. More sensitive and objective methods are needed to evaluate health status. Research has been focused on the use of acute phase proteins (APPs) to diagnose acute and chronic illness in cattle, because acute phase proteins respond very quickly when an inflammation event …

The Ipswich River Watershed, a 401 km2 watershed located in northeastern Massachusetts, has been observed to be undergoing increasing urbanization with resulting increases in nutrient loading, in particular, nitrogen. Nitrogen uptake occurs in a 1st-order process which is dependent on the concentration of nitrogen as well as the amount of time the water containing nitrogen remains within the wetland, which is described as the residence time distribution (RTD). During the winter months in the northern hemisphere, above 40 degrees latitude, a freezing event occurs that can last several months. Some lakes may have large portions of the shallow littoral zone frozen completely solid down into the sediments. Invertebrates affected by these events have developed survival strategies to cope with the harsh environment. The two main strategies for invertebrates in these areas are to either migrate to a different area and recolonize the original area when it becomes available again, or to burrow into the sediments and withstand the freezing. Scientific visualization can be viewed as a subset or a type of data visualization in which the data can be mapped to a geometric shape or spatial coordinate system. Geovisualization is a type of scientific visualization in which the data represented are earth data (e.g., bathymetry). As the need for exploring these scientific data sets increases, so will the need to support user interaction with the data. This requires creating user interfaces that allow users to carry out observational (e.g., compare and analyze different data sets) or experimental (e.g., data exploration) tasks on the data sets.

Immunohistochemical Localization Of The Neuropeptide Rfamide In The Hypostomal Nerve Net Of Hydra Vulgaris, Bailey Munro

This study examined the distribution and localization of the neuropeptide arginine-phenylalanine-amide (RFamide) in the hypostomal and tentacular nerve net of the cnidarian polyp Hydra vulgaris. Immunohistochemical techniques were used to visualize three RFamide-positive distinct nerve ring structures, nerves and neurites throughout the ectoderm of the hypostome and tentacles of ablated hypostomes. Two RFamide-positive circumhypostomal nerve rings are reported here: an inner ring located near the mouth and an outer ring located beneath the tentacle attachment sites. Both rings are consistent with rings labeled with anti-α-tubulin; the outer ring is also coincident with a GABAB receptor-antibody-labeled ring previously described …

Physical disruption of cellular membranes arising from interactions with engineered nanoparticles is an important, but poorly understood aspect of nanotoxicology and nanomedicine. Model cellular membranes (i.e. lipid bilayers) can be used to identify interaction mechanisms, and most studies have largely focused on lipid bilayers supported on solid planar or spherical substrates. While useful and informative, these systems do not accurately represent an intact cell membrane because they restrict the elastic motion of the bilayer and the capacity for mechanical changes. The overall survival rate for patients diagnosed with lung cancer is still extremely low and many affected patients are not eligible for first-line treatments such as surgery, chemotherapy, and radiation for the treatment due to the severe side effects associated with these therapies. Paclitaxel (PTX) along with imatinib mesylate are currently first-line therapies for many types of cancer (Kelly 2001, Schiller 2002). However, the successful delivery of paclitaxel faces many challenges due to its lipophilic nature and high protein affinity, which often results in low bioavailability. Despite concerns that the Louisiana Pine Snake (Pituophis ruthveni) has been extirpated from large portions of its historic range, only a limited number of studies on their movement patterns have been published. Winter movement patterns are of particular interest since it has been hypothesized that impacts of management practices would be reduced during the winter. Using radiotelemetry, we determined winter movement patterns of Louisiana Pine Snakes (11 males, 8 females) in 5 study areas (2 in Louisiana and 3 in Texas). Red crab (Chaceon spp.) fishery resources exist on both sides on the North Atlantic, and the fisheries that harvest these resources seek to maintain their sustainability. To be able to conduct fishery assessments with less uncertainty, resource managers need a better understanding of the life history characteristics of the species, more recent information on the abundance and distribution of the resource, and finally reliable estimates of the levels of exploitation and the effects of harvesting of the resources.
