In Vitro Efficacy Of Clinically Available Drugs Against Growth And Viability Of Acanthamoeba Castellanii Keratitis Isolate Belonging To The T4 Genotype, Abdul Mannan Baig, Junaid Iqbal, Naveed Ahmed Khan
Department of Biological & Biomedical Sciences
The effects of clinically available drugs targeting muscarinic cholinergic, adrenergic, dopaminergic, and serotonergic receptors; intracellular calcium levels and/or the function of calcium-dependent biochemical pathways; ion channels; and cellular pumps were tested against a keratitis isolate of Acanthamoeba castellanii belonging to the T4 genotype. In vitro growth inhibition (amoebistatic) assays were performed by incubating A. castellanii with various concentrations of drugs in the growth medium for 48 h at 30°C. To determine amoebicidal effects, amoebae were incubated with drugs in phosphate-buffered saline for 24 h, and viability was determined using trypan blue exclusion staining. For controls, amoebae were incubated with the …

The Capacity To Lead: Building A Collaborative Organization Through A Shared Vision And Improved Staff Capacity In An Inclusive New Jersey Preschool, Kathleen Mccabe-Odri
Theses and Dissertations
The purpose of my dissertation study was to examine my own leadership in the context of the community inclusive preschool that I own and operate along with my business partner. The focus was to build a collaborative organization through a shared vision and improved staff skill capacity. In order to implement the change process, I chose the specific learning environment of our instructional student centers (i.e., "zones") as the setting to conduct my action research project. The study describes both my personal leadership theory as well as an analysis of my leadership actions (i.e., theory in use) in order to …

A Phenomenological Study Of Falling Out Of Romantic Love, Joanni L. Sailor
A Phenomenological Study Of Falling Out Of Romantic Love, Joanni L. Sailor
The Qualitative Report
Romantic love is considered a necessary ingredient in marriage. In this study, the experience of falling out of romantic love with one’s spouse was examined. Eight individuals who had fallen out of romantic love with their spouse were interviewed. By using Moustakas’ Transcendental Phenomenological method, several themes emerged which provided a description of the experience of the phenomena. These themes included loss of trust, of intimacy, and of feeling loved; emotional pain; and negative sense of self. Gradual decline was identified as a slow, progressive deterioration of the relationship in which over time the romantic love decreased and eventually ended. …

Researchers across disciplines are increasingly utilizing electronic tools to collect, analyze, and organize data. However, when it comes to publishing their work, there are no common, well-established standards on how to make that data available to other researchers. Consequently, data are often not stored in a consistent manner, making it hard or impossible to find data sets associated with an article – even though such data might be essential to reproduce results or to perform further analysis. Data repositories can play an important role in improving this situation, offering increased visibility, domain-specific coordination, and expert knowledge on data management. As …
Water Soluble Polymer Solar Cells From Electrospray Deposition, Marshall Sweet
Water Soluble Polymer Solar Cells From Electrospray Deposition, Marshall Sweet
Theses and Dissertations
This dissertation reports the fabrication and characterization of thin films from the water soluble polymer sodium poly[2-(3-thienyl)-ethyloxy-4-butylsulfonate] (PTEBS) by electrospray deposition (ESD). Contiguous thin films were created by adjusting the parameters of the electrospray apparatus and solution properties to maintain a steady Taylor cone for uniform nanoparticle aerosolization and controlling the particle water content to enable coalescence with previously deposited particles. The majority of deposited particles had diameters less than 52 nm. A thin film of 64.7 nm with a root mean square surface roughness of 20.2 nm was achieved after 40 minutes of ESD. Hybrid Solar Cells (HSCs) with …

Dikar-Induced Synthetic Lethality In A Drosophila Model Of Cag Repeat Diseases Does Not Result From An Expression Feedback Loop, Daniel Camacho
Dikar-Induced Synthetic Lethality In A Drosophila Model Of Cag Repeat Diseases Does Not Result From An Expression Feedback Loop, Daniel Camacho
Honors Scholar Theses
Human CAG repeat diseases manifest themselves through the common pathology of neurodeneration. This pathological link is attributed to the property shared by all nine of these diseases: an expanded polyglutamine (polyQ) tract. The most evident result of polyQ expansion is protein aggregation, and it is believed that this phenomenon is partly responsible for conferring cytotoxic properties on the mutated protein. Apart from sequestering the mutated protein, cellular aggregates are able to incorporate native proteins via polyQ-mediated aggregation, thus disrupting important cellular pathways. Using Drosophila melanogaster as a disease model, researchers have been able to compile collections of these so-called disease …
From Demonic Possession To Conversion Disorder: A Historical Comparison, Laura Hatchman
From Demonic Possession To Conversion Disorder: A Historical Comparison, Laura Hatchman
Honors Scholar Theses
My thesis compares analyses of early-modern understandings of demonic possession in the West to discussion of modern cases of mass conversion disorder, specifically focusing on episodes involving large groups of affected individuals. My research on the early modern period includes a literature review of the famous seventeenth-century case of mass possession at Loudun and a lengthy examination of possession from the vantage point of key societal players. On a parallel track, I have identified and recent psychological and medical literature pertaining to modern cases of what is called “mass hysteria,” “epidemic hysteria,” “conversion disorder,” and “mass psychogenic illness.” By examining …
Investigations In Enhancing The Reproducibility Of Implantable Amperometric Glucose Biosensors For Mass Production, Dipesh Manharbhai Patel
Investigations In Enhancing The Reproducibility Of Implantable Amperometric Glucose Biosensors For Mass Production, Dipesh Manharbhai Patel
Honors Scholar Theses
Implantable glucose sensors for Diabetes management should possess several attributes such as linearity, sensitivity, selectivity, long life time and should elicit no negative tissue response. Based on this, the UConn implantable glucose sensor team has developed a 5-layer sensor architecture that simultaneously affords the aforementioned requirements. However, the large scale production of these 5-layer sensors is inhibited by the lack of high sensor-to-sensor reproducibility. Herein, we investigate the origin of sensor-to-sensor irreproducibility and develop methodologies for large-scale fabrication of these devices.
Pharmacological Inhibition Of Histone Methyltransferase G9a Affects Expression Of Citron Kinase Target Genes In Neural Stem Cells, Nicholas Heliotis
Pharmacological Inhibition Of Histone Methyltransferase G9a Affects Expression Of Citron Kinase Target Genes In Neural Stem Cells, Nicholas Heliotis
Honors Scholar Theses
Regulated repression of gene expression by post-translational modification of histones is required for normal development. The histone methyltransferase G9a is essential for embryonic development, and we have shown that phosphorylation of G9a by the CitK, a gene required for normal CNS development, gates gene repression and dimethylation of histone H3 at lysine 9 (H3K9me2) in neural progenitors. CitK and G9a co-localize to promoter regions of genes up-regulated in CitK null cells. CitK mutant progenitors lack H3K9me2 at promoter regions of up-regulated genes, and re-expression of CitK restores both repression of gene expression and H3K9me2 occupancy. In this thesis we examine …

A Detergent-Free Strategy For The Reconstitution Of Active Enzyme Complexes From Native Biological Membranes Into Nanoscale Discs, Ashley R. Long, Catherine C. O’Brien, Ketan Malhotra, Christine T. Schwall, Arlene D. Albert, Nathan N. Alder
Open Access Author Fund Awardees' Articles
Background
The reconstitution of membrane proteins and complexes into nanoscale lipid bilayer structures has contributed significantly to biochemical and biophysical analyses. Current methods for performing such reconstitutions entail an initial detergent-mediated step to solubilize and isolate membrane proteins. Exposure to detergents, however, can destabilize many membrane proteins and result in a loss of function. Amphipathic copolymers have recently been used to stabilize membrane proteins and complexes following suitable detergent extraction. However, the ability of these copolymers to extract proteins directly from native lipid bilayers for subsequent reconstitution and characterization has not been explored.
Results
The styrene-maleic acid (SMA) copolymer effectively …

The Role Of Gesture In Spatial And Non-Spatial Learning In Children And Adults, Gwendolyn Marrin, Amanda Olsen
The Role Of Gesture In Spatial And Non-Spatial Learning In Children And Adults, Gwendolyn Marrin, Amanda Olsen
Psychology Student Work
The purpose of this study was to identify whether gesture differentially affects spatial or non-spatial language comprehension and memory in both children and adults. Previous research (i.e. Goldin-Meadow, 1996, 2001; McNeill, 1992, 2005) examines only the role of gesture in learning without addressing development. For this study, participant’s baseline gesture rate is recorded and then stories containing either spatial or non-spatial components are read to the participants. Participants then receive factual and convergent comprehension questions. While answering, they either gesture naturally, are required to gesture, or are told not to gesture. A three-term inference problem task (Knauff & Johnson-Laird, 2002) …

Test Anxiety And Nursing Students., Brad Moore
Undergraduate Honors Theses
Test anxiety has been a problem for many individuals not only in the workforce, but also in many schools and colleges (Driscoll, Evans, Ramsey & Wheeler 2009). According to Driscoll et al., when compared to high school students and the general public (17%), nursing students are shown to have over double (55-60%) the rate of moderately high to high test anxiety. Cognitive test anxiety can account for a 7 to 8% drop in test grades, which can drop test score’s an entire letter grade (Cassady & Johnson, 2001). The purpose of this research is to explore the level of test …

Soliton Solutions Of The Nonlinear Schrödinger Equation., Erin Middlemas
Soliton Solutions Of The Nonlinear Schrödinger Equation., Erin Middlemas
Undergraduate Honors Theses
The nonlinear Schrödinger equation is a classical field equation that describes weakly nonlinear wave-packets in one-dimensional physical systems. It is in a class of nonlinear partial differential equations that pertain to several physical and biological systems. In this project we apply a pseudo-spectral solution-estimation method to a modified version of the nonlinear Schrödinger equation as a means of searching for solutions that are solitons, where a soliton is a self-reinforcing solitary wave that maintains its shape over time. The pseudo-spectral method estimates solutions by utilizing the Fourier transform to evaluate the spatial derivative within the nonlinear Schrödinger equation. An ode …
Reactive Intermediates In Hypoxia-Selective Dna Damage., Olivia Miller
Reactive Intermediates In Hypoxia-Selective Dna Damage., Olivia Miller
Undergraduate Honors Theses
A group of prospective drugs with the aromatic di-N-oxide (ANO) functionality as the common feature are currently undergoing testing for the ability to selectively target tumors surrounded by normal tissues. It has been long recognized that the mechanism of biological activity of these drugs involves DNA damage by free radical species generated through one-electron reduction, although the exact nature of the reactive intermediate responsible for DNA damage remains uncertain. It is believed, however, that one of the key factors defining, in particular, hypoxic selectivity of these drugs is the rate of N-O bond scission in the one-electron reduced intermediate. In …

Examining The Role Of L-Arginine In Tissues Of The Fetoplacental Unit And Endometrium, Jonathan Michael Greene
Theses and Dissertations
L-arginine is one of the most versatile amino acids due to the fact that it serves as a precursor for many molecules which have important roles in bodily functions including mammalian reproduction. The current studies sought to further examine the role that L-arginine has in mammalian reproduction utilizing both in vivo and in vitro approaches. In the first study, a novel bioluminescent murine pregnancy model was developed to monitor VEGFR2 transcription activity non-invasively in the fetoplacental unit. Secondly, the effect that dietary L-arginine supplementation has during mouse gestation was examined. L-arginine supplementation increased weight gain during the latter third of …
