Quantitative real-time PCR (qRT-PCR), a primary tool in gene expression analysis, requires an appropriate normalization strategy to control for variation among samples. The best option is to compare the mRNA level of a target gene with that of reference gene(s) whose expression level is stable across various experimental conditions. In this study, expression profiles of eight candidate reference genes from the diamondback moth, Plutella xylostella, were evaluated under diverse experimental conditions. An enduring controversy over the nature of ADHD complicates parents’ decisions regarding children likely to be diagnosed with the condition. Using a fallibilist perspective, this review examines how researchers construe ADHD and acknowledge the controversy. From a systematic literature search of empirical reports using parents of ADHD-diagnosed children as primary informants, 36 reports published between 1996 and 2008 (corresponding to 30 studies) were selected. Data on the studies’ characteristics and methodologies, definitions of ADHD, and extent of the acknowledgment of the ADHD controversy were extracted, as were data on a wide range of parental concerns and experiences. We present the case of an 8 year-old-girl with a 3-week history of visual conversion, inability to walk, intermittent urinary incontinence, and non-epileptic seizures. Although she had a history of shaken baby syndrome that resulted in hydrocephalus requiring bilateral ventriculoperitoneal (VP) shunt placement at 3 months of age, this current episode could not be attributed to any organic causes and hence she was diagnosed with conversion disorder with mixed presentation. Throughout the ages, treatment of the mentally ill has evolved with distinct periods of progression, stagnation and regression. These differences in time mirrored how society understood mental illness and the mentally ill person on both a biological and interpersonal level. At various points in history, treatment of the mentally ill included cruel and inhumane acts, while at other times, consisted of compassionate and benevolent care. Increasing numbers of studies are reporting phenotypic and functional heterogeneity within a variety of stem cell types as well as the existence of inter-convertible subsets within hematopoietic stem cells (HSCs), suggesting that these cells are alternating between several metastable states. The extent of heterogeneity and metastability in HSCs remains to be explored in depth. The metastability of HSCs is most likely positively and negatively regulated by currently unknown cell intrinsic and extrinsic mechanisms emanating from HSC niches. The host immune system plays an important role in the development and control of cancer, yet cancer cells often elaborate mechanisms to evade this regulation. Transforming growth factor beta (TGFβ) is a pleiotropic cytokine often overexpressed by cancers due to TGFβ’s growth-promoting effect on tumor cells and suppressive function on multiple cell types of the innate and adaptive immune systems. Genetic ablation of TGFβ signaling induces significant and toxic immune responses against host tissues, but when signaling is selectively ablated in cells of the adaptive immune system, such as CD8+ T cells, they become capable of mediating protection against various …

Electron Transfer By Excited Benzoquinone Anions: Slow Rates For Two-Electron Transitions, Matibur Zamadar, Andrew R. Cook, Anna Lewandowska-Andralojc, Richard Holroyd, Yan Jiang, Jin Bikalis, John R. Miller

Electron transfer (ET) rate constants from the lowest excited state of the radical anion of benzoquinone, BQ−•*, were measured in THF solution. Rate constants for bimolecular electron transfer reactions typically reach the diffusion-controlled limit when the free-energy change, ΔG°, reaches −0.3 eV. The rate constants for ET from BQ−•* are one-to-two decades smaller at this energy and do not reach the diffusion-controlled limit until −ΔG° is 1.5−2.0 eV. The rates are so slow probably because a second electron must also undergo a transition to make use of the energy of the excited state. Similarly, ET, from solvated electrons to neutral …

As a part of the overall program in the Grandy laboratory at Oregon Health & Science University (OHSU), studying the underlying chemical biology of methamphetamine (Meth) addiction, this dissertation reports on the development of six new thyronamine analogs which were synthesized and assayed against trace amine associated receptor 1 (TAAR1), giving preliminary results consistent with the analogs being inverse agonists. Due to highly variable TAAR1 expression levels in the assays, based on inter-assay response to control Meth stimulation as well as other possible factors, kinetic models were developed to qualitatively explain the assay results. Holocarboxylase synthetase (HLCS) is the sole human protein biotin ligase in the human proteome. Other than its role as a coenzyme to the five carboxylases in intermediary metabolism, HLCS and biotin are involved in epigenetic gene regulation and immune response. Previously, it was shown that (1) HLCS physically interacts with histone methyl transferase EHMT-1 which creates histone H3 lysine 9 methylation (H3K9me) marks and biotinylates lysine (K) 161 in EHMT-1, thereby strenghtening the HLCS/EHMT-1 interaction; (2) biotin depletion and HLCS knockdown cause a loss of H3K9me marks leading to de-repression of long terminal repeats (LTRs) and chromosomal abnormalities; and (3) …

Abstract

Endocrine disrupting substances (EDSs) have the potential to disrupt sensitive hormone pathways, including those involved in development and reproduction. Both fresh and estuarine water bodies receive inputs of EDSs from a variety of sources, including sewage effluent, industrial effluent, and agricultural runoff. Based on limited available literature, freshwater species appear to respond to lower levels of EDSs than estuarine or marine species. To address this potential difference, a short-term reproductive bioassay was carried out under low and high salinity conditions using mummichog (Fundulus heteroclitus), a euryhaline species that is native to the east coast of North America. …

Termites are highly eusocial insects and show a division of labor whereby morphologically distinct individuals specialize in distinct tasks. In the lower termite Reticulitermes flavipes(Rhinotermitidae), non-reproducing individuals form the worker and soldier castes, which specialize in helping (e.g., brood care, cleaning, foraging) and defense behaviors, respectively. Workers are totipotent juveniles that can either undergo status quo molts or develop into soldiers or neotenic reproductives. This caste differentiation can be regulated by juvenile hormone (JH) and primer pheromones contained in soldier head extracts (SHE). It has been reported that an individual’s initial level of response to a drug might be predictive of his or her future risk of becoming dependent, thus basal gene expression profiles underlying those drug responses may be informative for both predicting addiction susceptibility and determining targets for intervention. This dissertation research aims to elucidate genetic risk factors underlying acute alcohol and nicotine dependence phenotypes using mouse genetic models of addiction. Phenotyping, brain region-specific mRNA expression profiling, and genetic mapping of a recombinant inbred panel of over 25 mouse strains were performed in order to identify quantitative trait loci (QTL) harboring …

While on the surface mainstream discussions about race appear to encompass the values and ideologies associated with egalitarianism, subtle and at times not so subtle discussions continue to disparage groups and individuals who have been historically associated with the minority. The growing existence of hate groups illustrates that arguments for these extreme ideologies are not only present but for some individuals are gaining in acceptance. Analyzing the interaction between the lipid membranes and nanoparticles helps to determine beneficial and harmful effects of nanoparticles on cells. Since the point of entry of cells is their plasma membrane, interactions between nanorods and lipid membranes were studied using molecular dynamics simulations. The effect of nanorods on membrane morphology was investigated systematically as a function of nanorods interaction with the lipid membrane, their surface coverage and number surface density. Our results indicate that when adsorbed on the membrane, the nanorods may cluster into chains and may lead to strong membrane deformations due to curvature driven interaction between nanorods. At …

The multisubunit eukaryotic Mediator complex integrates diverse positive and negative gene regulatory signals and transmits them to the core transcription machinery. Mutations in individual subunits within the complex can lead to decreased or increased transcription of certain subsets of genes, which are highly specific to the mutated subunit. Recent studies suggest a role for Mediator in epigenetic silencing. Using white-opaque morphological switching in Candida albicans as a model, we have shown that Mediator is required for the stability of both the epigenetic silenced (white) and active (opaque) states of the bistable transcription circuit driven by the master regulator Wor1. Abstract: Nine mayfly species (Insecta: Ephemeroptera) that may be of conservation concern in South Carolina are discussed. Three such species associated with sand-bottomed streams are Acanthametropus pecatonica (Burks, 1953), Dolania americana Edmunds & Traver, 1959 and Homeoneuria dolani Edmunds, Berner & Traver, 1958. Three species of potential concern are associated with Hornleaf Riverweed (Podostemum ceratophyllum Michaux, 1803), and they include Barbaetis benfieldi Kennedy, 1985, Heterocloeon berneri (Muller-Liebenau, 1974) and Tsalia berneri (Allen & Edmunds, 1958). Ernest Everett Just, a pioneering American biologist, discovered the fundamental role of the environment in the development of embryos. His work led to the creation of the area of biology known as ecological developmental (Eco-Devo) biology. However, both his work and the context of his scientific contributions are not widely known. His work covered a diversity of fields of biology, including marine biology, cytology, and parthogenesis (asexual reproduction where growth and development of embryos occur without fertilization). The rapid expansion of computer technology in the past thirty years has impacted society in innumerable ways. Of interest for this discussion is the breath-taking change in measurement sciences that has occurred as the digital age dawned and has now become part of our daily lives. In particular, the effect on image-based, non-contacting measurements will be described and the continuing revolutionary impact that the resulting technology has engendered will be reviewed. Reduced glomerular filtration rate and increased albuminuria often develop in Type 2 Diabetes Mellitus (T2DM) and are predictive of chronic kidney and cardiovascular disease. Risk for renal disease in T2DM has been associated with cardiometabolic risk factors including, hypertension, dyslipidemia, and hyperglycemia. Weight loss is associated with improved outcomes in T2DM patients with existing renal disease and may also reduce risk in patients without renal disease via reduction of cardiometabolic risk factors.
