The delta smelt (Hypomesus transpacificus) is a small, pelagic fish endemic to the San Francisco Estuary (SFE) and protected under federal and state endangered species acts. This study examines the diet of adult delta smelt during their spawning migration in the winters of 2010 and 2012. Delta smelt and their zooplankton prey were sampled concurrently during a seasonal pulse of turbidity at sites along their migratory route from the low salinity zone in Suisun Bay to the fresher waters of the Sacramento/San Joaquin Delta. The research is testing the limits of detection and multiplexing with the use of three pathogens with the machine created from Luminex called Magpix. The pathogens are Salmonella enterica, Listeria monocytogenes, and Escherichia Coli O157. The gene that is being focused on for each pathogen is invA for Salmonella, hlyA for listeria, and hlyA for Escherichia Coli. In this research, we started with prepared probes that were attached to the magnetic microsphere beads. Then we extracted and amplify the DNA. Furthermore, the probes were combined to the amplified DNA and used this in Magpix which gave a corresponding MFI value.

Leatherback hatchling fitness on land is essential for their success in entering the ocean without being predated. Two ways fitness on land may be measured are by how fast hatchlings crawl as well as how fast their righting responses are when they are flipped onto their carapaces. Hatchlings from certain nests have slower righting response times along with slower crawl times, both of which would increase the odds of predation and hinder the success of hatchlings reaching the ocean.
Hatchling crawling speed was determined by timing 12 randomly selected hatchlings from each nest to crawl a distance of two meters. …

Ligament and tendon repair involves a complex series of coordinated events orchestrated by various cell types, cytokines and other factors. The repair process extends months to years and results in scar tissue mechanically inferior to native tissue. This is in clear contrast to tissue "regeneration" which would recapitulate the native tissue. Numerous approaches to stimulate a regenerative scenario have been attempted, including tissue-engineering, non-steroidal anti-inflammatories, ultrasonic, or electrical stimulation, but none have resulted in complete regeneration. In pursuit of a more regenerative outcome, it is important to understand the fundamentals of the normal healing process. Previous work from our lab …

This report focuses on Australian students' performance on the PISA items that have been released in Scientific Literacy. It includes an overview of the PISA Scientific Literacy Framework, Australian students' results in the 2006 international assessment, examples of responses and marking guides and the context behind achievement, e.g. attitudes, engagement and motivation, and recognition of environmental issues.

Adult American horseshoe crabs, Limulus polyphemus, possess endogenous circadian and circatidal clocks controlling visual sensitivity and locomotion, respectively. The goal of this study was to determine the types of activity rhythms expressed by juvenile horseshoe crabs (n = 24) when exposed to a 14:10 light/dark cycle (LD) for 10 days, followed by 10 days of constant darkness (DD). Horseshoe crab activity was recorded with a digital time-lapse video system that used an infrared-sensitive camera so animals could be monitored at night. In LD, 15 animals expressed daily patterns of activity, 6 displayed a circatidal pattern, and the remaining 3 were …

This exploration is rooted in the vision of a democratic classroom in which all voices are heard and takes place in an early years setting, or community of practice, in Dublin Ireland. The aim of the study is to explore what it means to belong to this particular community of practice from multiple perspectives. The objective is twofold: to respond to our ethical commitment as teachers, and to explore the sense of belonging in a community of practice. Evidence For A Novel Mechanism Of The Pak1 Interaction With The Rho-Gtpases Cdc42 And Rac, Yong Jae Shin, Eun Hye Kim, Adhiraj Roy, Jeong-Ho Kim

P21-activated kinase 1 (PAK1) is activated by binding to GTP-bound Rho GTPases Cdc42 and Rac via its CRIB domain. Here, we provide evidence that S79 in the CRIB domain of PAK1 is not directly involved in this binding but is crucial for PAK1 activation. S79A mutation reduces the binding affinity of PAK1 for the GTPases and inhibits autophosphorylation and kinase activity of PAK1. Thus, this mutation abrogates the ability of PAK1 to induce changes in cell morphology and motility and to promote malignant transformation of prostate epithelial cells. The contractile effects of neurotensin (NT) and cholecystokinin octapeptide (CCK-8) on isolated circular smooth muscle strips of chicken gallbladder were investigated. The NT (0.25-300 nM) produced concentration-dependent contractions on smooth muscle with an EC50 of 8.5 nM (95% confidence limits = 5.3-13.6 nM). In comparison, CCK-8 produced concentration-dependent contractions with an EC50 of 13 nM (95% confidence limits of 9-20 nM). There were no statistical differences in contractile responses when comparing NT and CCK-8 at equimolar concentrations. The NT appears to act directly on smooth muscle tissue in the chicken; the contractile responses were not blocked by 10 μM atropine …

Over the past two years, through an NSF RCN UBE grant, the ASBMB has held regional workshops for faculty members and science educators from around the country that focused on identifying: 1) core principles of biochemistry and molecular biology, 2) essential concepts and underlying theories from physics, chemistry, and mathematics, and 3) foundational skills that undergraduate majors in biochemistry and molecular biology must understand to complete their major coursework. Childhood obesity is a growing problem. As more researchers become involved in the study of parenting influences on childhood obesity, there appears to be a lack of agreement regarding the most important parenting constructs of interest, definitions of those constructs, and measurement of those constructs in a consistent manner across studies. This article aims to summarize findings from a working group that convened specifically to discuss measurement issues related to parental influences on childhood obesity. In the spring of 2013, the Maine Learning Technology Initiative (MLTI) adopted a new approach to its 1:1 device selection process. Rather than choosing a single vendor to provide a uniform statewide technology implementation, the Maine Department of Education opted to select a preferred solution and allowed schools the autonomy to choose from among four other approved bids. In anticipation that a number of schools would choose to provide iPads to students, the Maine Education Policy Research Institute (MEPRI) at the University of Southern Maine surveyed teachers at three Maine high schools currently using 1:1 iPads with their students.

Autosomal dominant leukodystrophy (ADLD) is an adult onset demyelinating disorder that is caused by duplications of the lamin B1 (LMNB1) gene. However, as only a few cases have been analyzed in detail, the mechanisms underlying LMNB1 duplications are unclear. We report the detailed molecular analysis of the largest collection of ADLD families studied, to date. We have identified the minimal duplicated region necessary for the disease, defined all the duplication junctions at the nucleotide level and identified the first inverted LMNB1 duplication. Social and behavioral research in public health is often intimately tied to profound, but frequently neglected, biological influences from underlying genetic, environmental, and epigenetic events. The dynamic interplay between the life, social, and behavioral sciences often remains underappreciated and underutilized in addressing complex diseases and disorders and in developing effective remediation strategies. Using a case-study format, we present examples as to how the inclusion of genetic, environmental, and epigenetic data can augment social and behavioral health research by expanding the parameters of such studies, adding specificity to phenotypic assessments, and providing additional internal control in comparative studies. Eastern hemlock (Tsuga canadensis) is in rapid decline because of infestation by the invasive hemlock woolly adelgid (Adelges tsugae; 'HWA') and, to a lesser extent, the invasive elongate hemlock scale (Fiorinia externa; 'EHS'). For many conifers, induced oleoresin-based defenses play a central role in their response to herbivorous insects; however, it is unknown whether eastern hemlock mobilizes these inducible defenses. We conducted a study to determine if feeding by HWA or EHS induced changes in the volatile resin compounds of eastern hemlock. This research concerns the process of college women's emotional labor in sexual decision-making and how women talk about their sexual decisions via their negotiation of dominant social discourses. The study seeks to determine how the college experience nurtures women's sexual subjectivity; how it affects their negotiation of sexual messaging and social dominant discourse; and how it impacts the emotional work involved in the process. The study was conducted at three college campuses that vary relative to institutional type (i.e., public vs. private), social class, and race.
