Tall Fescue Endophyte Effects On Tolerance To Water-Deficit Stress, Padmaja Nagabhyru, Randy D. Dinkins, Constance L. Wood, Charles W. Bacon, Christopher L. Schardl

Plant Pathology Faculty Publications

BACKGROUND: The endophytic fungus, Neotyphodium coenophialum, can enhance drought tolerance of its host grass, tall fescue. To investigate endophyte effects on plant responses to acute water deficit stress, we did comprehensive profiling of plant metabolite levels in both shoot and root tissues of genetically identical clone pairs of tall fescue with endophyte (E+) and without endophyte (E-) in response to direct water deficit stress. The E- clones were generated by treating E+ plants with fungicide and selectively propagating single tillers. In time course studies on the E+ and E- clones, water was withheld from 0 to 5 days, during which …

The Flexible Fairness: Equality, Earned Entitlement, And Self-Interest, Chunliang Feng, Yi Luo, Ruolei Gu, Lucas S. Broster, Xueyi Shen, Tengxiang Tian, Yue-Jia Luo, Frank Krueger

Behavioral Science Faculty Publications

The current study explored whether earned entitlement modulated the perception of fairness in three experiments. A preliminary resource earning task was added before players decided how to allocate the resource they jointly earned. Participants' decision in allocation, their responses to equal or unequal offers, whether advantageous or disadvantageous, and subjective ratings of fairness were all assessed in the current study. Behavioral results revealed that participants proposed more generous offers and showed enhanced tolerance to disadvantageous unequal offers from others when they performed worse than their presumed "partners," while the reverse was true in the better-performance condition. The subjective ratings also …

Biology And Control Of Common Purslane (Portulaca Oleracea L.), Christopher A. Proctor Sep 2013

Biology And Control Of Common Purslane (Portulaca Oleracea L.), Christopher A. Proctor

Department of Agronomy and Horticulture: Dissertations, Theses, and Student Research

Common purslane (Portulaca oleracea L.) is a summer annual with wide geographic and environmental distribution. Purslane is typically regarded as a weed in North America, but it is consumed as a vegetable in many parts of the world. One of the characteristics that make purslane difficult to control as a weed is its ability to vegetatively reproduce. Severed sections of purslane stem containing a node will produce adventitious roots from the cut end of the stem. Isoxaben and simazine were the only two effective preemergence herbicides for controlling purslane in our studies when applied at maximum or one-half maximum …

The proteasome is a key regulator of cellular protein homeostasis and is a clinically validated anticancer target. The immunoproteasome, a subtype of proteasome expressed mainly in hematopoietic cells, was initially recognized for its role in antigen presentation during the immune response. Recently, the immunoproteasome has been implicated in several disease conditions including cancer and autoimmune disorders, but many of the factors contributing to these pathological processes remain unknown. In particular, the codon 60 polymorphism of the PSMB9 gene encoding the β1i immunoproteasome catalytic subunit has been investigated in the context of a variety of diseases. Despite this, previous studies have …

An Exploration Of Language Used In Domestic Minor Sex Trafficking, Margot K. Goodnow Sep 2013

An Exploration Of Language Used In Domestic Minor Sex Trafficking, Margot K. Goodnow

Theses, Dissertations, and Projects

This theoretical investigation explored the effect of pejorative labels on victims of domestic minor sex trafficking. Presently, children involved in domestic minor sex trafficking are perceived either as "victims" in need of services, or as "criminals," juvenile prostitutes, deserving punitive action. Due to the age and compounded vulnerability of this population, understanding the negative effect this can have on victims' identity development is crucial to providing appropriate and deserved treatment. This study utilized relational-cultural theory and trauma-focused cognitive behavioral theory to understand the impact of the criminalization of complex trauma and to provide a framework for clinical interventions with this …

An Examination Of Marine Snow As A Pathway For Mercury Uptake In The Blue Mussel Mytilus Edulis, Veronica L. Ortiz Sep 2013

An Examination Of Marine Snow As A Pathway For Mercury Uptake In The Blue Mussel Mytilus Edulis, Veronica L. Ortiz

Master's Theses

Bioaccumulation of mercury (Hg), especially as methylmercury (CH3Hg), in the aquatic food web is a continuing public health concern. Extensive research has been conducted regarding the uptake of Hg by various aquatic animals via three main vectors: sediment, bioseston, and water, but virtually no research exists with respect to uptake of CH3Hgor inorganic mercury (Hg(II)) associated with marine snow. Marine snow composed of various organic and inorganic materials has been studied as an effective vertical transporter of nutrients and metals scavenged from the euphotic zone to the benthos. As various aquatic species utilize marine aggregates as …

Publications and Research

Centrosomal kinase Nek2 is overexpressed in different cancers, yet how it contributes toward tumorigenesis remains poorly understood. dNek2 overexpression in a Drosophila melanogaster model led to upregulation of Drosophila Wnt ortholog wingless (Wg), and alteration of cell migration markers—Rho1, Rac1 and E-cadherin (Ecad)—resulting in changes in cell shape and tissue morphogenesis. dNek2 overexpression cooperated with receptor tyrosine kinase and mitogen-activated protein kinase signaling to upregulate activated Akt, Diap1, Mmp1 and Wg protein to promote local invasion, distant seeding and metastasis. In tumor cell injection assays, dNek2 cooperated with Ras and Src signaling to promote aggressive colonization of tumors into different …

Pf2Α And Pge2 Their Effects On Cell Respiration And Steroidogensis In The Bovine Corpus Luteum, Charlotte B. Bacon Sep 2013

Pf2Α And Pge2 Their Effects On Cell Respiration And Steroidogensis In The Bovine Corpus Luteum, Charlotte B. Bacon

Doctoral Dissertations

Mitochondria are essential to the process of cell respiration and steroidogenesis. Both of these processes impact the viability of the corpus luteum, which is required to maintain early pregnancy. In cattle and other species, progesterone supports gestation, regulates embryogenesis and works in concert with estrogen to maintain the reproductive tract. Many investigators have sought to understand the processes that control regression of the corpus luteum. Two prostaglandins, PGF2a and PGE2, have been implicated in the functioning of this transient gland. It is accepted by most researchers that the primary luteolysin responsible for functional and structural demise of the corpus luteum …

Estrogen And Breast Cancer: Understanding The Complex Interaction One Gene At A Time, Victoria Harris, Dr. Chin-Yo Lin Sep 2013

Estrogen And Breast Cancer: Understanding The Complex Interaction One Gene At A Time, Victoria Harris, Dr. Chin-Yo Lin

Journal of Undergraduate Research

Estrogen is a steroid hormone that acts in the body in both beneficial and detrimental ways. It plays a crucial role in many human processes including coagulation, bone formation, and development of sexual organs. However, estrogen is also associated with the proliferation of breast cancer. Estrogen activates the estrogen receptor (ER), a ligand-dependent transcription factor that regulates gene expression by binding to specific DNA elements and promoting or repressing transcription.

Identifying The Mechanisms Of Estrogen Receptors In Human Breast Cancer Cells Through Study Of Direct Target Gene Expression, Megan Lundell, Dr. Chin-Yo Lin Sep 2013

Identifying The Mechanisms Of Estrogen Receptors In Human Breast Cancer Cells Through Study Of Direct Target Gene Expression, Megan Lundell, Dr. Chin-Yo Lin

Journal of Undergraduate Research

Estrogens, including estrodial (E2), are known to play a significant role in a number of developmental and physiological processes including the genesis and progression of breast cancer. The action of estrogens is mediated by estrogen receptors (ERs), which function as ligand-dependent transcription factors. Upon ligand binding, ERs interact with cis-regulatory elements of target genes either directly by binding to estrogen response elements or indirectly through interaction with transcription factor complexes and their respective binding sites.1 The primary objective of this project involved the knockdown, or reduction, of NCor, a co-repressor protein known to associate with ER in the presence of …

Understanding Cancer Metastasis: The Role Of Zyxin In Actin Dynamics, Nicholas H. Bishop Sep 2013

Understanding Cancer Metastasis: The Role Of Zyxin In Actin Dynamics, Nicholas H. Bishop

Journal of Undergraduate Research

My research aims to elucidate the protein interactions at cell-cell contacts (adherens junctions or AJs) by studying the fluorescence recovery of three known AJ proteins (zyxin, VASP, and actin) after photobleaching. At AJs, zyxin interacts with VASP (Vasioukhin et al., 2000) which in turn encourages the polymerization of actin (Vasioukhin et al., 2000). Mutation of certain regions of zyxin may reveal their role in zyxin-VASP binding and regulation. The effects of altering VASP activity should be evident in actin turnover rate. In Fluorescence Recovery after Photobleaching (FRAP), regions of cell-cell adhesions with fluorescently labeled zyxin, VASP, or actin are bleached …

Structure And Properties Of Dynamical Structure Functions Reconstruction Of Genetic Networks Using Gene Silencing And Overexpression Methods, Russell Howes, Dr. Sean Warnick Sep 2013

Structure And Properties Of Dynamical Structure Functions Reconstruction Of Genetic Networks Using Gene Silencing And Overexpression Methods, Russell Howes, Dr. Sean Warnick

Journal of Undergraduate Research

Network structure–the presence and absence of ‘connections’ among different variables in a dynamical system–is important in the identification and control of complex systems. Discovering structure from data can be difficult, and many current identification methods either concern themselves only with dynamic behavior (response of the system to an input), or try to infer structure without dynamics.

Species Composition Of A Desert Rock Pool Metacommunity, Cameron Kmetzsch, Russell B. Rader Sep 2013

Species Composition Of A Desert Rock Pool Metacommunity, Cameron Kmetzsch, Russell B. Rader

Journal of Undergraduate Research

Rock pools are one of the most unique and threatened aquatic habitats in the world. Changes in global climate could alter precipitation patterns and disrupt the annual cycle of filling and drying to which this community has evolved. Previous misconceptions assume that all rock pools are temporary habitats. Over a period of two years we have shown that at least 20 pools in three drainages have maintained constant wetted conditions with moderate seasonal fluctuations in water levels, despite air temperatures near 50º C (120º F) on the open hardpan. These “permanent” pools (Tinajas) are shaded within deeply incised channels. Tinajas …

Culture And Identification Of Bone Marrow Hematopoietic Cells, Joshua Hauser, Dr. Sandra Burnett Sep 2013

Culture And Identification Of Bone Marrow Hematopoietic Cells, Joshua Hauser, Dr. Sandra Burnett

Journal of Undergraduate Research

Successfully culturing and identifying bone marrow cells is a critical skill when doing stem cell, or hematopoietic research. This skill was especially important in Dr. Sandra Burnett’s research lab, with whom I conducted this research. Since the proper culture and identification of the cells was a vital part of any results or conclusions that were made in the lab, we deemed it imperative to determine whether or not we would be able to develop training protocols that would result in better and more consistent results. I strongly believed that this project would immediately impact our current and future research using …

Gene Expression Of Down-Regulated Estrogen Receptor Target Genes In Human Breast Cancer Cells Following Drug Rreatment, Ben Newman, Dr. Chin-Yo Lin Sep 2013

Gene Expression Of Down-Regulated Estrogen Receptor Target Genes In Human Breast Cancer Cells Following Drug Rreatment, Ben Newman, Dr. Chin-Yo Lin

Journal of Undergraduate Research

Breast cancer is the most prevalent type of cancer found in women and the majority of such cancers are estrogen receptor (ER) positive. This means the cancer is dependent on estrogen for growth. In such cancers, ER acts as a ligand-dependent transcription factor, changing gene expression when bound to estrogen. The altered expression of ER target genes in breast cancer is a likely cause of the uncontrolled cell growth.

Sinorhizobium Meliloti Adaptation In Alfalfa Bacterial Pathway Of Infection, Jacob Erickson, Dr. Joel Griffitts Sep 2013

Sinorhizobium Meliloti Adaptation In Alfalfa Bacterial Pathway Of Infection, Jacob Erickson, Dr. Joel Griffitts

Bacterial infection has been fought against successfully since the discovery of penicillin, and since then other antibiotics. Despite this, bacterial strains continue to mutate and find resistance to commonly used antibiotics. Gaining an understanding the genetic process of infection within bacteria will aid in the fight of harmful bacteria and the use of symbiotic bacteria.

A Trojan Horse Approach To Cancer Treatment: A2da And A2noon As Potential Triggered And Targeting Chemotherapeutic Drugs, Jeremy Koontz, Dr. Heidi Vollmer-Snarr Sep 2013

A Trojan Horse Approach To Cancer Treatment: A2da And A2noon As Potential Triggered And Targeting Chemotherapeutic Drugs, Jeremy Koontz, Dr. Heidi Vollmer-Snarr

Journal of Undergraduate Research

Despite the enormous amounts of time and money spent to research effective cures for cancer—the second leading cause of death in the United States —little is known today about how to kill cancer cells without harming normal cells. Current traditional forms of cancer treatment are chemotherapy, radiotherapy, and invasive surgery. However, the challenge with such treatments is they do not discriminate between normal cells and cancerous cells; these therapies struggle to balance toxicity and efficiency. In comparison, photodynamic therapy (PDT) offers a more favorable alternative to traditional methods of cancer treatment. In PDT drugs, called photosensitizers, are activated through specific …

Confirming Artificial Fungal Endophyte Inoculation Of Maize By Aflp And Sem, Alan Chambers, Dr. Brad Geary Sep 2013

Confirming Artificial Fungal Endophyte Inoculation Of Maize By Aflp And Sem, Alan Chambers, Dr. Brad Geary

Journal of Undergraduate Research

Plant microbe interactions have been shown to enhance plant growth (Nassar, 2005) and it is well established that fungal endophytes affect beneficial phenotypes like resistance to drought or insects (Bacon 1997). Plants have been selected for beneficial phenotypic traits for thousands of years, but selection for beneficial microbe interactions is under studied. We desire to investigate improving crop yield and quality, while minimizing the expenditure of natural resources, by growing maize artificially infected with fungal endophytes from plants adapted to stressful environments. We report our early success here.

Transition Metal As Tools To Probe The Electronic Properties Of Porphyrinoids, Jillian Worlinsky Sep 2013

Transition Metal As Tools To Probe The Electronic Properties Of Porphyrinoids, Jillian Worlinsky

Doctoral Dissertations

This thesis describes in a number of separate chapters a range of studies that measure the variation of specific physical properties with structural modification of the macrocycles, with a particular focus also on using their coordinated metals as probes. Specifically, the transition metal ions are used as tools for interrogation of the electronic properties of porphyrins and pyrrole-modified porphyrins. With the aim of better understanding the electronic and structural factors that govern the optical responses, axial binding strengths, fragmentation patterns and carbonyl stretching frequencies of meso-tetraarylporphyrins, a rational design of pyrrole-modified metalloporphyrins with designed optical properties is derived.

Patient Perceptions Of Shared Decision Making: What Does It Mean And How Does It Affect Patient Outcomes?, Laura Aubree Shay Sep 2013

Patient Perceptions Of Shared Decision Making: What Does It Mean And How Does It Affect Patient Outcomes?, Laura Aubree Shay

Theses and Dissertations

Introduction: Shared decision making (SDM) has been advocated as an optimal approach to medical decision-making. Yet, little is known about how patients perceive SDM and whether patient-defined SDM is associated with patient outcomes. Methods: This three-manuscript dissertation used a mixed-methods approach including a systematic literature review and both qualitative and quantitative research methods. The aims were to: (1) systematically review the patient outcomes studied in relation to SDM and identify under what measurement contexts SDM is associated with which types of patient outcomes; (2) use in-depth, qualitative interviews to develop a conceptual model of patient-defined SDM and compare this to …
