Enzymatic Characterization Of The Ammonia Tunnel In Helicobacter Pylori Asp-Trnaasn/Glu-Trnagln Amidotransferase, Liangjun Zhao Jan 2013

Enzymatic Characterization Of The Ammonia Tunnel In Helicobacter Pylori Asp-Trnaasn/Glu-Trnagln Amidotransferase, Liangjun Zhao

Wayne State University Dissertations

The Helicobacter pylori (H. pylori) Asp-tRNAAsn/Glu-tRNAGln amidotransferase (AdT) plays important roles in indirect aminoacylation and translational fidelity; however, its inter-domain communication and ammonia delivery mechanisms are not well understood. In the present study, we investigated the three activities of H. pylori AdT (glutaminase, kinase and transamidase) and used these reactions as probes to examine the inter-domain communication and ammonia delivery mechanisms between this enzyme's two isolated active sites. We adapted and optimized an assay to kinetically characterize a series of mutations at conserved positions throughout the putative AdT ammonia tunnel. The kinase assay enabled us …

The Effect Of Sensory Impairment On Cognitive Functioning And Functional Status In Octogenarians And Centenarians, Annalise Marie Rahman Jan 2013

The Effect Of Sensory Impairment On Cognitive Functioning And Functional Status In Octogenarians And Centenarians, Annalise Marie Rahman

Wayne State University Theses

Two theories predominate to explain the covariation of cognitive and sensory functions across the lifespan: The Common Cause hypothesis and the Sensory Deprivation hypothesis. It was hypothesized that the Common Cause hypothesis better accounted for the these relationships. This hypothesis was assessed by examining the associations between sensory functioning, cognitive functioning, functional status, and motor functioning in samples of octogenarians (n = 80) and centenarians (n = 244) drawn from the Georgia Centenarian Study, Phase 3, Project 3. Special attention was given to cross-sensory-modality associations. Hierarchical regressions were also utilized to determine whether inclusion of either measured or self-reported sensory …

The Influence Of Parent-Child Conflict And Stressful Experiences On The Health Of Youth With Asthma, Erin T. Tobin Jan 2013

The Influence Of Parent-Child Conflict And Stressful Experiences On The Health Of Youth With Asthma, Erin T. Tobin

Wayne State University Theses

Attaining a clear picture of everyday family interactions is essential for understanding how family stress and conflict adversely affects children's health, especially in the context of chronic illness. Using a naturalistic observation sampling method called the Electronically Activated Recorder (EAR), we sought to investigate the effects of daily interpersonal conflicts and parental stress on pediatric asthma outcomes. We collected data from 81 children, aged 10 to 17 (M age = 12.88), and their primary caregiver as part of a larger study. Each child completed a pulmonary function test and self-report questionnaires of asthma symptoms. Asthma-related medical information was abstracted from …

Impulsivity As A Correlate Of Waterpipe Tobacco Smoking, Dinah Ayna Jan 2013

Impulsivity As A Correlate Of Waterpipe Tobacco Smoking, Dinah Ayna

Wayne State University Theses

Waterpipe tobacco smoking has become increasingly popular in the past decade, yet no studies examined personality predictors of its use. This study is the first to assess impulsivity as a potential correlate for waterpipe tobacco smoking use and frequency. 43.2% reported ever using the waterpipe, with 64% of those reporting use less than once a month. Arab students were 3-5 times more likely to have used a waterpipe. Arab ethnicity moderated the relationship between waterpipe smoking and impulsivity such that self-reported (but not laboratory) impulsivity predicted lifetime waterpipe smoking for non-Arabs, but not for Arabs. Waterpipe users were also more …

Effects Of Tocotrienol Rich Fractions On Lipid Profiles In Hemodialysis Patients, Rami Hanna Jan 2013

Effects Of Tocotrienol Rich Fractions On Lipid Profiles In Hemodialysis Patients, Rami Hanna

Wayne State University Theses

Chronic hemodialysis (HD) patients have an increased risk of cardio vascular diseases (CVD) driven by dyslipidemia, oxidative stress and inflammation. Vitamin E isomers (tochopherols and tocotrienols) are fat-soluble anti oxidants. Tocotrienol isomers (T3) are fewer studies than tochopherol isomers, but they have multifaceted effects on oxidative stress, inflammation and lipid metabolism. We investigated the lipid modifying effects of tocotrienol rich fractions (TRF) on CKD patients receiving HD in a randomized, placebo-controlled, double-blind parallel trial on 81 patients (43M, 38F). Subjects (n=41) were given 220 mg/day of either TRF (180mg TRF, comprising of 34% áT3, 3%âT3, 50% ãT3, 13%äT3 and 40 …

Life Factors And Attendance Rates For Women Enrolled In A Parenting Program, Lilia Elizabeth Mucka Jan 2013

Life Factors And Attendance Rates For Women Enrolled In A Parenting Program, Lilia Elizabeth Mucka

Wayne State University Theses

Parenting interventions consistently have been shown to improve positive parenting effectiveness, child adjustment, and family functioning (Gardner et al., 2010). However, attendance rates reported in the literature tend to be low and dropout rates tend to be high, which likely diminishes the positive impact of such programs (Dumas et al., 2007). Parenting group success begins with attendance, therefore, the study aimed to understand which life factors were associated with attendance. Specifically, the study both qualitatively and quantitatively evaluated parents' responses to a brief intervention using MI techniques by using a coding system developed by the author to understand maternal expectations …

Investigation Of Physiochemical Properties Of A Novel Gradient Calcium Polyphosphate Bone Scaffold And Its Influence On Cellular Behavior, Liang Chen Jan 2013

Investigation Of Physiochemical Properties Of A Novel Gradient Calcium Polyphosphate Bone Scaffold And Its Influence On Cellular Behavior, Liang Chen

Wayne State University Theses

A good designed bone scaffold is crucial to bone tissue engineering. We have developed and characterized a novel gradient bone scaffold by combination of Calcium Polyphosphate (CPP) with different size of porogen (stearic acids). Compared with homogenous scaffold, the gradient bone scaffold with different pore size and porosity can better mimic natural bone structure. Directly perfusion flow bioreactor was developed. This 3D dynamic cell culture was better mimic the physiological condition for cell growth. It was beneficial to nutrition and oxygen delivery throughout the whole scaffold and was able to form shear stress. We wanted to investigate the effect of …

Impact Of Folate Depletion On Expression Of Folate Metabolizing Enzymes, Yizhen Wu Jan 2013

Impact Of Folate Depletion On Expression Of Folate Metabolizing Enzymes, Yizhen Wu

Wayne State University Theses

Folate is a water-soluble vitamin B that plays a critical roles in the de novo nucleotide synthesis, DNA repair, DNA methylation, and cellular growth. By involved in the one-carbon metabolic pathways, folate influences the thymidine monophosphate (dTMP) and the purine nucleotides synthesis, consequently affect the DNA synthesis and repair. Through the one-carbon pathway, the folate level also influences the DNA methylation, consequently affect the gene expression.

In this study, we inspected the change of gene expression of 14 genes which are related to the folate metabolism in two different cell lines. One is MEF cell line and the other one …

The Potential Impact Of Federal Laws On State Water Supplies, Brad B. Castleberry, Sara R. Thornton Jan 2013

The Potential Impact Of Federal Laws On State Water Supplies, Brad B. Castleberry, Sara R. Thornton

Texas A&M Journal of Property Law

States have long held the exclusive right to allocate their surface and groundwater supplies absent some express federal authority, usually via a contract with a federal agency for stored water in a federally sponsored and funded project. Over time, however, an emphasis on federal involvement has led to scrutiny of projects where there are no federal dollars being expended—only federal permits needed to implement projects by the states. This federal involvement can sometimes have extremely costly impacts to states, effectively creating an invisible dam of laws and regulations under the guise of protecting the environment. This paper will explore a …

Development Of Peptide Inhibitors Targeting Clostridium Difficile Toxins A/B And Characterizing The Regulatory Role Of A Putative Negative Regulator Tcdc In Clostridium Difficile Toxin Gene Expression, Sanofar Jainul Abdeen Jan 2013

Development Of Peptide Inhibitors Targeting Clostridium Difficile Toxins A/B And Characterizing The Regulatory Role Of A Putative Negative Regulator Tcdc In Clostridium Difficile Toxin Gene Expression, Sanofar Jainul Abdeen

Wayne State University Dissertations

Clostridium difficile infections cause one of the most common and vital hospitalacquired

diseases often associated with broad-spectrum antibiotic usage. TcdA and TcdB

are the key virulence factors involved in major patho-physiology. While standard

antibiotics provide some respite, due to the high relapse rates and the emergence of more

severe disease presentations, antibiotics alone have often proven to be suboptimal.

Therefore there is a desperate need to develop an effective non-antimicrobial

therapeutics. Part of this work focuses on identification and further characterization of

peptide therapeutic that target the major virulence factor TcdA/TcdB. Towards

development of mechanistic-based anti-toxin agent, phage display was …

The Role Of Tissue Sound Speed As A Surrogate Marker Of Breast Density, Mark Sak Jan 2013

The Role Of Tissue Sound Speed As A Surrogate Marker Of Breast Density, Mark Sak

Wayne State University Dissertations

Breast density is one of the strongest predictors of breast cancer risk as women with the densest breasts have a three- to five-fold increase in risk compared to women with the least dense breasts. Breast density is currently measured by using mammography, the current gold standard for breast imaging. There are many shortcomings to using mammography to measure breast density, including the use of ionizing radiation. Ultrasound tomography (UST) does not use ionizing radiation and can create tomographic breast sound speed images. These sound speed images are useful because breast density is proportional to sound speed. The purpose of this …

An Exploration Of The Relationships Among Demographics, Risk Factors, Perceived Self-Efficacy, And Fall Prevention Behaviors In Community-Dwelling Thai Older Adults, Kanyarat Ubolwan Jan 2013

An Exploration Of The Relationships Among Demographics, Risk Factors, Perceived Self-Efficacy, And Fall Prevention Behaviors In Community-Dwelling Thai Older Adults, Kanyarat Ubolwan

Wayne State University Dissertations

Background: Worldwide, falls among older adults lead to significant physical, psychological, and social problems. Studies have described fall prevalence, risk factors, and the impact of falls in Thailand. Although fall prevention requires significant self-care abilities, insufficient knowledge exists about how to prevent falls in Thai community settings or the predictors of fall prevention behaviors among Thai Older adults.

Purpose: Orem's Self-Care Theory was used: 1) to examine the relationships among the basic conditioning factors (BCFs) of age, gender, falls, comorbidity, mental and physical health status, self-care agency (i.e., self-efficacy), and fall prevention self-care behaviors and 2) to determine the predictors …

Facilitation Of Language Acquisition Viewed Through An Interpretative Lens: The Role Of Authenticity, Melanie Ann Harper Jan 2013

Facilitation Of Language Acquisition Viewed Through An Interpretative Lens: The Role Of Authenticity, Melanie Ann Harper

Wayne State University Dissertations

A paradigm is the conceptual framework or lens one uses to view reality. The field of speech-language pathology is traditionally rooted in the empirical paradigm, which believes that language can be fragmented into isolated skills and taught in a hierarchal fashion. This belief has resulted in service delivery models that remove students from naturalistic contexts for decontextualized exercises. Progress in language acquisition is measured objectively. The empirical belief is exemplified by the accountability requirements in special education law (e.g., IEP). It is compounded by the realities of public school speech-language pathologists (SLPs), such as high caseload numbers, multiple buildings, and …

Vascular Depression: An Early Warning Sign Of Frailty, Daniel Paulson, Peter A. Lichtenberg Jan 2013

Vascular Depression: An Early Warning Sign Of Frailty, Daniel Paulson, Peter A. Lichtenberg

Psychology Faculty Research Publications

Objectives: Frailty is a common geriatric disorder associated with ADL impairment, hospitalization, and death. Phenomenological evidence suggests that late-life depression (Katz, 2004), particularly vascular depression, may be a risk factor for frailty. This study tests that hypothesis.

Methods: We identified a sample of stroke-free women over the age of 80 from the Health and Retirement Survey. The sample included 984 respondents in 2000 (incidence sample). Of these, 459 were non-frail at baseline and still alive in 2004 (prevalence sample). Frail respondents experienced at least three of the following: wasting, exhaustion, weakness, slowness, and falls. Vascular depression was represented using two …

Victimization, Urbanicity, And The Relevance Of Context: School Routines, Race And Ethnicity, And Adolescent Violence, Anthony A. Peguero, Edwardo L. Portillos, Jun Sung Hong, Juan Carlos GonzáLez, Lindsay L. Kahle, Zahra Shekarkhar Jan 2013

Victimization, Urbanicity, And The Relevance Of Context: School Routines, Race And Ethnicity, And Adolescent Violence, Anthony A. Peguero, Edwardo L. Portillos, Jun Sung Hong, Juan Carlos GonzáLez, Lindsay L. Kahle, Zahra Shekarkhar

Social Work Faculty Publications

The United States is undergoing a historical racial and ethnic demographic shift. There is limited criminological research exploring if and how these changes influence variation in the relationship between routine activity theory and adolescent violence. Although the link between routine activities and victimization has been tested and well established, criminologists have questioned if routine activities can explain adolescent violence across different social contexts. Prior research demonstrates that there are potential nuances in the theoretical connections between routine activities and victimization, particularly when considering race and ethnicity. This study builds on previous research by questioning if the elements of routine activities …

Impact Of Dual Disorders, Trauma, And Social Support On Quality Of Life Among Women In Treatment For Substance Dependence, Suzanne Brown, Min Kyoung Jun, Meeyoung Oh Min, Elizabeth M. Tracy

Social Work Faculty Publications

Objective Women with dual disorders report lower levels of social support than women with substance dependence alone, and lower levels of social support have been associated with lower quality of life among individuals with substance use disorders. However, little is known about the impact of trauma symptoms and violence exposure on quality of life for women with dual disorders. The purpose of this study was to examine the impact of dual disorders, trauma, and social support related to recovery on various domains of quality of life among women in substance abuse treatment.

Methods This study utilized multiple standardized measures and …

(Pre)Determined Occupations: The Post-Colonial Hybridizing Of Identity And Art Forms In Third World Spaces, Amanda Alexander, Manisha Sharma Jan 2013

(Pre)Determined Occupations: The Post-Colonial Hybridizing Of Identity And Art Forms In Third World Spaces, Amanda Alexander, Manisha Sharma

Journal of Social Theory in Art Education

In this article, we present the effects of globalization on art forms in Peru and on teacher identity in India while exploring hybridization as an ongoing global paradigm in both contexts (Bhabha, 1994; Said, 1979). Peruvian art forms are continuously shifting as global cultures meld and become more technologically connected, which ultimately brings about questions of authenticity. The identities of Indian art educators are evolving, and shifting indicating an assemblage or structure containing many parts working together to perform a particular function. In realizing its function, the structure can be named or its form made visible (Deleuze & Guattari, 1987). …

Life, Heartbeat, Birth: A Medical Basis For Reform, David F. Forte Jan 2013

Life, Heartbeat, Birth: A Medical Basis For Reform, David F. Forte

Law Faculty Articles and Essays

This Article does not revisit the moral, legal, and constitutional critiques of the Court’s position [in Roe v. Wade]. The voluminous commentaries on the flaws in the Court’s opinions speak for themselves. Rather, this Article seeks to ground an expansion of the protection available to the unborn on the implicit principles underlying current Supreme Court doctrine, refined and modified by recent medical research on nature of pregnancy and human pre-natal development. It will argue that the State’s compelling interest in the protection of what the Court has called “potential life” ripens at an earlier point in time than what the …

Climate Change Geoengineering: Philosophical Perspectives, Legal Issues, And Governance Frameworks, William C. G. Burns, Andrew L. Strauss Jan 2013

Climate Change Geoengineering: Philosophical Perspectives, Legal Issues, And Governance Frameworks, William C. G. Burns, Andrew L. Strauss

School of Law Faculty Publications

The international community is not taking the action necessary to avert dangerous increases in greenhouse gases. Facing a potentially bleak future, the question that confronts humanity is whether the best of bad alternatives may be to counter global warming through human-engineered climate interventions. In this book, eleven prominent authorities on climate change consider the legal, policy, and philosophical issues presented by geoengineering. The book asks: When, if ever, are decisions to embark on potentially risky climate modification projects justified? If such decisions can be justified, in a world without a central governing authority, who should authorize such projects and by …

The Pea Aphid Uses A Version Of The Terminal System During Oviparous, But Not Viviparous, Development, Ryan D. Bickel, Hillary C. Cleveland, Joanna Barkas, Caitlin C. Jeschke, Amelie A. Raz, David L. Stern, Gregory K. Davis

Biology Faculty Research and Scholarship

Background: In most species of aphid, female nymphs develop into either sexual or asexual adults depending on the length of the photoperiod to which their mothers were exposed. The progeny of these sexual and asexual females, in turn, develop in dramatically different ways. The fertilized oocytes of sexual females begin embryogenesis after being deposited on leaves (oviparous development) while the oocytes of asexual females complete embryogenesis within the mother (viviparous development). Compared with oviparous development, viviparous development involves a smaller transient oocyte surrounded by fewer somatic epithelial cells and a smaller early embryo that comprises fewer cells. To investigate …

The Evodevoci: A Concept Inventory For Gauging Students’ Understanding Of Evolutionary Developmental Biology, Kathryn E. Perez, Anna Hiatt, Gregory K. Davis, Caleb Trujillo, Donald P. French, Terry Mark, Rebecca M. Price

Biology Faculty Research and Scholarship

The American Association for the Advancement of Science 2011 report Vision and Change in Undergraduate Biology Education encourages the teaching of developmental biology as an important part of teaching evolution. Recently, however, we found that biology majors often lack the developmental knowledge needed to understand evolutionary developmental biology, or “evo-devo.” To assist in efforts to improve evo-devo instruction among undergraduate biology majors, we designed a concept inventory (CI) for evolutionary developmental biology, the EvoDevoCI. The CI measures student understanding of six core evo-devo concepts using four scenarios and 11 multiple-choice items, all inspired by authentic scientific examples. Distracters were designed …

Response Of Macroarthropod Assemblages To The Loss Of Hemlock (Tsuga Canadensis), A Foundation Species, Tara E. Sackett, Sydne Record, Sharon Bewick, Benjamin Baiser, Nathan J. Sanders, Aaron M. Ellison Jan 2013

Response Of Macroarthropod Assemblages To The Loss Of Hemlock (Tsuga Canadensis), A Foundation Species, Tara E. Sackett, Sydne Record, Sharon Bewick, Benjamin Baiser, Nathan J. Sanders, Aaron M. Ellison

Biology Faculty Research and Scholarship

In eastern North American forests, eastern hemlock (Tsuga canadensis) is a foundation species. As hemlock is lost from forests due to the invasive hemlock woolly adelgid (Adelges tsugae) and preemptive salvage logging, the structure of assemblages of species associated with hemlock is expected to change. We manipulated hemlock canopy structure at hectare scales to investigate the effects of hemlock death on assemblages of ants, beetles, and spiders in a New England forest. Relative to reference hemlock stands, both in situ death of hemlock and logging and removal of hemlock altered composition and diversity of beetles and spiders, and logging increased …
