Membrane Association Via An N-Terminal Amphipathic Helix Is Required For The Cellular Organization And Function Of Rnase Ii, Feng Lu, Aziz Taghbalout

UCHC Articles - Research

The subcellular localization of the exoribonuclease RNase II is not known despite the advanced biochemical characterization of the enzyme. Here we report that RNase II is organized into cellular structures that appear to coil around the Escherichia coli cell periphery and that RNase II is associated with the cytoplasmic membrane by its aminoterminal amphipathic helix. The helix also acts as an autonomous transplantable membranebinding domain capable of directing normally cytoplasmic proteins to the membrane. Assembly of the organized cellular structures of RNase II required the RNase II amphipathic membrane-binding domain. Coimmunoprecipitation of the protein from cell extracts indicated that RNase …

Ring Domain, David J. Hall Jan 2013

Ring Domain, David J. Hall

Protein Domains

Ring domain #1CHC. The RING finger is a specialized type of Zn finger consisting of 40–60 residues that binds two atoms of zinc, and is involved in mediating protein—protein interactions. Many zinc fingers bind nucleic acids. The presence of a RING finger domain is a characteristic of RING-class E3 ubiquitin protein ligases capable of transferring ubiquitin from an E2 enzyme to a substrate protein.

Sh2 Domain, David J. Hall Jan 2013

Sh2 Domain, David J. Hall

Protein Domains

SH2 domain #1BFJ. Src-homology 2 (SH2) domains are modules of ~100 amino acids that bind to specific phospho tyrosine (pY) containing peptide motifs. Conventional SH2 domains have a conserved pocket that recognizes pY, and a more variable pocket that binds 3-6 residues C-terminal to the pY and confers specificity.

Sh3 Domain, David J. Hall Jan 2013

Sh3 Domain, David J. Hall

Protein Domains

SH3 domain #1NEB. Src-homology 3 (SH3) domains bind to Pro-rich peptides that form a left-handed poly-Pro type II helix, with the minimal consensus Pro-X-X-Pro. Each Pro is usually preceeded by an aliphatic residue. Each in the aliphatic-Pro pair binds to a hydrophobic pocket on the SH3 domain.

Ig Domain, David J. Hall Jan 2013

Ig Domain, David J. Hall

Protein Domains

Ig domain #2CKN. This particular domain is named for the first protein in which it was found, the immunoglobulin. An immunoglobulin is a antibody. Antibodies are generated by our immune system to recognize the specific size, shape and charge of pathogens. This domain is also found on the extracellular portion of many receptors including the interleukin-1 family of receptors.

Beta Barrel, David J. Hall Jan 2013

Beta Barrel, David J. Hall

The protein is routinely used to visualize a variety of biological processes. The beta barrel domain is a beta sheet wrapped around the fluorescent active site to provide structure.

Evolutionary Responses To Invasion: Cane Toad Sympatric Fish Show Enhanced Avoidance Learning, Georgina Caller, Culum Brown Jan 2013

Evolutionary Responses To Invasion: Cane Toad Sympatric Fish Show Enhanced Avoidance Learning, Georgina Caller, Culum Brown

Aquaculture Collection

The introduced cane toad (Bufo marinus) poses a major threat to biodiversity due to its lifelong toxicity. Several terrestrial native Australian vertebrates are adapting to the cane toad’s presence and lab trials have demonstrated that repeated exposure to B. marinus can result in learnt avoidance behaviour. Here we investigated whether aversion learning is occurring in aquatic ecosystems by comparing cane toad naı¨ve and sympatric populations of crimson spotted rainbow fish (Melanotaenia duboulayi). The first experiment indicated that fish from the sympatric population had pre-existing aversion to attacking cane toad tadpoles but also showed reduced attacks on native tadpoles. The second …

Social Contact Theory: A Framework For Understanding Aids-Related Stigma, Brandon K. Attell Jan 2013

Social Contact Theory: A Framework For Understanding Aids-Related Stigma, Brandon K. Attell

The Journal of Public and Professional Sociology

How do sociologists understand the social stigma that is attached to individuals living with HIV/AIDS? In this article, I offer four criticisms of the AIDS-related stigma scholarly literature. Furthermore, I propose a new framework for understanding AIDS-related stigma: Allport’s social contact theory. I discuss both the benefits and drawbacks of utilizing social contact theory to examine AIDS-related stigma, as well as how the theory could be employed in both quantitative and qualitative research.

Making A Case For Multi-Disciplinary Analysis, Matthew J. Richard, Michael G. Noll, Catherine Oglesby, Mark Patrick George, Tracy Woodard

The Journal of Public and Professional Sociology

In recent years, I have been drawn to inter-disciplinary approaches to scholarship. Specifically, I have been attracted to “mind studies”—an alluring amalgam of Cognitive Science, Neuroscience, Developmental Psychology, Psychoanalysis, Social Psychology, and Anthropology—as offering more satisfying explanations of human action. In this paper I want to explore more deeply and expound further upon the benefits of multi-disciplinary research. To do so, I’ve invited colleagues of mine who work in other disciplines to view an ethnographic film about a poor Appalachian family and to identify specific issues in it that they would develop further in their classes. My working assumption is …

A Content Analysis Of Salvia Divinorum Use On Youtube, Brent A. Paterline, Maria J. Albo

The Journal of Public and Professional Sociology

While peer groups remain the primary place where learning about drugs takes place, recently, researchers have recognized that channels of mass communication (e.g. movies, television, the Internet) may also play an important role in the learning process. For the last several decades, studies have demonstrated that there is a positive relationship between mass media use and drug use. The purpose of this study is to examine the use of a new recreational drug, Salvia divinorum, on YouTube. Salvia divinorum is a naturally occurring hallucinogenic drug from the mint family indigenous to Mexico and Central and South America. When smoked, the …

Background: Demosponges are challenging for phylogenetic systematics because of their plastic and relatively simple morphologies and many deep divergences between major clades. To improve understanding of the phylogenetic relationships within Demospongiae, we sequenced and analyzed seven nuclear housekeeping genes involved in a variety of cellular functions from a diverse group of sponges. Methodology/Principal Findings: We generated data from each of the four sponge classes (i.e., Calcarea, Demospongiae, Hexactinellida, and Homoscleromorpha), but focused on family-level relationships within demosponges. With data for 21 newly sampled families, our Maximum Likelihood and Bayesian-based approaches recovered previously phylogenetically defined taxa: Keratosap, Myxospongiaep, Spongillidap, Haploscleromorphap (the …

Chemically Modified Proteins As Functional Biomaterials, Vindya K. Thilakarathne

Doctoral Dissertations

The goal of this thesis was to explore how to improve the functionality of proteins as biomaterials by chemical modification of the surface of the protein. In order to achieve this goal protein surfaces were decorated with small amine molecules and large synthetic polymers. The introduced functionality by the facile modification, allowed the protein to behave as a successful bio catalysts, bio sensor and a useful scaffolding platform.

Thorough characterization was performed using various biochemical, bioanalytical biophysical methods to evaluate the structural integrity, enzymatic activity , thermodynamic and kinetic stability of chemically modified proteins/enzymes. Improved stability and reversible thermal denaturation …
