The Influence Of Anthropogenic Development Of Water On Coyotes And Kit Foxes In The Great Basin And Mojave Deserts, Lucas Keith Hall Mar 2013

The Influence Of Anthropogenic Development Of Water On Coyotes And Kit Foxes In The Great Basin And Mojave Deserts, Lucas Keith Hall

Theses and Dissertations

Anthropogenic provisioning of water (water developments) to enhance abundance and distribution of wildlife is a common management practice in arid regions where water is limiting. Despite the long-term and widespread use of water developments, little is known about how they influence distribution, competition dynamics, and behavior of native species. To elucidate the potential influences of water developments on native species, we tested hypotheses concerning the occurrence and behavior of native kit foxes (Vulpes macrotis). First, we tested the indirect effect of water hypothesis (IEWH) which proposes that water developments negatively affect the arid-adapted kit fox by enabling a …

Application Of Genome Reduction, Next Generation Sequencing, And Kaspar Genotyping In Development, Characterization, And Linkage Mapping Of Single Nucleotide Polymorphisms In The Grain Amaranths And Quinoa, Scott Matthew Smith

Theses and Dissertations

The grain amaranths (Amaranthus sp.) and quinoa (Chenopodium quinoa Willd.) are important seed crops in South America. These crops have gained international attention in recent years for their nutritional quality and tolerance to abiotic stress. We report the identification and development of functional single nucleotide polymorphism (SNP) assays for both amaranth and quinoa. SNPs were identified using a genome reduction protocol and next generation sequencing. SNP assays are based on KASPar genotyping chemistry and were detected using the Fluidigm dynamic array platform. A diversity screen consisting of 41 amaranth accessions showed that the minor allele frequency (MAF) of the amaranth …

Continuous Monitoring Reveals Multiple Controls On Ecosystem Metabolism In A Suburban Stream, Jake J. Beaulieu, Clay P. Arango, David A. Balz, William D. Shuster Mar 2013

Continuous Monitoring Reveals Multiple Controls On Ecosystem Metabolism In A Suburban Stream, Jake J. Beaulieu, Clay P. Arango, David A. Balz, William D. Shuster

Biology Faculty Scholarship

1. Primary production and respiration in streams, collectively referred to as stream ecosystem metabolism, are fundamental processes that determine trophic structure, biomass and nutrient cycling. Few studies have used high‐frequency measurements of gross primary production (GPP) and ecosystem respiration (ER) over extended periods to characterise the factors that control stream ecosystem metabolism at hourly, daily, seasonal and annual scales.

2. We measured ecosystem metabolism at 5‐min intervals for 23 months in Shepherd Creek, a small suburban stream in Cincinnati, Ohio (U.S.A.).

3. Daily GPP was best predicted by a model containing light and its synergistic interaction with water temperature. Water …

New Hampshire EPSCoR

To examine whether stream nitrogen concentrations in forested reference catchments have changed over time and if patterns were consistent across the USA, we synthesized up to 44 yr of data collected from 22 catchments at seven USDA Forest Service Experimental Forests. Trends in stream nitrogen presented high spatial variability both among catchments at a site and among sites across the USA. We found both increasing and decreasing trends in monthly flow-weighted stream nitrate and ammonium concentrations. At a subset of the catchments, we found that the length and period of analysis influenced whether trends were positive, negative or non-significant. Trends …

Hinds, Lars Bellner, Dov Goldstein, Stephen J . Peterson, Joseph I. Shapiro M.D., Nader G. Abraham

Biochemistry and Microbiology

Introduction:

Heme oxygenase (HO), a major cytoprotective enzyme, attenuates oxidative stress and obesity. The canonical Wnt signaling cascade plays a pivotal role in the regulation of adipogenesis. The present study examined the interplay between HO-1and the Wnt canonical pathway in the modulation of adipogenesis in mesenchymal stem cell (MSC)-derived adipocytes.

Methods:

To verify the role of HO-1 in generating small healthy adipocytes, cobalt protoporphyrin (CoPP), inducer of HO-1, was used during adipocyte differentiation. Lipid accumulation was measured by Oil red O staining and lipid droplet size was measured by BODIPY staining.

Results:

During adipogenesis in vitro, differentiating pre-adipocytes display transient …

Cantilever-Based Optical Interfacial Force Microscope In Liquid Using An Optical-Fiber Tip, Byung I. Kim, Luke Smith, Thanh Tran, Steven Rossland, Erik Parkinson

Physics Faculty Publications and Presentations

We developed a novel cantilever-based optical interfacial force microscope (COIFM) to study molecular interaction in liquid environments. The force sensor was created by attaching a chemically etched optical-fiber tip to the force sensor with UV epoxy, and characterized by imaging on a calibration grid. The performance of the COIFM was then demonstrated by measuring the force between two oxidized silicon surfaces in 1 mM KCl as a function of distance. The result was consistent with previously reported electrical double layer forces, suggesting that a COIFM using an optical-fiber tip is capable of measuring force in a liquid environment.

H3k4 Methyltransferase Set1 Is Involved In Maintenance Of Ergosterol Homeostasis And Resistance To Brefeldin A, Paul F. South, Kayla M. Harmeyer, Nina D. Serratore, Scott D. Briggs

Faculty Publications

Set1 is a conserved histone H3 lysine 4 (H3K4) methyltransferase that exists as a multisubunit complex. Although H3K4 methylation is located on many actively transcribed genes, few studies have established a direct connection showing that loss of Set1 and H3K4 methylation results in a phenotype caused by disruption of gene expression. In this study, we determined that cells lacking Set1 or Set1 complex members that disrupt H3K4 methylation have a growth defect when grown in the presence of the antifungal drug Brefeldin A (BFA), indicating that H3K4 methylation is needed for BFA resistance. To determine the role of Set1 in …

Hematopoietic Stem Cells And Progenitors In Murine Autoimmune Arthritis, Kwadwo Asare Oduro Jr. Mar 2013

Hematopoietic Stem Cells And Progenitors In Murine Autoimmune Arthritis, Kwadwo Asare Oduro Jr.

All Theses and Dissertations (ETDs)

Although the physiological role of stem cells in generating and maintaining a functional biological system is widely appreciated, very little is known about the behavior of these cells in pathological conditions. If and how these primitive cells are affected by a disease process or may contribute to the disease process has not been adequately explored. We have studied the Hematopoietic System, with particular emphasis on Hematopoietic Stem Cells: HSCs) and Progenitors, in a spontaneous murine autoimmune arthritis model. We found that these mice have a systemic increase in myeloid cells, which correlated with an increased frequency of granulocyte-monocyte progenitors relative …

Experimental And Natural Warming Elevates Mercury Concentrations In Estuarine Fish, Jennifer A. Dijkstra, Kate L. Buckman, Darren Ward, David W. Evans, Michele Dionne, Celia Y. Chen

Dartmouth Scholarship

Marine food webs are the most important link between the global contaminant, methylmercury (MeHg), and human exposure through consumption of seafood. Warming temperatures may increase human exposure to MeHg, a potent neurotoxin, by increasing MeHg production as well as bioaccumulation and trophic transfer through marine food webs. Studies of the effects of temperature on MeHg bioaccumulation are rare and no study has specifically related temperature to MeHg fate by linking laboratory experiments with natural field manipulations in coastal ecosystems. We performed laboratory and field experiments on MeHg accumulation under varying temperature regimes using the killifish, Fundulus heteroclitus. Temperature treatments …

Degree Of Handedness, But Not Direction, Is A Systematic Predictor Of Cognitive Performance, Eric Prichard, Ruth Propper, Stephen D. Christman

Department of Psychology Faculty Scholarship and Creative Works

A growing body of evidence is reviewed showing that degree of handedness (consistent versus inconsistent) is a more powerful and appropriate way to classify handedness than the traditional one based on direction (right versus left). Experimental studies from the domains of episodic memory retrieval, belief updating/cognitive flexibility, risk perception, and more are described. These results suggest that inconsistent handedness is associated with increased interhemispheric interaction and increased access to processes localized to the right cerebral hemisphere.
