Invertebrate animal models such as the nematode Caenorhabditis elegans (C. elegans) are increasingly used in nanotechnological applications. Research in this area covers a wide range from remote control of worm behavior by nanoparticles (NPs) to evaluation of organismal nanomaterial safety. Despite of the broad spectrum of investigated NP-bio interactions, little is known about the role of nanomaterials with respect to aging processes in C. elegans. We trace NPs in single cells of adult C. elegans and correlate particle distribution with the worm's metabolism and organ function. By confocal microscopy analysis of fluorescently labeled NPs in living worms, we identify two …

Longitudinal studies are a useful tool for investigating the course of chronic diseases. Many chronic diseases can be characterized by a set of health states. We can improve our understanding of the natural history of the disease by modeling the sequence of visited health states and the duration in each state. However, in most applications, subjects are observed only intermittently. This observation scheme creates a major modeling challenge: the transition times are not known exactly, and in some cases the path through the health states is not known.

The ability of high-risk neuroblastoma to survive unfavorable growth conditions and multimodal therapy has produced an elusive childhood cancer with remarkably poor prognosis. A novel phenomenon enabling neuroblastoma to survive selection pressure is its capacity for reversible adaptive plasticity. This plasticity allows cells to transition between highly proliferative anchorage dependent (AD) and slow growing, anoikis-resistant anchorage independent (AI) phenotypes. Both phenotypes are present in established mouse and human tumors. The differential gene expression profile of the two cellular phenotypes in the mouse Neuro2a cell line delineated pathways of proliferation in AD cells or tyrosine kinase activation/ apoptosis inhibition in AI …

Objective

The objective of this study was to determine whether the detection of Aggregatibacter actinomycetemcomitans (Aa) correlates with the clinical and immunoinflammatory profile of Localized Aggressive Periodontitis (LAP), as determined by by 16S rRNA gene-based microarray.

Subjects and Methods

Subgingival plaque samples from the deepest diseased site of 30 LAP patients [PD ≥ 5 mm, BoP and bone loss] were analyzed by 16S rRNA gene-based microarrays. Gingival crevicular fluid (GCF) samples were analyzed for 14 cyto/chemokines. Peripheral blood was obtained and stimulated in vitro with P.gingivalis and E.coli to evaluate inflammatory response profiles. Plasma lipopolysaccharide (LPS) levels were also measured. …

Originally conducted by the Gulf of Maine Council on the Marine Environment from 1993 to 2011, the Gulfwatch Program examined trends in the water quality of the Gulf of Maine by monitoring toxic contaminant concentrations in the tissues of shellfish. Starting in 2012 the Piscataqua Region Estuaries Partnership (PREP) continued this program in the Piscataqua Region. Each year, PREP collects blue mussels at three sites: Dover Point, NH (NHDP), Clark Cove on Seavey Island, ME (MECC), and Hampton-Seabrook Harbor (NHHS). A growing number of studies are evaluating retinal progenitor cell (RPC) transplantation as an approach to repair retinal degeneration and restore visual function. To advance cell-replacement strategies for a practical retinal therapy, it is important to define the molecular and biochemical mechanisms guiding RPC motility. We have analyzed RPC expression of the epidermal growth factor receptor (EGFR) and evaluated whether exposure to epidermal growth factor (EGF) can coordinate motogenic activity in vitro. Using Boyden chamber analysis as an initial high-throughput screen, we determined that RPC motility was optimally stimulated by EGF concentrations in the range of 20-400 ng/ml, with decreased …

This dissertation describes the physiological role of the Capping Protein- CARMIL interaction in migrating cells. I establish the CARMIL-CP complex as a key regulator of lamellipodial actin assembly and of lamellipodial dynamics. Membrane ruffling at the leading edge of motile cells and macropinocytosis were also found to be dependent on the CARMIL1-CP interaction. This is consistent with macropinocytosis and ruffling being dependent on a functional and dynamic lamellipodium.: Kerr and Teasdale, 2009).

Cone-rod homeobox: CRX) protein is a "paired-like" homeodomain transcription factor that is essential for regulating rod and cone photoreceptor transcription. Mutations in human CRX are associated with the dominant retinopathies Retinitis Pigmentosa: RP), Cone-Rod Dystrophy: CoRD) and Leber Congenital Amaurosis: LCA), with variable severity. The goal of my dissertation project was to develop and characterize animal models to understand genetic mechanisms of phenotypic diversity in CRX-associated disease. L-rhamnose (Rha) is a deoxy-hexose sugar commonly found in nature. L-Rha catabolic pathways were previously characterized in various bacteria including Escherichia coli. Nevertheless, homology searches failed to recognize all the genes for the complete L Rha utilization pathways in diverse microbial species involved in biomass decomposition. Moreover, the regulatory mechanisms of L-Rha catabolism have remained unclear in most species. A comparative genomics approach was used to reconstruct the L-Rha catabolic pathways and transcriptional regulons in the phyla Actinobacteria, Bacteroidetes, Chloroflexi, Firmicutes, Proteobacteria, and Thermotogae. The genre of short story has been viewed by scholars as being very controversial and conflicting. The debates are on the proper way of defining short stories, their classification with supporting examples, and their actual distinctive features. This paper acknowledges the need to analyze literature into genres and even sub-genres, but shows how controversial literary genres can be. It mainly focuses on controversies pertaining to short story as a genre of Kiswahili literature. This paper is about teachers’ attitudes towards teaching sexuality education (SE) in Morogoro rural and urban districts. It is based on a study which employed mixed method design and used both qualitative and quantitative approaches. It involved a sample of 126 teachers residing from Morogoro rural and urban schools. Data were obtained through interviews and questionnaire. It was revealed that a majority of the teachers supported the teaching of SE in schools with more support of HIV and AIDS contents than pregnancy and contraceptives. Kimboza forest reserve in Morogoro region, is an important biodiversity hotspot among the Eastern Arc forests of Tanzania. High population pressure and encroachment threatens its conservation values. This is based on a study which aimed at investigating the composition, diversity, and structure of the tree community in the forest. All trees with DBH ≥ 10cm were inventoried from 0.72ha in a total of 18 plots of 20m × 20m placed randomly from the forest edge to the interior. The transition from colonialism to independence and Ujamaa had great impacts on society—politically, economically and socially. This is discussed in relation to the novel Village in Uhuru (1969) by Gabriel Ruhumbika. Within the backdrop of independence, he explores the social changes that occur in the small village of Chamambo. Cattle are reared in diverse environments and collecting phenotypic body temperature (BT) measurements to characterize BT variation across diverse environments is difficult and expensive. To better understand the genetic basis of BT regulation, a genome-wide association study was conducted utilizing crossbred steers and heifers totaling 239 animals of unknown pedigree and breed fraction. Private Flashbulb Memories: The Case Of Coming Out Memories, Gabrielle Weber

Participants (N = 83) from Butler University and communities in Indianapolis, IN answered a three part questionnaire regarding their experience revealing their homosexual identity to their parents that included: a narrative, a set of probe questions, and a short demographic section. The narrative and probed recall data were scored in a similar fashion to flashbulb memory narratives with canonical features. The memory of revealing homosexuality to parents displayed flashbulb-like qualities. We found that those most confident in their probed recall answers were those with moderate arousal and few recounts. Also, those reporting moderate affect and fewer recounts had better memory …

The accumulation of pollutants near the shoreline can result in low quality coastal water with negative effects on human health. To understand the role of mixing by tidal flows in coastal water quality, we study the nearshore Lagrangian circulation of four recreational beaches in Miami, FL, who, despite close proximity, have varying microbial levels. Specifically, we reveal Lagrangian Coherent Structures (LCSs), i.e., distinguished material curves which shape global mixing patterns and thus act as skeletons of the Lagrangian circulation. We show how these material curves, which shape circulation and mixing patterns, can be used to explain the incongruous states of …
