Investigation Of Ghrelin Acylation By Ghrelin O-Acyltransferase, Edward P. Prybolsky May 2013

Investigation Of Ghrelin Acylation By Ghrelin O-Acyltransferase, Edward P. Prybolsky

Renée Crown University Honors Thesis Projects - All

Ghrelin is a peptide hormone involved in hunger signaling and other physiological processes. To become active, ghrelin first must be acylated by ghrelin O-acyltransferase (GOAT). This enzyme carries out a post-translational modification that attaches an octanoyl group to the side chain of the serine 3 (S3) residue of ghrelin. With ghrelin linked to hunger signaling, memory, and other physiological processes, GOAT has been identified as a potential drug target. However, inhibitor design is difficult because not much is known regarding the active site structure and catalytic mechanism of GOAT. This study reports the design of a novel fluorescence-based assay that …

Developing An Experimental System For Studying Early Events In Speciation In Drosophila, Maria Poplawska May 2013

Developing An Experimental System For Studying Early Events In Speciation In Drosophila, Maria Poplawska

Renée Crown University Honors Thesis Projects - All

Speciation occurs when two populations of a species can no longer reproduce, either because of (1) pre-mating reproductive isolation, for example, populations having non-overlapping habitats, or evolving different courtship behaviors, or (2) post-mating pre-zygotic (PMPZ) isolation, where mechanical (e.g., incompatible differences in genitalia), or gametic (e.g., sperm and egg become incompatible) differences renders the strains unable to produce offspring. Previous work has suggested that when incipient species begin to experience reproductive isolation, a phenomenon called “reinforcement” can accelerate the process, by the rapid evolution of new pre- mating or PMPZ barriers. While the occurrence of reinforcement has been studied for …

Where Does The Water Come From? A Temporal Geochemical Characterization Of Water Sources In The Lives Of Storms, Mallory Ringham May 2013

Where Does The Water Come From? A Temporal Geochemical Characterization Of Water Sources In The Lives Of Storms, Mallory Ringham

Renée Crown University Honors Thesis Projects - All

An analysis of the ion chemistry, δ18O, and δ2H values of precipitation may allow for the characterization of their water sources. As water evaporates, it retains a signature of its source in the ions that it carries and in the amount of fractionation of the oxygen and hydrogen isotopes. Precipitation samples for coastal, lake effect, and continental sources were collected over the months of October- January in Syracuse, New York throughout the duration of each storm, including one hurricane, one thunderstorm, two lake effect snowstorms, and seven rain events. These samples were run through IC, ICP-OES, …

Metal-Citrate Transport In The Gram-Positive Bacterium Kineococcus Radiotolerans, Eleanor Robertson May 2013

Metal-Citrate Transport In The Gram-Positive Bacterium Kineococcus Radiotolerans, Eleanor Robertson

Renée Crown University Honors Thesis Projects - All

Little is known about metal-citrate transport in Gram-positive bacteria. Previously, all such transporters identified have belonged to the citrate-metal hydrogen symport (CitMHS) family. Presented herein is our research on the predicted metal-citrate transport system in the Gram-positive bacteria Kineococcus radiotolerans. Given the high alkaline natural environment of this organism, it was hypothesized such a metal-citrate transporter may utilize an antiport mechanism, which to date has not been observed. Herein it is shown that K. radiotolerans has a calcium-citrate transporter. Preliminary results also support the hypothesis that this transporter operates through hydroxide antiport, rather than hydrogen symport, making this the …

The Role Of Cell Adhesion Molecules In Cyst Breakdown And Follicle Formation In The Mouse Neonatal Ovary, Dana Senderoff May 2013

The Role Of Cell Adhesion Molecules In Cyst Breakdown And Follicle Formation In The Mouse Neonatal Ovary, Dana Senderoff

Renée Crown University Honors Thesis Projects - All

The total number of oocytes that an individual will have available for reproduction is defined at birth. This established oocyte pool is essential to fertility, as a female loses oocytes throughout her life and never makes more. In the mouse, germ cells form in the fetus and migrate to the gonad, where they undergo mitosis and are referred to as oogonia. These cells are in germ line cysts, which are clusters of germ cells connected by intercellular bridges that form by incomplete cytokinesis. Germ cells in the human also develop in clusters with characteristics of cysts. The cells within the …

Development Of Protein Farnesyltransferase Variants With Altered Substrate Selectivity, Susan Zhang May 2013

Development Of Protein Farnesyltransferase Variants With Altered Substrate Selectivity, Susan Zhang

Renée Crown University Honors Thesis Projects - All

Post-translational modifications play an essential role in regulating protein structure and function. Enzymes catalyzing these modifications must often recognize and modify multiple substrate proteins from among a plethora of non-substrates with similar structures and amino acid sequences. For example, protein farnesyltransferase (FTase) catalyzes the addition of an isoprenoid group to a cysteine near the C-terminus of a substrate protein and is proposed to modify a pool of substrates numbering more than one hundred. We seek to understand the interactions in the FTase active site that engender substrate selectivity. By mutating two residues within FTase, we have developed FTase variants with …

Design Of Metalloproteins For Catalysis And Bioimaging, Krystyna Zhezherya

Renée Crown University Honors Thesis Projects - All

Abstract

Metalloenzymes constitute a large subclass of proteins with diverse functions including metal-sensing, signal transduction pathways, and metal transport. In this report, I aim to study the intricate relationship between structure and function as it relates to metalloproteins in the fields of catalysis and bioimaging. For catalysis, I used existing platforms in addition to de novo design to study small scaffolds with varying secondary structure, and their function in catalyzing the hydrolysis of a p-nitrophenol ester. For bioimaging, I used an existing enzyme known as human carbonic anhydrase I, hCA, to develop a bioimaging agent that can be fused to …

The Heteronormative Objectification Of Women In The Disney Princess Films: A Study Of Brand Advertising And Parents’ Perceptions, Melanie Greenblatt May 2013

The Heteronormative Objectification Of Women In The Disney Princess Films: A Study Of Brand Advertising And Parents’ Perceptions, Melanie Greenblatt

Renée Crown University Honors Thesis Projects - All

Although the Disney Princess franchise portrays its ten princess movies as tales of “happily ever after,” they are filled with negative stereotypes of all kinds; this study in particular focuses on those addressed towards females. Each of these princesses lives in a patriarchal society and her sole desire is to find a man who guides her to happiness. The Princess brand has immense popularity even with these unconstructive stigmatizations of women. This study explores these movies while investigating, through qualitative research, the potency of these stereotypes and whether the parents of the target audience (defined by Disney as ages three …

Structural And Functional Studies On The Role Of The Retinoblastoma Binding Protein Five (Rbbp5) In The Histone Methyltransferase Activity Of The Mixed Lineage Leukemia (Mll1) Core Complex, Melody Sanders

Biology - Theses

Cells employ elaborate mechanisms to introduce structural and chemical variation into chromatin in the form of covalent post-translational modifications. Covalent modifications of histones contribute to the dynamic states of chromatin structure that govern nearly all of DNA-coupled processes such as transcription, replication, and repair. The mechanism by which covalent modifications of histones contribute to these activities remains a central question to understanding genome regulation and its dysfunction in human disease. Although there is extensive literature documenting the identification of many of the enzymes that place histone modifications, far less is known about how the enzymes are targeted and how their …

Influences To Post-Graduation Career Aspirations And Attainment In Stem Doctoral Candidates And Recipients, Deborah Barry

Science Teaching - Dissertations

As the realities of the academic job market have forced some PhD recipients to accept less-preferable position types, there has been increasing concerns that these students are not prepared for their careers, especially in STEM fields. However, aside from the labor market, few studies have explored the influences on career aspiration and attainment among doctoral degree holders. This study utilized the socialization theory framework to identify aspects of the doctoral education process that are predictive of the likelihood of certain career aspirations among science and engineering doctoral candidates and career attainment among STEM doctoral recipients by utilizing nationally representative datasets: …

Maternal Obesity Alters Fetal Development Due To Impaired Placental Function And Has Lasting Effects On Adult Offspring, Kristin Ann Norwood May 2013

Maternal Obesity Alters Fetal Development Due To Impaired Placental Function And Has Lasting Effects On Adult Offspring, Kristin Ann Norwood

Department of Animal Science: Dissertations, Theses, and Student Research

Obesity is an epidemic in many developed nations and maternal obesity can result in developmental alterations in offspring that have long-lasting effects. Two experiments were conducted to determine the impact of maternal obesity on placental development and early embryonic growth and muscle development. Experiment one utilized obese Lethal Yellow (LY) and normal weight C57BL/6 (B6) dams to assess how maternal obesity alters skeletal muscle development in mid-gestational embryos. Embryos from LY dams exhibited decreased embryo and placental weights as well as an increase in the myogenic marker desmin. Furthermore, the adipogenic marker PPARG expression was predominately localized to the neural …

Increasing Liana Abundance And Basal Area In A Tropical Forest: The Contribution Of Long‐Distance Clonal Colonization, Suzanne R. Yorke, Stefan A. Schnitzer, Joseph Mascaro, Susan G. Letcher, Walter P. Carson

Biological Sciences Faculty Research and Publications

Recent evidence suggests that liana abundance and biomass are increasing in Neotropical forests, representing a major structural change to tropical ecosystems. Explanations for these increases, however, remain largely untested. Over an 8‐yr period (1999–2007), we censused lianas in nine, 24 × 36 m permanent plots in old‐growth and selectively logged forest at La Selva Biological Station, Costa Rica to test whether: (1) liana abundance and basal area are increasing in this forest; (2) the increase is being driven by increased recruitment, decreased mortality, or both; and (3) long‐distance clonal colonization explains the increase in liana abundance and basal area. We …

Listeria Monocytogenes Cholecystitis: A Possible New Syndrome., Jackrapong Bruminhent, Timothy K Lynch, Jonathan Gefen, Jerome Santoro

Division of Internal Medicine Faculty Papers & Presentations

: The U.S. Food and Drug Administration recently added potentially fatal Listeria monocytogenes infection to the list of opportunistic infections that can occur in patients who receive tumor necrosis factor inhibitor therapy. In this study, the first reported case of L monocytogenes cholecystitis associated with etanercept use is described. It also appears that tumor necrosis factor inhibitor therapy likely increases the risk for Listeria cholecystitis. Clinicians need to be aware of this association when selecting antimicrobial therapy for these patients.

Evaluation Of Wetland Plants For Susceptibility To Species Of Phytophthora Present In Runoff Water At Ornamental Plant Nurseries, Garrett Ridge

All Theses

Competition between agricultural producers and urban communities for high quality water is increasing; therefore, growers need to adopt water conservation and reuse practices to ensure an adequate supply of irrigation water. One concern with recycled irrigation water is the potential to recirculate propagules of oomycete pathogens. Constructed wetlands are a biologically-based treatment option for nutrient and chemical remediation that potentially also could filter pathogen propagules. The goal of this study was to assess the susceptibility of plants routinely used in constructed wetlands to infection by species of Phytophthora commonly found at ornamental plant nurseries. In laboratory and greenhouse experiments, an …

Exploration Of Mouth Shading And Lighting In Cg Production, Chaoren Li May 2013

Exploration Of Mouth Shading And Lighting In Cg Production, Chaoren Li

All Theses

The lighting and shading of human teeth in current computer animation features and live-action movies with effects are often intentionally avoided or processed by simple methods since they interact with light in complex ways through their intricate layered structure. The semi-translucent appearance of natural human teeth which result from subsurface scattering is difficult to replicate in synthetic scenes, though two techniques are often implemented. The first technique is to create an anatomically correct layered model, and render the teeth with both theoretically and empirically derived optical parameters of human teeth using physical subsurface materials. The second technique largely takes advantage …

Assessment And Characterization Of In Situ Rotator Cuff Biomechanics, Erika Trent May 2013

Assessment And Characterization Of In Situ Rotator Cuff Biomechanics, Erika Trent

All Theses

Rotator cuff disease is a degenerative disorder that is a common, costly, and often debilitating, ranging in severity from partial thickness tear, which may cause pain, to total rupture, leading to loss in function. In current clinical practice, treatment decisions are frequently made through subjective assessment of pain and range of motion combined with qualitative assessment of x-ray images, magnetic resonance images (MRI), or ultrasound images. Treatment of disease may include physical therapy, surgery, or a combination of both. Often, the final determination of the best course of action is at the discretion of the clinician, and is based on …

Development Of An Ex Vivo Tissue Testing System For Three Dimensional Biomechanical Analysis Of Abdominal Aortic Aneurysm, Bradley Johns May 2013

Development Of An Ex Vivo Tissue Testing System For Three Dimensional Biomechanical Analysis Of Abdominal Aortic Aneurysm, Bradley Johns

All Theses

Abdominal aortic aneurysms (AAA) are localized, progressive dilations of the aortic wall and are the 13th most common cause of death in the United States (~15,000 per year) and surgery is usually recommended when the aneurysm is 5 cm or larger in diameter. Because of this clinical parameter, previous studies of AAA biomechanics have utilized a one-dimensional analysis that focuses solely on changes in wall diameter and have attempted to model geometric changes with mathematical formulas, generally based on the LaPlace equation. This is not sufficient however, as the mechanical behavior of the tissue at sites of aneurysm have been …

Evaluating Mechanical Performance Of Hydrogel-Based Adhesives For Soft Tissue Applications, Nitin Balakrishnan May 2013

Evaluating Mechanical Performance Of Hydrogel-Based Adhesives For Soft Tissue Applications, Nitin Balakrishnan

All Theses

According to the National Center for Health Statistics, an estimated 22 million women have undergone a hysterectomy procedure in the United States. The most common complication during hysterectomies is accidental laceration of the urinary bladder during the surgery with incidence between 0.2-8.3% with the current gold standard wound repair method being sutures. Yet, sutures come with their own limitations in that they necessitate use of a catheter and collection bag during healing due to preventing proper distention of the bladder tissue at normal pressures. The long-term goal of our study is to eliminate the need for suturing by creating a …

Measuring Arousal Through Physiological Responses To Packaging Designs: Investigating The Validity Of Electrodermal Activity As A Measure Of Arousal In A Realistic Shopping Environment, Daniel Hutcherson May 2013

Measuring Arousal Through Physiological Responses To Packaging Designs: Investigating The Validity Of Electrodermal Activity As A Measure Of Arousal In A Realistic Shopping Environment, Daniel Hutcherson

All Theses

The validity of electrodermal activity is investigated in the context of packaging design evaluation as a sensitive measure of arousal. Analysts agree that purchasing decisions are subconscious and emotional decisions, contrasted against the popular belief that consumers make purely rational decisions. To understand the personal and rapid character of a consumer's purchasing decisions we must find methods of which to measure and interpret consumer reactions to various packaging designs. Focus groups are discussed as antiquated research methods and new, advanced technologies are outlined as physiological responses. Past literature displays methodological approaches to using electrodermal activity measures in consumer studies, however …

Bioelectrochemical Reduction Of Carbon Dioxide To Acetate Using A Microbial Consortium Derived From The Cow Rumen, Ryan Hammonds May 2013

Bioelectrochemical Reduction Of Carbon Dioxide To Acetate Using A Microbial Consortium Derived From The Cow Rumen, Ryan Hammonds

All Theses

Pure cultures of chemoautotrophic microorganisms have previously been reported to produce acetate from carbon dioxide using reductive electric current. We hypothesized that electrofuel precursors could be produced in a similar manner from a consortium consisting of microbial populations acting as a stable community. This approach might offer improved efficiency and selectivity, greater sustainability, a more diverse group of end products, and more opportunities for community optimization in electrofuel production than is possible with pure cultures. To evaluate this concept, bovine rumen contents were enriched for autotrophic, anaerobic microbes for several generations using hydrogen and carbon dioxide for growth and energy. …
