Background: There is increasing epidemiologic evidence that arsenic exposure in utero, even at low levels found throughout much of the world, is associated with adverse reproductive outcomes and may contribute to long-term health effects. Animal models, in vitro studies, and human cancer data suggest that arsenic may induce epigenetic alterations, specifically by altering patterns of DNA methylation.
Objectives: In this study we aimed to identify differences in DNA methylation in cord blood samples of infants with in utero, low-level arsenic exposure.

The recent overpopulation of jellyfish occurring in the Barnegat Bay of New Jersey has been of growing concern. One prolific species, Chrysaora quinquecirrha, has been the subject of genetic research to determine the relatedness amongst populations and therefore determine its spread. In this study, a partial nucleotide sequence of the 45S ribosomal DNA cassette from C. quinquecirrha genomic DNA of Barnegat Bay was identified and compared to other cnidarian species. Approximately 44.8% of the cassette was identified, 3465 bp total. A partial 18S rDNA sequence was generated of 1772 bp (96% total). Complete ITS1, 5.8S rDNA, and ITS2 sequences were …

This thesis includes two studies on childhood obesity. This first study investigates whether childhood obesity rates affect their academic achievement scores by using a school-level panel data set on Arkansas 4th and 6th grades. The main results indicate that childhood obesity rates do not significantly affect academic achievement scores. Controls for education inputs such as library volume, educational expenditures per student and teacher salary show consistently significant positive relationship to students' test scores in both whole sample and in subsample analyses by socio-economic and minority status. Protein and oil are the major chemical constituents of soybean seed that affect the quality of soyfood, feed, and oil products. Therefore, soybean cultivars with high protein and/or high oil are desirable for the soyfood and feed markets. Use of molecular-marker-assisted selection will facilitate the breeding process of such cultivar development. The objectives of this research were to identify new quantitative trait loci (QTL) and confirm previously reported QTL associated with seed protein and oil content by using simple sequence repeat (SSR) markers and single nucleotide polymorphism (SNP) markers. Genome sequencing technology provides new and promising tests for clinical practice, including whole genome sequencing, which measures an individual’s complete DNA sequence, and whole exome sequencing, which measures the DNA for all genes coding for proteins. These technologies make it possible to test for multiple genes in a single test, which increases the efficiency of genetic testing. However, they can also produce large amounts of information that cannot be interpreted or is of limited clinical utility. This additional information could be distracting for patients and clinicians, and contribute to unnecessary healthcare costs. The potential for genomic sequencing to improve care …

Fatigue is a common and distressing symptom of childhood cancer that is often viewed as an inevitable part of cancer treatment and under assessed in clinical practice. Yet, cancer-related fatigue can disrupt normal childhood development and family functioning, as well as, signal serious complications of cancer and its treatment. Little is known about fatigue and quality of life among children undergoing cancer treatment in outpatient settings. This qualitative descriptive study explored parents' observations and interpretations of cancer-related fatigue and how it shaped their child's day-to-day quality of life during cancer treatment. Two experiments were conducted to investigate the effects of collection method and diet on estimating digestibility values. In Exp. 1, 24 barrows were fed either a corn-soybean meal diet (CSBM) or CSBM with 20% dried distillers grains with solubles (DDGS). Fecal collections were based on exact timing (Day collection) or marker-to-marker collection (MM) methods during a 4-d period. Diets contained 0.5% of titanium dioxide (TiO2) for estimating digestibility using the index method. Recently, corn has become expensive and this has led to high feed costs and decreased profitability. High priced corn has allowed for the consideration of other ingredients as replacements to reduce diet cost. In much of the Corn Belt, supplies of wet or modified distillers grains and crop residues are abundant. These ingredients were investigated as corn replacements. Studies were conducted to identify methods for treating crop residues to improve digestibility and value in finishing diets based on corn grain and corn wet distillers grain with solubles (WDGS). Bacteria are well known to obtain tolerance to antibiotics by forming multicellular structures, known as biofilms, and by entering dormancy and forming persister cells. Both mechanisms allow bacteria to tolerate antibiotics at concentrations hundreds to thousands of times higher than the lethal dose for regular planktonic cells of the same genotype. Persister formation increases in biofilms; thus, effective control of persister cells, especially those in biofilms is critically important to infection control. Understanding the genetic and environmental factors responsible for differences in size, morphology and behavior can aid in determining how alternative male mating strategies evolve in natural populations. One important environmental factor is nutrition, as it underlies growth of both body size and morphological traits that are linked to alternative mating strategies in many animals. In sailfin mollies, Poecilia latipinna , male size is fixed at maturity, highly variable within populations and correlated with dorsal fin morphology and expression of alternative male mating behaviors. Though many HPLC methods have been developed and reported in literature for vitamin analysis for the past two decades, applying certain methods directly from literature more than often fails to reproduce the results reported due to many variables of liquid chromatography. This issue was targeted in this project through the examination of chromatographic behaviors of water-soluble vitamins in order to help the analysts better modify methods from literature or even develop new methods from scratch to fit their analytical need with the resources available (e.g., columns, detectors, etc.) in their lab.

Timor-Leste, which gained its independence in 2002, has accumulated $13 billion in its petroleum fund in less than a decade. Many physiological processes of Daphnia are negatively affected at low calcium concentrations. The concentration of calcium within lake water influences how Daphnia populations will survive and reproduce in natural environments. An experiment was designed to test the effects of calcium concentration and food level on the growth and reproduction of a Daphnia hybrid. Infectious disease can influence organisms at all levels of ecological organization, from individuals to ecosystems. Likewise, the ecosystems where pathogens exist directly influence their success. Recent theoretical studies have tied disease prevalence to biotic factors such as genetic diversity, biodiversity, and host behavior, and abiotic factors that include temperature and increased nutrient concentrations. Olanzapine (OLZ) is an effective first-line treatment for schizophrenia and bipolar disorder. The benefits of OLZ are countermanded by side effects such as weight gain, glucose intolerance, dyslipidemia, and liver injury. These effects impact not only antipsychotic medication compliance, but also increase the health risks to patients. Food safety knowledge, attitudes, and behaviors of Native American families with young children were studied using a mixed methods approach. Quantitative and qualitative data was collected together, analyzed separately, and compared. A food safety knowledge survey was created using the FightBac!™ concepts. A total of 102 participants completed the knowledge survey during survey validation and as part of focus groups. Anthropogenic disturbances have altered the natural flow regimes of large rivers, and have led to declines in species that rely on elements of natural flow. Similarly, shovelnose sturgeon distribution has diminished in the last 100 years due to habitat alteration, overharvest, and water contamination. To fully understand the status and viability of a fish population, basic knowledge of a fish’s reproductive strategy is needed. Ames dwarf (df/df) mice are deficient in anterior pituitary hormones: growth hormone (GH), thyroid stimulating hormone (TSH), and prolactin (PRL) due to a spontaneous, homozygous mutation of prop1[superscript df] gene. These dwarf mice exhibit characteristics such as delayed growth and development coupled with delayed aging, increased lifespan, overall increased insulin sensitivity, as well as resistance to certain diseases and cancers. The mutant mice possess low blood glucose, low serum insulin, and lower body temperature. Their enhanced longevity (about 40-60% longer lifespan than normal mice) is associated with their GH deficiency and disruption in the somatotropic axis (GH/IGF-1 hormonal pathway) as …

The antimicrobial properties of a chitosan-coated vacuum-packaged pouch were tested against Listeria monocytogenes (LM) inoculated on frankfurters. A 1.5% chitosan solution was successfully coated onto a corona-treated poly/nylon based film and then heat-sealed to form a pouch. An initial 5 log CFU/mL population of LM was used to inoculate a frankfurter in the pouch, which was then vacuum-sealed and stored at 4¡ and 13¡ C for a period of 8 weeks, with bacteria enumeration performed on weekly intervals.
