Functional Environmental Screening Of A Metagenomic Library Identifies Stla; A Unique Salt Tolerance Locus From The Human Gut Microbiome, Roy D. Sleator, Et. Al.

Functional environmental screening of metagenomic libraries is a powerful means to identify and assign function to novel genes and their encoded proteins without any prior sequence knowledge. In the current study we describe the identification and subsequent analysis of a salt-tolerant clone from a human gut metagenomic library. Following transposon mutagenesis we identified an unknown gene (stlA, for “salt tolerance locus A”) with no current known homologues in the databases. Subsequent cloning and expression in Escherichia coli MKH13 revealed that stlA confers a salt tolerance phenotype in its surrogate host. Furthermore, a detailed in silico analysis was also …

When John Barber (Statesville, N.C.) walked across the stage to receive his diploma during Gardner-Webb University’s fall commencement, it was an achievement for which he had long hoped. Barber, once homeless, survived extreme circumstances and in 2009, he enrolled in the GWU GOAL (Greater Opportunity for Adult Learners) program. He received a Bachelor of Science in Business Administration degree on Dec. 16.

Abstract Burnout is a syndrome consisting of emotional exhaustion (EE) and depersonalization (DPZ) (Yong & Yue, 2007). Teachers who fall victim to burnout are likely to be less sympathetic toward students, have a lower tolerance for classroom disruption, be less apt to prepare adequately for class, and feel less committed and dedicated to their work (Betoret, 2006; Byrne, 1991; Fisher, 2011). The purpose of this phenomenological study was to explore the burnout experiences among secondary teachers and the ways they cope with the adverse conditions. DNA microarrays are a potentially disruptive technology in the medical field, but their use in such settings is limited by poor reliability. Microarrays work on the principle of hybridization and can only be as reliable as this process is robust, yet little is known at the molecular level about how the surface affects the hybridization process. This work uses advanced molecular simulation techniques and an experimentally-parameterized coarse-grain model to determine the mechanism by which hybridization occurs on surfaces and to identify key factors that influence the accuracy of DNA microarrays. Identification Of The Binding Partners For Hspb2 And Cryab Reveals Myofibril And Mitochondrial Protein Interactions And Non-Redundant Roles For Small Heat Shock Proteins, Kelsey Murphey Langston

Small Heat Shock Proteins (sHSP) are molecular chaperones that play protective roles in cell survival and have been shown to possess chaperone activity. As such, mutations in this family of proteins result in a wide variety of diseases from cancers to cardiomyopathies. The sHSPs Beta-2 (HspB2) and alpha-beta crystalline (CryAB) are two of the ten human sHSPs and are both expressed in cardiac and skeletal muscle cells. A heart that cannot properly recover or defend against stressors such as extreme heat or cold, oxidative/reductive stress, and heavy metal-induced stress will constantly struggle to maintain efficient function. The majority of schools use punitive and reactive strategies to deal with maladaptive behaviors. This descriptive study was a replication of Wilmott (2012) and occurred in a multi-tiered system of supports (MTSS) high school. Professional development was provided to 22 teachers from a high school located in the western United States. This included 61% males and 39% females. Measures included the frequency of praise notes and ODRs, the quality of praise notes based on a praise note rubric, and a social validity questionnaire. The ODR and praise note frequencies were compared using the Spearman correlational coefficient that resulted in a …

There is considerable variability in response to behavioral weight control treatment. Identifying pretreatment health behaviors that predict an individual’s weight loss success has the potential to assist with assignment into more appropriate treatment settings (i.e., standard behavioral program versus comprehensive home environment program) that are likely to yield greater weight loss outcomes over time. Participants (N= 201; 48.9 ± 10.5 years; 78.1% women) enrolled in a study comparing a standard behavioral weight loss program (BWL; n=99) to a home-environment focused weight loss program (BWL+H; n=102) completed baseline questionnaires assessing individual control variables (i.e., dietary restraint, self-weighing) and control over the …

Electronic consults (e-consults) are types of communications systems used between physicians and their patients to communicate outside the medical office. While nothing can replace the traditional face-to-face interaction, e-consults can be used to help manage patient queue, share non-urgent information, and provide an alternative avenue of communication while recording electronic copies. Literature reveals that lack of a standardized reimbursement system, uncertain workloads, and perceived risks to patient confidentiality are barriers to mainstream adoption of e-consult systems. Until the late 1960s, serious scholarship involving the impact of popular music as political expression was lacking. Studies in rhetorical criticism tended to focus upon speeches, essays, and historical documents. Yet in the 1970s, protest songs were given more attention by scholars. However, this trend was not continued into the 2000s. By analyzing the ideological themes within the lyrics, this thesis examined the rhetorical function of protest songs performed during the 2000s. Background: Evaluation of task related outcomes within geriatric and fall-prone populations is essential not only for identification of neuromuscular deficits, but also for effective implementation of fall prevention strategies. As most tasks and activities of daily living are performed at submaximal force levels, restoration of muscle strength often does not produce the expected benefit in functional capacity. However, it is known that muscular control plays a key role in the performance of functional tasks, but it remains unclear to what degree muscular control and the associated neuromuscular noise (NmN) is age-related, particularly in the lower-extremities.

The goal of turfgrass nitrogen (N) fertilization is to supply sufficient N for aesthetically and functionally high-quality turfgrass without accumulation of excessive soil N, which can facilitate nitrate (NO3-) leaching losses. Currently, subjective measures (e.g., assumed N needs of turfgrass species; visual preference of turfgrass quality; historical N rates) remain the primary means used to guide N fertilization of turf. This makes it is more likely that excessive N will be applied to turfgrass. Today, 72% of individuals with intellectual and developmental disabilities (IDDs) in the United States live with a caregiver (Braddock & Rizzolo, 2013). In many families, siblings of individuals with IDDs are the default long-term care providers (Heller et al., 2008). Despite the fact that research has begun to recognize and explore the roles of family members in care, little is known about sibling support providers (SSPs).
