Remote sensing can provide long-term and large-scale products helpful for ecosystem model evaluation. The authors compare monthly gross primary production (GPP) simulated by the Community Land Model, version 4 (CLM4) at a half-degree resolution with satellite estimates of GPP from the Moderate Resolution Imaging Spectroradiometer (MODIS) GPP product (MOD17) for the 10-yr period January 2000–December 2009. The assessment is presented in terms of long-term mean carbon assimilation, seasonal mean distributions, amplitude and phase of the annual cycle, and intraannual and interannual GPP variability and their responses to climate variables. Algorithms For Optimizing Cross-Overs In Dna Shuffling, Lu He, Alan M. Friedman, Chris Bailey-Kellogg

DNA shuffling generates combinatorial libraries of chimeric genes by stochastically recombining parent genes. The resulting libraries are subjected to large-scale genetic selection or screening to identify those chimeras with favorable properties (e.g., enhanced stability or enzymatic activity). While DNA shuffling has been applied quite successfully, it is limited by its homology-dependent, stochastic nature. Consequently, it is used only with parents of sufficient overall sequence identity, and provides no control over the resulting chimeric library.
Results: This paper presents efficient methods to extend the scope of DNA shuffling to handle significantly more diverse parents and to generate more predictable, optimized libraries. …

The disappearance of the federally endangered American burying beetle (Nicrophorus americanus Olivier) over most of its range is poorly understood, which is why the identification and evaluation of conservation measures is important. The presence and distribution of all carrion beetles in Nebraska was first reported to help understand where American burying beetles (ABBs) occur to aid in creating a habitat suitability model for the Sandhills ecoregion and to locate suitable experiment areas. There are 18 Nebraska counties with records of ABB presence in the last 15 years. The final habitat suitability model performed well with an AUC value above …

Vertebrates are associated with trillions of bacteria, with the densest populations residing in the large intestine. The symbiosis between vertebrates and their gut microbiota has resulted in important implications of the gut microbiome on host health. Diet is an important factor that shapes gut microbiota composition, and because of the interplay between host-microbiome-diet, dietary strategies that modulate gut microbiome structure are deemed a relevant tool to improve host health. River otters (Lontra canadensis) are native to Nebraska but were extirpated by the early 1900s. The Nebraska Game and Parks Commission (NGPC) reintroduced river otters during 1986–1991 to restore the species in the state but little is known regarding the habitat needs and status of this high profile threatened species. To provide information for management I conducted research to determine home range, habitat use, overnight movement distance, and annual survival of river otters in the central Platte River of Nebraska.
I trapped, implanted telemetry transmitters, and tracked 18 river otters during 2006–2009. I obtained 996 locations and constructed …

Nuclear Magnetic Resonance (NMR) has proven to be an excellent tool to probe the structure of molecules by gaining insight into nuclear interactions. Used in conjunction with theoretical calculations, NMR has the potential to elucidate these nuclear interactions and how they affect structure, bonding and dynamics on a molecular scale. The first two projects presented herein study the peroxide explosive hexamethylene triperoxide diamine (HMTD). Alzheimer's disease (AD) is an age progressive neurodegenerative brain disorder, affecting 37 million people worldwide. Cleavage of amyloid precursor protein by ?- and ?-secretase produces the amyloid-beta (A?) protein, which significantly contributes to AD pathogenesis. The A? aggregates, formed at the surface of neurons and intracellularly, cause neurotoxicity and decrease synaptic function. Inhibiting or degrading A? accumulation is a key goal for development of new AD treatments. Natural Killer (NK) cells are a subset of lymphocytes that regulate adaptive immune responses and utilize "missing self" recognition to activate anti-tumor and anti-viral cytotoxicity. Clinical research, as well as murine and ex vivo models, have shown that a variety of NK cell applications have proven useful as immunotherapeutic treatments for patients with hematologic malignancies. However, the selective expansion of NK cells to yield relevant amounts of these lymphocytes has been a major hurdle in the development of methods for clinical therapeutic use. Over the past several years, increasing public emphasis on preventing child maltreatment has resulted in substantial changes to Florida's child abuse and neglect central registry. Many of these recent changes, aimed at preventing child maltreatment, have resulted in over one million false, unsubstantiated, and inconclusive reports of child abuse and neglect within the last decade. The purpose of this study is to examine adult literacy beyond its constraints as a social problem and instead consider the implications of illiteracy as a particular form of lived experience, analogous to women's oppression at large. Through a complex system of meaning making, the knowledge accrued by illiterate adults is qualitatively different, and examining these differences in terms of their correlation to coping mechanisms developed in the face of social alienation and diminished professional prospects yields a greater understanding of class privilege and how nontraditional learners fit into a larger social structure. The main objective in this research was to examine the extent to which gender and gender biases influence monetary benefits received, including jury award amounts, in workplace sexual harassment claims. Two methods were utilized to explore the discrepancies in monetary benefits received based on gender differences. We investigated three methods for estimating centre of pressure excursions, as measured using a portable pressure sensor matrix, in order to deploy similar technology into the homes of older adults for longitudinal monitoring of postural control and falls risk. We explored the utility of these three methods as markers of falls risk in a cohort of 120 community dwelling older adults with and without a history of falls (65 fallers, 55 non-fallers). The host range of poxviruses is highly regulated and is limited to certain hosts. A number of genes in different poxviruses have been identified that contribute to cell tropism. The ankyrin (ANK) repeat genes encode for proteins characterized by the repetitive motifs in its amino acid sequence. These genes have been found to play a role in determining the ability of poxviruses to replicate in certain cell types. A new class of geminally-substituted nitroso compounds, i.e., α-benzotriazoyl nitroso derivatives, is presented. These compounds display a rather different behavior than other related nitroso compounds bearing a geminal electron-withdrawing group. An unexpected and spontaneous oxidation to the nitro analog is observed in solution.

This thesis focuses on an adaptive quadratic spiking model of a motoneuron that is both versatile in its ability to represent a range of experimentally observed neuronal firing patterns as well as computationally efficient for large network simulation. The objective of research is to fit membrane voltage data to the model using a parameter estimation approach involving simulated annealing. By manipulating the system dynamics of the model, a realizable model with linear parameterization (LP) can be obtained to simplify the estimation process. This study was conducted to gather information about why collegiate women discontinue participation in sports. Women at a Midwestern Division III institution participated in this study. Three groups of women were interviewed through focus groups: women no longer playing a sport, women currently playing collegiate athletics, and a mix of women currently playing a college sport and women no longer involved. The women were asked questions about sport involvement and answered collectively. Women provided responses that fell into 5 main categories: psychological, physical, educational, social, and attachment. Understanding subsurface structure by studying microseismicity influences a wide range of activities, including energy extraction, aquifer storage, carbon sequestration, and seismic hazard assessment. Identifying individual fractures in a larger fault system is key to characterizing, understanding, and potentially mitigating risks of natural or induced seismicity.
A year-long study associated with a carbon dioxide (CO2) sequestration project was conducted at the Aneth oil field in southeast Utah to record microseismicity at a single downhole geophone array. AutoDock is a well-known and powerful software program used to model ligand to receptor binding interactions. The cytochrome P450 (CYP) superfamily of heme enzymes play an important role in the metabolism of a large number of endogenous and exogenous compounds, including most of the drugs currently on the market. Inhibitors of CYP enzymes have important roles in the treatment of several disease conditions such as numerous cancers and fungal infections in addition to their critical role in drug-drug interactions. Vascular endothelial dysfunction is one of the earliest recognizable events under hyperglycemic conditions. It is characterized by decreased endothelium-derived nitric oxide (NO) bioavailability and increased oxidative stress, such as superoxide and hydrogen peroxide (H2O2) overproduction. However, the real-time changes in blood NO and H2O2 levels under acute hyperglycemia have not been evaluated. In this study, acute hyperglycemia (200 mg/dl, 400 mg/dL, and 600 mg/dL) was induced by intravenous infusion of 20%, 30%, and 50% D-glucose respectively for 180 min. Young children who display extreme levels of disruptive behaviors are at increased risk for later academic difficulties, poor social relationships and adolescent delinquency, making early intervention efforts a priority. Studies evaluating Child-Parent Relationship Therapy (CPRT) have demonstrated its effectiveness in improving parental empathy, improving parental acceptance, decreasing parenting stress, and decreasing perceived child problem behavior. These outcomes, however, must be evaluated in light of several research limitations (e.g., lack of randomization, use of non-standardized measures, lack of treatment integrity data). Well conducted studies that assess the effectiveness of CPRT on specific presenting problems are needed to evaluate better the impact, …

Marital quality is a broad measurement of perceptions of satisfaction, happiness, and stability by partners in an established relationship. Marital quality has been relatively understudied among Mexican Americans, a population that warrants the inclusion of cultural constructs in any model concerning relationship outcomes. Therefore, acculturation differences between Mexican American couples were conceptualized as a distal context for understanding marital quality. Traditional gender role values and communication style (warmth and hostility) were included as proximal contexts. Data from Conger’s California Families Project were utilized; results indicated that while most measures of acculturation did not impact marital quality, language use interacted with …

In recent decades, a new branch of psychology has emerged—ecopsychology. This field looks at the complex relationship between human beings and the natural world. Previous research has shown the importance of direct experiences in nature for the promotion of human health and well-being. However, it is still not understood why these experiences in nature carry so much importance.
It is possible that the concept of connectedness to nature (CTN) plays a role in this relationship between nature and wellness. CTN is defined as feelings of close affiliation with the natural world in a physical, cognitive, and emotional manner. Previous research …

Anaerobic digestion has great potential as an energy source. Not only does it provide an effective method for waste mitigation, but it has the potential to generate significant quantities of fuel and electricity. In order to ensure efficient digestion and biomass utilization, however, the system must be continuously maintained at elevated temperatures. It is technically feasible to supplement such a system with outside energy, but it is more cost effective to heat the system using only the produced biogas.
