Modeling Environmental Contamination Of Antibiotic-Resistant Bacteria In Hospitals, Lei Wang Aug 2012

Modeling Environmental Contamination Of Antibiotic-Resistant Bacteria In Hospitals, Lei Wang

Open Access Dissertations

Methicillin-resistant Staphylococcus aureus (MRSA) is a bacterium that causes infections in different parts of the body. It is tougher to treat than most strains of Staphylococcus aureus or staph, because it is resistant to some commonly used antibiotics. In this work, we investigate the role of environmental contamination on the clinical epidemiology of antibiotic-resistant bacteria in hospitals. A compartmental model is constructed to describe the transmission characteristics of MRSA in hospital setting. The deterministic epidemic model includes five compartments: colonized and uncolonized patients, contaminated and uncontamincated health care workers (HCWs), and bacterial load in environment. Basic reproduction number R0 is …

Active Induction Of Experimental Autoimmune Encephalomyelitis By Mog35-55 Peptide Immunization Is Associated With Differential Responses In Separate Compartments Of The Choroid Plexus, Nivetha Murugesan, Debayon Paul, Yen Lemire, Bandana Shrestha, Shujun Ge, Joel S. Pachter

UCHC Articles - Research

Background

There is increasing awareness that, aside from producing cerebrospinal fluid, the choroid plexus (CP) might be a key regulator of immune activity in the central nervous system (CNS) during neuroinflammation. Specifically, the CP has recently been posited to control entry of sentinel T cells into the uninflamed CNS during the early stages of neuroinflammatory diseases, like multiple sclerosis (MS) and its animal model experimental autoimmune encephalomyelitis (EAE). As the CP is compartmentalized into a stromal core containing fenestrated capillaries devoid of typical blood–brain barrier properties, surrounded by a tight junction-expressing choroidal epithelium, each of these compartments might mount unique …

Modeling Sleep Fragmentation In Populations Of Sleep Hypnograms, Bruce J. Swihart, Naresh M. Punjabi, Ciprian M. Crainiceanu Aug 2012

Modeling Sleep Fragmentation In Populations Of Sleep Hypnograms, Bruce J. Swihart, Naresh M. Punjabi, Ciprian M. Crainiceanu

Johns Hopkins University, Dept. of Biostatistics Working Papers

We introduce methods for the analysis of large populations of sleep architectures (hypnograms) that respect the 5-state 20-transition-type structure defined by the American Academy of Sleep Medicine. By applying these methods to the hypnograms of 5598 subjects from the Sleep Heart Health Study we: 1) provide the firrst analysis of sleep hypnogram data of such size and complexity in a community cohort with a 4-level comorbidity; 2) compare 5-state 20-transition-type sleep to 3-state 6-transition-type sleep for a check of feasibility and information-loss; 3) extend current approaches to multivariate survival data analysis to populations of time-to-transition processes; and 4) provide scalable …

A Phase I Bayesian Adaptive Design To Simultaneously Optimize Dose And Schedule Assignments Both Among And Within Patients, Thomas M. Braun, Jin Zhang Aug 2012

A Phase I Bayesian Adaptive Design To Simultaneously Optimize Dose And Schedule Assignments Both Among And Within Patients, Thomas M. Braun, Jin Zhang

The University of Michigan Department of Biostatistics Working Paper Series

In traditional schedule or dose-schedule finding designs, patients are assumed to receive their assigned dose-schedule combination throughout the trial even though the combination may be found to have an undesirable toxicity profile, which contradicts actual clinical practice. Since no systematic approach exists to optimize intra-patient dose-schedule as- signment, we propose a Phase I clinical trial design that extends existing approaches that optimize dose and schedule solely among patients by incorporating adaptive variations to dose-schedule assignments within patients as the study proceeds. Our design is based on a Bayesian non-mixture cure rate model that incorporates multiple administrations each patient receives with …

Flow Valve Diagnostics For Label-Free, Quantitative Biomarker Detection: Device Fabrication, Surface Modification, And Testing, Danielle Scarlet Mansfield Aug 2012

Flow Valve Diagnostics For Label-Free, Quantitative Biomarker Detection: Device Fabrication, Surface Modification, And Testing, Danielle Scarlet Mansfield

Theses and Dissertations

Diseases are often diagnosed by detection of disease-specific biomarkers in fluid samples. However, many state-of-the-art detection methods require a lab with complex machinery, trained operators, and/or lengthy analysis time. In contrast, point-of-care (POC) devices are brought to the patient's location, they are easy to use, and results are obtained almost immediately. Many current POC devices are too difficult to be used without a skilled assistant, and although many are able to detect analytes above a threshold value, they give little or no quantitative information. This work presents the development of polymer-based microfluidic devices capable of sensing and quantifying biomarkers in …

Formation Of Cyclopentene Nitro-Ester And Derivatives, Kirpal S. Bisht, Pasha M. Khan, Alberto Van Olphen, Ruizhi Wu Aug 2012

Formation Of Cyclopentene Nitro-Ester And Derivatives, Kirpal S. Bisht, Pasha M. Khan, Alberto Van Olphen, Ruizhi Wu

USF Patents

Disclosed is a method of synthesizing new optically pure heterocyclic compounds using Pd(0) catalyzed intramolecular cyclizations. Analogs of cyclopentanes, like isoxazoline-2-oxide and furan, with similar framework to the cyclopentanes act as anti-HIV and anticancer agents which opens a whole new field for application of these compounds. Starting from a meso-diol, optically pure compounds were prepared without utilizing chiral ligands at any stage of the synthesis. The stereochemical outcome of the product (>99% ee) was influenced by desymmetrization catalyzed by Pseudomonas cepacia lipase and the stereoselective nature of the palladium catalyzed transformations.

Mathematics Faculty Publications

Background: Despite decades of new discoveries in biomedical research, the overwhelming complexity of cells has been a significant barrier to a fundamental understanding of how cells work as a whole. As such, the holistic study of biochemical pathways requires computer modeling. Due to the complexity of cells, it is not feasible for one person or group to model the cell in its entirety. Results: The Cell Collective is a platform that allows the world-wide scientific community to create these models collectively. Its interface enables users to build and use models without specifying any mathematical equations or computer code - addressing …

Characterization Of The Cellular And Organellar Dynamics That Occur With A Partial Depletion Of Mitochondrial Dna When Arabidopsis Organellar Dna Polymerase Ib Is Mutated, John D. Cupp Aug 2012

Characterization Of The Cellular And Organellar Dynamics That Occur With A Partial Depletion Of Mitochondrial Dna When Arabidopsis Organellar Dna Polymerase Ib Is Mutated, John D. Cupp

Theses and Dissertations

Plant mitochondrial genomes are large and complex, and the mechanisms for maintaining mitochondrial DNA (mtDNA) remain unclear. Arabidopsis thaliana has two DNA polymerase genes, polIA and polIB, that have been shown to be dual localized to mitochondria and chloroplasts but are unequally expressed within primary plant tissues involved in cell division or cell expansion. PolIB expression is observed at higher levels in both shoot and root apexes, suggesting a possible role in organelle DNA replication in rapidly dividing or expanding cells. It is proposed that both polIA and polIB are required for mtDNA replication under wild type conditions. An Arabidopsis …

Synthesis And Biological Evaluation Of Various Derivatives Of A Broad-Spectrum Anticancer Nucleoside, Jadd R. Shelton Aug 2012

Synthesis And Biological Evaluation Of Various Derivatives Of A Broad-Spectrum Anticancer Nucleoside, Jadd R. Shelton

Theses and Dissertations

Recently the Peterson lab discovered a promising anticancer adenosine derivative-- 2´,3´-bis-O-tert-butyldimethylsilyl-5´-deoxy-5´-[N-(methylcarbamoyl)amino]-N6-(N-phenylcarbamoyl)adenosine. This compound showed selective toxicity against human colon cancer cells in vitro with LC50's = 6--10 µM. It was hypothesized that the lead compound exerted its cytotoxic effects by interacting with a protein kinase. A systematic Structure Activity Relationship (SAR) was undertaken in an attempt to increase the kinase-binding affinity of the lead compound. Many regions of the lead compound were examined: the N6-phenyl urea moiety, the 5´-N-methyl urea group, the 2´,3´-bis-O-TBS groups, the nucleobase, and the ribose sugar. Results of these studies produced some promising new derivatives. In …

The Forgotten Medium: The Impact Of Reading Violent Literature On Aggressive Thoughts, Mckay Robert Stevens Aug 2012

The Forgotten Medium: The Impact Of Reading Violent Literature On Aggressive Thoughts, Mckay Robert Stevens

Theses and Dissertations

Media violence in television, film, video games, and music has been linked to increased aggression. Only in recent years have researchers begun to investigate the impact that reading violent literature can have on individuals. The present study exposed individuals to either a violent or nonviolent story and measured aggressive cognitions. No significant effect was found for story type on aggressive cognitions but a gender effect for aggressive word completions was found. Discussion centers on possible differences between media types as well as future suggestions for investigations into violent literature.

Pairwise Protein Expression Classifier For Candidate Biomarker Discovery For Early Detection Of Human Disease Prognosis, Parminder Kaur, Daniela Schlatzer, Kenneth Cooke, Mark R. Chance Aug 2012

Pairwise Protein Expression Classifier For Candidate Biomarker Discovery For Early Detection Of Human Disease Prognosis, Parminder Kaur, Daniela Schlatzer, Kenneth Cooke, Mark R. Chance

Faculty Scholarship

Background: An approach to molecular classification based on the comparative expression of protein pairs is presented. The method overcomes some of the present limitations in using peptide intensity data for class prediction for problems such as the detection of a disease, disease prognosis, or for predicting treatment response. Data analysis is particularly challenging in these situations due to sample size (typically tens) being much smaller than the large number of peptides (typically thousands). Methods based upon high dimensional statistical models, machine learning or other complex classifiers generate decisions which may be very accurate but can be complex and difficult to …

Fillial Responsibility: A Survey Across Time And Oceans , Andrea Rickles-Jordan Aug 2012

Fillial Responsibility: A Survey Across Time And Oceans , Andrea Rickles-Jordan

Marquette Elder's Advisor

In the United States, Medicare and Medicaid are helping fund the cost of caring for the elderly. However, long before these programs existed, several states enacted statutes requiring children to care for their elderly parents. These filial responsibility statutes still exist in many states today, although their effectiveness and use has been called into question by opponents. To understand the current need for these statutes, this article first looks at the reasons the statutes were created. The article then follows their growth throughout the history of the world and the United States. As further means of comparison, the article examines …

Bringing Age Discrimination And Disability Discrimination Together: Too Few Intersections, Too Many Interstices, Leslie Pickering Francis, Anita Silvers Aug 2012

Bringing Age Discrimination And Disability Discrimination Together: Too Few Intersections, Too Many Interstices, Leslie Pickering Francis, Anita Silvers

Marquette Elder's Advisor

Old age and disability would seem to have adverse features in common. Membership in either group suggests depleted capability, decreased social contribution, significant fragility, and heightened susceptibility to maltreatment by other people. Yet some have argued that no wrong occurs when advanced age is made disadvantageous, for the elderly have had a fair chance at the goods of life. By contract, this argument goes, there is never a fair chance for people with disabilities. This article challenges the view that takes discrimination based on disability and discrimination based on age as so discrepant that they do not deserve similar efforts …

Old Lawyers, Blue Eyes, And The Medicalization Of Aging, Michael J. Myers Aug 2012

Old Lawyers, Blue Eyes, And The Medicalization Of Aging, Michael J. Myers

Marquette Elder's Advisor

The article examines the "medicalization" of the aging process and the pitfalls of the increased medical care and myths about what constitutes "healthy" later in life. Financial reward and fear of death drive the increasing amount and extensiveness of care for chronic disease encountered in older adults, sometimes to an unnecessary end. The author puts forth the theory that older adults are now part of a weakened generation, rooted in a refusal to "suffer their reality" and ignore the fact that aging is not a disease, that is, not until it is placed under a doctor's care.
