The cosmopolitan make-up of the American society has yielded cultural hybrid offspring and this cultural hybridity features strongly in contemporary American fiction. Amy Tan and Mary Yukari Waters are both Asian-Americans who portray such hybridity in their short stories which depict the shifting identities of the self. But do the internal categories of gender, race and ethnicity help in the coherence or do they add to the fragmentation of diverse identities? Success of the cattle industry in Latin America is impeded by the common vampire bat, Desmodus rotundus, through decreases in milk production and mass gain and increased risk of secondary infection and rabies. We used ecological niche modeling to predict the current potential distribution of D. rotundus and the future distribution of the species for the years 2030, 2050, and 2080 based on the A2, A1B, and B1 climate scenarios from the Intergovernmental Panel on Climate Change. Science education reform has prompted scientists, engineers, and politicians alike to act on the growing need for improved educational programs in the STEM fields. Informal science programs that provide an educational experience outside of a traditional classroom and involve students on real-life and hands-on experiments are among these initiatives. It is hypothesized that informal science programs may increase students interest and performance in science and related subjects. Candida albicans, a fungal pathogen of humans, regulates its morphology in response to many environmental cues and this morphological plasticity contributes to virulence. Farnesol, an autoregulatory molecule produced by C. albicans, inhibits the induction of hyphal growth by inhibiting adenylate cyclase (Cyr1). The role of farnesol and Cyr1 in controlling the maintenance of hyphal growth has been less clear. Here, we demonstrate that preformed hyphae transition to growth as yeast in response to farnesol and that strains with increased cyclic AMP (cAMP) signaling exhibit more resistance to farnesol. Microorganisms numerically dominate terrestrial biodiversity, and play important biochemical and geochemical roles in the environments they inhabit. To understand structure and function of complex ecosystems, it is essential to identify primary drivers of microbial diversity and community structure. Historically, the study of microbial ecology was reductive, limited to microbes able to be cultured and enumerated. Microbes meeting this criterion were thought to comprise the dominating members of the environments they were isolated from, however, estimates suggesting up to 99% of the endogenous species are uncultivable with existing methodologies; a concept that reflects experimental failure, rather than a verifiable conclusion. Chilodonella uncinata, like all ciliates, contains two distinct nuclei in every cell: a germline micronucleus and a somatic macronucleus. The macronucleus develops from the zygotic nucleus through a series of chromosomal rearrangements. Macronuclear development in C. uncinata yields a nucleus with highly amplified gene-sized chromosomes. The macronucleus is transcriptionally active during vegetative growth while there is no expression in the micronucleus except during a brief period following conjugation. Gene family evolution in ciliates occurs through complex processes including gene duplication and an alternative processing of scrambled genes. Quorum sensing in bacteria is a process of cell-to-cell communication facilitated by small molecules called autoinducers. Interspecies quorum sensing is facilitated by the autoinducer AI-2. The enzyme LuxS catalyzes the formation of AI-2 from S-ribosyl homocysteine (SRH) in a wide variety of bacterial species. Inhibition of LuxS would therefore inhibit interspecies quorum sensing. The goal of this project is to establish biochemical assays for the evaluation of small molecules as potential LuxS inhibitors. The first assay is a conventional colorimetric assay that utilizes Ellman’s reagent to quantify the homocysteine byproduct of DPD production by LuxS. For this assay purified …

The objectives of this study were to develop a transcriptomic reference resource and to characterize polymorphism between isolates of Erysiphe necator (syn. Uncinula necator), grape powdery mildew. The wine and fresh fruit markets are economically vital to many countries worldwide, and E. necator infection can cause severe crop damage and subsequent financial loss. Most of the publicly available sequence data for Erysiphales are from research done on Blumeria graminis f. sp. hordei, barley powdery mildew, which occupies a distinct clade within the Erysiphales. We obtained 641,601 sequencing reads from a Roche 454–FLX next–generation sequencer (RNA–Seq) and performed multiple assemblies using …

The chemical constituents of Ochrosia oppositifolia have been studied. The compounds were extracted from the bark and leaves of this plant by using n-hexane,dichloromethane and methanol as solvents. The crudes were subjected to extensive chromatographic techniques like preparative thin layer chromatography, column chromatography and high performance liquid chromatography (HPLC). A total of six compounds were obtained. Structural elucidation was established through several spectroscopic methods, such as 1D-NMR (1H, 13C, DEPT, NOE), 2D-NMR (COSY,NOESY, HMQC, and HMBC), UV, IR, and MS (GCMS, LCMS and HRMS) and comparison with the published data. This paper offers and initial examination of representations of Asian American mixed race during the long period of Asian exclusion and enforced anti-miscegenation when racial mixing was legally proscribed. I argue that this literature must be understood through two categories Orientalist texts that produce and reproduce dominant discourses about Asian American mixed race and cosmopolitan texts written by mixed-race figures circling in vanguard cliques of art and literature. After tracking the representations of early Asian American mixed race in U.S. literature, this paper offers an analysis of the writing of Asian American mixed-race authors themselves in order to expand …

This paper focuses on the contradictions and tensions inherent in self-identification and oppositional identities. In the past twenty years, Hapa has emerged as an oppositional identity used by mixed-race Asian Americans. While many scholars have noted the tension of the term for its linguistic appropriation from the Native Hawaiian language, fewer scholars have concentrated their examinations on the ways in which the term reproduces the very notions it aims to subvert. This paper concentrates on the areas of contradiction inherent in finding and using "a word of power" and making that word into a recognizable identity.

It is an act of courage or foolhardiness to produce theatre in the heart of the film world, depending on your point of view and how large the houses turn out to be. In the fall of 2005, I produced Three Sisters in a 60-seat theatre in Burbank, California (home of Disney and Warner Bros.). The odds were stacked even higher against the show's success when I stipulated that the main characters, the upper-class and highly educated Russian Prozorov siblings, had to be played by hapa actors. Background: Streptococcus pneumoniae is a leading cause of invasive bacterial disease in developed and developing countries. We studied the loop-mediated isothermal amplification (LAMP) technique to assess its suitability for detecting S. pneumoniae nucleic acid in cerebrospinal fluid (CSF).
Methodology/Principal Findings: We established an improved LAMP assay targeting the lytA gene (Streptococcus pneumoniae [Sp] LAMP). Understanding the dynamics of respiratory gasses gives critical insight into the metabolic processes of a biological system. An efficient and effective means of measuring respiration is essential to understanding aspects of the biosphere. Many methods have been developed for measuring changes in CO2 and O2, both as integrated systems and as individual components. Many experiments use an alkali trap with subsequent titration as an inexpensive method for CO2 measurement. Haney et al. β2 adrenergic receptor (β2AR) is a prototypical G-protein coupled receptor that stimulates the classic cAMP-protein kinase A (PKA) signaling pathway. Recent studies indicate that the cAMP-PKA activities are spatiotemporally regulated in part due to dynamic association of β2AR with phosphodiesterase 4D (PDE4D), a group of cAMP degradation enzymes. Here, we demonstrate that in cardiomyocytes, palmitoylation of β2AR, the covalent acylation of cysteine residue 341, plays a critical role in shaping subcellular cAMP-PKA activities in cardiomyocytes via regulating β2AR association with arrestin/PDE4D. Fish farming is one of the fastest growing sectors of agriculture, attracting considerable attention to the question of whether existing farming regulations and animal welfare laws are adequate to deal with the expanding role of fish in feeding humans. The role of fish as model organisms in scientific research is also expanding -- a majority of research biology departments now keep zebrafish for the purposes of genome biology, and they are used widely used for basic neuroscience research. The purpose of the current study was to examine whether (a) schizotypy is positively related to negative affect and negatively related to positive affect, (b) whether individuals with high levels of schizotypy use more maladaptive coping behaviors than those low in schizotypy, (c) that maladaptive coping behaviors will mediate the relationship between schizotypy and negative affect, and (d) that adaptive coping behaviors will mediate the relationship between schizotypy and positive affect. A sample of 435 undergraduate participants completed self-report measures including The Brief Cope, The Schizotypal Personality Questionnaire, and The Positive and Negative Affect Schedule. The main purpose of this research is to study the effectiveness of group REBT in reducing irrational beliefs in two groups of Iranian female adolescents living in Kuala Lumpur. A number of 40 Iranian female adolescents participated in the study. The participants were assigned to two experimental and two control groups randomly. This study used the pre-test and post-test, by using Jones’ Irrational Beliefs Test (Jones, 1969). Therapeutic sessions were held in 5 consecutive weeks, with two sessions a week.
