The pseudo-glycosyltransferase VldE catalyzes non-glycosidic C-N coupling between an unsaturated cyclitol and a saturated aminocyclitol with the conservation of the stereochemical configuration of the substrates to form validoxylamine A 7′-phosphate, the biosynthetic precursor of the antibiotic validamycin A. To study the molecular basis of its mechanism, the three-dimensional structures of VldE from Streptomyces hygroscopicus subsp. limoneus was determined in apo form, in complex with GDP, in complex with GDP and validoxylamine A 7′-phosphate, and in complex with GDP and trehalose. The structure of VldE with the catalytic site in both an "open" and "closed" conformation is also described. With these …

The purposes of this research were to determine which consumers tend to purchase fresh organic food and which consumers tend to purchase packaged organic food, which type of messaging organic food distributors rely on to entice customers and which messages work most effectively to promote organic food consumption, and which channels different organic consumers use to gain knowledge about organic food. Implications for organic food marketing are discussed.

Development Of Scn Connectivity And The Circadian Control Of Arousal: A Diminishing Role For Humoral Factors?, Andrew J. Gall, William D. Todd, Mark S. Blumberg

The suprachiasmatic nucleus (SCN) is part of a wake-promoting circuit comprising the dorsomedial hypothalamus (DMH) and locus coeruleus (LC). Although widely considered a "master clock," the SCN of adult rats is also sensitive to feedback regarding an animal's behavioral state. Interestingly, in rats at postnatal day (P)2, repeated arousing stimulation does not increase neural activation in the SCN, despite doing so in the LC and DMH. Here we show that, by P8, the SCN is activated by arousing stimulation and that selective destruction of LC terminals with DSP-4 blocks this activational effect. Novel Discretization Schemes For The Numerical Simulation Of Membrane Dynamics, Kyle F. Kolsti

Motivated by the demands of simulating flapping wings of Micro Air Vehicles, novel numerical methods were developed and evaluated for the dynamic simulation of membranes. For linear membranes, a mixed-form time-continuous Galerkin method was employed using trilinear space-time elements, and the entire space-time domain was discretized and solved simultaneously. For geometrically nonlinear membranes, the model incorporated two new schemes that were independently developed and evaluated. Time marching was performed using quintic Hermite polynomials uniquely determined by end-point jerk constraints. The single-step, implicit scheme was significantly more accurate than the most common Newmark schemes. For a simple harmonic oscillator, the scheme …

The Manduca Sexta species of moth serves as a source of biological inspiration for the future of micro air vehicle flapping flight. The ability of this species to hover in flapping flight has warranted investigation into the critical material, structural, and geometric properties of the forewing of this biological specimen. A rigorous morphological study of the Manduca Sexta forewing was conducted to characterize the physical and material properties of the biological forewing for the purpose of developing an advanced parametric three dimensional model finite element analysis (FEA) model. The Ciona intestinalis voltage-sensitive phosphatase, Ci-VSP, was the first Voltage-controlled Enzyme (VEnz) proven to be under direct command of the membrane potential. The discovery of Ci-VSP conjugated voltage sensitivity and enzymatic activity in a single protein. These two facets of Ci-VSP activity have provided a unique model for studying how membrane potential is sensed by proteins and a novel mechanism for control of enzymatic activity. These facets make Ci-VSP a fascinating and versatile enzyme. Ci-VSP has a voltage sensing domain (VSD) that resembles those found in voltage-gated channels (VGC). The VSD resides in the N-terminus and is formed by four …

What The Wild Things Are: A Critique On Clare Palmer’S “What (If Anything) Do We Owe Wild Animals?”, Joel P. Macclellan
Between the Species
In this critique of “Clare Palmer’s “What (if anything) do we owe wild animals?”, I develop three points. First, I consider the case study which opens her essay and argue that that there are good empirical reasons to think that we should assist domesticated horses and not wild deer. Then, I critique Palmer’s claim that “wildness is not a capacity”, arguing that wildness connotes certain capacities which wild animals generally have and which domesticated animals generally lack. Lastly, I develop what I call the “supererogation problem” against Palmer’s preferred contextualist view, claiming that while the contextualist view doesn’t obligate us …
