CD4+ T cell-mediated immunity plays a central role in determining the immunopathogenesis of viral infections. However, the role of CD4+ T cells in EV71 infection, which causes hand, foot and mouth disease (HFMD), has yet to be elucidated. We applied a sophisticated method to identify promiscuous CD4+ T cell epitopes contained within the sequence of the EV71 polyprotein. Fifteen epitopes were identified, and three of them are dominant ones. The most dominant epitope is highly conserved among enterovirus species, including HFMD-related coxsackieviruses, HFMD-unrelated echoviruses and polioviruses. Carboxypeptidase A6 (CPA6) is a member of the A/B subfamily of M14 metallocarboxypeptidases that is expressed in brain and many other tissues during development. Recently, two mutations in human CPA6 were associated with febrile seizures and/or temporal lobe epilepsy. In this study we screened for additional CPA6 mutations in patients with febrile seizures and focal epilepsy, which encompasses the temporal lobe epilepsy subtype. Mutations found from this analysis as well as CPA6 mutations reported in databases of single nucleotide polymorphisms were further screened by analysis of the modeled proCPA6 protein structure and the functional role of the mutated amino acid. …

This action research project describes the methods an African-American female instructor used when introducing biology-related video clips with a multicultural component to predominantly white pre-service elementary students. Studies show that introducing multiculturalism into classrooms is crucial for students and teachers. Multicultural theoretical frameworks that focus on social change arises out of an acknowledgement of achievement disparities prevalent around the world. Students viewed video clips and answered questions regarding their thoughts and views on the information in the clip; including issues relating to race, diversity, and social injustice, nature of science (NOS) and nature of science inquiry (NOSI). Exercise can increase skeletal muscle sensitivity to insulin, improve insulin resistance and regulate glucose homeostasis in rat models of type 2 diabetes. However, the potential mechanism remains poorly understood. In this study, we established a male Sprague-Dawley rat model of type 2 diabetes, with insulin resistance and β cell dysfunction, which was induced by a high-fat diet and low-dose streptozotocin to replicate the pathogenesis and metabolic characteristics of type 2 diabetes in humans. Situated at the northwest end of Santa Cruz County and occupying circa 30 square miles of sharply contrasted terrain, the Scott Creek Watershed concentrates within its geomorphological boundaries, at least 10-12% of California's flora, both native and introduced. This study explored the use of a dichotomous key as a scaffolding tool in the museum setting. The dichotomous key was designed as a scaffolding tool to help students make more detailed observations as they identified various species of birds on display. The dichotomous key was delivered to groups of fifth and seventh graders in two ways: on a mobile platform and by museum educators. Data was collected in the forms of pre- and post-testing and observations to compare the two methods. Prenatal Genetic Diagnosis Using Transcervically Derived And Immunomagnetically Isolated Trophoblast Cells, Swati Bajpayee

Trophoblast cells migrate from the placenta into the endocervical canal early in the first trimester and can be collected non-invasively by transcervical sampling (TCS), potentially providing fetal DNA for prenatal genetic diagnosis. Experiments were conducted to separate fetal cells from maternal cells within the TCS specimens and use the fetal cells to perform genetic analysis. Trophoblast cells were efficiently isolated from TCS specimens during weeks 5-18 of gestation using HLA-G antibody coupled to magnetic nanoparticles. Immunofluorescence microscopy revealed that 99% of isolated cells expressed the chorionic gonadotropin β subunit (β-CG), while the non-bound cell fraction expressed none. Polyploidization instantly doubles all genome content by combining two genomes that have markedly different methylation and gene expression levels. This process may be accompanied by genetic and epigenetic changes in each genome. Sequencing of the transcriptome (RNA-seq) and the methylome (bisulfite treated libraries whole genome libraries) were used to measure gene expression and methylation levels of genic regions of allopolyploid cotton petals and petals of their diploid relatives. Many differentially expressed genes detected by RNA-seq were consistent with expression levels previously detected by microarrays. RNA-seq results also reconfirmed the presence of general polyploid gene expression trends like expression level dominance …

RNA-seq transcriptome analysis of Chenopodium quinoa at different water treatment levels was conducted in a greenhouse study using four water treatments (field capacity to drought) on a valley ecotype quinoa (variety Ingapirca) and an Altiplano Salares ecotype quinoa (variety Ollague). Physiological results support the earlier findings that the Salares ecotypes display greater tolerance to drought-like stress conditions than the valley ecotypes (as determined by growth rate, photosynthetic rate, stomatal conductance, and stem water potential). cDNA libraries from root tissue sample for each treatment x variety combination were sequenced using Illumina Hi-Seq technology in an RNA-seq experiment. The objective of this study was to use pedological evidence in conjunction with Geographic Information Systems, and soil physical and chemical analyses as means to better understand the agricultural landscape surrounding the ancient Maya city of Uci. Specifically, the query of this thesis is to determine whether there is an association between settlement density and soil resources, and what relationship if any there is between the ancient sacbe of Uci and its surrounding agricultural potential. Dopamine cells within the ventral tegmental area of the brain are involved in motivation and reward. Drugs of abuse target these dopamine cells altering their activity and plasticity resulting in addiction. While dopamine cell activity is primarily involved in addiction, the GABA neurons in the VTA have also been shown to have an indirect role. By decreasing the activity of the inhibitory GABA inputs onto dopamine neurons abusive drugs can indirectly increase dopamine cell activity resulting in addictive behaviors. The 52 kDa intermediate filament protein desmin plays an important role in force transmission in skeletal muscle by connecting myofibrils at Z-lines and to the sarcolemma. Desmin content in muscle adapts to contractile activity and may be involved in cellular signaling mechanisms responsible for muscle growth. Purpose: To compare signaling responses of the mTOR pathway in wild type (WT) vs desmin knock out (KO) mice. It is widely assumed that current phylogenetic methods are fairly accurate at recovering the evolutionary relationships among different species, but evaluating the relative success of this enterprise is a difficult task. This study addresses some fundamental questions associated with generating phylogenetic trees. The complete genomes of five species of Steinernema were sequenced and assembled. Genes were predicted in AUGUSTUS and orthologous genes were found from those data using OrthoMCL. I aligned 3890 genes in MAFFT and eliminated poorly aligned positions with GBlocks. Wyoming big sagebrush (Artemisia tridentata Nutt.) is an important native shrub in the Great Basin because of its wide distribution where it dominates over 60 million ha and provides essential habitat and forage for many varied species.
