Use Of Real-Time Pcr To Detect Surimi Adulteration In Vegetarian Foods, Ching-Yang Cheng, Yeu-Ching Shi, Sin-Rong Lin, Chi-Chiang Chou, Chin-Cheng Huang

Vegetarian foods are abundant in the domestic market. It is found that animal ingredients have been added into vegetarian products to improve the texture and taste. Surimi adulteration in vegetarian food is an area of concern for vegetarians. The α-skeletal actin gene of aquatic animals as novel specific primers and probe of Actinopterygii in 36 fish species are tested using real-time PCR method. It was found that all of the 36 fish species of Actinopterygii showed positive results. The 10 surimi products, 10 surimi-based products containing fish components showed positive results. Cysteine residues in insulin degrading enzyme have been reported as non-critical for its activity. We found that converting the twelve cysteine residues in rat insulin degrading enzyme (IDE) to serines resulted in a cysteine-free form of the enzyme with reduced activity and decreased activation by polyanions. Mutation of each cysteine residue individually revealed cysteine 904 as the key residue required for maximal activity and polyanion activation, although other cysteines affect polyanion binding to a lesser extent. Based on the structure of IDE, Asn 575 was identified as a potential hydrogen bond partner for Cys904 and mutation of this residue also …

Early evaluation of new drug entities for their potential to cause mitochondrial dysfunction is becoming an important task for drug development. Multi-parametric high-content screening (mp-HCS) of mitochondrial toxicity holds promise as a lead in-vitro strategy for drug testing and safety evaluations. In this study, we have developed a mp-HCS and multi-parametric data analysis scheme for assessing cell responses to induced mitochondrial perturbation. The mp-HCS measurements are shown to be robust enough to allow for quantitative comparison of biological systems with different metabolic pathways simulated by alteration of growth media. The development of new small molecule-based therapeutic drugs requires accurate quantification of drug bioavailability, biological activity and treatment efficacy. Rapidly measuring these endpoints is often hampered by the lack of efficient assay platforms with high sensitivity and specificity. Using an in vivo model system, we report a simple and sensitive liquid chromatography-tandem mass spectrometry assay to quantify the bioavailability of a recently developed novel cyclin-dependent kinase inhibitor VMY-1-103, a purvalanol B-based analog whose biological activity is enhanced via dansylation. We characterized soil communities in the Mojave Desert across an elevation gradient. Our goal was to test the hypothesis that as soil quality improved with increasing elevation (due to increased productivity), the diversity of soil prokaryotes and nematodes would also increase. Soil organic matter and soil moisture content increased with elevation as predicted. Soil salinity did not correlate to elevation, but was highest at a mid-gradient, alluvial site. Soil nematode density, community trophic structure, and diversity did not show patterns related to elevation. Similar results were obtained for diversity of bacteria and archaea. Effect Of Mulberry Lees Addition On The Quality Of Bread And Anthocyanin Content, Jean-Yu Hwang, Wen-Chieh Sung

Mulberry lees were added to bread dough at levels of 10%, 20%, and 30%. Loaves were prepared using the straight dough procedure. Bread volume, crust and crumb color, crumb texture, flavor and overall acceptability were evaluated. Anthocyanin content of crumbs was determined for 5 days storage duration. The volume of bread was decreased (p < 0.05) as the mulberry lees addition amount increased. Overall acceptability of 10% mulberry lees addition bread was highest among all breads. There was no significant difference in crumb texture except the firmness of 30% mulberry lees addition was higher than other groups. Addition of mulberry lees increased the anthocyanin content of crumb. Oxidation of anthocyanin occurs in crumbs during storage. Mulberry lees could be added to bread within 20% level for enhancing the nutritional quality and characteristics of bread.

Certain marine water quality or ecosystem model parameters vary in space and time because of different plankton taxonomic compositions over a large domain. The same parameter vectors can result in suboptimal calibration. In the present paper, a data-driven model based on an artificial neural network is developed to inverse the values of model parameters dynamically. All training data used are calculated using numerical water quality models from the results of multiparameter matching design cases such that physical properties are not disturbed. While chemical defenses can determine plant persistence in terrestrial ecosystems and some marine macroalgae, their role in determining seagrass persistence in areas of intense grazing is unknown. As a first step toward determining if concentrations of feeding deterrents in seagrasses increase following herbivore attacks, we conducted 4 experiments using a common macrograzer (sea urchin Lytechinus variegatus) and 2 phylogenetically divergent seagrass species (Thalassia testudinum and Halodule wrightii). Macrograzer impacts on production of phenolic acids and condensed tannins varied somewhat idiosyncratically with season, urchin density, and distance from urchin damage. There have developed a variety of microsystems that harness energy and convert it to mechanical motion. Here we developed new autonomous biochemical motors by integrating metal-organic framework (MOF) and self-assembling peptides. MOF is applied as an energy-storing cell that assembles peptides inside nanoscale pores of the coordination framework. The robust assembling nature of peptides enables reconfiguring their assemblies at the water-MOF interface, which is converted to fuel energy. Re-organization of hydrophobic peptides could create the large surface tension gradient around the MOF and it efficiently powers the translation motion of MOF. Multivariate Gaussian mixtures are a class of models that provide a flexible parametric approach for the representation of heterogeneous multivariate outcomes. When the outcome is a vector of repeated measurements taken on the same subject, there is often inherent dependence between observations. However, a common covariance assumption is conditional independence---that is, given the mixture component label, the outcomes for subjects are independent. In this paper, we study, through asymptotic bias calculations and simulation, the impact of covariance misspecification in multivariate Gaussian mixtures. The Gene Ontology (GO) has become the internationally accepted standard for representing function, process, and location aspects of gene products. The wealth of GO annotation data provides a valuable source of implicit knowledge of relationships among these aspects. We describe a new method for association rule mining to discover implicit co-occurrence relationships across the GO sub-ontologies at multiple levels of abstraction. Prior work on association rule mining in the GO has concentrated on mining knowledge at a single level of abstraction and/or between terms from the same sub-ontology. This study investigated the impact of atmospheric metal deposition on natural phytoplankton communities at open-ocean and coastal sites in the Sargasso Sea during the spring bloom. Locally collected aerosols with different metal contents were added to natural phytoplankton assemblages from each site, and changes in nitrate, dissolved metal concentration, and phytoplankton abundance and carbon content were monitored. Addition of aerosol doubled the concentrations of cadmium (Cd), cobalt (Co), copper (Cu), iron (Fe), manganese (Mn), and nickel (Ni) in the incubation water. Over the 3-day experiments, greater drawdown of dissolved metals occurred in the open ocean water, whereas little metal drawdown …

AMP-activated protein kinase (AMPK) is a master regulator of energy metabolism in skeletal muscle; AMPK induces muscle protein degradation but the underlying mechanisms are unclear. Myostatin is a powerful negative regulator of skeletal muscle mass and growth in mammalian species. We hypothesized that AMPK stimulates myostatin expression, which provides an explanation for the negative role of AMPK in muscle growth. The objective of this study is to demonstrate that AMPK stimulates myostatin expression using C2C12 cells as a model. Activation of AMPK by 5-aminoimidazole-4-carboxamide-1-β- d-riboruranoside (AICAR.) dramatically increased the mRNA expression and protein content of myostatin in C2C12 myotubes, and …
