Active uptake of Na+ at the gill by fish is fundamental to their osmoregulation and thus survival in freshwater environments. Studies to date have demonstrated several different mechanisms by which fish can accomplish this important physiological function. This dissertation provides a comparative characterization of Na+ uptake in the coastal pupfish, Cyprinodon variegatus variegatus (Cvv), the closely related Lake Eustis pupfish, C. v. hubbsi (Cvh), and the endangered desert pupfish, C. macularius (Cm). When acclimated to slightly saline freshwater (7 mM Na+) all three fish use a low affinity Na+/H+ exchanger (NHE) for apical Na+ uptake. Cvv also uses a Na+:K+:2Cl- …

Introduction:

The patient was a 64-year-old male presented with a 2 day history of increasing fevers and altered mental status. He underwent orthotopic liver transplant for cryptogenic cirrhosis, probably secondary to non-alcoholic steatohepatitis, 45 days before. The head and neck imaging showed pancytopenia with WBC 0.6 x 109 /L., hemoglobin 6.8 g/dl and platelet 29 x 109 /L.

Mechanism Of Action Studies On A New Class Of Anticancer Nucleosides, Megan E. Browning

We have completed mechanism of action studies on a new class of anticancer nucleosides typified by a novel nucleoside discovered in our lab, MAP-870. In order to study the mechanism of MAP-870, several experiments were completed on a colorectal adenocarcinoma cell line, HT-29, including trypan blue cell count, sulforhodamine B assays, flow cytometry of cell cycle, propidium iodide incorporation, and phosphatidylserine externalization, Caspase-Glo3/7 assays, DNA fragmentation gel, cyclophilin A release gel, PAMPA, and confocal imaging. Sulforhodamine B assays show that MAP-870 does indeed cause growth inhibition and cell death in the model tested. PAMPA assays show that MAP-870 does not …

Cytoplasmic dynein and kinesin-1 are microtubule-based motors with opposite polarity that transport a wide variety of cargo in eukaryotic cells. Many cellular cargos demonstrate bidirectional movement due to the presence of ensembles of dynein and kinesin, but are ultimately sorted with spatial and temporal precision. To investigate the mechanisms that coordinate motor ensemble behavior, we built a programmable synthetic cargo using three-dimensional DNA origami to which varying numbers of DNA oligonucleotide-linked motors could be attached, allowing for control of motor type, number, spacing, and orientation in vitro. Arsenic and cadmium (Cd+2) are well established environmental pollutants. Human exposure to arsenite (As+3) and Cd +2 have been associated with many types of cancers including skin, bladder, liver, prostate, stomach and lung. Epidemiological studies have associated Cd+2 exposure with breast cancer development while As +3 has not been directly associated with breast cancer development, although both As+3 and Cd+2 have been implicated in estrogen receptor mediated gene transcription and endocrine disruption. This paper discusses the results of improved optical probe that works according to optical fluorescence and absorption phenomena for measuring the phytoplankton suspension concentrations. Measurements are made on the Scenedesmus sp. culture. The laboratory test has shown that range of concentration from 104 up to around 1 x 106 cells/mL; fluorescence intensity at λ = 685 nm; and logarithmic of transmission intensity at λ = 405 nm are proportional to the cell concentration linear with proportional constant γ = 4 × 10-5 and β = –2 × 10-7 mL/cell respectively.

This dissertation is divided into three chapters, each discussing the study of different regulatory molecules involved in sensory neurogenesis occurring in the trigeminal placode. Chapter one is a spatiotemporal description of Notch pathway genes in chick opV placode by stage-specific expression analysis, showing expression of many Notch pathway genes and effectors in the opV placode. Notch pathway gene expression is primarily confined to the ectoderm with highest expression of these genes at the beginning stages of peak neuronal differentiation. We propose a method for measuring the conjectural errors that inexpert consumers make relative to experts in using observable product characteristics as surrogate indicators of a valued unobservable characteristic. Observations on the unobservable characteristic, available to the researcher but not consumers, are used to divide the data into high- and low-quality subsamples. Separate hedonic estimation on the subsamples enables measurement of the relative valuations and conjectures of experts and non-experts with respect to indicators under the assumption that consumers sort across quality grades based on their appraisal expertise. The amyloid fibrils formed by islet amyloid polypeptide (IAPP) are associated with type II diabetes. One of the proposed mechanisms of the toxicity of IAPP is that it causes membrane damage. The fatal mutation of S20G human IAPP was reported to lead to early onset of type II diabetes and high tendency of amyloid formation in vitro. Characterizing the structural features of the S20G mutant in its monomeric state is experimentally difficult because of its unusually fast aggregation rate. Bertrand Russell was an interesting and, at times, delightfully insightful thinker who lived a colourful life and fought for many important causes. Certainly he was, and still is, an inspiring figure for some philosophers, humanists, social activists, and educators. Unfortunately, on several occasions, he demonstrated only a limited understanding of the depth, complexity, and subtlety of John Dewey's pragmatic project for philosophy, education, and society. Doubly unfortunate, the Russellian critique of John Dewey articulated by Michael Rockier carries on this misinformed tradition. Thus, I contend, the Russellian criticisms offered by Rockier are at best superficial, at worst seriously misleading.

High-performance affinity chromatography (HPAC) was used to examine the changes in binding that occur for the sulfonylurea drug glibenclamide with human serum albumin (HSA) at various stages of glycation for HSA. Frontal analysis on columns containing normal HSA or glycated HSA indicated glibenclamide was interacting through both high affinity sites (association equilibrium constant, K_a, 1.4–1.9 × 10⁶ M⁻¹ at pH 7.4 and 37°C) and lower affinity sites (K_a, 4.4– 7.2 × 10⁴ M⁻¹).
