The t(15;17) translocation is found in nearly 98% of acute promyelocytic leukemia: APL, FAB subtype M3) cases and results in the fusion of the promyelocytic leukemia: PML) gene with the retinoic acid receptor alpha: RARA) gene. The fusion product, PML-RARA, encodes a functionally altered transcription factor that is the initiating event in APL. To better understand the transcriptional changes associated with APL pathogenesis, we compared the gene expression profiles of APL samples to those of other acute myeloid leukemia FAB subtypes and of enriched normal human promyelocytes. Osteoclasts are essential for skeletal homeostasis: Teitelbaum, 2000). These macrophage-lineage cells function by generating a polarized microenvironment between themselves and bone wherein skeletal matrix is degraded. This resorptive compartment is isolated from the general extracellular space by an actin ring which encompasses the ruffled border, a convoluted plasma membrane structure formed by its fusion with lysosome-related vesicles containing an electrogenic H+ATPase, a chloride channel, LAMP-1, and cathepsin K. In consequence, resorption of bone reflects secretion of HCl to mobilize mineral, and cathepsin K to degrade the collagen-rich organic matrix, into the resorptive space: Stenbeck, 2002; Zhao et al., 2008). The generation of a functional antigen receptor gene in developing lymphocytes requires that the second exon be assembled through a process known as V(D)J recombination, a process that necessarily involves the generation and repair of DNA double-strand breaks made by the Rag endonuclease. Double strand breaks incurred during G1 of the cell cycle activate ATM, a PI3-kinase-like kinase that, in response to genotoxic DNA damage, is known to phosphorylate hundreds of proteins with unique and diverse functions. Notably, deficiencies in ATM lead to ataxia-telangiectasia, a syndrome characterized by lymphopenia, genomic instability and a predisposition to tumors involving antigen receptor loci …

"The noncatalytic reformation of glycerin in supercritical water was studied in a Haynes 282 tubular reactor. In order to determine which parameters were the most influential, a 2³ experimental matrix was conducted, with temperatures of 500 and 700⁰C, water/glycerin molar ratios of 3:1 and 13:1, and residence times of 30 and 90 seconds, all at a pressure of 24 MPa. It was found that temperature had the largest effect on the two gasification parameters deemed most important, gasification percentage and hydrogen yield. Cellular response to osmotic stress is critical for survival and involves volume control through the regulated transport of osmolytes [1-3]. Organelles may respond similarly to abrupt changes in cytoplasmic osmolarity [4-6]. The plastids of the Arabidopsis thaliana leaf epidermis provide a model system for the study of organellar response to osmotic stress within the context of the cell. An Arabidopsis mutant lacking two plastid-localized homologs of the bacteria mechanosensitive channel MscS (MscS-like [MSL] 2 and 3) exhibits large round epidermal plastids that lack dynamic extensions known as stromules [7]. In bacteria, MscS-type mechanosensitive channels serve to protect cells from lysis as they swell during extreme osmotic stress. We recently showed that two MscS homologs from Arabidopsis thaliana serve a similar purpose in the epidermal plastids of the leaf, indicating that the plant cell cytoplasm can present a dynamic osmotic challenge to the plastid. MscS homologs are predicted to be targeted to both plastids and mitochondrial envelopes and have been found in the genomes of intracellular pathogens. Here we discuss the implications of these observations, and propose that MS channels provide an essential mechanism for osmotic adaptation to both intracellular …

"Research into polymer-clay nanocomposites (PCN's) has been ongoing for decades as a result of the property enhancements offered by clay. To fully exploit these property enhancements, organically modified clays (organoclays) are utilized to promote clay delamination by reducing the disparity between the hydrophilicity of the clay and the hydrophobicity of the highly used polyolefin polymer. This dissertation attempts to answer questions from two different areas of biology, ecology and neuroscience, using physics-based techniques. In Section 2, suitability of three competing random walk models is tested to describe the emergent movement patterns of two species of primates. The truncated power law (power law with exponential cut off) is the most suitable random walk model that characterizes the emergent movement patterns of these primates. "The Yilgarn Craton in Western Australia hosts hundreds of shallow ephemeral hypersaline lakes, the majority of which have acid to neutral pH values. As part of a multidisciplinary study of the evolution of hypersalinity and acidity in the region, three drill cores were studied for their palynofacies and palynomorph contents in order to characterize palynofloral response to environmental changes. Drill cores from two acid lakes, Prado Lake (PL1-09 and PL2-09) and Twin Lake West (TLW1-09), and the neutral-alkaline Gastropod Lake (GLE1-09) south of Norseman recovered Miocene to Holocene sediments of the Revenge and Polar Bear formations. In this project, solid phase microextraction (SPME) fibers, protected in perforated steel tubes, were used as in situ passive samplers to measure the treatment of activated carbon (AC) in polycyclic aromatic hydrocarbon (PAH) contaminated sediment. "An additive manufacturing process called Freeze-form Extrusion Fabrication (FEF) was developed in this study to fabricate complex ceramic parts that require the use of sacrificial materials. The aqueous paste of alumina (Al₂O₃) was used as the main material, with solids loadings as high as 50 vol. % and water as the main liquid medium. Methyl cellulose, with a 10 vol. % solids loading and 90 vol. % water content, was used as the sacrificial material. This thesis describes a method which safely and accurately evaluates rockfall on rock outcrops through the use of Light Detection and Ranging (LiDAR) data. Author's abstract: Some researchers assert that cultural display rules may explain differences in perceiving emotions (Matsumoto, Yoo, & Chung, 2010). The current study examined the display rule of masking within the Southern culture of honor. It was hypothesized that masking within the culture of honor negatively affects emotion perception sensitivity, particularly in the speed and accuracy of recognizing anger. Southern undergraduate students were primed with the culture of honor and then presented with the Emotional Expression Multimorph Task. Author's abstract: Carbohydrate supplementation has been associated with increased performance in aerobic and anaerobic activity. Honey, specifically, has shown to increase performance in aerobic exercise as well as provide other health benefits. The purpose of this study was to see if honey, a natural substance, has a positive effect on anaerobic performance, rates of perceived exertion, and gastrointestinal distress. Subjects consisted of 18 healthy, Division 1 collegiate male soccer players (19.39 ± 0.85 yr, 75.39 ± 7.97 kg). Diarrhea can be caused by a range of pathogens, including several bacteria. Conventional diagnostic methods, such as culture, biochemical tests, and enzyme-linked immunosorbent assay (ELISA), are laborious. We developed a 7-plex PCR-Luminex assay to simultaneously screen for several of the major diarrhea-causing bacteria directly in fecal specimens, including pathogenic Aeromonas, Campylobacter jejuni, Campylobacter coli, Salmonella, Shigella, enteroinvasive Escherichia coli (EIEC), Vibrio, and Yersinia. We included an extrinsic control to verify extraction and amplification. This study investigated health behavior as a possible mediator of the link between hostility and poor physical and psychological health. The relations of health behavior, character strength, and virtuous behavior to hostility and health were also examined. A sample of 689 undergraduate participants (69.4% female) completed a series of on-line assessments that measured hostility, physical health, psychological health, health behavior, social support, character strength, and virtuous behavior. Health behavior was examined and found to be a significant partial mediator of the relations between hostility and physical and psychological health. Methamphetamine (METH) is a highly addictive psychostimulant drug of abuse, causing hyperthermia and neurotoxicity at high doses. Currently, there is no clinically proven pharmacotherapy to treat these effects of METH, necessitating identification of potential novel therapeutic targets. Earlier studies showed that METH binds to sigma receptors in the brain at physiologically relevant concentrations, where it acts in part as an agonist. AC927 (1-(2-phenethyl)piperidine oxalate), CM156 (3-(4-(4-cyclohexylpiperazin-1-yl)butyl)benzo[d]thiazole-2(3H)-thione) and SN79 (6-acetyl-3-(4-(4-(4-florophenyl)piperazin-1-yl)butyl)benzo[d]oxazol-2(3H)-one) were synthesized as putative sigma receptor antagonists with nanomolar affinity and selectivity for sigma receptors over 29-80 other binding sites. Basal type/triple negative breast cancer (BTBC) represents a distinct tumor group that is characterized by an aggressive disease phenotype and poor clinical outcome. Patients with this subtype cannot benefit from the available anti-hormone and anti-HER2 targeted therapies (e.g. tamoxifen and herceptin). Therefore, characterizing potential therapeutic targets is urgently needed. We reasoned that the Src-homology 2 domain-containing tyrosine phosphatase 2 (SHP2) could serve as a therapeutic target in BTBC, because of its critical regulation of epidermal growth factor receptor (EGFR) which is overexpressed in BTBC. Nightmares have been linked with several negative consequences, including death by suicide. Further, nightmare sufferers report that nightmares have a significant negative impact on their lives. To address this problem, several nightmare treatments have been studied, with Imagery Rehearsal Therapy (IRT) having the greatest amount of empirical support. The prohibition approach to narcotics control has been the dominant policy paradigm of the United States (US) and the global narcotics regime under the United Nations (UN) for more than a century. In recent years, however, key questions have been raised on the existing prohibition policy approach, which many consider as ineffective and unchanging from a broader perspective. Problem
Although most students who attend Oakwood University are self-identified Seventh-day Adventist Christians, from general observation of their activities, conversations, and attitudes displayed on any given Friday night and/or Saturday (i.e., Sabbath), it would be easy to conclude that many neither enter nor exit their collegiate experience with a holistic understanding or experience of Sabbath and Sabbathkeeping.
Methodology

Samples were collected in 35 states as part of a national monitoring system to detect multiple diseases in feral swine (Sus scrofa). During March 2009 through December 2010, we collected serum samples from 4,479 feral swine from 13 states, and 159 animals tested were seropositive for brucellosis. No difference in likelihood of infection was found between males and females, but adults were more likely than sub-adults or juveniles to be exposed to brucellosis. Feral swine sampled during winter months also were more likely to be seropositive than animals sampled during other seasons. Apparent prevalence varied among states, and …

Monoamine transporter (MAT) proteins are responsible for regulating cellular signal transduction through control of neurotransmitter reuptake in the synapse, and are therefore relevant to diseases including addiction, psychosis, anxiety and depression. MATs, specifically the serotonin transporter (SERT or 5-HTT), norepinephrine transporter (NET), and dopamine transporter (DAT), serve as the principal targets for antidepressant drugs, such as SSRIs (selective serotonin reuptake inhibitors), NRIs (norepinephrine reuptake inhibitors) and TCAs (tricyclic antidepressants), as well as psychostimulant drugs of abuse such as cocaine and the amphetamines. This study sought to explore the lived experience of parenting a child adopted from an institution in Eastern Europe. A hermeneutic, phenomenologic method following the Utrecht School was used. The setting for this study was a southern state on the east coast of the United States. The sample consisted of 11 participants, nine mothers and two fathers, self- identified as adoptive parents to children from institutions in Eastern Europe. Semi-structured interviews, with thematic analysis of transcriptions were conducted. Five major themes were identified: choosing-being chosen, weaving the fabric, the importance of doing, remaining mindful, and looking-in, looking out. Conclusions were …

Unconventional natural gas extraction from the Marcellus Shale requires millions of gallons of water to fracture shale and release natural gas from the formation. This process produces water with high levels of total dissolved solids (TDS); and, efforts to recycle these fluids has stimulated microbial growth in produced water. The objective of this study was to analyze the ionic composition of and characterize microorganisms from Marcellus produced water samples. A semi-synthetic culture medium was designed with high TDS to enrich for halophilic microbes, which yielded robust cultures that were able to grow over a wide range of salinities. DNA extracted …
