Three dimensional micro- and nano-structures are commonly used in the field of Photonics, Optoelectronics, Sensors and Biological applications. Although numerous physical models are developed, a major challenge has been in their fabrication which is commonly limited to conventional layer-by-layer techniques. In this dissertation, a novel method for fabricating three dimensional structures using Electron Beam Lithography (EBL) will be presented.

In the United States there are 939 million acres of land held by ranchers and farmers. Conservation of this agricultural landscape is critical to the protection of biodiversity. Management of agricultural land is also associated with the success of biodiversity protection efforts in protected areas, including parks and preserves, as well as outside protected areas. Sustained viability of the New York farming sector will enhance and secure biodiversity on farmland, as viable farms will remain part of the landscape.

Non-melanoma skin cancers (NMSC) comprise more than one third of all cancers, and are increasing worldwide. The most common NMSC are Basal Cell Carcinoma (BCC) and Squamous Cell Carcinoma (SCC). Studies on humans and animals suggest that sunlight and arsenic play important roles in the pathogenesis of these skin malignancies. However, the number of epidemiological studies focusing on NMSC in workers exposed to ultraviolet (UV) radiation or airborne arsenic is limited, and the findings are contradictory.

Candida albicans is a fungal pathogen that causes serious infections among immune-compromised patients and premature infants. C. albicans can become drug resistant, therefore, identifying new antifungal drug targets is an important goal. Here, we study a peptidyl-prolyl cis/trans isomerase called Ess1 as a potential drug target. Ess1 is conserved among pathogenic fungi, and therefore, potential inhibitors of Ess1 should display a broad spectrum of activity. We confirm that Ess1 is essential for growth in Candida albicans, but unlike the previously published find, deleting one copy of the C. albicans ESS1 gene did not affect morphogenetic switching. α-Amino-3-hydroxy-5-methyl-4-isoxazolepropionic acid (AMPA) receptors are one of the three subtypes of ionotropic glutamate receptors. AMPA receptors mediate fast synaptic neurotransmission in the central nervous system (CNS). Over-activation of calcium permeable AMPA receptors causes intracellular calcium overload, which leads to neurodegeneration and cell death. As such, AMPA receptors have been implicated in a number of neurological disorders and diseases, such as epilepsy, amyotrophic lateral sclerosis (ALS), and Parkinson's disease. 2,3-Benzodiazepine derivatives (or GYKI compounds) are a group of structurally similar compounds synthesized as inhibitors of AMPA receptors, and they have been used as potential drug candidates for the treatment of various …

This thesis examines the divisions and boundaries made by the mechanisms of separation that authorize a false perception of land, animals, blacks and women as commodities in William Faulkner's "Go Down, Moses." Asserting that "Go Down, Moses" describes mixing as well as divisions, this study demonstrates that the dichotomous boundaries imposed upon nature and humans repeatedly fail to function. Wilderness and plantation land can never be separated as they exist in mixture. And the racial boundary between whites and blacks is destabilized and blurred by characters like Lucas Beauchamp and Tomey's Turl, who engage in resistance against the society whose …

Improved understanding of health risks from short- and long-term exposure to fine particulate matter (PM2.5) constituents may explain seasonal and geographic heterogeneity in PM2.5-health associations and inform control efforts targeting PM sources. Few studies have examined PM species health effects; most have been limited by their exposure assessments and modeling approaches. The goals of this project were to improve the PM exposure assessment and explore relationships between PM2.5 species and health in acute and chronic contexts.

A thermodynamic and structural analysis of DNA nanostructures was performed, following the replacement of the unpaired thymine bases within a central loop to determine their effect on the self-assembly process. Specifically this study focused on DNA cross-tile nanostructures. Self-assembly of DNA cross-tiles occurs by means of Watson-crick base-pairing interactions between designed single-stranded DNAs from which lattice structures assemble through sticky-end cohesion. These cross-tile structures were constructed from nine single-stranded DNAs and consisted of a central loop containing 16 unpaired thymine bases, four shell strands, and four arms. Modifications were introduced to replace the central unpaired thymine base loop. The modifications …

Coordinated synthesis and assembly of the cytoskeletal network contribute significantly to morphological changes during axon outgrowth. Previous studies demonstrated that heterogeneous ribonucleoprotein K (hnRNP K), an RNA binding protein, binds to the 3'-untranslated regions of all neurofilament triplet subunits, the most abundant components of the axonal cytoskeleton. These findings raised the hypothesis that hnRNP K post-transcriptionally mediates the coordinated expression of axonal cytoskeletal components. In my thesis, I test this hypothesis during both axonal development and regeneration.

In this thesis 6 and 18 MV photon beams for a Varian linear accelerator were simulated using a general purpose Monte Carlo code MCNP5 (Monte Carlo N-Particle Transport Code). The potential of MCNP5 is explored to calculate the dose in high dose gradient regions. In addition, MCNP5 was used to model another geometry, low energy focused x ray beams created with polycapillary optics, to explore their potential to provide a different skin-sparing technique for radiation therapy.

This dissertation analyzes the boundaries between the nonprofit and for-profit sectors using network analysis on interlocking directorates. Drawing upon data that contains over 26,000 directors and 1000 organizations, it seeks to identify the nature of the network between the largest organizations in American society and addresses how it might explain recent marketization trends in the nonprofit sector. The “biznification” of the nonprofit sector has caused some alarm amongst nonprofit scholars and this dissertation adds another layer to the discussion by looking at the directors of the nonprofit and for-profit sectors. This thesis argues against the interpretations of Sigmund Freud's Oedipus complex and Jacques Lacan's graph of sexuation as either misogynistic or anti-woman. The Oedipus complex has often been considered and taught as an explanation for binary sexual difference and heterosexual identity. This thesis will dispute such considerations through re-reading Freud himself among other literary scholars. Thus, the critiques that treat the complex as hetero-normative and homogeneous will be disproved. Additionally, psychoanalysts and academic critics alike accuse Lacan of a phallocentric framework. Because of this phallocentrism, Lacan is doubly accused of misogyny and patriarchy. Through reading his work on feminine sexuality, …

The purpose of this project is to compare the relative robusticity between native populations in New York State from the Archaic, Woodland, and Historic time periods. Musculoskeletal markers are used to determine any similarities and differences in robusticity between ages, sexes, and time periods. Relative robusticity is also assessed in terms of upper and lower limbs to further investigate any habitual activity patterns that can be discerned between groups. It is hypothesized that the Archaic populations would be comparatively more robust than the Woodland and Historic periods. During attempts to develop aptamers that bind to the nucleocapsid protein of HIV-1 (i.e. NCp7) with high affinity, a heretofore unreported property of the protein--the ability to mediate degradation of nucleic acids, was discovered. Using λ DNA as a model nucleic acid system, it was shown that NCp7-mediated degradation of nucleic acids is non-specific, depends on incubation time, the concentration of NCp7, and the presence of divalent and monovalent cations. It was further demonstrated that degradation can be abrogated if NCp7 is pre-incubated with NCp7-inhibitors. Lyophilization of NCp7 can induce irreversible changes in its secondary structure that result in loss …

This research project aims to provide an in-depth analysis of women in the landscaping field. It is important to expand on the literature on nontraditional blue-collar occupations, as more women are moving into these jobs in this 21st century. According to the United States Bureau of Labor Statistics (2001), the landscape and horticultural services industry is ranked 11th in the top twenty fastest growing service industries, almost two times the average for job growth. This occupation represents a significant, viable and fruitful arena for sociological investigation, as well a fantastic occupational option for women.

Postsynaptic [Ca2+]i has been proposed to play an important role in both synaptic plasticity and synaptic homeostasis. Postsynaptic Ca2+ signals appear to regulate synaptic transmission at the Drosophila larval NMJ; however, they have not been characterized at a NMJ. We were interested in examining the postsynaptic Ca2+ signals in this system and determining its influence on synaptic strength. The muscle Ca2+ transients were recorded by injecting the muscle fibers with Ca2+ indicators; discrete postsynaptic Ca2+ transients were observed along the NMJ during evoked and spontaneous transmitter release. University students (N=1,924) at a large public university were surveyed to measure their nutrition knowledge, dietary practices, and ecological literacy. A quantitative measure of ecological literacy was developed for this purpose, reflecting familiarity with nature, awareness of one's personal impact on local and global ecologies, and personal lifestyle choices that demonstrate civic responsibility for the natural environment. The association between nutrition knowledge and dietary practices was compared to the association between ecological literacy and dietary practices. Knowledge and application of theory are integral to the practice of psychotherapy. Despite the focus on theory in clinical training, little is known about how psychotherapy trainees experience, learn about, and develop a theoretical orientation. This phenomenological study used grounded theory methods to examine 15 counseling and clinical psychology doctoral students' experiences developing a theoretical orientation. The specific purpose of the study was to understand in depth these trainees' perspectives on and attributions about learning and developing expertise with a specific theoretical orientation.

DNA polymerases are essential for genome replication and DNA repair in all living organisms. Precise DNA replication is critical for the preservation of genomic stability. Any insult, endogenous/exogenous, to cellular DNA requires properly functioning repair polymerases. In eukaryotes, DNA polymerase beta, a small enzyme (39 kDa), plays an important role in DNA repair during the base excision repair pathway. Pol beta catalyzes the incorporation of nucleotides in small stretches (1-6 nucleotides) of damaged double-stranded DNA. Should gap-filling synthesis by pol beta be compromised, mutations in genomic DNA accumulate, which are frequently linked to human diseases, including cancers. For this reason, …

Manipulation and control of electron current in a graphene p-n junction (e.g. electron waveguiding, reflection, focusing) is directly determined by the spatial gradient of the Fermi level across the junction. Sharp Fermi level gradients are associated with negative index `lensing' of electrons in graphene while broader gradients are predicted to form reflective boundaries. Quantitative metrology of the Fermi level gradient at p-n junctions is thus essential to determine device performance, validate models for device design and switch architectures, and quantitatively determine the impact of defects on device function and leakage.

This study identifies specific patterns of risk and protective factors using a person-centered approach (cluster analysis) in low-income ethnic-minority early adolescents, and then used variable centered approaches to examine how these patterns are related to developmental outcomes 6 years later. The present study used data from Welfare, Children, and Families: A Three Cities Study, and comprised of 939 African American and Latino adolescents ages 10-14 at the initial assessment. The primary purpose of this study was to identify reflective function in Head Start teachers. Reflective function (RF) is a measure of a cognitive-emotional capacity that has been measured through interviews with parents and is linked to the parent's ability to create physical and psychological safety for his/her child (Slade, 2005). This study is the first to apply this measure to teacher interviews. This thesis explores my personal experience of having an absent biological father and my attempt to try to understand why my father left my family before I was born. While my biological father was physically absent my entire childhood, there are other children who have fathers who are both physically and emotionally absent for different portions of their lives. Effectiveness Of An Online Personalized Feedback Intervention For Young Adult Problem Drinkers, Tracey Rocha

This study investigated the effectiveness of a two-component, personalized feedback intervention for reducing alcohol use among adults, aged 18-35, who meet criteria for problem drinking. All recruitment and intervention procedures were conducted over the internet. The study compared the effects of personalized social norms feedback alone, to social norms feedback plus feedback about the individual's level of risk and harm, and to an educational intervention (control group). The interaction of age with the effectiveness of each component of the intervention was also examined.
