Design, Synthesis, And In Vitro And In Vivo Biological Studies Of A 3′-Deoxythymidine Conjugate That Potentially Kills Cancer Cells Selectively, Qiong Wei, Dejun Zhang, Anna Yao, Liyi Mai, Zhiwei Zhang, Qibing Zhou

Medicinal Chemistry Publications

Thymidine kinases (TKs) have been considered one of the potential targets for anticancer therapeutic because of their elevated expressions in cancer cells. However, nucleobase analogs targeting TKs have shown poor selective cytotoxicity in cancer cells despite effective antiviral activity. 3′-Deoxythymidine phenylquinoxaline conjugate (dT-QX) was designed as a novel nucleobase analog to target TKs in cancer cells and block cell replication via conjugated DNA intercalating quinoxaline moiety. In vitro cell screening showed that dT-QX selectively kills a variety of cancer cells including liver carcinoma, breast adenocarcinoma and brain glioma cells; whereas it had a low cytotoxicity in normal cells such as …

The Removal Of Pluto From The Class Of Planets And Homosexuality From The Class Of Psychiatric Disorders: A Comparison, Peter Zachar, Kenneth S. Kendler Jan 2012

The Removal Of Pluto From The Class Of Planets And Homosexuality From The Class Of Psychiatric Disorders: A Comparison, Peter Zachar, Kenneth S. Kendler

Psychiatry Publications

We compare astronomers' removal of Pluto from the listing of planets and psychiatrists' removal of homosexuality from the listing of mental disorders. Although the political maneuverings that emerged in both controversies are less than scientifically ideal, we argue that competition for "scientific authority" among competing groups is a normal part of scientific progress. In both cases, a complicated relationship between abstract constructs and evidence made the classification problem thorny.

Detection Of Horizontal Transfer Of Individual Genes By Anomalous Oligomer Frequencies, Jeff Elhai, Hailan Liu, Arnaud Taton Jan 2012

Detection Of Horizontal Transfer Of Individual Genes By Anomalous Oligomer Frequencies, Jeff Elhai, Hailan Liu, Arnaud Taton

Study of Biological Complexity Publications

Background

Understanding the history of life requires that we understand the transfer of genetic material across phylogenetic boundaries. Detecting genes that were acquired by means other than vertical descent is a basic step in that process. Detection by discordant phylogenies is computationally expensive and not always definitive. Many have used easily computed compositional features as an alternative procedure. However, different compositional methods produce different predictions, and the effectiveness of any method is not well established.

Results

The ability of octamer frequency comparisons to detect genes artificially seeded in cyanobacterial genomes was markedly increased by using as a training set those …

Study of Biological Complexity Publications

The Human Microbiome Project (HMP) aims to characterize the microbial communities of 18 body sites from healthy individuals. To accomplish this, the HMP generated two types of shotgun data: reference shotgun sequences isolated from different anatomical sites on the human body and shotgun metagenomic sequences from the microbial communities of each site. The alignment strategy for characterizing these metagenomic communities using available reference sequence is important to the success of HMP data analysis. Six next-generation aligners were used to align a community of known composition against a database comprising reference organisms known to be present in that community. All aligners …

Shared Resistance To Aging And Als In Neuromuscular Junctions Of Specific Muscles, Gregorio Valdez, Juan C. Tapia, Jeff W. Lichtman, Michael A. Fox, Joshua R. Sanes

Anatomy and Neurobiology Publications

Normal aging and neurodegenerative diseases both lead to structural and functional alterations in synapses. Comparison of synapses that are generally similar but respond differently to insults could provide the basis for discovering mechanisms that underlie susceptibility or resistance to damage. Here, we analyzed skeletal neuromuscular junctions (NMJs) in 16 mouse muscles to seek such differences. We find that muscles respond in one of three ways to aging. In some, including most limb and trunk muscles, age-related alterations to NMJs are progressive and extensive during the second postnatal year. NMJs in other muscles, such as extraocular muscles, are strikingly resistant to …

Spxa1 Involved In Hydrogen Peroxide Production, Stress Tolerance And Endocarditis Virulence In Streptococcus Sanguinis, Lei Chen, Xiuchun Ge, Xiaojing Wang, Jenishkumar R. Patel, Ping Xu

Philips Institute for Oral Health Research Publications

Streptococcus sanguinis is one of the most common agents of infective endocarditis. Spx proteins are a group of global regulators that negatively or positively control global transcription initiation. In this study, we characterized the spxA1 gene in S. sanguinis SK36. The spxA1 null mutant displayed opaque colony morphology, reduced hydrogen peroxide (H2O2) production, and reduced antagonistic activity against Streptococcus mutans UA159 relative to the wild type strain. The ΔspxA1 mutant also demonstrated decreased tolerance to high temperature, acidic and oxidative stresses. Further analysis revealed that ΔspxA1 also exhibited a ~5-fold reduction in competitiveness in an animal model of …

Augmenter Of Liver Regeneration (Alr) Promotes Liver Outgrowth During Zebrafish Hepatogenesis, Yan Li, Muhammad Farooq, Donglai Sheng, Chanchal Chandramouli, Tian Lan, Nilesh K. Mahajan, R. Manjunatha Kini, Yunhan Hong, Thomas Lisowsky, Ruowen Ge

Biochemistry and Molecular Biology Publications

Augmenter of Liver Regeneration (ALR) is a sulfhydryl oxidase carrying out fundamental functions facilitating protein disulfide bond formation. In mammals, it also functions as a hepatotrophic growth factor that specifically stimulates hepatocyte proliferation and promotes liver regeneration after liver damage or partial hepatectomy. Whether ALR also plays a role during vertebrate hepatogenesis is unknown. In this work, we investigated the function of alr in liver organogenesis in zebrafish model. We showed that alr is expressed in liver throughout hepatogenesis. Knockdown of alr through morpholino antisense oligonucleotide (MO) leads to suppression of liver outgrowth while overexpression of alr promotes liver growth. …

Analysis Of Embryoid Bodies Derived From Human Induced Pluripotent Stem Cells As A Means To Assess Pluripotency, Steven D. Sheridan, Vasudha Surampudi, Raj R. Rao

Chemical and Life Science Engineering Publications

Human induced pluripotent stem cells (hiPSCs) have core properties of unlimited self-renewal and differentiation potential and have emerged as exciting cell sources for applications in regenerative medicine, drug discovery, understanding of development, and disease etiology. Key among numerous criteria to assess pluripotency includes the in vivo teratoma assay that has been widely proposed as a standard functional assay to demonstrate the pluripotency of hiPSCs. Yet, the lack of reliability across methodologies, lack of definitive clinical significance, and associated expenses bring into question use of the teratoma assay as the “gold standard” for determining pluripotency. We propose use of the in …

Genetic Variation In Spag16 Regions Encoding The Wd40 Repeats Is Not Associated With Reduced Sperm Motility And Axonemal Defects In A Population Of Infertile Males, David R. Nagarkatti-Gude, Giulia Collodel, Lori D. Hill, Elena Moretti, Michela Geminiani, Zhibing Zhang, Jerome F. Strauss

Obstetrics and Gynecology Publications

Background SPAG16 is a critical structural component of motile cilia and flagella. In the eukaryotic unicellular algae Chlamydomonas, loss of gene function causes flagellar paralysis and prevents assembly of the “9 + 2” axoneme central pair. In mice, we have previously shown that loss of Spag16 gene function causes male infertility and severe sperm motility defects. We have also reported that a heterozygous mutation of the human SPAG16 gene reduces stability of the sperm axonemal central apparatus.

Methods In the present study, we analyzed DNA samples from 60 infertile male volunteers of Western European (Italian) origin, to search for novel …

Obstetrics and Gynecology Publications

Hyperandrogenism is characteristic of women with polycystic ovary syndrome (PCOS). Ovarian theca cells isolated from PCOS follicles and maintained in long-term culture produce elevated levels of progestins and androgens compared to normal theca cells. Augmented steroid production in PCOS theca cells is associated with changes in the expression of genes for several steroidogenic enzymes, including CYP11A1, which encodes cytochrome P450 cholesterol side-chain cleavage. Here, we further examined CYP11A1 gene expression, at both the transcriptional and post-transcriptional level in normal and PCOS theca cells propagated in long-term culture utilizing quantitative RT-PCR, functional promoter analyses, and mRNA degradation studies. The minimal …

The innate ability of the human cell to silence endogenous retroviruses through RNA sequences encoding microRNAs, suggests that the cellular RNAi machinery is a major means by which the host mounts a defense response against present day retroviruses. Indeed, cellular miRNAs target and hybridize to specific sequences of both HTLV-1 and HIV-1 viral transcripts. However, much like the variety of host immune responses to retroviral infection, the virus itself contains mechanisms that assist in the evasion of viral inhibition through control of the cellular RNAi pathway. Retroviruses can hijack both the enzymatic and catalytic components of the RNAi pathway, in …

The Role Of Ezh2 In The Regulation Of The Activity Of Matrix Metalloproteinases In Prostate Cancer Cells., Yong Jae Shin, Jeong-Ho Kim Jan 2012

The Role Of Ezh2 In The Regulation Of The Activity Of Matrix Metalloproteinases In Prostate Cancer Cells., Yong Jae Shin, Jeong-Ho Kim

Biochemistry and Molecular Medicine Faculty Publications

Degradation of the extracellular matrix (ECM), a critical step in cancer metastasis, is determined by the balance between MMPs (matrix metalloproteinases) and their inhibitors TIMPs (tissue inhibitors of metalloproteinases). In cancer cells, this balance is shifted towards MMPs, promoting ECM degradation. Here, we show that EZH2 plays an active role in this process by repressing the expression of TIMP2 and TIMP3 in prostate cancer cells. The TIMP genes are derepressed by knockdown of EZH2 expression in human prostate cancer cells but repressed by overexpression of EZH2 in benign human prostate epithelial cells. EZH2 catalyzes H3K27 trimethylation and subsequent DNA methylation …

Development And Application Of A Hydroclimatological Stream Temperature Model Within The Soil And Water Assessment Tool, Darren L. Ficklin, Yuzhou Luo, Iris T. Stewart-Frey, Edwin P. Maurer Jan 2012

Development And Application Of A Hydroclimatological Stream Temperature Model Within The Soil And Water Assessment Tool, Darren L. Ficklin, Yuzhou Luo, Iris T. Stewart-Frey, Edwin P. Maurer

Civil, Environmental and Sustainable Engineering

We develop a stream temperature model within the Soil and Water Assessment Tool (SWAT) that reflects the combined influence of meteorological (air temperature) and hydrological conditions (streamflow, snowmelt, groundwater, surface runoff, and lateral soil flow) on water temperature within a watershed. SWAT currently uses a linear air-stream temperature relationship to determine stream temperature, without consideration of watershed hydrology. As SWAT uses stream temperature to model various in-stream biological and water quality processes, an improvement of the stream temperature model will result in improved accuracy in modeling these processes. The new stream temperature model is tested on seven coastal and mountainous …

Low-Tech Coconut Shell Activated Charcoal Production, Ami Cobb, Mikel Warms, Edwin P. Maurer, Steven Chiesa

Civil, Environmental and Sustainable Engineering

Water treatment technologies in the developing world typically focus on removing two types of impurities from water sources: suspended solids and microbial pathogens. However, as industrialization and high-input agriculture has expanded into the developing world, chemical impurities such as pesticides, herbicides, and fertilizers have found their way into drinking water supplies and have been linked to severe health-related issues. Activated carbon has the capacity to remove these problematic chemicals from water sources. A simple, inexpensive, and effective activated carbon production process using local agricultural waste byproducts was assessed for the community of Bluefields, Nicaragua. Coconut shell charcoal was produced on …

Mathematical Modeling And Simulation Of Biologically Inspired Hair Receptor Arrays In Laminar Unsteady Flow Separation, John R. Singler, Belinda A. Batten, Benjamin T. Dickinson

Mathematics and Statistics Faculty Research & Creative Works

Bats possess arrays of distributed flow-sensitive hair-like mechanoreceptors on their dorsal and ventral wing surfaces. Bat wing hair receptors are known to play a significant role in flight maneuverability and are directionally most sensitive to reversed flow over the wing. in this work, we consider the mechanics of flexible hair-like structures for the time accurate detection and visualization of hydrodynamic images associated with unsteady near surface flow phenomena. a nonlinear viscoelastic model of a hair-like structure coupled to an unsteady nonuniform flow is proposed. Writing the hair model in nondimensional form, we identify five dimensionless groups that govern hair behavior. …

Latino Parental Acceptability Towards The Human Papilloma Virus Vaccine, Linda M. Kondilis

Theses and Dissertations

This study examined parental acceptability of the Human Papilloma Virus (HPV) vaccination among Latino parents. The role that acculturation may play in Latino parents' acceptability of the vaccine was also examined. A survey conceptualized using the Health Belief Model and two acculturation measures were administered to participants (N = 130). Approximately 70% of parents endorsed that they would vaccinate their daughters with the HPV vaccine. A regression analysis revealed that perception of susceptibility, perception of seriousness, perception of benefit, and cues to action significantly predicted parental acceptability, with 57% of the variance accounted by these variables. Acculturation was not a …

Dredging And Shipping Impacts On Southeast Florida Coral Reefs, Brian K. Walker, David S. Gilliam, Richard E. Dodge, Joanna Walczak

Marine & Environmental Sciences Faculty Proceedings, Presentations, Speeches, Lectures

Many coastal regions have experienced extensive population growth during the last century. Commonly, this growth has led to port development and expansion as well as increased vessel activity which can have detrimental effects on coral reef ecosystems. In southeast Florida, three major ports built in the late 1920’s along 112 km of coastline occur in close proximity to a shallow coral reef ecosystem. Recent habitat mapping data were analyzed in GIS to quantify the type and area of coral reef habitats impacted by port and shipping activities. Impact areas were adjusted by impact severity: 100% of dredge and burial areas, …
